Captain John C. Maitha enlisted for military service as a combat medic in 2002. He volunteered and was selected for service with the 75th Ranger Regiment and received follow on medical training at Fort Bragg, North Carolina. CPT Maitha served in 3d Battalion, 75th Ranger Regiment as a platoon medic, company senior medic, and medical platoon sergeant where he deployed nine times in support of Operation Iraqi and Enduring Freedom. Additionally, he served on the 75th Ranger Regimental staff as the Medical Force Modernization NCO.    CPT Maitha earned a Bachelor and a Master of Science degrees in Physician Assistant Studies from the Interservice Physician Assistant Program in San Antonio, Texas, and then commissioned as an officer in 2016. He served as an Aeromedical Physician Assistant in 3rd Squadron, 17th Cavalry Regiment located at Hunter Army Airfield, Georgia. Then he was assessed and selected to serve as the 1st Battalion, 75th Ranger Regiment Physician Assistant where he deployed three times in support of Operation Freedom's Sentinel. He is currently the Field Craft Training Officer in Charge at the Combat Medic Specialist Training Program, Joint Base San Antonio, Texas.    CPT Maitha's military education includes the Senior Leader Course, Basic Officer Leader Course, Captains Career Course, Medical Management of Chemical and Biological Casualties, Medical Effects of Ionizing Radiation course, Army Flight Surgeon Course (Long), Tactical Combat Medicine Course, Jumpmaster School, Ranger School, Special Operations Combat Medic Course and Airborne School.   CPT Maitha's awards and decorations include the Bronze Star Medal with V device (2 Oak Leaf Clusters), Bronze Star Medal (2 Oak Leaf Clusters), Purple Heart, Meritorious Service Medal (2 Oak Leaf Clusters), Joint Service Commendation Medal, Ranger Tab, Combat Medical Badge, Expert Field Medical Badge, Army Basic Flight Surgeon Badge, Senior Parachutist Badge.He is happily married with four children.

Humans have been capable of leaving planet earth for just 60 years. Since that time, we have learned a lot about how to keep humans safe and healthy in space. However, our knowledge remains primitive. To become multiplanetary, physicians, scientists, engineers, and other disciplines will need to take space medicine to the next level. This will be the greatest preventative medicine challenge ever undertaken. Morbidity and MortalitySpace can kill you in 1000 different ways. Explosions, lethal radiation doses, and environmental control failures can all result in fatality. Those who spend long amounts of time in space can have long term vision issues, higher rates of cancer, and back problems. Abstract: DNA double-strand breaks, marked by Ionizing Radiation-Induced (Repair) Foci (IRIF), are the most serious DNA lesions, dangerous to human health. IRIF quantification based on confocal microscopy represents the most sensitive and gold standard method in radiation biodosimetry and allows research of DSB induction and repair at the molecular and a single cell level. In this study, we introduce DeepFoci - a deep learning-based fully-automatic method for IRIF counting and its morphometric analysis. DeepFoci is designed to work with 3D multichannel data (trained for 53BP1 and {gamma}H2AX) and uses U-Net for the nucleus segmentation and IRIF detection, together with maximally stable extremal region-based IRIF segmentation. The proposed method was trained and tested on challenging datasets consisting of mixtures of non-irradiated and irradiated cells of different types and IRIF characteristics - permanent cell lines (NHDF, U-87) and cell primary cultures prepared from tumors and adjacent normal tissues of head and neck cancer patients. The cells were dosed with 1-4 Gy gamma-rays and fixed at multiple (0-24 h) post-irradiation times. Upon all circumstances, DeepFoci was able to quantify the number of IRIF foci with the highest accuracy among current advanced algorithms. Moreover, while the detection error of DeepFoci remained comparable to the variability between two experienced experts, the software kept its sensitivity and fidelity across dramatically different IRIF counts per nucleus. In addition, information was extracted on IRIF 3D morphometric features and repair protein colocalization within IRIFs. This allowed multiparameter IRIF categorization, thereby refining the analysis of DSB repair processes and classification of patient tumors with a potential to identify specific cell subclones. The developed software improves IRIF quantification for various practical applications (radiotherapy monitoring, biodosimetry, etc.) and opens the door to an advanced DSB focus analysis and, in turn, a better understanding of (radiation) DNA damaging and repair. It’s a safe bet that Professor Michael Short’s 22.01 Introduction to Nuclear Engineering and Ionizing Radiation is the only course at MIT where students are encouraged to bring their toenail clippings to class. In this episode, Professor Short discusses one of the core principles of his teaching philosophy: the importance of making abstract concepts tangible by means of hands-on activities. Want to know how much gold or arsenic is in your body? Bombard the aforementioned toenail clippings with neutrons in a reactor and see what gamma rays they give off! Want to know whether the stone in your ring is a genuine diamond or just a cubic zirconia? Put it under the electron microscope! Professor Short also emphasizes the value of opening “knowledge gaps”—awakening his students’ curiosity by focusing on interesting questions they don’t yet know the answer to. Later, Professor Short describes how he designed the course with a built-in mechanism for collecting real-time feedback so that he can respond immediately to students’ concerns. We take you inside the core of America’s biggest nuclear power plant and trace what went wrong at Fukushima to try to figure out: when will the next meltdown happen? Background: Erufosine is a promising anticancer drug that increases the efficacy of radiotherapy in glioblastoma cell lines in vitro. Moreover, treatment of nude mice with repeated intraperitoneal or subcutaneous injections of Erufosine is well tolerated and yields drug concentrations in the brain tissue that are higher than the concentrations required for cytotoxic drug effects on glioblastoma cell lines in vitro. Methods: In the present study we aimed to evaluate the effects of a combined treatment with radiotherapy and Erufosine on growth and local control of T98G subcutaneous glioblastoma xenograft-tumours in NMRI nu/nu mice. Results: We show that repeated intraperitoneal injections of Erufosine resulted in a significant drug accumulation in T98G xenograft tumours on NMRI nu/nu mice. Moreover, short-term treatment with 5 intraperitoneal Erufosine injections caused a transient decrease in the growth of T98G tumours without radiotherapy. Furthermore, an increased radiation-induced growth delay of T98G xenograft tumours was observed when fractionated irradiation was combined with short-term Erufosine-treatment. However, no beneficial drug effects on fractionated radiotherapy in terms of local tumour control were observed. Conclusions: We conclude that short-term treatment with Erufosine is not sufficient to significantly improve local control in combination with radiotherapy in T98G glioblastoma xenograft tumours. Further studies are needed to evaluate efficacy of extended drug treatment schedules.

MicroRNA expression profiles in human cancer cells after ionizing radiation

Introduction: MicroRNAs are regulators of central cellular processes and are implicated in the pathogenesis and prognosis of human cancers. MicroRNAs also modulate responses to anti-cancer therapy. In the context of radiation oncology microRNAs were found to modulate cell death and proliferation after irradiation. However, changes in microRNA expression profiles in response to irradiation have not been comprehensively analyzed so far. The present study's intend is to present a broad screen of changes in microRNA expression following irradiation of different malignant cell lines. Materials and methods: 1100 microRNAs (Sanger miRBase release version 14.0) were analyzed in six malignant cell lines following irradiation with clinically relevant doses of 2.0 Gy. MicroRNA levels 6 hours after irradiation were compared to microRNA levels in non-irradiated cells using the "Geniom Biochip MPEA homo sapiens". Results: Hierarchical clustering analysis revealed a pattern, which significantly (p = 0.014) discerned irradiated from non-irradiated cells. The expression levels of a number of microRNAs known to be involved in the regulation of cellular processes like apoptosis, proliferation, invasion, local immune response and radioresistance (e. g. miR-1285, miR-24-1, miR-151-5p, let-7i) displayed 2 - 3-fold changes after irradiation. Moreover, several microRNAs previously not known to be radiation-responsive were discovered. Conclusion: Ionizing radiation induced significant changes in microRNA expression profiles in 3 glioma and 3 squamous cell carcinoma cell lines. The functional relevance of these changes is not addressed but should by analyzed by future work especially focusing on clinically relevant endpoints like radiation induced cell death, proliferation, migration and metastasis.

The yield of his+ reversions in the Ames Salmonella tester strain TA2638 has been determined for 60Co γ rays, 140 kV X rays, 5.4 keV characteristic X rays, 2.2 MeV protons, 3.1 MeV α particles, and 18 MeV/U Fe ions. Inactivation studies were performed with the same radiations. For both mutation and inactivation, the maximum effectiveness per unit absorbed dose was obtained for the characteristic X rays, which have a dose averaged linear energy transfer (LET) of roughly 10 keV/μm. The ratio of the effectiveness of this radiation to γ rays was 2 for inactivation and about 1.4 for the his+ reversion. For both end points the effectiveness decreases substantially at high LET, i.e., for the α particles and the Fe ions. The composition of the bottom and the top agar was the one recommended by Maron and Ames [Mutat. Res. 113, 173-215 (1983)] for application in chemical mutagenicity tests. The experiments with the less penetrating radiations differed from the usual protocol by utilization of a technique of plating the bacteria on the surface of the top agar. As in an earlier study [Roos et al., Radiat. Res. 104, 102-108 (1985)] greatly enhanced yields of mutations, relative to the spontaneous reversion rate, were obtained in these experiments by performing the irradiations 6 h after plating, which differs from the conventional procedure to irradiate the bacteria shortly after plating.
