His research utilises high-resolution topographic models of the seafloor and complementary geophysical and geospatial datasets to understand the geomorphic evolution and process landform relationship of the deep sea and the continental shelves. We are still talking about Antarctica, the continent that keeps on giving! Alan and Thom discuss trying to stop working momentarily, constructing a treehouse, and acquiring a shark. In the news, we rattle off a list of newly discovered species with some very cool (but hard to pronounce) names. There has been a lot of squiddy news. Footage of divers swimming with a giant squid has resurfaced; in an exclusive for the podcast, Alan has recorded more amazing Magnapinna (bigfin or elbow squid) footage. And the biggest bit of news: the colossal squid has been seen alive in its natural habitat for the first time! Thom and Kat were part of the press conference. Megalodon (the not-deep-sea and very extinct shark) has been reassessed based on what we do know. It was likely longer and slimmer than we thought, and we have estimations for their speed and size at birth. We are all back home, and we promise to stop bothering the poor continent.   Thom couldn't talk about it until after the press release, but the Schmidt Ocean Institute cruise he was on had to look at the seabed under a 150-meter-thick ice shelf right as it moved out of the way. We talked to the science leads on that cruise, Patricia Esquete and Sasha Montelli. More cable cutting in our news updates, blobfish being voted fish of the year, and the tongue-eating louse potentially being invertebrate of the year. We don't want to say we influence the news, but it seems a little spooky. Substandard and falsified (SF) antimicrobials are a neglected global health problem and have been implicated as drivers of antimicrobial resistance (AMR). Substandard medicines are authorized medical products that fail to meet either their quality standards or their specifications, whereas falsified medicines are those that deliberately and fraudulently misrepresent their identity, composition or source. In this episode of the Epigenetics Podcast, we talked with Weiwei Dang from Baylor College of Medicine about his work on molecular mechanisms of aging and the role of H3K36me3 and cryptic transcription in cellular aging. The team in the Weiwei Dang lab explored the connection between histone marks, specifically H4K16 acetylation and H3K36 methylation, and aging. Dr. Dang describes how the lab conducted experiments by mutating H4K16 to determine its effect on lifespan. They observed that the mutation to glutamine accelerated the aging process and shortened lifespan, providing causal evidence for the relationship between H4K16 and lifespan. They also discovered that mutations in acetyltransferase and demethylase enzymes had opposite effects on lifespan, further supporting a causal relationship. Weiwei Dang then discusses their expanded research on aging, conducting high-throughput screens to identify other histone residues and mutants in yeast that regulate aging. They found that most mutations at K36 shortened lifespan, and so they decided to follow up on a site that is known to be methylated and play a role in gene function. They discovered that H3K36 methylation helps suppress cryptic transcription, which is transcription that initiates from within the gene rather than at the promoter. Mutants lacking K36 methylation showed an aging phenotype. Next, the interview focuses on her work in reconstituting the MLL core complex and the insights gained from this process. In a recent discussion with CancerNetwork®, Joleen Hubbard, MD, research collaborator with Mayo Clinic and deputy director for clinical research at Allina Health Cancer Institute in Minneapolis, Minnesota, discussed new opportunities for patients with colorectal cancer (CRC).   In the discussion, Hubbard highlighted the potential of trastuzumab deruxtecan-nxki (Enhertu; T-DXd) in patients with HER2-expressing metastatic CRC based on studies including the phase 2 DESTINY-CRC01 trial (NCT03384940), which assessed the efficacy and safety of the agent in those who progressed after 2 or more prior regimens.1  These trials may play a role in continuing HER2 inhibition downstream after the FDA approval of trastuzumab (Herceptin) plus tucatinib (Tukysa) in patients with metastatic HER2-positive CRC.2   Hubbard also discussed the phase 3 MOUNTAINEER-03 study (NCT03043313) and its effects in the CRC space.3 This trial assessed the safety and efficacy of frontline tucatinib and trastuzumab in patients with treatment-refractory, RAS wild-type, HER2-positive metastatic CRC. Primary endpoints for this analysis showed a clinically meaningful overall response rate of 38.1% and a median duration of response of 12.4 months. Additionally, the treatment combination was well tolerated.   She said she is “optimistic” that moving HER2-directed therapy to the first-line setting, as seen in the MOUNTAINEER-03 study, may help outcomes for patients with metastatic CRC.  “It's a very exciting space,” Hubbard said. “Because it's only 5% to 8% of patients [who have CRC], it may not get as much attention, but there's 150,000 new cases of [CRC] diagnosed each year. This is true on a macro level but even more on a microscopic level, with many different kinds of microbes of various groups with fascinating life strategies. And despite being microscopic, with enough of them around, they can affect the whole planet's climate in significant ways. In this study, one protist species gets most of its nutrients from photosynthesis, but what it can't get from the sun, it takes from prey microbes by force. To catch its prey, it creates an intricate bubble of mucus called a mucosphere, and waits for other microbes to swim into it, thinking it is food, and get stuck. In this episode of the Epigenetics Podcast, we talked with Alistair Boettiger from Stanford University about his work on the analysis of 3D chromatin structure of single cells using super-resolution imaging. Alistair Boettiger and his team focus on developing advanced microscopy techniques to understand gene regulation at the level of 3D genome organization. They have developed Optical Reconstruction of Chromatin Architecture (ORCA), a microscopy approach to trace the 3-dimensional DNA path in the nucleus with genomic resolution down to 2 kb and a throughput of ~10,000 cells per experiment. Over the years, I've noticed that people veer away from supplements with complicated-sounding names like nicotinamide riboside (NR). That could be what's kept people in the dark so much about this specific form of vitamin B3. I have to admit that I ignored it for a while, when research papers began emerging several years ago, as well. But the health and fitness benefits are too good to ignore, which is why I decided it was time to write a guide covering what it is, how you might benefit from using it, how much to use, and a few other details about NR. You'll quickly see how it could be helpful for you or someone you know. What is Nicotinamide Riboside (NR)? Nicotinamide Riboside (NR) is a naturally occurring form of vitamin B3. As a precursor to nicotinamide adenine dinucleotide (NAD+), a critical coenzyme found in all living cells, NR maintains cellular energy and supports various biological processes.Canto, C., Menzies, K. J., & Auwerx, J. (2015). NAD+ metabolism and the control of energy homeostasis: a balancing act between mitochondria and the nucleus. In this episode of the Epigenetics Podcast, we caught up with Melissa Harrison from the University of Wisconsin-Madison to talk about her work on the “Pioneer” Transcription Factors - Zelda and Grainyhead - and their role at the maternal-to-zygotic transition. The Harrison lab studies how differentiation and development are driven by coordinated changes in gene expression. To do this, the targets of choice are the transcription factors Zelda and Grainyhead that bind to the genome at specific and crucial points in development and differentiation. These specialised transcription factors have the ability to bind to DNA in the context of nucleosomes which defines regulatory elements and leads to subsequent binding of additional classical transcription factors. Marco Trizzino and his team focus on characterising transposable elements and how they affect gene regulation, evolution and ageing in primates.
