Mikki Tal, PhD, is a Principal Scientist at MIT's Department of Biological Engineering and Associate Scientific Director of the MIT Center for Gynepathology Research. BUFFALO, NY — June 25, 2025 — A new #research paper was #published in Aging (Aging-US) Volume 17, Issue 5, on May 28, 2025, titled “Investigating telomere length in progeroid syndromes: implications for aging disorders.” In this study, led by first author Luma Srour and corresponding authors Yosra Bejaoui and Nady El Hajj, from Hamad Bin Khalifa University, Qatar Foundation, researchers investigated whether shortened telomeres, a marker of cellular aging, are present across various rare genetic disorders that mimic early aging, known as progeroid syndromes. The study found that telomere shortening, also called telomere attrition, is not a shared characteristic of all these disorders. This finding is important because it challenges the belief that telomere loss is a common thread in premature aging and could help refine how these syndromes are studied and treated. Progeroid syndromes are rare conditions that cause individuals to display symptoms of aging far earlier than expected. While some of these syndromes share signs of typical aging, others arise from very different genetic alterations. Researchers focused on comparing telomere length in individuals with several of these syndromes to better understand how aging develops at the cellular level. Telomeres are protective ends of chromosomes that shorten as cells divide, acting as a biological clock linked to aging and disease. Using DNA methylation data from blood samples, the team analyzed telomere length in patients with six progeroid syndromes: Werner Syndrome, Hutchinson-Gilford Progeria Syndrome, Berardinelli-Seip Congenital Lipodystrophy type 2, Dyskeratosis Congenita, Cerebroretinal Microangiopathy with Calcifications and Cysts, and Wiedemann-Rautenstrauch Syndrome. They found significant telomere shortening only in classical Werner Syndrome, Berardinelli-Seip Congenital Lipodystrophy type 2, and Dyskeratosis Congenita. Other syndromes, including the widely studied Hutchinson-Gilford Progeria Syndrome, did not show this pattern. “To investigate whether progeroid syndromes have telomere attrition, we calculated telomere length using the DNAmAge web-based calculator.” The findings suggest that telomere shortening cannot be used as a universal marker for all forms of premature aging. In fact, some syndromes linked to DNA repair problems showed telomere damage, while others with different genetic mutations did not. These results indicate that the underlying biology of each syndrome is crucial in determining whether telomere shortening plays a role. Researchers also compared telomere lengths in people with genetic variants known to protect against age-related diseases. Those with protective genes had longer telomeres than people with progeroid syndromes. This study challenges assumptions about aging in rare disorders and highlights the need for more personalized approaches in aging research. By identifying which syndromes involve telomere attrition, it opens new avenues for treating or delaying aging-related symptoms. Future research may explore other biological factors behind premature aging and how genetic differences influence the aging process. The most debilitating one is gut pain around the mid-lower right side of my large intestine(?). Often, it's at it WORST in the AM and causes me to wake up around 4-5. After being awake for 1.5 hours, I fall back to sleep. Was told its my appendix and almost had it removed in '22-'23. Doctors I've met with recently said if it were my appendix, it would hurt more. Been diagnosed with hiatal hernia, LPR, IBS, slow gut motility, stool burden, diverticulosis etc. To cut a long story short, not looking for a diagnosis, just wondering if you've seen this in your practice. Namely, early morning gut discomfort on the right side (colon?) which often subsides as the morning progresses or is relieved via bowel movements. My journey with PCOS began after the birth of my third child. My issues started post-pregnancy, with the onset of thyroid problems after each of my three children's births, all delivered via emergency C-sections. After my youngest was born prematurely, I began experiencing excruciating pain caused by ovarian cysts. The pain from the cysts was debilitating. I vividly remember collapsing in my bathroom from the stabbing pain, thinking something was seriously wrong with my C-section scar. After multiple episodes and many frustrating visits to multiple doctors, I was told to just go on birth control or take painkillers - band aids that didn't address the root issue. The worst advice I got (from a male doctor no less) — to get pregnant again to avoid cysts — was the catalyst for me to take control and find natural solutions myself. I began researching the female cycle, eating for my cycle and focusing on hormone patterns. This included implementing seed cycling and matching my foods to the different phases of my cycle. The next layer was focusing on my excess estrogen. I worked on detoxing and supporting my liver and gut health. Along with this came a focus on stress and cortisol. Over the years, I've continued to research and implement tools to support my hormone levels. This consistency has paid off - I haven't had a cyst in over 12 years! As I approach menopause, I am focused on building muscle and maintaining my metabolic health. PCOS is a metabolic issue, and it requires ongoing management, especially as hormonal changes come with age. Here are some takeaways if you are struggling with PCOS: Addressing PCOS requires looking at your overall health, not just reproductive symptoms. This includes diet, stress management, and supporting your body's natural detox processes. Regular hormone testing, such DUTCH testing (a comprehensive dried urine test for hormones), will give you the info you need to pin-point certain hormone levels and see what's working. Your body's needs change over time, especially with life events and aging. Be prepared to adjust and adapt with your body. Everyone with PCOS has a unique experience. I think the name should really be “Please Confirm this Ovary Syndrome.” I'm telling you, I find that there's no condition that's more misdiagnosed than PCOS. Today on The Egg Whisperer Show podcast, I'm talking about PCOS, or Polycystic Ovary Syndrome. Did you know that celebrities like Jillian Michaels and Posh Spice both have PCOS? A few PCOS facts: PCOS does not equal infertility. You may have a harder time getting pregnant if you have PCOS, because you may not be ovulating frequently or regularly. It's difficult only because it's hard to know when you're ovulating. When you do ovulate with PCOS, you are likely to ovulate multiple eggs, making you very fertile! Every time you ovulate, you make a cyst of ovulation. Ovulating multiple eggs is what creates multiple cysts, which is where you get the name: Poly (multiple) cystic (cystic). Some symptoms may include: hair loss of the scalp, hirsutism (or hair on your body in areas like the chin, upper abdomen, lower abdomen or back), high testosterone levels, high insulin levels, higher estrogen levels, and higher BMI. You can help manage PCOS by a paleo-like or mediterranean diet, taking birth control pills which can lower testosterone, or other testosterone-lowering drugs, and supplements. I am not sure if this has attributed to my conditions, but I do take a digestive enzyme along with HCL with each meal. I am not sure if I am taking enough, but I take what I can afford. My GI doctors have been stumped by my conditions, but did offer antibiotics for my LIMO, which I am considering because my insurance will cover it and I am out of money. I know you've done many episodes on melatonin and are generally a proponent of its usage in moderate amounts. I typically take 2-3 mg per night. But there are a lot of people in the health space who do not think taking melatonin is a good idea. I hear them usually say something about it being a hormone and not wanting to introduce external hormones to the body if you can help it. But I've always found their rationale to be vague. I know you've done so much research on melatonin. Could you articulate the “steel man” argument for why taking 1-5mg of melatonin every night may not be a good idea? Jessica: One more question on melatonin - Would you advise someone stop taking melatonin when they become pregnant? That seems to be the conventional wisdom because there is little research on it. But, in theory, it seems like melatonin could be useful for calming the mother's central nervous system, improving sleep, providing antioxidants, etc. — all of which would be favorable to the baby.
