This week we review a recent large scale study on catecholaminergic polymorphic ventricular tachycardia (CPVT) in children which is based on two large international registries. How do outcomes differ between those symptomatic CPVT patients treated with or without an ICD? Who amongst symptomatic CPVT pediatric patients likely warrant an ICD? What sort of ICD might be optimal - subcutaneous or transvenous? Are there ways to program ICD's in CPVT patients that may reduce inappropriate shocks? Why should the ICD be rarely employed in the CPVT pediatric patient? These are amongst the questions reviewed this week on the podcast. Association Between Neighborhood Deprivation and DNA Methylation in an Autopsy Cohort

BUFFALO, NY- May 8, 2024 – A new research paper was published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 8, entitled, “The association between neighborhood deprivation and DNA methylation in an autopsy cohort.” Previous research has found that living in a disadvantaged neighborhood is associated with poor health outcomes. Living in disadvantaged neighborhoods may alter inflammation and immune response in the body, which could be reflected in epigenetic mechanisms such as DNA methylation (DNAm). In this new study, researchers Lindsay Pett, Zhenjiang Li, Sarina Abrishamcar, Kenyaita Hodge, Todd Everson, Grace Christensen, Marla Gearing, Michael S. Kobor, Chaini Konwar, Julia L. MacIsaac, Kristy Dever, Aliza P. Wingo, Allan Levey, James J. Lah, Thomas S. Wingo, and Anke Hüls from Emory University, University of British Columbia, BC Children's Hospital Research Institute, Centre for Molecular Medicine and Therapeutics, and Atlanta VA Medical Center used robust linear regression models to conduct an epigenome-wide association study examining the association between neighborhood deprivation (Area Deprivation Index; ADI), and DNAm in brain tissue from 159 donors enrolled in the Emory Goizueta Alzheimer's Disease Research Center (Georgia, USA). “We found one CpG site (cg26514961, gene PLXNC1) significantly associated with ADI after controlling for covariates and multiple testing (p-value=5.0e-8).” Effect modification by APOE ε4 was statistically significant for the top ten CpG sites from the EWAS of ADI, indicating that the observed associations between ADI and DNAm were mainly driven by donors who carried at least one APOE ε4 allele. Four of the top ten CpG sites showed a significant concordance between brain tissue and tissues that are easily accessible in living individuals (blood, buccal cells, saliva), including DNAm in cg26514961 (PLXNC1). This study identified one CpG site (cg26514961, PLXNC1 gene) that was significantly associated with neighborhood deprivation in brain tissue. She was curious about how they thought about life, the way that they approached it, and their stories and experiences.  Jasmine completed her counselling education in B.C. and worked in organizations before starting her own private practice. She knew that she loved helping youth and their families and wanted to make a difference.     Starting and growing a private practice  Jasmine built her Canadian private practice in South Surrey. She felt lucky in the fact that her practice has been successful from when it opened its doors, so she was able to continue developing it over the years, with some minor financial dips here and there.  Jasmine was also able to get some advice and help from her husband, who has completed a masters degree in business.  In terms of building a successful business, Jasmine noticed a desperate need in an area of her community and decided to set up her practice there.  Now Jasmine has five associates, and everyone is fully booked, including her!  What's on the horizon?  After Jasmine's business quickly scaled, she noticed that something wasn't feeling quite right.  Now that her business has grown and is on the path to continue doing so, Jasmine wants to be intentional about structuring her business so that it doesn't get overwhelmingly busy and run away with her.  Therefore, Jasmine's now decided to actually scale back a bit and to not continue growing the business, without intention. Her Canadian private practice is successful now, and she's focusing on maintaining that success at this level so that she can remain a successful niche business without getting swept away.  Jasmine's advice for listeners  Remember the power of vulnerability and the importance of being authentic, especially as a leader. Dr. William Gibson discusses his research in this area, as well as what he has learned from studying the genetics behind very unique forms of diabetes.Dr. Gibson is an Investigator at the BC Children's Hospital, a Senior Clinician Scientist at the Laboratory for Obesity Genetics and Indirect Calorimetry (LOGIC) at BC Children's Hospital Research Institute. In addition, he is a Professor in the Department of Medical Genetics, Faculty of Medicine at the University of British Columbia.

Risers and thrivers: Be inspired by my dear friend, Genevieve McLean!   She's an inspirational "3" time cancer survivor and thriver, now living life to the fullest and married to Kirk McLean, a retired NHL goaltender and a true legend here in Vancouver!    Genevieve first faced AML (leukaemia) as a child, when she did only 1/2 of the treatment protocol at BC Children's Hospital and left to pursue alternative medicine. It was a success.    She was in remission for 9 years, then relapsed. She only learned of the relapse after an injury endured while snowboarding and went for a regular routine check up. As a young adult, she was treated at VGH and had a bone marrow transplant, with thanks to her incredible brother who served as her donor, her lifeline.    Her 3rd time facing cancer was now 7 years ago, only 6 months after she first met Kirk.   A 6X5inch adrenal tumour was discovered and fortunately, a surgical procedure to excise the Tumor was 100% successful. Despite all that she's faced, she's also professionally modelled, has achieved a degree in business, and has worked for the Yellow Pages Group for almost 15 years as a top sales rep in media sales!   She's also given back to the cancer community in every way she can- she's volunteered at Camp Good Times (summer camp for kids with cancer) and has sat on the board of the Michael Cuccione Foundation.     No matter what you are facing today, hold on to hope and actively seek out inspirational role models like Genevieve!     Tag your friends who may need inspiration and follow along here @60daystolive2012 for more stories of hope, resilience and inspiration to be empowered to overcome your hard days and to live today with more vitality and joy!
