BUFFALO, NY — April 1, 2025 — A new #research paper was #published in Aging (Aging-US) on March 29, 2025, as the #cover of Volume 17, Issue 3, titled “Differential senolytic inhibition of normal versus Aβ-associated cholinesterases: implications in aging and Alzheimer's disease.” In this study, a research team from Dalhousie University, led by Sultan Darvesh, discovered that certain anti-aging compounds, known as senolytics, can block harmful brain enzymes linked to Alzheimer's disease (AD) without affecting healthy ones. Senolytics are compounds that help clear out damaged or “zombie” cells that build up with age and contribute to inflammation and tissue dysfunction. This work provides new insight into how AD-related damage can be precisely targeted, leading the way for safer treatments that protect memory and brain health in older adults. Alzheimer's disease is one of the most common causes of memory loss and dementia. A hallmark of the disease is the buildup of sticky protein clumps in the brain, known as amyloid-beta plaques. Two enzymes—acetylcholinesterase (AChE) and butyrylcholinesterase (BChE)—are found near these plaques. While these enzymes play important roles in brain function, they can also contribute to AD progression when they attach to plaques. Drugs that target these enzymes are already used to help with memory, but they often block both harmful and healthy forms, which can cause unwanted side effects. To investigate a better solution, researchers tested six compounds that are known for their anti-aging or brain-boosting properties. They wanted to know if these compounds could block only the harmful AChE and BChE enzymes forms linked to Alzheimer's disease. Using brain tissue samples from AD patients and enzyme activity assays, they discovered that compounds such as dasatinib and nintedanib, both senolytics, were able to block the forms of AChE and BChE associated with amyloid-beta plaques. These compounds did not affect normal brain enzymes, though. “We show that the selected senolytics and nootropic inhibit ChEs associated with plaques but not the enzymes associated with normal neural elements.” The study also used computer modeling to explore how these compounds interact with the enzymes. The models showed that the enzymes change shape when near plaques, making them easier for certain compounds to target. This change may explain how the drugs can selectively affect only the diseased areas of the brain. While not all compounds worked equally well, the findings offer a new strategy for treating AD. By focusing on the differences between healthy and diseased enzyme forms, researchers may be able to design more precise and effective therapies. This selective approach could improve memory, reduce inflammation, and avoid the side effects of AD's current treatments. In summary, this research opens new possibilities for treating Alzheimer's disease in a more targeted way. It also highlights how discoveries in aging and brain health can work together to create better therapies for neurodegenerative diseases. Six conditions cause the majority of deaths in the world, these include dementia and cardiovascular disease. Traditionally, we've looked at these conditions through the lens of what goes wrong individually, such as blocked arteries around the heart or tangles of proteins that impair electrical signalling in the brain. But the common denominator is actually ageing.As grandiose and esoteric as that sounds, ageing and the processes that govern ageing are what we need to be tackling if we want to better manage or even prevent these conditions of older age.And with me to discuss why and how we age is Richard Faragher. He's Professor of Biological Gerontology at the University of Brighton, and has dedicated his career to understanding the ageing process and what we can do to improve this inevitable phase of life.As you can tell from today's episode, Richard is a passionate advocate for older people and can wax lyrical about the history of the gerontology field.A few definitions before we dive into the episode I think will be helpful for people who are new to the research.There are agreed “hallmarks” of ageing that are a description of processes, such as metabolism and DNA repair functions, that go awry as we grow older. Ageing is caused by these changes at the molecular level that lead to downstream effects like disease and degeneration.Cell senescence is what happens at the end of a cell's lifespan, where they become dysfunctional and need to be removed by immune cells. However in older age, it can become more troublesome for your immune system to clear away these senescent cells and increasing inflammation ensues.Senolytics are compounds or drugs that have been designed to selectively eliminate these senescent cells. And the hope is that they can mitigate a range of age-associated conditions like arthritis, cataracts, dementia and more.Trigger warning. We discuss a graphic analogy for senescent cells using suicide, so I just wanted to give a heads up for anyone who might find that analogy uncomfortable.

BUFFALO, NY- October 22, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science), Volume 16, Issue 19 on September 26, 2024, entitled, “Use of the senolytics dasatinib and quercetin for prevention of pelvic organ prolapse in a mouse animal model.” Pelvic organ prolapse is a common condition among women in the U.S., with a 13% estimated risk of requiring surgery by age 80. Senolytic agents like dasatinib and quercetin (D+Q) target age-related cellular senescence and reduce senescent cell activity in various disease processes. In their paper, researchers Erryn Tappy, Haolin Shi, Jessica Pruszynski, and Maria Florian-Rodriguez from the University of Texas Southwestern Medical Center, Department of Obstetrics and Gynecology in Dallas, utilized a mouse model of pelvic organ prolapse, Fibulin-5 knockout (Fbln-5-/-) mice, to assess the ability of D+Q to prevent development of prolapse. The D+Q injections administered did not result in significant differences in prolapse development but did reduce cellular senescence markers in Fbln-5-/- mice. This suggests senolytic agents may help mitigate the role of cellular senescence in tissue dysfunction associated with prolapse. The researchers suggest that further studies are needed to determine optimal timing, dosage, and delivery of senolytics for prolapse prevention. BUFFALO, NY- March 6, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 4, entitled, “Exploring the effects of Dasatinib, Quercetin, and Fisetin on DNA methylation clocks: a longitudinal study on senolytic interventions.” Senolytics, small molecules targeting cellular senescence, have emerged as potential therapeutics to enhance health span. However, their impact on epigenetic age remains unstudied. In this new study, researchers Edwin Lee, Natàlia Carreras-Gallo, Leilani Lopez, Logan Turner, Aaron Lin, Tavis L. Mendez, Hannah Went, Alan Tomusiak, Eric Verdin, Michael Corley, Lishomwa Ndhlovu, Ryan Smith, and Varun B. Dwaraka from the Institute For Hormonal Balance, TruDiagnostic, Buck Institute for Research on Aging, and Cornell University aimed to assess the effects of senolytic treatments on DNA methylation (DNAm), epigenetic age, and immune cell subsets. “[...] this study aims to comprehensively assess the impact of senolytic drugs on epigenetic aging through two longitudinal studies to address our research objective. The initial investigation focuses on a combination treatment of Dasatinib and Quercetin, while the subsequent phase incorporates Fisetin into the treatment regimen.” In a Phase I pilot study, 19 participants received Dasatinib and Quercetin (DQ) for 6 months, with DNAm measured at baseline, 3 months, and 6 months. Significant increases in epigenetic age acceleration were observed in first-generation epigenetic clocks and mitotic clocks at 3 and 6 months, along with a notable decrease in telomere length. However, no significant differences were observed in second and third-generation clocks. Building upon these findings, a subsequent investigation evaluated the combination of DQ with Fisetin (DQF), a well-known antioxidant and antiaging senolytic molecule. After one year, 19 participants (including 10 from the initial study) received DQF for 6 months, with DNAm assessed at baseline and 6 months. Remarkably, the addition of Fisetin to the treatment resulted in non-significant increases in epigenetic age acceleration, suggesting a potential mitigating effect of Fisetin on the impact of DQ on epigenetic aging. “Furthermore, our analyses unveiled notable differences in immune cell proportions between the DQ and DQF treatment groups, providing a biological basis for the divergent patterns observed in the evolution of epigenetic clocks. Dr Shade explains how each of these factors plays a role in promoting longevity and discusses the importance of supplementation and lifestyle choices in optimising these pathways. A new research paper was published in Aging (Aging-US) Volume 15, Issue 7, entitled, “Characterization of the HDAC/PI3K inhibitor CUDC-907 as a novel senolytic.” The accumulation of senescent cells has an important role in the phenotypical changes observed in aging and in many age-related pathologies. Thus, the strategies designed to prevent these effects, collectively known as senotherapies, have a strong clinical potential. Senolytics are a type of senotherapy aimed at specifically eliminating senescent cells from tissues. Several small molecule compounds with senolytic properties have already been identified, but their specificity and range of action are variable. Because of this, potential novel senolytics are being actively investigated. Given the involvement of HDACs and the PI3K pathway in senescence, researchers Fares Al-Mansour, Abdullah Alraddadi, Buwei He, Anes Saleh, Marta Poblocka, Wael Alzahrani, Shaun Cowley, and Salvador Macip from the University of Leicester, Najran University and Universitat Oberta de Catalunya hypothesized that the dual inhibitor CUDC-907, a drug already in clinical trials for its antineoplastic effects, could have senolytic effects. “Here, we show that CUDC-907 was indeed able to selectively induce apoptosis in cells driven to senesce by p53 expression, but not when senescence happened in the absence of p53.” Consistent with this, CUDC-907 showed senolytic properties in different models of stress-induced senescence. Their results also indicate that the senolytic functions of CUDC-907 depend on the inhibitory effects of both HDACs and PI3K, which leads to an increase in p53 and a reduction in BH3 pro-survival proteins. They don't just brush plaque away, but release antimicrobials to kill bad bacteria. If you want to outsource brushing your teeth to robots. Part of the problem with teeth brushing is that toothbrushes don't account for differences in the shape or spacing of different people's teeth.The new system solves that issue by effectively shapeshifting. It's made up of iron oxide nanoparticles, which can be assembled into different formations and controlled using magnetic fields. The particles can be arranged into the shape of bristles to brush plaque from tooth surfaces, or thinner floss-like strands to scrub between teeth. Iron oxide is known to activate hydrogen peroxide, triggering a reaction that produces free radicals that kill bacteria and the sticky biofilms they form on teeth. The system was tested on a flat slab of artificial tooth-like material, then a 3D-printed tooth model, and finally on real human teeth. The tests showed that these microrobots could efficiently remove plaque and biofilms, reducing pathogenic bacteria below detectable levels. Also showed they could precisely control the bristle stiffness and length by adjusting the magnetic field.This allowed the tips to be made firm enough to clean the teeth but still soft enough to be gentle on gums. The researchers say that the customizability of this microrobot system could clean people's teeth better, while its hands-free potential could allow people with reduced fine motor skills to still take care of their oral health. New drug increases human life span by 30%, according to Mayo Clinic | Brighter Side News (19:47) Scientists may never find the elixir of life, but they are finding new ways to extend our life spans. Mayo Clinic researchers say senolytic drugs can boost a key protein in the body that may protect older people against aspects of aging and a range of diseases. Senolytics developed at Mayo Clinic and given once clear the bloodstream of senescent or "zombie" cells.These cells contribute to multiple diseases and negative aspects of aging. Diseases including atherosclerosis (a form of heart disease), diabetes, and lung disease. As you age, these zombies build up in the body. They can make it difficult for your body to repair tissue, and they can release chemicals that harm normal cells nearby. This study shows that the removal of senescent cells significantly boosts the production of a protective protein called a-klotho.The protein a-klotho is important to maintaining good health, as it tends to decrease with age, and especially decreases in multiple diseases, including Alzheimer's, diabetes and kidney disease. James Kirkland, M.D., Ph.D., a Mayo Clinic internist and senior author of the study., talked on these Senolytics drugs:“We show that there is an avenue for an orally active, small-molecule approach to increase this beneficial protein and also to amplify the action of senolytic drugs." Animal studies have shown that decreasing a-klotho in mice shortens life span and increasing a-klotho in mice by inserting a gene that causes its production increases life span by 30%. This study shows that senolytics increased a-klotho in humans with idiopathic pulmonary fibrosis, a senescence-associated disease that leads to frailty, serious breathing difficulties and death. Tesla will open up Superchargers to non-Tesla electric vehicles in the US later this year - The Verge (24:23) Tesla plans to open up its Supercharger network to non-Tesla electric vehicles in the US in late 2022, according to a White House memo. The company has been allowing non-Tesla EVs to use its Supercharger plugs in several cities in Europe as part of a limited pilot program but has been quiet about when US charging stations would be available to non-Tesla EV owners. The memo stated: “Later this year, Tesla will begin production of new Supercharger equipment that will enable non-Tesla EV drivers in North America to use Tesla Superchargers.” Tesla did not respond to an email seeking confirmation of these plans (Elon Musk disbanded the company's PR office in 2019). It's not clear whether the White House purposefully scooped Tesla's own announcement on this project. Tesla's Supercharger network is often held up as the best possible example of an EV charging network: fast, reliable, and plentiful. This news is in line with what Tesla's CEO Elon Musk has, for years, about opening up his Superchargers to other companies' electric vehicles. If the US version of the pilot program works the same as it does in Europe, non-Tesla EV owners will need to download the most recent version of Tesla's smartphone app in order to access the stations. They will be prompted to select “Charge Your Non-Tesla” in order to find the closest participating Supercharger site. Tesla owners can still use the stations just as normal.

Mitochondrial decline is linked to senescence cells. Senescence cells stop dividing and do nothing. As we age, we may have 5-20% senescence cells in different tissues. This causes inflammation and causes the mitochondria to function improperly, thus increasing reactive oxygen species within cells. Nuclear DNA is usually packed in histones, which preserve genetic material. However, mitochondrial DNAs lack histones, making it prone to oxidative stress and mutations. As we age, our epigenetic processes become less efficient; it becomes a vicious cycle with more reactive oxygen species and less capacity to deal with them. [08:23] How Else Do Our Bodies Respond to Ageing Ageing declines autophagy, which recycles cellular organelles. When there's nothing to clean up dysfunctional mitochondria, problems with energy generation arise. Athletes who have exercised intensely their whole life will have worn-off tissues and lactic acid buildup. Remember, exercise is good, since we want some level of oxidative stress; however, we don't want more oxidative stress than what our body can cope with. Lisa shares how running ultramarathons accelerated her ageing process, wear and tear, and inflammation. [12:29] How to Support Autophagy and Athletic Performance? Intermittent fasting is when people have a certain feeding window a day. After eating, mTOR switches off and AMPK is activated which allows for autophagy. If you want to build muscle, Dr. Elena recommends eating after exercising so you can activate mTOR which helps with muscle growth. You need to create a cycle going of mTOR and AMPK since autophagy recycles cells back into amino acids, which can help cells build new proteins. Don't create a feeding window that's too narrow; you need to have enough fuel for your body. Eat chicken, meat, and foods rich in amino acids. Make sure you're sleeping properly, as this can affect your muscle growth. [18:30] Supplements to Support Autophagy and Longevity You can also support autophagy through supplements like berberine. Dr. Elena recommends adding supplements to specifically target senescence cells. Senolytics help kill senescence cells while senomorphics modify the senescence cells. Quercetin added with Vitamin C can help kill senescence cells. By itself, quercetin can produce toxic metabolites, so it needs Vitamin C to serve as an antioxidant. She also recommends taking their quercetin product with NAC and acetylcysteine to boost glutathione, a potent antioxidant. Quercetin has many benefits including anti-inflammatory properties, and can be used to combat cardiovascular disease, fight seasonal allergies, and can fight cancer. [26:51] How to Take Quercetin Quercetin can be found naturally in fruits and vegetables like strawberries and onions. If you've never taken compounds for their senescent action before, start by taking quercetin at 500 mg every day. After a month of taking it, check in on how you feel. You can also implement detox protocols with a higher dosage of quercetin(around 2 - 2 ½ grams). If you want to implement this protocol, start with 5 days first. Quercetin can be combined with 500mg of chlorella to boost the detoxifying reaction. People with kidney diseases should consult with a doctor before taking quercetin. [32:43] How NMN Can Help Athletes In addition to mitochondrial decline, athletes can experience loss of endurance and muscle loss as they age. Dr. Elena recommends increasing your fasting window depending on your goals. You want more oxygenation in your tissues to preserve endurance and muscle. [36:37] How to Optimise NMN NMN functions optimally when it's attached to a methyl group. Unfortunately, our body's methylation process also declines with age. [39:11] Longevity is Your Biological Age, Not Your Chronological Age Many people over 40 are insulin resistant. Insulin resistance is a gateway to biological decline. You can check your insulin resistance by checking your waist to hip ratio. After the age of 40, it's much easier to experience hormonal dysfunction. We need to make a conscious effort to improve our health. Ageing can cause issues to spiral if left uncontrolled. [42:50] Be Careful With Your Food 80% of adults in the west are insulin resistant. The food industry worsens the condition by offering so many processed carbohydrates. Many people have false ideas about what a healthy diet is. You can't have too many carbohydrates and sugar, even if they're coming from fruits. Cut down on carbohydrates, avoid processed foods, and take good oils. [45:21] Start with the Basics Start with the basics. You don't need to add supplements immediately. Just eat clean and good food. There's no such thing as a universal diet. Taking cold showers in the morning can improve hormonal balance. You can start with just 10 seconds of cold exposure. Manage your stress levels and make sure you move regularly. You don't need to be perfect. [50:47] How Some People Live to 100 There were two hypotheses surrounding centenarians: good habits and the absence of high-risk genes. However, some centenarians were found to have bad habits and mutations for Alzheimer's disease. Centenarians have genes that mitigate risks from bad mutations. They also have low IGF 1 levels. When people are young, IGF 1 can improve muscle and tissue growth, but it can be harmful when people grow older. Centenarians' genetic combination allowed for IGF to decrease as they grow older. Listen to the episode to learn more about how centenarians' hormonal processes are being regulated. Commit to regularly regulating and balancing your hormones. Dr. Tohru Minamino is Professor and Chairman of the Department of Cardiovascular Biology and Medicine, Juntendo University Graduate School of Medicine (https://juntendo-cvbm.com/en/about.html). He also serves as Director of the Cardiovascular Medicine, Juntendo University Hospital. He received his MD from the Chiba University Graduate School of Medicine in 1989 and his PhD from Faculty of Medicine, the University of Tokyo in 1997. Dr. Minamino is a medical cardiologist and research scientist focusing on molecular mechanisms of aging. He started his major research focusing on cardiovascular aging at Harvard Medical School (1997–2000), and his research interests have currently been growing in the biology of aging including metabolic pathways of longevity and senolysis. He has published more than 100 papers including in Nature, Nature Medicine, Cell, Cell Metabolism, and Lancet. Dr. Minamino has won several awards including Satoh Memorial Award in Japanese Circulation Society, and Erwin von Bälz Award (1st prize).
My guests in today's episode are Dr. Irina and Michael Conboy of the Department of Bioengineering at the University of California Berkeley. Their groundbreaking research focuses on tissue rejuvenation via plasma dilution and heterochronic parabiosis, where the Conboys took a young mouse and an older mouse and sutured them together, such that the animals shared blood and environment. The old mice became rejuvenated & younger. They've also pioneered a process called the Neutral Blood Exchange or Therapeutic Blood Exchange, through which dilution of old blood plasma can lead to true rejuvenation in the brain & other tissues of mice to a younger state!! Further, we touch upon Irina's work showing how decreased Oxytocin levels with age contributes to poor muscle function. Dr. Judith Campisi, PhD (https://www.buckinstitute.org/lab/campisi-lab/) is a biochemist, cell biologist, and Professor of Biogerontology at the Buck Institute for Research on Aging. Dr. Campisi received a PhD in biochemistry from the State University of New York at Stony Brook and completed her postdoctoral training in cell cycle regulation at the Dana-Farber Cancer Institute and Harvard Medical School. As an assistant and associate professor at the Boston University Medical School, she studied the role of cellular senescence in suppressing cancer and soon became convinced that senescent cells also contributed to aging. She joined the Lawrence Berkeley National Laboratory as a senior scientist in 1991 working with Dr. Mina Bissell. In 2002, she started a second laboratory at the Buck Institute. At both institutions, Dr. Campisi established a broad program to understand the relationship between aging and age-related disease, with an emphasis on the interface between cancer and aging. Dr. Campisi is a member of the National Academy of Sciences and a fellow of the American Association for the Advancement of Science. Dr. Campisi has received numerous awards for her research, including two MERIT awards from the National Institute on Aging and awards from the AlliedSignal Corporation, Gerontological Society of America, and American Federation for Aging Research. She is a recipient of the Longevity prize from the IPSEN Foundation, the Bennett Cohen award from the University of Michigan, and the Schober award from Halle University, and she is the first recipient of the international Olav Thon Foundation prize in Natural Sciences and Medicine. Dr. Campisi currently serves on advisory committees for the Alliance for Aging Research, Progeria Research Foundation, and NIA's Intervention Testing Program. She is also an editorial board member for more than a dozen peer-reviewed journals. Dr. Campisi is a scientific founder of Unity Biotechnology, a California-based company focused on developing senolytic therapies for age-related pathologies. She has served on the scientific advisory boards of the Geron Corporation, Sierra BioScience, and Sangamo Biosciences.

Andy Koff is the Founder of Atropos Therapeutics. A company with a new approach to tackling senescent cells, one fo the biggest hallmarks of aging.Atropos Therapeutics uses a bio marker to identify when a cell starts transitioning from a quiescent cell into a senescent cell. Using this bio marker, Atropos aims to be able to prevent the cell from transitioning into a senescent cell, they've dubbed this type of technology a "senosuppressant"For 25 years or so, starting with his post-graduate work at the Fred Hutchinson Cancer Center with Jim Roberts, and into his faculty position at Memorial Sloan Kettering Cancer Center, Andy has used a multitude of approaches, biochemical, genetic and cellular, to identify the proteins and the mechanisms that control how cells choose between duplicating themselves, known as proliferating, or remaining in a non-proliferating state. This decision is incorrectly made in virtually all cancer cells, thus providing the framework to explain much about how drugs work in oncology.About five years ago, Andy’s attention shifted to another decision…how cells maintain themselves in the non-proliferating state and what might be the consequences to not being able to do so. Professor Michael Lisanti, Chair of Translational Medicine at the University of Salford joins David Gornoski and physicist Weiping Yu to discuss his work using antibiotics to destroy senescent cells-dysfunctional aging cells-as well as cancer stem cells. These generic, very inexpensive antibiotics have studies showing antiviral effects in other viruses. Dr. Lisanti proposes that clinical trials should be conducted using these antibiotics for COVID-19 coronavirus. Senescent cells in the lungs act as host receptors for COVID-19. If anti-aging drugs and senolytics can be used to destroy senescent cells, this may be a promising area of research for virologists.
