Abstract: Myogenous temporomandibular disorders (TMD-M) are the most prevalent group of painful orofacial conditions, and the second most frequent among musculoskeletal pain conditions. TMD-M is associated with an increased responsiveness of nerves innervating the masseter (MM), temporal (TM), medial pterygoid closing (MPM) and lateral pterygoid gliding muscles (LPM). Treatment of this disorder remains difficult and is further complicated by each muscle having diverse and functionally distinct nerve innervation. This study examined expression of sensory markers in MM, TM and LPM of adult male common marmosets, a type of non-human primate. Using immunohistochemistry, we found that masticatory muscles were predominantly innervated with A-fibers (NFH+). All C-fibers (pgp9.5+/NFH-) observed in masticatory muscles were peptidergic (CGRP+) and lacked antibody labeling for mrgprD, trpV1 and the silent nociceptive marker, CHRNA3. The proportion of C- to A-fibers was highest in LPM, while MM had a minimal percentage (6-8%) of C-fibers. Interestingly, C-fibers in masticatory muscle may have myelin sheath, since many NFH- nerves were labeled with GFAP+. A-fiber types were also dissimilar among these muscles. Thus, there are substantially more peptidergic A-fibers (CGRP+/NFH+) in TM and LPM compared to MM. Almost all A-fibers in MM expressed trkC, with some of them having trkB and parvalbumin (PV). In contrast, a lesser number of TM and LPM nerves expressed trkC, lacked trkB and had fewer PV+ fibers in LPM. Along with sensory fibers, the masticatory muscles contain sympathetic fibers (tyrosine hydroxylase; TH+), which are located around blood vessels. This TH expression was absent in trigeminal neurons. Overall, the masticatory muscles of male marmosets have distinct expression patterns when compared to each muscle of the jaw and cutaneous fibers innervated by DRG neurons. Interview with KB Boyce Singer-Song writer Bassist, Drummer, and activist Queer Rebel Productions: our mission is to showcase queer and trans artists of color, connect generations, and honor our histories with art for the future. www.queerrebels.org Music https://thehomobiles.com https://www.leftfordeadrecords.com/nasty-facts Nasty Facts Nastyfacts were fronted by bassist/lead singer/songwriter Cherl Boyze, an 18 year oldQueer POC with the rest of the band being in their mid-teens or younger in 1981 when their three-song, 7-inch debut, Drive My Car, was released on the independent Jimboco label. The record was a word-of-mouth hit and sold strongly in the U.S. despite group members being too young to tour. John Peel played it repeatedly on his legendary indie- centric BBC show and helped get it released in the UK. Over 40 years after their release these songs still capture the imaginations of music lovers the world over, showing up on OG punk compilations like Killed By Death while rabid collectors pay $300 for original copies,and counterfeiters regularly sell out bootleg pressings. The reason for the EP's continued appeal is nicely captured in Steve Gardner's (editor of Noise For Heroes / Another Tuneless Racket fanzines) description of the music contained here in this re-release's liner notes: “The drums were crazed and hammering, as wild and primal in their own way as Rat Scabies on “New Rose”. The bass motored up and down, laying out a great melody but reinforcing the rhythm as well. The guitar was power-chording nirvana, and when the lead break came it was perfect. The vocals weren't the hyper-aggro thing that was the stock-in-trade most of the furious punks of the day; the lead was handled deftly by Cherl Boyze, who, I wrote in NFH #9, was ‘black, female, and at 18 the oldest member of the band”. She didn't snarl, but she didn't croon, either, and when her male bandmates added backing vocals the result was pop/punk magic. And finally, the production had all kinds of wild bad driving sound effects that just elevated everything.'” Nastyfacts were part of a Brooklyn teen-punk scene that included the Speedies and The Stimulators. Their core line-up consisted of Cherl Boyze on bass and lead vocals, guitarists Brad Craig and Jeff ;Range Tischler with Genji Searizak & Siraisi on drum; these last three also sang back-up. They began playing music together while attending St. Ann's School in Brooklyn and after a year of playing covers locally began building a following in Manhattan playing amateur nights at CBGBs and then regular slots at Max's Kansas City and other venues that'd host all-ages shows. They soon accumulated a full album's worth of solid original material, but unfortunately only made studio recordings of the three amazing songs that make up Drive My Car.

R., Macarron, V., Hubrich, J., D'Este, E. Abstract: Neurofilaments are one of the main cytoskeletal components in neurons and they can be found in the form of oligomers at pre- and postsynapses. How their presence is regulated at the postsynapse remains widely unclear. Here we systematically quantified by immunolabeling the occurrence of the neurofilament isoform triplet neurofilament light (NFL), medium (NFM), and heavy (NFH) at the postsynapse with STED nanoscopy together with markers of synaptic strength and activity. Our data shows that within dendritic spines neurofilament isoforms rarely colocalize with each other and that they are present to different extents, with NFL being the most abundant isoform. The amount of the three isoforms correlates with markers of postsynaptic strength and presynaptic activity to varying degrees: while NFL shows moderate correlation to both synaptic traits suggesting its involvement in synaptic response, NFM and NFH were only correlated on a low level. By quantifying the presence of neurofilaments at the postsynapse within the context of the synaptic status, this work sheds new light on the regulation of synaptic neurofilaments and their possible contribution to psychiatric disorders. Abstract: Mechanosensitive bladder sensory neurons are critical for sensing bladder distention, but their role in bladder pain and bladder pathology is poorly understood, due in part, to the challenges of identifying mechanoreceptors in tissue sections. A lot of what is known about how disease alters sensory innervation of the bladder comes from studies that traditionally focus on nociceptive nerve terminals. In seeking tools to characterize the role of non-nociceptive afferents in the bladder, we first examined neurofilament heavy (NFH), a marker for medium to large-diameter myelinated A fibers, in combination with the common marker for peptidergic nociceptors, calcitonin gene-related peptide (CGRP). While there was partial overlap between NFH and CGRP, 87% of NFH fibers were CGRP-negative, underscoring the abundance of non-nociceptive A fibers nerve terminals in the bladder. Two mouse lines that have been used for genetic labeling of mechanoreceptors of the skin were tested for their ability to label bladder afferents. Once crossed to Cre-dependent reporter lines, tyrosine kinase B (TrkB) TrkBCreER2 mice can be used to label A-delta mechanoreceptors while receptor tyrosine kinase ret protooncogene (Ret) RetCreER2 mice can label a combination of A-beta mechanoreceptors and non-peptidergic nociceptors in adult mice (late Ret). Both mouse lines produced successful labeling of bladder nerve terminals demonstrating partial overlap with NFH and minimal overlap with CGRP. Thus, we have identified new genetic strategies to investigate CGRP-negative subpopulations of bladder afferents that remain largely uncharacterized in studies that target peptidergic nociceptors. These tools can help elucidate the role of mechanosensitive afferents in bladder pathophysiology and urologic chronic pain. Copy rights belong to original authors. Abstract: Blood-based biomarkers can be informative of brain disorders where protein aggregation play a major role. The proteome of plasma and circulating protein aggregates (CPA) reflect the inflammatory and metabolic state of the organism and can be predictive of system-level and/or organ-specific pathologies. CPA are enriched with heavy chain neurofilaments (NfH), key axonal constituents involved in brain aggregates formation and biomarkers of the fatal neurodegenerative disorder amyotrophic lateral sclerosis (ALS). Here we show that CPA and brain protein aggregates (BPA) from ALS differ in protein composition and appear as a combination of electron-dense large globular and small filamentous formations on transmission electron microscopy. CPA are highly enriched with proteins involved in the proteasome and energy metabolism. Compared to the human proteome, proteins within aggregates show distinct and tissue-dependent chemical features of aggregation propensity. The use of a TMTcalibrator proteomics workflow with ALS brain as calibrant reveals 4973 brain-derived low-abundance proteins in CPA, including the products of translation of 24 ALS risk genes. 285 of these (5.7%) are regulated in ALS CPA including FUS (p

After a chance meeting while coaching young athletes I stayed in touch with Eric Bostrom for two reasons. First, he had a clean energy. I know that sounds pretty damn woo-woo, but as I've gotten older I've just learned to trust my gut more when it comes to meeting people. Eric was soft spoken but from the moment I met him there was an obvious intensity to him. That leads to the second reason: He was a champion motorcycle racer. You don't get to be a champion by being unfocused, and God knows I love the folks who can dial in focus to the white hot intensity of a thousand burning suns. Getting on a motorcycle (for me) represents the fastest way to transition from the normal humdrum of life into an experience that puts you at mortal peril. One minute you're straddling an inert piece of metal and plastic in front of your house, the next you're in traffic, whipping along at speeds humans just weren't meant to attain, surrounded by heavy and fast beasts that basically don't notice or care whether you exist. Bloody dangerous, and open to anyone crazy enough to swing a leg over and twist the throttle. Racing motorcycles is a few steps further, and something I've always thought far too dangerous for me. Eric is one of those guys who raced at a bunch of levels, who chased victory hard for years because that's how he paid the bills, and who squeaked through the injury cycle of professional motorsports and out the other side with a combination of skill and luck. On that far side of competition he created a solution for the neck and back pain that had come from the crashing, dings, and dents that are a part of hurtling through space with basically no protection. At the end of the show we talk about the Backmate, which we use at the Paleo Treats office. Not only does the thing work and provide relief, it stands as a shining example of how to ROCK a Kickstarter campaign. You'll notice some common themes with Eric and Brian Enos. Competition forces you to get better. The more skilled you become, the better you are at listening to your environment. You don't need to try harder to go faster, you need to think better. This show with Roy Harris skips most of his (DEEP!) martial arts history and jumps into two things he's passionate about and equally stoked on: Teaching and Human Movement. Roy has the kind of resume you might build if you were writing a character for Hollywood that needed to be able to fight anybody, anywhere, with anything, but never needed to. He's known for his Jiu Jitsu courses, so we jump way into how he came up with his teaching style, what he thinks is important with teachers and students, and (of course) we can't avoid his love for Filipino Martial Arts. David Easton builds "museum quality, visual masterpiece walls that happen to support the roof." His medium is rammed earth, he's been doing this for 40 years, and if you've ever loved any kind of architecture or building you'll dig this show. This is closer to Paleo than many of our other podcasts in the sense that rammed earth building is a "locavore" (or loca-building) method. Typically you'll use what's on site (the earth) to pour into forms, tamp into rock, remove the forms and enjoy the heck out of one of the most beautiful walls you'll ever see. I was introduced to David Easton through his book, The Rammed Earth House, many moons ago, even going so far as to build a rammed earth wall for a garden in our backyard back in '09. As a long time fan of rammed earth & Easton's work, I'm super stoked to have him on the show to talk about one of our mutually favorite subjects: Rammed earth. Listen for this quote, it's one of my favorite from any of these podcasts: "Every day in some form or another, I touch earth. I don't mean I walk on it, everybody does that. I mean the earth that you pick up and touch, it's all different, you know. Some of it's granular and coarse, and some of it's silky and soft. One of my favorite guests, Roz Savage combines the fierce heart of an adventurer with the ability to describe beautifully the wildest & loneliest places on the planet.As a Guinness World Record holder (first woman to row solo across 3 oceans), Roz is a shining example of someone following her dream. She started out as a money hungry young professional, saying “I was laboring under the illusion that money was going to make me happy.” Working in London alongside the rest of the sharks (and whales) cured her of that drive; she realized it was an illusion, and in a famous story wrote down two obituaries. First, the one she was headed for, and second, the one she wanted.Staring her in the face was her whole future, and it wasn't one she wanted. “I realized to my great embarrassment and shame that the dream that I'd been chasing for 11 years was not the right dream.”She was ““…actually quite terrified of the implications of realizing I was living the wrong dream. I had to self-sabotage because I didn't have the courage to change.”So, cutting ties with job, relationships, and material possessions, she set out to row across the Atlantic and began to live the path she actually wanted.Along the way she learned when to use the word fail (or really, what to replace it with) as well as what the right questions to ask were.
