Deloitte believes that consumers and consumer companies are moving towards a more utopian future, despite the dystopian impacts that exist today. Consumers are demanding reliable, safe, convenient, sustainable, socially just, and personalized goods and services, and companies are responding by leveraging technology to build more powerful and relevant consumer connections while making commitments to a broader set of stakeholders. Together, they are "buying into better," which means fusing profit with purpose and addressing all of the things that are important, including impacts on the environment and society. Deloitte's approach to understanding the future of the consumer industry involved engaging individuals and having over a thousand conversations to identify the market shifts that will have consequences on businesses in the next five to ten years. Instead of taking a crystal ball approach, they focused on the next five to ten years and identified three primary business dimensions impacted by six forces with nine implications. Deloitte recognizes that human centricity is critically important, and that a lot of what's important to humans goes beyond the products and services provided. The impact that companies make on the environment and society is also important to consumers, who are looking to place their dollars where they can have an impact on change. The framework for thinking about the future across three business dimensions: markets, models, and mechanics has been developed. The six forces impacting markets, models, and mechanics are shifting and broadening consumer bases, diversifying life journeys, exponential technology and AI, everything-as-a-service climate change, and geopolitical turmoil. Models and mechanics serve the market by delivering the unique customer experience or the unique product. The three Msms have emerged as an easy way to engage client executives and identify opportunities. The implications of the three business dimensions, buried throughout the three dimensions, affect organizations. Mass-to-micro is an example of how the three Msms are related. The report emphasizes the importance of organizations outsourcing parts of their business to increase efficiency and meet demand. It also highlights the need for long-term strategic plans as current business models may not work in the future. The six forces in the marketplace are rapidly changing and converging, creating new risks and opportunities for organizations. Digital goods and services are growing exponentially, and organizations must be mindful of these changes to take advantage of growth opportunities. Matthew Charnetski, MSMS, NRP, CHSOS, CHSE is the Director of Simulation-Based Education and Research for Dartmouth Health (DH) in Lebanon, NH. His winding path took him around the world several times between Iowa, Africa, Antarctica, Kazakhstan, Arkansas, New Hampshire and almost every bit in between. Matthew's simulation career has been largely academic focused primarily in medical education prior to arriving at DH in 2019. He is a graduate of the Master's in Medical and Healthcare Simulation Program at Drexel University and is actively pursuing his PhD in Health Professions Education at Maastricht University in Maastricht, Netherlands. Matthew is also adjunct faculty in the Massachusetts General Hospital Institute of Health Professions Master's in Health Professions Education Program in the Simulation Operations Track. Matthew is involved extensively with The Society for Simulation in Healthcare and The International Nursing Association for Clinical Simulation and Learning. For SSH, Matthew is Chair of the Hospital-Based Simulation Programs Section and a member of the Internal Relations Committee and Chair of the Renewal Technology Subcommittee. He serves as a member of the Diversity Equity and Inclusion Task Force and serves as a Director-At-Large for the SSH Board of Directors where he is board liaison to the Education Committee. Matthew is a member of the Standards Committee for INACSL and serves as the Chair of the Operations Standard Subcommittee. He is currently acting as a co-planning chair for SimGHOSTS planning this falls S3 conference in Singapore. His recent research interests largely focus on cultural considerations in and around healthcare simulation. His PhD work surrounds the role that culture plays in the adaptation and transfer of simulation curricula in transborder education partnerships. Matthew has contributed book chapters on simulation methodologies, moving simulation centers, and cultural considerations in simulation. Abstract: Markov state models (MSMs) have been widely applied to study the kinetics and pathways of protein conformational dynamics based on statistical analysis of molecular dynamics (MD) simulations. These MSMs coarse-grain both configuration space and time in ways that limit what kinds of observables they can reproduce with high fidelity over different spatial and temporal resolutions. Despite their popularity, there is still limited understanding of which biophysical observables can be computed from these MSMs in a robust and unbiased manner, and which suffer from the space-time coarse-graining intrinsic in the MSM model. Most theoretical arguments and practical validity tests for MSMs rely on long-time equilibrium kinetics, such as the slowest relaxation timescales and experimentally observable time-correlation functions. Here, we perform an extensive assessment of the ability of well-validated protein folding MSMs to accuractely reproduce path-based observable such as mean first-passage times (MFPTs) and transition path mechanisms compared to a direct trajectory analysis. We also assess a recently proposed class of history-augmented MSMs (haMSMs) that exploit additional information not accounted for in standard MSMs. We conclude with some practical guidance on the use of MSMs to study various problems in conformational dynamics of biomolecules. In brief, MSMs can accurately reproduce correlation functions slower than the lag time, but path-based observables can only be reliably reproduced if the lifetimes of states exceed the lag time, which is a much stricter requirement. Abstract: Abnormal changes in the neuronal microtubule-associated protein Tau, such as hyperphosphorylation and aggregation, are considered hallmarks of Alzheimer disease. Hyperphosphorylation is thought to take place before aggregation, and it is often assumed that phosphorylation predisposes Tau towards aggregation. However, the nature and extent of phosphorylation has remained ill-defined. Tau protein contains up to 85 potential phosphorylation sites, many of which can be phosphorylated by various kinases because the unfolded structure of Tau makes them accessible. However, limitations in methods have led to conflicting results regarding the overall degree of phosphorylation of Tau in cells. Here we present results from a new approach, that is based on native mass spectrometry analysis of intact Tau expressed in Sf9 cells which reveals Tau in different phosphorylation states. The extent of phosphorylation is remarkably heterogeneous with up to ~20 phosphates per molecule and distributed over 51 sites (including all P-sites published so far and additional 18 P-sites). The medium phosphorylated fraction Pm showed overall occupancies centered at 8 Pi ( 5 Pi) with a bell-shaped distribution, the highly phosphorylated fraction Ph had 14 Pi ( 6 Pi). The distribution of sites was remarkably asymmetric (with 71% of all P-sites located in the C-terminal half of Tau). All phosphorylation sites were on Ser or Thr residues. Other known posttranslational modifications of Tau were near or below our detection limit (e.g. acetylation, ubiquitination). Many biological processes including protein folding, stability, immune response, and receptor activation are regulated by glycosylation. Fucosylation of proteins regulates such processes and is associated with various diseases including autoimmunity and cancer. Mass spectrometry efficiently identifies structures of fucosylated glycans or sites of core fucosylated N-glycopeptides but quantification of the glycopeptides remains less explored. We performed experiments that facilitate quantitative analysis of the core fucosylation of proteins with partial structural resolution of the glycans and we present results of the mass spectrometric SWATH-type DIA analysis of relative abundances of the core fucosylated glycoforms of 45 glycopeptides derived from 18 serum proteins in liver disease of different etiologies. Our results show that a combination of soft fragmentation with exoglycosidases is efficient at the assignment and quantification of the core fucosylated N-glycoforms at specific sites of protein attachment. In addition, our results show that disease-associated changes in core fucosylation are peptide-dependent and further differ by branching of the core fucosylated glycans. While this separation resolves peptides with high efficiency, glycans are resolved poorly if at all. Consequently, glycopeptides consisting of the same peptide bearing different glycan structures will all possess very similar retention times and co-elute. While this has previously been viewed as a disadvantage, we show that shared retention time can be used to map multiple glycan species to the same peptide and location. In combination with MSMS and pseudo MS3, we have constructed a detailed mass-retention time database for Spike. This database allows any ESI-TOF equipped lab to reliably identify and quantify spike glycans from a single overnight elastase protein digest in less than 90 minutes. Darkrooms, glory holes, pleasure pens… in episode 7 of The Steam Room, we get frank about sex clubs. What are sex clubs? We spoke to Danie Hamman, who has been the owner of Camp David in Pretoria since it first opened its doors in 2003. Hamman calls Camp David “heaven for gays”. Historically, spaces where men could safely engage in sex with other men were rare, and restricted to spaces like public bathrooms due to the criminal nature of homosexuality and homosexual acts. Hamman felt that the trouble MSMs had with the authorities, along with several instances of gay bashing, necessitated the opening of a space where men could engage in sexual activities with other men safely, securely, and without the treat of attacks for whatever reason. Hamman makes it clear, however, that Camp David is not a brothel, and paid sex as well as drug use is strictly forbidden, and no under-18s are allowed. Jean Nel agrees with Hamman, adding that these spaces are organised to cater for specific needs within the community of men who have sex with other men. According to Bruce Little, content creator at the Anova Health Institute, sex clubs were created to provide a service to men who make the lifestyle choice to engage in thrilling and group sex activities. “A sex club is a convenient way to make sure that a certain group of men meet at a certain place at a certain time so that they can engage in group sex,” says Little. Who goes to sex clubs? Danie Hamman says that the clientele at Camp David range from gay men, to bisexual men and other men who have sex with men. Quite often, these men have specific sexual preferences and fetishes, including S&M and kinky sex. Fisting, rimming, and the chance to live out your most intimate fantasies are all on the menu at Camp David. Jean Nel agrees, and says that there is not just one type of man that frequents sex clubs like Camp David, but that all the men who do, are sexually liberated and looking to satisfy their ...

Once nearly eradicated in the United States, syphilis is back on the rise - mostly among gay and bisexual men. Rates reached an all-time low in 2000, and of the roughly 6,000 cases, only around 7% were among gay men; it was a concern almost exclusively for straight people. But better treatments for HIV lead to complacency about safe sex, perhaps especially among younger men who didn't witness the AIDS crisis of the 1980s first-hand. Now, men who have sex with men (known in medical research as MSMs) account for a full 91% of all national cases. And those nationwide numbers are reflected at home. In Louisville, rates jumped from 13.2 per 100,000 residents in 2009 to 27.7 in 100,000 - well above the national average of 18. Statewide, reported syphilis infections have almost doubled since 2009. To help explain why this is happening, and what can help, we talk this week with Dr. Kraig Humbaugh, Deputy Director of the Kentucky Cabinet for Health and Family Services. Dr. Humbaugh explains how exactly the disease is transmitted, and how you can protect yourself and your partners. Syphilis is treated with penicillin, and responds well to treatment - but you have to know you have it. Also this week, we talk to playwright and friend-to-the-show Basil Kreimendahl, whose absurdist play "Sidewinders" runs in Louisville through May 23rd, produced by Looking for Lilith Theatre Company. Kreimendahl says she set the play on a frontier in the American West to evoke a lawless place where people made their own rules - much like people are making their own rules today about gender identity. "It's just sort of about being a human being, and about naming things," she says. "You know there's a lot of power in naming things and having a name for who and what you are, and it sort of explores, what if there isn't a word for what you are that fits perfectly? How does that affect you?" "Sidewinders" runs through May 23rd at OPEN. (Full Disclosure: Our producer Laura Ellis is in the cast, and joins us in this segment to talk about her character and work on the play) In our Juicy Fruit segment, we talk about First Lady Michelle Obama's commencement speech at Tuskegee University in which she described how the public's perception of her as First Lady was colored by race and racism. Remember the "terrorist fist jab?" The New Yorker cover showing her with an afro and machine gun? "Obama's baby mama?" FLOTUS name-checked them all, and talked about the doubts they raised in her mind. "I had a lot of sleepless nights," she said, "worrying about what people thought of me, wondering if I might be hurting my husband’s chances of winning his election, fearing how my girls would feel if they found out what some people were saying about their mom." Obama also laid out the can't-win situation black women face today, especially in the public eye. "Was I too loud, or too angry, or too emasculating?
