BUFFALO, NY - April 15, 2025 – A new #research paper was #published in Oncotarget, Volume 16, on April 4, 2025, titled “Association between two single nucleotide polymorphisms of the Prostaglandin-Endoperoxide Synthase 1 and 2 genes and cell proliferative prostatic diseases in Lebanon." The team of researchers led by first author Brock J. Sheehan and corresponding author Ruhul H. Kuddus, from Utah Valley University, discovered that a specific genetic variation in the PTGS2 gene is associated with a higher risk of benign prostate hyperplasia (BPH), a common condition in aging men. The study, which focused on Lebanese men, suggests that the C allele of the -765 G>C polymorphism in the PTGS2 gene may increase risk to this non-cancerous but problematic prostate condition. This finding could help identify men at greater risk earlier and lead to better treatment choices. Benign prostate hyperplasia and prostate cancer are two common conditions that involve abnormal cell growth in the prostate gland. While prostate cancer is malignant and potentially life-threatening, BPH is a non-cancerous enlargement that can still significantly affect quality of life. Both conditions are widespread in older men, with BPH affecting over 70% of men above 60. Researchers have long suspected that inflammation-related genes may play a role in their development. In this study, the focus was to study PTGS1 and PTGS2, genes that help produce enzymes involved in inflammation. Using DNA samples from 168 Lebanese men, including 61 with prostate cancer, 51 with BPH, and 56 healthy controls, the researchers analyzed two common gene variants. They found no link between the PTGS1 variant and either condition. However, the PTGS2 variant showed a strong association with BPH. Men carrying the C version of this gene were more than twice as likely to have BPH compared to those without it. While a similar trend was observed in men with prostate cancer, the results were less conclusive. "The C allele of SNP-765G>C of the PTGS2 gene was significantly associated with an increased risk of BPH (OR = 2.30, p-value = 0.01)." This is the first study to report a genetic link between the C allele of the -765 G>C polymorphism in the PTGS2 gene and BPH in Lebanese men. It builds on earlier findings that associated this gene variant with various cancers, including prostate, colon, and stomach cancers. Although based on a relatively small and specific population, the study offers new insight that could help improve genetic screening and guide prevention strategies. The research also points to the potential benefits of COX-2 inhibitors—drugs already used to treat prostate conditions—which may be more effective for men with certain PTGS2 gene types. Further studies in larger and more diverse groups are needed to confirm these results and explore how this gene variant influences prostate disease. In the future, simple genetic tests could help identify men at higher risk before symptoms appear, allowing for earlier and more personalized care. The woolly mammoth, a giant furry elephant-like creature that roamed the Ice Age, is one of the top candidates. They're also trying to bring back the Tasmanian tiger, a wolf-like marsupial that disappeared in the 1930s. In this episode, we discuss breaking up complex research projects into phases. This is a great way to make progress on a research question that might otherwise seem daunting. We'll talk about how to identify the mother of Elsie, a woman born in 1820, as an example of how to set up phases for research. We'll also discuss how to determine which phases are necessary and how to write research objectives. We wrap it up with a discussion of Diana's Clemsy Cline project phases. The Clemsy Cline project sought to identify the father of Mary "Clemsey" Cline. Clemsy was born about 1817 in Alabama and died after 1880 in Wise County, Texas. She married William Henderson Weatherford in about 1839 in Arkansas. Phase 1 of the project focused on finding a candidate for Clemsy's father. Diana analyzed census records from 1850, 1860, and 1880 and concluded that she was most likely born between 1817 and 1820 in Alabama. Her father would have been born before 1800 in Virginia. The researchers also found that Clemsy was associated with a number of people in Arkansas, including Jacob Cline, Mahala Cline, and John Cline. These individuals were all living in the same area at the time that Clemsy was born, and they may have been able to provide some clues about her father's identity. Phase 2 of the project focused on exploring the land patents of Clemsy Cline's FANs. Diana identified three Cline individuals who patented land in Izard County, Arkansas, between 1838 and 1850: John C. Cline, Mahala Cline, and John Cline. Diana believes that these land patents may provide some clues about the father of Clemsy Cline. Phase 3 of the Clemsy Cline project is currently ongoing andseeks to test the hypothesis that John C. Cline was the biological father of Clemsy Cline. Using DNA evidence and documentary research, Diana has found a cluster of Clines in a network graph. This cluster includes descendants of Mahala Cline Shockley and descendants of John C. Cline through his 1850 census family with probable second wife, Milky Cline. The death of Christchurch woman Mellory Manning is one of two cold cases police are keen to crack, looking to a new DNA tool. Police confirm they're trialling Forensic Investigative Genetic Genealogy testing, which compares DNA of unidentified suspects with genetic profiles on genealogy websites. Mellory's brother Rob Manning says this technology is a positive step forward for everyone in society. Using DNA evidence, the scientific journal Nature publishes findings that put to rest a centuries-old mystery: Was Sally Hemings, an enslaved woman at Monticello, the mother of six of Thomas Jefferson's children? Until then, the historical consensus had been this: “The Jefferson-Hemings relationship can be neither refuted nor substantiated.” Jefferson's white descendants were more categorical: they flatly denied it. But now the truth was out. Why was this story denied for so long, and what does that say about whose version of history is believed? Join Dr. Gladden on this conversation with Ryan Smith about epigenetic age testing and DNA methylation. Ryan has a biochemistry degree from the University of Transylvania. He went to medical school and started clinical rotations before realizing he didn't want to be a doctor. That's when he pivoted to the world of pharmacy, focusing on peptide synthesis, and opened up Tailor Made Compounding.
