Charles Brenner is the Alfred E Mann Family Foundation Chair in Diabetes and Cancer Metabolism in the Department of Diabetes & Cancer Metabolism. His laboratory focuses on disturbances in nicotinamide adenine dinucleotide (NAD), the central catalyst of metabolism, in diseases and conditions of metabolic stress. Among his most significant discoveries, he identified nicotinamide riboside as an unanticipated vitamin precursor of NAD. He also developed a quantitative metabolomic technology that allowed him to discover that the NAD system is disturbed by many diseases and conditions of metabolic stress including diabetes and cancer. Specifically, Dr. Brenner and colleagues have found that in animal models of fatty liver, type 2 diabetes, diabetic and chemotherapeutic neuropathy, central brain injury, heart failure, postpartum and coronavirus infection, the NAD system is disturbed. In these models, provision of nicotinamide riboside is highly protective. Dr. Brenner led the team that conducted the first clinical trial of nicotinamide riboside establishing safe oral availability and has participated in trials establishing safety and potential activities of nicotinamide riboside in clearance of liver fat and depression of common markers of inflammation. He continues to research how the NAD system is dysregulated as a function of postpartum, coronavirus infection, inflammatory conditions, diabesity and specific malignancies with the dual goals of defining mechanisms of biological regulation and developing safe preventative and therapeutic interventions for people. Question: Is whole food vitamin C superior to natural because it is part of a tyrosinase complex? Short Answer: Vitamin C is nearly ubiquitously distributed in plant tissues, and is never bound to any enzyme as a structural complex. Vitamin C promotes absorption of iron from plant foods, inhibits copper absorption, and de-loads copper from ceruloplasmin, which may play a role in distributing copper to tissues. Vitamin C is not capable of destroying ceruloplasmin. These functions follow directly from vitamin C as an electron donor and there is no evidence whatsoever that whole food vitamin C behaves differently in these respects than synthetic vitamin C. However, daily needs in most contexts are 2-400 milligrams of vitamin C per day, which is below the dose shown to potentially cause problems with copper. Getting this from whole foods or whole food supplements is better than using synthetic vitamin C because it avoids GMO corn and Chinese synthetics and provides a host of other beneficial constituents alongside the vitamin C. Question: What Is the Real Issue With Seed Oils? Short Answer: The main issue with seed oils is that they present an oxidative liability. They do not acutely cause oxidative stress, but their polyunsaturated fatty acids (PUFAs) are more vulnerable than any other macronutrient to oxidative damage. Oxidative stress can increase because of nutrient deficiencies, toxins, infections, other sources of inflammation, alcohol, or smoking, and it will inevitably increase as a function of aging. As oxidative stress increases, more PUFAs in the tissues mean more damage. At least 0.6 milligrams of vitamin E should be gotten per gram of PUFA in the diet, but vitamin E cannot fully protect against PUFA, so their intake should be moderated to the very low levels needed, as obtained by eating fatty fish once or twice a week, eating eggs daily, and eating 4-8 ounces of liver per week. Additional secondary problems with them include residual solvents and heat damage prior to intake, but the main issue is that we do not want to increase our tissue PUFA content more than needed. Question: How useful is hair trace mineral analysis (HTMA) for nutritional testing? Short Answer: Hair trace mineral analysis is included as an optional add-on in the comprehensive nutritional screening from Testing Nutritional Status: The Ultimate Cheat Sheet, because it can capture data for some ultra-trace minerals for which there are no better-validated tests, and it might capture a pattern that might not be picked up as quickly with blood work, such as a mineral transport issue. However, its utility is limited by the fact that hair mineral content is not well validated as a test for any specific mineral, is generally anti-validated when there is enough science on a mineral (such as zinc, where hair zinc does not go down in deficiency), and should not be used as a central piece of data without corroboration from other more well-validated tests, which exist for most of the nutrients. Over the years, I've noticed that people veer away from supplements with complicated-sounding names like nicotinamide riboside (NR). That could be what's kept people in the dark so much about this specific form of vitamin B3. I have to admit that I ignored it for a while, when research papers began emerging several years ago, as well. But the health and fitness benefits are too good to ignore, which is why I decided it was time to write a guide covering what it is, how you might benefit from using it, how much to use, and a few other details about NR. You'll quickly see how it could be helpful for you or someone you know. What is Nicotinamide Riboside (NR)? Nicotinamide Riboside (NR) is a naturally occurring form of vitamin B3. As a precursor to nicotinamide adenine dinucleotide (NAD+), a critical coenzyme found in all living cells, NR maintains cellular energy and supports various biological processes.Canto, C., Menzies, K. J., & Auwerx, J. (2015). NAD+ metabolism and the control of energy homeostasis: a balancing act between mitochondria and the nucleus. So wie der Motor sein Benzin braucht, so benötigt der Körper für einen funktionierenden Stoffwechsel (Umwandlung der Stoffe, der Körper ist ein biologisch-chemisches Labor) die passenden Zutaten. Da wir durch die Evolution mit allen organischen Lebewesen verwandt sind benötigen wir in erster Linie Wärme und Licht, Sauerstoff und Wasser. Weiterhin müssen wir unserem System sekundäre Brennstoffe zuführen (Sauerstoff ist der primäre Brennstoff allen Lebens), wie Kohlehydrate, Eiweiße und Fette. Darüber hinaus benötigt der Körper die Zufuhr von diversen Zutaten, ohne die der besagte Stoffwechsel nicht funktionieren kann, die deshalb (hauptsächlich über die Nahrung) zugeführt werden müssen. Weiterhin können wir allerdings mit Nahrungsergänzungsmitteln die Zusammensetzung optimieren, die Qualität erhöhen, die nötigen Zutaten zum biologisch-chemischen Labor bewußt und absichtlich zuführen. Das setzt natürlich auch ein fundiertes Wissen voraus, das man sich dann aneignen muss, und auch Disziplin, da eben nicht alles im Leben immer so einfach geht. Das Coenzym Nicotinamid-Ribosid wirkt ähnlich wie der Katalysator im Autos, es trägt in den Zellen die Elektronen. Die Anteile, die keine Elektronen aufgeladen haben, bekommen aus der Nahrung dann ihren Teil, die diese dann an die Mitochondrien weitergeben. Mitochondrien finden sich in fast jeder Zelle unseres Körpers, besonders in Muskeln und im Hirn, sind dabei verantwortlich für die Zusammenführung von Energie (aus Sauerstoff und Nahrung). Besonders im höheren Alter regelt der Körper den Stoffwechsel weiter herunter, ganz einfach weil die Kraft hierfür fehlt. Durch Zuführung von Nicotinamid-Ribosid erscheint ein Anti-Aging-Effekt einzutreten, die Beschaffenheit der Mitochondrien wird verbessert und die Herstellung von neuen Zellen vereinfacht. So kommt die Energieproduktion wieder in die Gänge, vergleichbar mit einem Holzofen, in dem ein neues Stück Holz eingelegt wird. Auch werden durch Nicotinamid-Ribosid als Nahrungsergänzungsmittel die Zellen verjüngt, die Oxidation von Fettsäuren in ein normales Mass gebracht. Besonders in Fällen von Alzheimer und Parkinson kann die Einnahme von Nicotinamid-Ribosid als Nahrungsergänzungsmittel die Beschwerden verringern, die Aktivitäten des Gehirns normalisieren, da es nervenschützende Effekte aufweist. Our guest today is Dr. Charles Brenner, the Roy J. Carver Chair of Biochemistry at the University of Iowa. Charles is one of the world’s leading experts on nicotinamide adenine dinucleotide, commonly referred to as NAD, which is an essential molecule found in every living cell. In 2004, he discovered the nicotinamide riboside kinase pathway, which leads to a special form of vitamin B3. We talk to Charles about his research into NAD and why he believes supplementation with NR could help people age better. In addition to his work at the University of Iowa, he is also the chief scientific advisor for ChromaDex, which markets the NR supplement Tru Niagen.
