Nebi (s.a.v.)'in bu konudaki görüşü şöyleydi: “Bir kişinin yiyeceği iki kişiye, iki kişinin yiyeceği dört kişiye, dört kişinin yiyeceği ise sekiz kişiye yeter” Hz. Âişe (r.anhâ) şöyle derdi: “Nebiyy-i Ekrem (s.a.v.) hiçbir zaman doyasıya yemek yememiştir. Ailesiyle birlikte otururken onlardan yiyecek bir şey istemez, bir şeyi canının çektiğini de söylemezdi. Önüne getirileni yer, bir yemeği sevmediğini söylemez, kendisine sunulan içeceği alıp içerdi.” Lokman Hakîm, hikmetlerinden birinde oğluna şöyle demiştir: “Yavrucuğum! Abstract: The Sigma 2 receptor ({sigma}2R) was described pharmacologically more than three decades ago, but its molecular identity remained obscure until recently when it was identified as transmembrane protein 97 (TMEM97). We and others have shown that {sigma}2R/TMEM97 ligands produce analgesia in mouse neuropathic pain models with a time course wherein analgesic onset is 24 hours following dosing. We sought to understand this unique anti-neuropathic pain effect by addressing two key questions: do these {sigma}2R/TMEM97 compounds act selectively via the receptor, and what is their downstream mechanism on nociceptive neurons. Using male and female conventional knockout (KO) mice for Tmem97, we find that a novel {sigma}2R/TMEM97 binding compound, FEM-1689, requires the presence of the gene to produce analgesia in the spared nerve injury model in mice. Using primary mouse dorsal root ganglion (DRG) neurons, we demonstrate that FEM-1689 inhibits the integrated stress response and promotes neurite outgrowth via a {sigma}2R/TMEM97-specific action. We extend the clinical translational value of these findings by showing that FEM-1689 reduces ISR and p-eIF2 levels in human sensory neurons and that it alleviates the pathogenic engagement of ISR by methylglyoxal. We also demonstrate that {sigma}2R/TMEM97 is expressed in human nociceptors and satellite glial cells. These results validate {sigma}2R/TMEM97 as a promising target for further development for the treatment of neuropathic pain. Copy rights belong to original authors. Abstract: Mitogen activated protein kinase interacting kinases (MNK) 1 and 2 are serine-threonine protein kinases that play an important role in translation of mRNAs through their phosphorylation of the RNA 5 prime-cap binding protein, eukaryotic translation initiation factor (eIF) 4E. These kinases are downstream targets for mitogen activated protein kinases (MAPKs), extracellular activity regulated protein kinase (ERK) and p38. MNKs have been implicated in the sensitization of peripheral nociceptors of the dorsal root and trigeminal ganglion (DRG and TG) using transgenic mouse lines and through the use of specific inhibitors of MNK1 and MNK2. While specific knockout of the Mknk1 gene suggests that it is the key isoform for regulation of nociceptor excitability and nociceptive behaviors in mice, both MKNK1 and MKNK2 genes are expressed in the DRG and TG of mice and humans based on RNA sequencing experiments. Single cell sequencing in mice suggests that Mknk1 and Mknk2 may be expressed in different populations of nociceptors. We sought to characterize mRNA expression in human DRG and TG for both MNK1 and MNK2. Our results show that both genes are expressed by nearly all neurons in both human ganglia with expression in other cell types as well. Our findings provide evidence that MNK1 and MNK2 are expressed by human nociceptors and suggest that efforts to pharmacologically target MNKs for pain would likely be translatable due its conserved expression in both species. Alberta and the legendary pediatric cardiology investigator, Dr. David Sahn. In this work on tricuspid valve disorders with TR and their diagnosis in fetal life, we learn about our first understandings of this complex physiology and problem and use this as starting point to speak about Professor Sahn, who passed just a few weeks ago. Dr. Hornberger was one of his most important mentees and was inspired to become a fetal cardiologist from her work with this master of echocardiography. Here Dr. Hornberger share her thoughts on Ebstein's anomaly but also Dr. Sahn and the meaningful impact a mentor can have in shaping a person's entire life and career. Jace “The Machine” Guymon is one-of-a-kind! He eats, sleeps, breathes all things hunting! McCade had the opportunity to meet Jace the fall of 2016, when he had an archery elk tag in Utah. Jace had never met McCade before, but invited him to tag along while he checked some trail cameras. He told McCade to bring his bow. The next day, Jace helped McCade bone-out and pack-out the 6x6 bull he was able to harvest. Jace showed his true colors, and was all smiles the entire day! In this episode, McCade recaps an incredibly rare opportunity, that both he and Zach had recently. In summary, only two words are needed: Buffalo Hunt!!! It was totally random, and definitely unplanned, but the two of them were able to help a once-in-a-lifetime buffalo tag holder, harvest his trophy buffalo!! Zach and McCade originally took a trip to the Henry Mountains of Utah, to film and photograph big rutting Mule Deer bucks, Thanksgiving Weekend. They did get a lot of footage of some pretty great bucks, too! You have to tune in, and listen to the crazy story unfold! Be on the lookout for the video of the buffalo hunt, on their YouTube Channel, in the very near future. As he recaps some input on topics to discuss, from the SAHN social media platforms, McCade also drops some words of wisdom, and talks about being kind to others.
