Summary In dieser Gastvorlesung geht es um den Einsatz von Künstlicher Intelligenz (KI) im Studium und in der Praxis. Gerade ging im australischen Hobart die 21. Welt-Windenergie-Konferenz zu Ende. Neben technischen Themen wurde dort sehr viel über die Themen Bürgerbeteiligung und Bürgerenergie gesprochen, in vielen Ländern heute maßgebliche Faktoren beim Ausbau der Windenergie. In den ersten Jahren sicherte eine lokale Verankerung den Fortschritt bei der Windkraftnutzung, vor allem in Dänemark und Deutschland. Mein Gast Heinrich Bartelt ist einer der ersten Windpioniere Deutschlands und hat verschiedene Verbände mit aufgebaut, um die energiepolitischen Rahmenbedingungen im Sinne der Erneuerbaren Energien zu beeinflussen. Mit ihm spreche ich darüber, wie lokale Initiativen die moderne Windindustrie gestartet haben und was in den vergangenen 40 Jahren erreicht wurde. Er kehrt gerade von der Welt-Windenergie-Konferenz zurück, bei der auch diskutiert wurde, was heute die maßgeblichen Herausforderungen sind und was der Windsektor von den frühen Pionieren und ihren Erfolgen lernen kann.

Abstract: In thermogenic adipocytes, uncoupling protein-1 (UCP1) is a key mediator of non-shivering thermogenesis (NST) by uncoupling the electron transport chain from FoF1-ATP synthase-mediated ATP production. While regulatory mechanisms of UCP1 are important for NST, it is unknown whether also the activity of ATP synthase is modulated during NST. Here, we show a critical role of Inhibitory Factor 1 (IF1), an inhibitor of ATP synthase, for brown adipocyte energy metabolism. In mice, IF1 protein content is diminished in brown adipose tissue of mice after 5 days of cold exposure. Additionally, the capacity of ATP synthase to generate mitochondrial membrane potential (MMP) through ATP hydrolysis (the so-called reverse mode) was higher in mitochondria isolated from cold-adapted mice compared to mice housed at room temperature. While silencing of IF1 in cultured brown adipocytes did not affect MMP, IF1 overexpression resulted in an inability of mitochondria to sustain MMP upon adrenergic stimulation. The effects of IF1 overexpression on MMP were blunted when UCP1 was silenced or when a mutant IF1, incapable of binding to ATP synthase, was used. In brown adipocytes, IF1 ablation was sufficient to increase mitochondrial lipid oxidation and the cellular dependency on glycolysis to produce ATP. Conversely, IF1 overexpression blunted mitochondrial respiration without causing energetic stress, leading to a quiescent-like phenotype in brown adipocytes. In conclusion, our data show that the cold-induced downregulation of IF1 facilitates the reverse mode of ATP synthase and enables proper bioenergetic adaptation of brown adipose tissue to effectively support NST. Abstract: Glutamatergic projection neurons of the lateral habenula (LHb) drive behavioral state modulation by regulating the activity of midbrain monoaminergic neurons. Identifying circuit mechanisms that modulate LHb output is of interest for understanding control of motivated behaviors. A small population of neurons within the medial subnucleus of the mouse LHb express the GABAergic synthesizing enzyme GAD2, and they can inhibit nearby LHb projection neurons; however, these neurons lack markers of classic inhibitory interneurons and they co-express the vesicular glutamate transporter VGLUT2. To determine the molecular phenotype of these neurons, we genetically tagged the nuclei of GAD2-positive cells and used fluorescence-activated nuclear sorting to isolate and enrich these nuclei for single nuclear RNA sequencing (FANS-snRNAseq). Our data confirm that GAD2+/VGLUT2+ neurons intrinsic to the LHb co-express markers of both glutamatergic and GABAergic transmission and that they are transcriptionally distinct from either GABAergic interneurons or habenular glutamatergic neurons. We identify gene expression programs within these cells that show sex-specific differences in expression and that are implicated in major depressive disorder (MDD), which has been linked to LHb hyperactivity. Finally, we identify the Ntng2 gene encoding the cell adhesion protein Netrin-G2 as a marker of LHb GAD2+/VGLUT+ neurons and a gene product that may contribute to their target projections. These data show the value of using genetic enrichment of rare cell types for transcriptome studies, and they advance understanding of the molecular composition of a functionally important class of GAD2+ neurons in the LHb.
