Thirty Years After Discovery of SA-β-gal: Researchers Revisit the Hallmarks of Cellular Senescence

BUFFALO, NY — June 5, 2026 — A new #review was #published in Volume 18 of Aging on May 15, 2026, titled “Blue period – features of senescence 30 years after beta-galactosidase.” The review was led by first author Chisaka Kuehnemann and corresponding author Christopher D. Wiley from Tufts University. Cellular senescence has emerged as one of the most important biological processes linked to aging and age-related disease. Senescent cells stop dividing in response to stress or damage, yet they remain metabolically active and release a variety of signaling molecules that can influence surrounding tissues. Over the past three decades, evidence has increasingly shown that the accumulation of these cells contributes to chronic inflammation, tissue dysfunction, and many degenerative conditions associated with aging. In this review, the authors examine how the field has evolved since the landmark discovery of senescence-associated beta-galactosidase (SA-β-gal) in 1995. That finding provided one of the first practical methods for identifying senescent cells and helped establish that these cells accumulate in aging tissues. Since then, researchers have identified numerous additional characteristics of senescence and developed new approaches to study their role in health and disease. The review highlights several major features now recognized as hallmarks of senescent cells. These include stable proliferative arrest, increased lysosomal activity, secretion of inflammatory and signaling molecules collectively known as the senescence-associated secretory phenotype (SASP), mitochondrial dysfunction, alterations in nuclear architecture, accumulation of metals and lipofuscin, and enhanced survival despite exposure to cellular stress. Your skin has a biological age and for most people, it's not the same as their chronological one. Mayo Clinic published the SkinSpan framework this year specifically to quantify it, and the 12 Hallmarks of Aging now have a direct application to dermal tissue. This raises an uncomfortable question: if you're already optimizing everything below the neck with NAD IVs, peptides, and red light, why is the skincare on your face still operating on a paradigm from 1987?In this new episode of the Biohacking Beauty Podcast, we give you a preview of the keynote we're delivering at Dave Asprey's Beyond Conference and walk you through the 3-pillar framework that replaces traditional anti-aging skincare with longevity science in mind.Because the results you can feel today and results you can measure in ten years are not the same thing. Longevity Science Explained: Mitochondria, DNA Damage, Senescent Cells & Peptides with Greg MacphersonHost Vicki Nguyen introduces One Body, One Life, a longevity-focused show sponsored by Jomei's Fine Foods, and interviews Greg Macpherson, a New Zealand pharmacist and founder of SRW Laboratories. Greg shares how working with residential care patients and discovering “The Hallmarks of Aging” shaped his focus on healthspan versus lifespan, using a Mount Everest analogy and noting healthspan is typically about 10 years shorter than lifespan. He highlights key drivers of aging including DNA damage (with lifestyle strategies like “sunscreen for your cells”), personal viral burden and immune fatigue (including attention to absolute lymphocyte count), mitochondrial decline and targeted antioxidants like mitoquinol, and chronic inflammation linked to oxidative stress. They discuss cellular senescence, fasting, time-restricted eating, exercise, and compounds like fisetin and quercetin, plus emerging areas like stem cells, peptides, biological age testing via DNA methylation, and future cellular “reprogramming,” along with Greg's top lifestyle practices and where to find him (srw.co).

The One System Behind ALL 14 Hallmarks of Aging — And How to Target It | Courtney Van Bussum

Your immune system isn't just fighting off colds. It's the foundational system that influences all 14 hallmarks of aging — and when it declines, everything downstream accelerates. In this episode, I sit down with Courtney Van Bussum, COO of Longevity Launch Labs, to dive deep into immunosenescence — the aging of the immune system — and why it starts far earlier than most people realise. We unpack what Stanford's landmark 1000 Immunomes Project revealed about the real driver of biological aging, why your supplement stack might be missing the most important piece, and how we built Re:juvenate Pro (Cell Restore in the US) to target immune aging at its root. We get into the fire station analogy that makes the immune system finally make sense, why 70-80% of your immune system lives in your gut and what that means for barrier health, the science behind four synergistic ingredients — Immune Defense Protein, Immunel colostrum extract, carnosic acid from NZ rosemary, and kawakawa — and why standard colostrum supplements aren't cutting it. We also cover why NSAIDs are impairing your recovery, what CXCL9 is and why most practitioners have never heard of it, Isobel's lymphocyte count recovery story, the immune age calculator app, and why "boost your immune system" is the wrong approach — modulation is everything. This is not another colostrum product. This is upstream immune intervention. In this episode of the Future of Fitness Podcast, host Eric Malzone sits down with Chris Mirabile, founder of NOVOS, to unpack the science and business of longevity. Chris shares how surviving a brain tumor as a teenager reshaped his perspective on health and pushed him to study aging at a biological level. The conversation dives into what longevity actually means beyond buzzwords—how it differs from simply living a healthy lifestyle, why healthspan matters just as much as lifespan, and the science behind targeting the biological "hallmarks of aging." Chris also explains why some popular trends like hormone optimization and peptides are more complex than they seem, and how NOVOS approaches longevity by addressing the root cellular mechanisms that drive aging, disease risk, and long-term performance. Together, they dive into Louise's recent research exploring how psilocybin, the active compound in psychedelic mushrooms, may influence aging biology at the cellular and organismal level.Louise shares the story behind her curiosity about psilocybin, sparked by conversations with a friend and fueled by a lack of scientific answers. She explains how her team overcame regulatory hurdles to study the effects of psilocybin on human cells and aging mice. Their findings showed that psilocybin extended cellular lifespan and reduced hallmarks of aging, such as oxidative stress and DNA damage. In mice, regular dosing improved survival, reversed visible signs of aging, and affected organs beyond the brain. The conversation also covers the challenges of translating these findings into humans, the need for more research on dosing and safety, and the importance of funding in moving this field forward. Louise encourages listeners to stay curious, think beyond established paths, and keep an open mind as new questions and discoveries emerge in the science of aging.Guest-at-a-Glance

A secure relationship isn't one where nothing ever goes wrong — it's one where the foundation is strong enough to hold the hard stuff. For many people (especially those with anxious attachment), insecurity doesn't come from being “too sensitive,” but from being in dynamics that lack safety, consistency, or clarity. In this episode, I break down five key qualities that tend to be present in secure relationships, and how they actually feel on a nervous system level.I cover:What emotional safety really looks like (and what it doesn't)Why trust is about reliability and consistency, not just honestyHow secure couples approach conflict and repair after ruptureWhat it means for a relationship to be a secure base rather than a constant projectWhy shared vision and felt commitment are essential for long-term securityWhether you're assessing your current relationship, healing after an insecure one, or wanting to understand what you're moving towards, this episode offers a grounded framework for what relational security is built on — and what helps it endure.Explore my couples course, Secure TogetherFree resources for building secure attachment

In this episode of the Ageless Future Podcast, host Cade Archibald reframes aging as a modular, systems-based process rather than a simple function of time. Drawing on the Hallmarks of Aging and the geroscience framework, Cade explores how different biological systems—immune function, mitochondrial health, stem cell signaling, cellular senescence, vascular elasticity, brain plasticity, and nutrient sensing—decline at different rates and influence one another. He explains how chronic inflammation acts as a unifying driver of dysfunction and highlights practical strategies to target specific aging bottlenecks, including nutrition, exercise, mitochondrial optimization, peptides, detoxification, and metabolic support. The core message: aging is not inevitable or uniform—it's engineerable. By focusing on system repair instead of surface-level anti-aging, we can enhance resilience, cognitive function, recovery, and long-term vitality.AGELESS FUTURE:Book Comprehensive Labs: https://agelessfuture.com/longevity-labs/FREE copy of The Peptide Blueprint: https://agelessfuture.com/blueprintSign up for future Health Accelerator Challenges calls LIVE! https://us02web.zoom.us/webinar/register/WN_YZsiUMOzSyqcE8IinC5YEQ#/registrationBooks: https://www.amazon.com/Books-Regan-Archibald/s?rh=n%3A283155%2Cp_27%3ARegan%2BArchibaldArticles: https://medium.com/search?q=Regan+ArchibaldLIKE/FOLLOW/SUBSCRIBE AGELESS FUTURE:YouTube -https://www.youtube.com/@ReganArchibald / https://www.youtube.com/@Ageless.FutureLinkedIn: https://www.linkedin.com/in/regan-archibald-ab70b813Instagram: https://www.instagram.com/ageless.future/Facebook: https://www.facebook.com/AgelessFutureHealth/DISCLAIMER: This video is for educational purposes only and does not provide medical advice, diagnosis, or treatment.  Many of the molecules discussed in this video are research compounds and are not approved by the U.S. Food and Drug Administration (FDA) for any specific medical use, indication, or condition. Angela talks to Dr. Ash Kapoor, a longevity specialist and cellular health expert, who shares his approach to health that focuses on understanding and addressing the root causes of disease and aging, particularly through the lens of the 12 Hallmarks of Aging.  Dr. Kapoor talks about the fact that true longevity is not just about extending lifespan, but about maximising independence, purposefulness, and fulfilment, explaining that modern stress depletes the body's primary energy source (ATP), forcing it to steal from the immune, hormonal, and nervous systems KEY TAKEAWAYS: Longevity Redefined: True longevity is defined not merely by living longer, but by being independent, purposeful, and pursuing fulfilment until an achievable age of 123. The Root Cause of Decline: All 126,000 known diseases can be traced back to the 12 Hallmarks of Aging, a breakdown in the body's cellular systems that is often driven by chronic modern stress A Strategy for Optimisation: Dr. Kapoor's approach involves a multi-stage strategy of detoxifying (cleaning the liver and gut), repairing (restoring hormones and nutrients, and re-establishing circadian rhythm), and renewing the body's cellular environment The Power of Inner Energy: The core problem is operating on stress-based energy (like cortisol and immune/hormonal reserves) instead of the primary mitochondrial energy (ATP) TIMESTAMPS AND KEY TOPICS: 0:01:25: Defining Longevity: What true "living" means beyond just existing. 0:03:00: The Science of the 12 Hallmarks of Aging and the control we have over them. 0:08:48: The link between the external world (success) and internal depletion (energy crash). 0:18:25: The fundamental strategy: Detox, Repair, and Renew in the correct order. 0:25:05: Understanding hormone depletion: Why the body "steals" hormones and the concept of "restorative hormone therapy." Most conversations about longevity focus on tactics. This episode breaks down the biology underneath them. You'll hear a systems level analysis of how insulin signaling, mTOR, mitochondrial function, inflammation, and nutrient sensing interact to determine biological age, disease risk, and long term human performance. This is not surface level biohacking. It is a deep dive into the mechanisms that actually control aging, metabolism, muscle preservation, and brain optimization, and why most anti aging strategies fail when they ignore these foundations. This all-time classic episode from 2024, has host Dave Asprey joined by Mark Hyman, a practicing physician and one of the most respected leaders in functional medicine. Dr. Hyman is the founder and senior advisor of The UltraWellnessCenter, the founder and former head of strategy and innovation at the Cleveland Clinic Center for Functional Medicine, and a long time advocate for treating root causes instead of managing disease. With over 30 years of clinical experience, his work focuses on restoring mitochondrial health, optimizing metabolism, reversing chronic disease, and slowing biological aging using science backed interventions. Together, Dave and Mark dismantle common myths around protein, carnivore diets, ketosis, fasting, supplements, and longevity drugs. They explain why sugar accelerates aging faster than protein, how mTOR must cycle instead of staying suppressed, and why muscle, mitochondria, and metabolic flexibility matter more than calorie restriction. The conversation also covers nootropics, sleep optimization, neuroplasticity, AI driven diagnostics, and how functional medicine reframes aging as a reversible biological process rather than an inevitable decline. You'll learn: • How insulin and nutrient sensing control aging speed • Why mitochondria sit at the center of longevity and disease • The truth about mTOR, fasting, and protein intake • How ketosis and metabolic flexibility protect long term health • Where supplements and nootropics actually fit in brain optimization • Why functional medicine outperforms symptom based care • How AI and advanced testing are reshaping modern medicine Thank you to our sponsors! - MASA Chips | Go to https://www.masachips.com/DAVEASPREY and use code DAVEASPREY for 25% off your first order. -ARMRA | Go to https://tryarmra.com/ and use the code DAVE to get 15% off your first order. -OneSkin | Try OneSkin for 15% off with code DAVE at https://www.oneskin.co/DAVE. Lessons in Longevity: Skincare That Goes Beyond Anti-Aging Featuring Smitha Rao of Parëva Beauty

In this episode of Skin Anarchy, Dr. Ekta Yadav sits down with Smitha Rao—biotechnologist, longtime beauty innovator, and founder of Pareva Beauty—for a grounded, science-first conversation on what skin longevity actually means. Part of the Lessons in Longevity series, this episode challenges the tired idea of “anti-aging” and reframes skincare as long-term biological support rather than surface-level correction. With nearly two decades in biotech and beauty, Smitha brings a rare perspective shaped by molecule development, patents licensed across major brands, and leadership roles behind more than 200 products. That experience revealed a growing disconnect: while consumers want clarity and real science, the industry often defaults to hype, fear-based claims, and trend chasing. Pareva Beauty was born as a response—built on transparency, rigor, and the belief that longevity is about extending skin function and resilience, not reversing time.At the center of the discussion is Molecur 3™, Pareva's proprietary, plant-based technology inspired by the Hallmarks of Aging. Smitha explains how targeting inflammation, cellular stress, and age-related decline—rather than isolated “anti-aging” claims—creates a more realistic and sustainable path for skin health. The conversation also dives into plant-derived exosomal signaling, why it differs from human-derived systems, and how it offers a safer, scalable approach for topical longevity skincare.Together, Dr. Ekta and Smitha unpack why fewer, better-designed products outperform complex routines, why transparency matters more than vague “clean” labels, and how longevity skincare must earn trust through data—not promises.Listen to the full episode to hear how Smitha Rao is helping redefine the future of skincare—where science, honesty, and long-term skin health come first.Shop Parëva Beauty and use code SA20 for 20% through 3/17.CHAPTERS:(0:02) - Introduction, Lessons in Longevity & Smitha Rao(1:21) - From Biotech to Beauty Innovation(2:50) - Why Pareva Was Created & The Longevity Gap(3:33) - Defining Skin Longevity & Extending Your Prime(5:38) - Rejecting Anti-Aging & Reframing Aging Quality(8:51) - Molecure 3™: Hallmarks of Aging Inspiration(10:27) - Plant Stem Cells, NAD⁺ & Exosome Signaling(14:00) - Exosomes Explained: Plant vs Human-Derived(22:51) - Simplifying Skincare: Fewer Steps, Better ResultsPlease fill out this survey to give us feedback on the show!Don't forget to subscribe to Skin Anarchy on Apple Podcasts, Spotify, or your preferred platform.Reach out to us through email with any questions.Sign up for our newsletter!Shop all our episodes and products mentioned through our ShopMy Shelf!*This is a paid collaboration Hosted on Acast. In this episode of "Women of Impact," Lisa Bilyeu welcomes Dr. Orion Taraban, psychologist, YouTube creator, and author of the bestselling book "The Value of Others." Dr. Taraban brings a refreshingly candid — and sometimes controversial — perspective to the complex world of romantic relationships, power dynamics, and emotional safety. Together, they pull back the curtain on common traps women fall into, how to recognize manipulation (especially from narcissists or cluster B personalities), and the essential mindsets needed to reclaim your relationship power once and for all. Dr. Taraban and Lisa set the groundwork by tackling why no one can technically manipulate your emotions — unless you allow them to. This is essential listening for anyone looking to understand how to protect themselves while dating—or simply elevate their personal relationships to a new level of autonomy. In this episode, Josh unpacks the four layers of anxiety—psychological, emotional, cognitive, and behavioral—highlighting why avoidance is the key feature that transforms ordinary worry into disorder. He explains why he continues to treat OCD and PTSD as anxiety conditions despite their DSM-5 reclassification, and he draws important distinctions between worries versus worrying and thoughts versus thinking. The discussion explores health anxiety, illness anxiety, and the impact of modern contributors such as wearables, social media, and the COVID era, while weaving in real-world case studies and Josh's structured assessment approach. Josh also breaks down evidence-based treatments, from exposure therapy and cognitive-behavioral therapy (CBT) to acceptance and commitment therapy (ACT), along with the role of medication, lifestyle factors, and how shifting from avoidance to committed action can build long-term resilience.
