Former Yale professor Morgan Levine shares 3 ways to change your diet to extend your life. Dr. Morgan Levine, author of "True Age," explains how the impact of diet on aging and longevity is determined by three factors: quantity, quality, and timing of food intake. First, caloric restriction, typically a 20% reduction in overall calorie intake, has been linked to increased lifespan in several animal models. However, the positive effects may stem from avoiding overeating rather than restriction per se. Second, a plant-based diet, with less animal products and refined sugars and more fruits, veggies, and whole foods, seems beneficial for aging and longevity. Third, fasting, or limiting eating to specific time windows, might mimic the benefits of caloric restriction by causing 'hormesis', mild stressors that boost resilience to aging-related changes. The optimal diet is likely individual, influenced by factors like genetics and age, and should be monitored through measures of biological age and personal well-being. Future research into aging biomarkers could further our understanding of diet's role in the aging process.  In this episode of the Epigenetics Podcast, we caught up with Morgan Levine from Altos lab to talk about her work on Epigenetic Clocks and Biomarkers of Ageing. The Levine Lab focuses on deciphering mechanisms that lead to epigenetic ageing, which can be measured by epigenetic clocks. Epigenetic clocks were first described in 2011 by Bocklandt et al.. Later-on, the Horvath and the Hannum clock were described by using a combination of CpGs to calculate biological/epigenetic age in contrast to chronological age. The Levine Lab themselves worked on generating an advanced version of an Epigenetic clock, called "DNAm PhenoAge" that will now be used, and not only in human samples. Dr. Morgan Levine is an Assistant Professor of Pathology and Epidemiology at the Yale School of Medicine. Morgan runs the lab for Aging in Living Systems at Yale. Her labs works on Epigenetic Clocks, Epigenetic reprogramming, cellular aging trajectories, and in trying to understand the causes of Alzheimer's disease. I can't even begin to tell you how much I learned in today's conversation! We extensively discussed Morgan's work on developing a new Epigenetic clock called the SystemsAge clock, which is very cool and can test the biological age of different organs & systems in your body. We also spoke of different features of these clocks, interventions tested, what we know so far on reversing your biological age, some of the limitations of the clocks, why she is so excited about epigenetic reprogramming, and some of her work around Alzheimer's disease. Most people are interested in knowing their biological age and epigenetic clocks are a way to test it. Dr. Morgan Levine, PhD (https://www.morganlevinelab.com/) is Assistant Professor of Pathology and Epidemiology, at the Yale School of Medicine. Dr. Levine is also a member of both the Yale Combined Program in Computational Biology and Bioinformatics, and the Yale Center for Research on Aging. Her work relies on an interdisciplinary approach, integrating theories and methods from statistical genetics, computational biology, and mathematical demography to develop biomarkers of aging for humans and animal models using high-dimensional -omics data. As PI or co-Investigator on multiple NIH-, Foundation-, and University-funded projects, she has extensive experience using systems-level and machine learning approaches to track epigenetic, transcriptomic, and proteomic changes with aging and incorporate this information to develop measures of risk stratification for major chronic diseases, such as cancer and Alzheimer's disease. Dr. Levine's work also involves development of systems-level outcome measures of aging, aimed at facilitating evaluation for gero-protective interventions. A number of the existing biological aging measures she has developed are being applied in both basic and observational research.

Today I'm speaking with Morgan Levine. Morgan is an assistant professor at Yale's Pathology department and runs a lab that aims to discover both the mechanisms that drive aging and interventions to slow or reverse that process. The lab also develops tools to measure biological age, which we will get into in our discussion of epigenetic clocks. Aging has long been assumed to be our insurmountable human fate. But as research of the past years has uncovered aging's malleability in some model organisms, the hope to slow or reverse the aging process in humans has become an increasingly attractive goal. And given that age is the greatest risk factor for developing many of the most common lethal diseases today, longevity research could potentially be extremely impactful both for prolonged health- and lifespans. “For me being able to quantify aging is one of the most important endeavours in the field. There is a lot of emphasis on intervening in aging, to either reverse or slow it, but I would argue that there's no way to test if you did that if you can't directly measure the thing that you're trying to intervene in.”In this session, Morgan Levine, Assistant Professor at Yale, gave a sneak peek into the new epigenetic clock they are developing that is able to probe into multiple organ systems, as well as a new approach how to calculate clocks that is much more reliable, enabling to generate insights from methylation clocks with much smaller samples required. The second talk was given by Jamie Justice, Assistant Professor at Wake Forest, that covered the current ways and strides the longevity field is making towards validating biomarkers of aging through clinical trials, showing on examples of a few senolytic trials they made. We are all used to feeling younger than our years and occasionally older than our birth date would suggest.  In effect, we have two ages - chronological and biological. Chronological age simply states the number of years that we have been alive, whereas biological age is a measure of how well our bodies are performing, in relation to the rest of the population.  So how is it determined?  LLAMA host Peter Bowes delved into the science with Elysium Health, which has developed Index, a DNA test that tracks how fast we have been aging, based on our epigenome, which regulates the way genes work.  In this episode, Peter meets Dr. Morgan Levine, an assistant professor of pathology at Yale School of Medicine and an advisor to Elysium Health.
