Abstract: Brain endothelial cells (BECs) play an important role in maintaining central nervous system (CNS) homeostasis through blood-brain barrier (BBB) functions. In addition to actively regulating material exchange between the circulation and CNS, BECs express low baseline levels of adhesion receptors, which limits entry of leukocytes. Supporting cells in the neurovascular unit, such as astrocytes and pericytes, are crucial for promoting this immune privilege phenotype in BECs. However, the molecular mediators governing this phenotype are unclear. Here, we explored how BBB immune privilege is influenced by IQGAP2, a scaffold protein associated with elevated barrier function. Using single-cell RNA sequencing and immunohistology, we show that BECs from mice lacking Iqgap2 exhibit a profound inflammatory signature, including extensive upregulation of adhesion receptors and antigen-processing machinery. Further, in zebrafish and mice, loss of Iqgap2 increases infiltration of peripheral leukocytes into the brain. Overall, our results implicate IQGAP2 as an essential regulator of BBB immune privilege. Seth Shostak is a renowned astronomer and science communicator who is best known for his work on the search for extraterrestrial intelligence (SETI). As the Senior Astronomer at the SETI Institute, a non-profit research organization based in Mountain View, California, Shostak has been at the forefront of SETI research for over three decades. Stu's career in television and broadcasting dates back to the early 1970s when he began as an audience coordinator for game shows and sitcoms. In 1982, he received his performing break when he was given an opportunity to do the audience warm-up at the popular situation comedy, "Diff'rent Strokes". He did so well that he was assigned regular warm-up duties the following season on the Ricky Schroder sitcom, "Silver Spoons", a position he held for two years. Stu has also done warm-ups for "The Jeffersons", "Mama's Family","Amen", and many others. Behind the scenes, he's been the owner/operator of Shokus Video, a company dedicated to the preservation of 1950s TV shows on DVD and video tape, since 1979. He was also the producer and director of "The Jack La Lanne Show Commemorative Special" home video, as well as Lucille Ball's personal film archivist for the last ten years of her life. He produced, wrote, and directed a bonus featurette for Shout! Factory's "Leave it to Beaver - The Complete Series" DVD box set that starred the series' Ken Osmond and Frank Bank (Eddie and Lumpy), along with supplying a generous amount of vintage archival footage and several "Stu's Show" interviews for the release. He most recently hosted an on-camera bonus video and co-produced Shout! Factory's "Dennis the Menace Season One" DVD set. Today on Too Opinionated we visit with Television historian, archivist and podcast pioneer Stu Shostack! Award-winning filmmaker CJ Wallis' latest documentary STU'S SHOW, chronicling the relationship between legendary actress Lucille Ball and TV historian Stu Shostak, premieres on digital May 2, 2022 from Upstream Flix. The story of TV historian and archivist Stu Shostak, the documentary features screen-legends Tony Dow (“Leave it to Beaver”), Michael Cole ("The Mod Squad"), the late Ed Asner (“Lou Grant”), Butch Patrick (“The Munsters”), Academy Award Winner Margaret O'Brien, Geri Jewell and many, many more. Shostak got his start handing out tickets to Norman Lear sitcom tapings to people in Hollywood and parlayed that into doing audience warm-ups prior to tapings for shows such as “All In The Family” and “One Day At A Time”. The film also indirectly tells the story of television legend Lucille Ball who, contrast to popular belief, was an extremely kind and generous person who treated those around her like family. Ball is hired to hold Q&A's at a Los Angeles based College which provides a window of opportunity that alters the course of Stu's life. Shostak's encyclopedic knowledge of Ball's career earns his way into becoming an essential part of her small inner circle as her archivist and assistant to her husband Gary Morton. After Ball passes away, Shostak pioneers what we know now as “podcasting”, hosting internet shows interviewing celebrity cast and crew of the golden age of television. He also co-produces the widely successful LOVING LUCY conventions which welcomed prior cast, crew and superfans from around the world to come together for a few days to celebrate their love of I Love Lucy. At one of these Loving Lucy conventions Stu meets Jeanine Kasun, a music teacher and Lucy super-fan, who noticed Shostak hosting game shows, events and trivia contests. The two speak on the phone at length and eventually begin dating until Jeanine suffers a brain aneurysm and the two, alongside the legends of television, enter into a war with the medical industry to keep her alive. Dr. Seth Shostak is the senior astronomer at the SETI Institute. He claims to have developed an interest in extraterrestrial life at the tender age of 10, when he first picked up a book about the solar system. This innocent beginning eventually led to a degree in radio astronomy, and now, as senior astronomer, Shostak is an enthusiastic participant in the Institute's SETI observing programs. In addition, he is keen on outreach activities: interesting the public – and especially young people – in science in general, and astrobiology in particular. He's co-authored a college textbook on astrobiology, and has written three trade books on SETI.
