FDA 483 #10 – Non-Viable Particle Counting

Learning Objectives Discuss the importance of particulate in relation to microorganisms Explain why 0.5 micron and larger sized particles are of great importance Discuss 3 ways that we can minimize the number of particles in our cleanroom In todays 483 we're going to be looking at non-viable particle counts otherwise and more accurately known as total particle counts and why they're so important. So let's take a look at the 483 observation first then we'll dive into it a little deeper. In this 483 it's noted that there was an out of specification result for environmental monitoring samples. I've talked in a previous podcast and posts about viable particles, but this week let's focus on non-viables. In observation 2 under section "d" it talks about not further investigating sterility failures for two particular compounds. The investigation documentation did not include and evaluation of production batch record reviews, EM for hoods and rooms (non-viable or viable particulates, personnel monitoring, press plates), cleaning logs for the room, qualification of the hoods, retain samples or previous OOS. FDA 483 Observation 2 So why does it mention specifically looking at non-viable samples, aside from all of the others? Let's take a quick look at a table that's from USP , which is actually from an international document, ISO 14644. I'm sure you've seen this before but if you haven't here it is. But what exactly do all of these numbers mean and why are they so important? In the left column you have ISO classes which are the classes of different areas of the clean room. Your primary engineering controls or your hoods are typically ISO class 5 and your secondary engineering controls or your room are typically ISO class 7 or you may have an anteroom that's ISO class 8. In the right hand column you have the limits of the particle counts per cubic meter, meaning if you have particle counts higher than the numbers in this column corresponding to the particular ISO class that area fails and cannot be classified as such. One thing to note here, at the bottom of the table in small writing it specifically mentions the numbers pertain to particles of a particular size 0.5 micron or larger. Why are 0.5 micron sized particles of importance? Well, there's one thing you should know about microorganisms. Rarely are microorganisms free floating by themselves. They actually travel on particles, and if you haven't already guessed they're usually attached to particles that are 0.5 micron or larger. One other interesting point worth mentioning and this may seem like common sense but the larger the particle, the more dense it is and the heavier it is. These larger sized particles are the ones that tend to settle on surfaces. Smaller, lighter particles can stay free floating in the air but it's the job of our engineering controls to push them away from our critical areas and hopefully push them down toward the ground where they're not a threat to our preparations. Now, if you have high particle counts in your cleanroom or primary engineering control does that mean that every one of those particles has a microorganism attached to it? No, not necessarily. However, taking particle counts is a great, general way of looking at the state of cleanliness of your cleanroom. Another question is, do we know the percentage of particles that have microorganisms attached to them? To be completely honest, I'm not sure if there's been a study done to make that correlation but one thing that's for certain is that the higher the particle counts in your classified areas, the higher the chance for contamination. One point I really want to drive home is that when we get our cleanroom certified and we're looking at the numbers of particle counts, you can glean a lot of information from that report. If you have high particle counts in a particular area, you can look inside your room and see what might be causing those high particle counts. How do particles get inside our cleanroom? GREAT question, they travel on materials that are brought into the cleanroom, equipment, and of course people that are going in and out of the cleanroom. Ok, great...we know what particle sizes we're looking for, why those particular particle sizes are important (because they carry microorganisms) and we know how they get in the cleanroom. The next question is how do we keep them OUT of the cleanroom? This comes down to a few simple and easy practices that you can do on a daily basis to minimize the number of particles you're bringing into the cleanroom. First, people. It's inevitable that people are going to be in the cleanroom since they're the ones performing the compounding. It comes down to how these people are gowned, how much skin is visible, and their general behavior while inside the cleanroom. As I said before if our engineering controls are doing their job properly they're hopefully pushing particles out of our primary engineering controls and the secondary engineering controls are pushing them down to the ground. This is where good daily practices come into play. As a cleanroom operator, we should have an awareness at all times all of the things I've just gone over. Just being in the cleanroom, we're disrupting the flow of air, so we want to do our best to disrupt this airflow as little as possible and we do that by moving slowly, methodically and purposefully. You don't want your operators rushing around, using fast hand movements or shuffling their feet which only puts the particles that may be on the ground back into the air. Another way of minimizing particle counts is ALWAYS wiping off materials with disinfectants prior to bringing them into the cleanroom and also prior to putting them into our primary engineering controls. This is absolutely critical. Using a wipe saturated in disinfectant and using unidirectional strokes to wipe off the materials prior to bringing them into the cleanroom drastically reduces the amount of particulate you're putting into your controlled environment. Lastly, having cleaning protocols that remove particulate from the cleanroom is also of utmost importance. According to USP , at minimum we should be cleaning our floors on a daily basis. Again, the floor, if our engineering controls are doing their job properly, is where a lot of these particles will reside. So cleaning the floors on a daily basis removes particles as well. We should also be wiping off and disinfecting our critical surfaces frequently. In particular your primary engineering control should be cleaned prior to every use and according to USP they should be cleaned and disinfected every 30 minutes during continuous use. I'm actually going to reference the 2019 revision of USP which is currently in an appeal process because of the BUD sections of the standard. However, this table should not be disregarded as it is a great guide for what surfaces should be cleaned and how often it should be performed. USP 2019 Revision Table 8 Keeping particulate down to a minimum is of utmost importance. WHY? Just to review, particles are what microorganisms travel on. So if you're able to control the number of particles, you're able to control the level of microorganisms. Review this with your operators on a regular basis, it should absolutely be a part of your regular training.

I am so excited to have you here and share with you today the secret to crushing your goals. Now I know we all have goals for our businesses and life but lets be honest sometimes you may find yourself struggling to reach them or staying focus. First off know you are not alone.For instance, this is exactly how I felt when writing this episode. I knew the message but could not for the life of me find the words. It was extra frustrating in the fact that I managed to carve out not one but two whole consecutive hours to dedicate to it that weren’t at the crack of dawn or the wee hours of the night! I freaking miracle, right?! And yet there I was at the end of those two hours nowhere near to being done. Seriously despite my best efforts I barely had an intro done and not even recorded! As I packed up my things, I was so frustrated I wanted to throw things. In my mind I was screaming, “I just wasted two hours of my life! Do you know what else I could have spent that time on?! Are you kidding me? Oh, my goodness.” However, despite my frustration I put a smile on my face as I entered the building to pick up my kids. It wasn’t their fault that I just wasted two hours of my life. I punched in the code and opened the door to my one-year old’s room and watch as her face just lit up as she saw it was me. And if that moment was perfect already, she proudly let go of the bench she was holding on to and walked confidently to me! This was a big deal!! She has been walking like a champ while holding onto our hands for the past few weeks but has taken very few steps solo. No more! Now she is a fearless walking little girl. I was just beaming with pride as I knelt there hugging her. And then it clicked. In that moment I knew exactly how to write this episode because it was all about moments just like this. You know those moments where everything just clicks into place, the fog has lifted and the view is clear. Yes, I am talking about the aha moments or clarity. For the last month I have watched my little girl tirelessly try to walk, no matter how many times she fell she would get back up and try again. She knew exactly what she wanted, perfect clarity. That is it! That is the secret to not getting stuck, being lost or unfocused. Its clarity because when you know EXACTLY what you want you take it to a new level. Now you can do this for anything in your life but let’s use your business as an example. In your business you need to get clear, crystal clear, like in a deep, real authentic way about:Why you’ve created this business?Who you really want to work with?What you want to do for them?Where you want to take them?How you are going to get them there?Why they should choose you?Now if you are like me and struggle with this a little imagine you are having this conversation with your toddler whose favorite question is WHY? Just keep asking yourself why and before you know it you will find the root of the answer to those questions. When you know the answers to these questions – in a deep way – not surface stuff, but really digging in and laying it down. getting yourself ridiculously clear on YOUR message and YOUR mission in the world.Then, only then can you truly begin creating the business of your dreams. Because there will be no question left in your mind. Now, you can  get to work before you get clarity, we do that all the time. But let me tell you..  being CLEAR moves things forward so much faster because you won’t be wasting your time doing stuff that isn’t perfectly aligned with where you want to go and How you want your business to contribute to the world.It’s like staying with that job that pays the bills and is alright but doesn’t make you excited to go to work every day!Honestly, feeling freaking amazing and over the moon inspired about your business is the biggest motivator. A MUCH bigger motivator than “making money”  So that’s why I think that for your business or ANY big goal that you want to accomplish you need to take the time to get CLARITY, to get clear about what it is that you really want, deep at the core of your being. Today we are talking about hip-hop, the rap. I don't know what else I'd call it.   Matthew: The rap.   Becky: The rap.   Matthew: I mean you are talking to the two white people in the room talking about hip-hop. That is what this episode is.   Becky: I know. Oh, this is going to go down horribly. Although I do love my 80s, rap and I love the old Run DMC stuff before Aerosmith. Who else is in there? I am trying to think. A tribe called Quest. Although I cannot remember if they were 80s or not. It all runs together now for me. Then, of course, Public Enemy. I don't think that was 80s. Maybe they were 80s. Oh, my God. Yeah. Oh, there is a lot in there. 3rd Bass. That is right; I pull out 3rd Base, which you will never know. But the one guy in 3rd Base, a white guy is now like a baseball historian at Cooperstown, if I remember correctly.   Matthew: That is a turn career.   Becky: Yeah, Pete Nice. Was it Pete Nice? Oh I don't think it was Pete Nice. I cannot remember who it was now.   Matthew: Was it was not Pete Townsend? Becky: No, now I am going to have to look it up. Who were the members of 3rd Base? Yeah, so that is where I am coming from.   Matthew: Interesting. Mine, you know. Like, that is all I really need to say. We actually had a very interesting discussion at the end of the last episode talking about where does R&B begin versus hip-hop specifically.   Becky: Yes.   Matthew: I approach hip-hop from the more R&B side. So I am thinking Beyoncé, Lemonade.   Becky: All right, okay.   Matthew: To an extent, Drake, although he is not my favourite.   Becky: Oh God!   Matthew: And then smaller artists, particularly from the HBO show Insecure, has some very good hip-hop…   Becky: See I don’t know that.   Matthew: References. TT the artists. What is the name of the song? Is featured in it. She is great. Now I will have to introduce you to it. Then, of course, where would we be? But two people, two white people talking about hip hop. Also, listen to the entirety of Hamilton and needed to get said. There it is. It has been said we can now glaze past it.   Becky: I only know the Alexander Hamilton [Making sound 00:2:56]. I don't know anything else.   Matthew: That is all you need to know. That is what the musical is.   Becky: Yeah, I. Oh, man. I think I was right with Pete Nice. What did I say? Oh, my God.   Matthew: You did say Pete Nice.   Becky: Yeah. There is MC Serch and Pete Nice, but I feel like. Yeah. Pete Nice. Baseball historian, I had it right the first time.   Matthew: Well, with a band name like 3rd Base, you kind of have to.   Becky: They had a song called The Cactus.   Matthew: Why?   Becky: I can't even remember. I just remember The Cactus. I am sure I still have that CD somewhere. But yeah, The Cactus.   Matthew: I love.   Becky: I cannot even remember. It is all gone. It is so bad; they did have a big hit. What is compelling women to devour and double-tap this worldview and fueling its presence in bible studies and book clubs across the country? In this episode, I chat with Anne Kennedy about the theology of Rachel Hollis. We examine Rachel’s theology (the nature of her religious and spiritual beliefs) based on her prolific writing and speaking. It’s important to note this is not a personal attack on Ms. Hollis, but rather an evaluation of her words and teachings against what we find in God’s word. For when someone offers answers to the foundational questions of life: why are we here? what is our purpose? how can we be happy? etc. you can be certain they are offering you a worldview, and you should carefully consider how they answer each of those questions. If we don’t or can’t hold these teachings up to the light, we’ll easily prey to a false gospel or a message that is less than the one God has given us in the Bible.

Do you follow people on social media whose bodies you are completely envious of? And does seeing their posts of their rocking body make you feel worse about your own? Before you praise someone for what their body looks like, first ask yourself: Are they taking a healthy approach to fitness? What do you really know about what is behind that “perfect” body? We shouldn’t celebrate or be envious or inspired by someone’s perfect-looking body until we know what is really behind their fitness success. Why? Just because someone looks great, doesn’t mean they are healthy. People will take extreme measures to get fit, disregarding their health altogether. They can be the fittest person in the world—with perfectly sculpted legs and six-pack abs, able to do 500 burpees in a row and squat an insane amount of weight—but that does not necessarily mean they are healthy. In this episode, I explain what it means to be fit, what it means to be healthy and how, in some cases, there can be a huge difference. Plus, I talk you through the long-term damage you can do to your body when you prioritize being fit over being healthy. The trope to start all great action movies. You thought he was out of the military/government agency/shadow ops squad/paramilitary assassination group/illuminati, but they’ve got to bring him in one last time, and now it's personal. Why does it work? 2 things. Quick character history. He’s retired? That means he was good enough at what he did to do it a long time. Then, carte blanche to kill any and everyone. It’s bad enough they had to drag him out of retirement, but then there’s that extra push. Dead dog, dead wife, kidnapped daughter. As soon as that happens, he could bomb a nursery and you would still be rooting for him.Speaking of nurseries, please, for the love of the god I don’t believe in, leave your kids there and don’t take them out to restaurants. You want to go out? Get a sitter. Can’t afford one? Then you can’t afford to go out either. Can’t find one in time? Tough luck. Pick up your phone and use Grubhub or Ubereats or what have you and eat in. Where people can’t hear your baby cry. Or, buy me an appetizer. Deal? You buy me an appetizer, and your kid gets one good cry. He does it again? Entree. Third time? Kid gets a punt in the head. Or you do. Build a positive healthy self-image by helping others without expecting anything in return and choose the people you hang out with carefully. Step 7: This one is an interesting one. Step 7 is to do something for someone else, without expecting any favor in return. Sales fellows we are building a healthy self-image so that we can become a better salesperson, a better husband and a better human being overall. Here are some quick examples of how we can do something for someone else. We can sign up as a volunteer at Salvation Army or Red Cross. We can donate blood. We can visit a sick person at their residence of the hospital. We can opt to babysit for a single parent, giving them a break to go out and do things they like to do. Become a big brother to an orphan. Visit the senior’s home and read them a book. If you know a graduate entering the job market, we can enhance their interview skills, or work on their resume making it more presentable. Just remember you do not want any compensation or the person or persons you are assisting should be in a position to return any favor back to you. Believe me, when you will give something without expecting anything in return, yes the recipient will feel amazing, that is true, however the feeling you will get is completely indescribable. Gratitude. You will realize that you how fortunate your are and you will not complain. You will be grateful for little things and you will feel that you have a lot to be thankful for in your life. Being selfless will make you feel good about yourself. You will feel that you are somebody, somebody who can make a contribution in other peoples lives. Now let’s move on to the step 8 out of 15 in building a healthy self-image. Step 8: Choose your associates carefully John Lee Dumas uses this quote over and over that “You are the average of five people you hang out with.” Hang out with a high moral character who look on the bright side of life in all aspects, how much will you benefit from it? A good example supposes you are a person who is always late and you start hanging out with people who are punctual and start everything on time. Guess what? Two three times that you notice, hey they do not wait for me, you will also become punctual and develop a positive habit. Hang out with someone who exercises regularly, chances are you will push yourself too and start exercising. In Asian culture, our parents always scan our friends closely. They say you are known by the company you keep. It is absolutely true. You acquire much of the thinking, manners, and characteristics of the people you are around. Trust me if all the other professionals in the world took sales training and had to sell for living for three months, our country will become even greater. Why? Just think about it. In the sales profession, what do you hear the most from trainers, coaches, managers, and associates? Yes, “You can do it”. They make you believe that YOU CAN DO IT. Be careful and pick out those people who are optimistic and enthusiastic about life and I guarantee you will see those traits in yourself over a period of time. In his book “Think and Grow Rich” Napoleon Hill has a full chapter on this subject. He calls it “The Mind Group” Napoleon Hill brings three key benefits of handing out with the crowd. 1. Infinite Intelligence 2. Accumulated Experience 3. Experiment and Research Quote By John C. Maxwell an American author who has written a number of books on leadership. Excellent content to consume. There are two kinds of learning: Experience, which is gained from your own mistakes, and Wisdom which is learned from the mistakes of others. Sales fellows when you feel good about yourself, everyone around you feels good. Which means you have a happy workplace, happy customers, happy managers, happy life at home! Christine Egan is a cancer survivor with an incredible story to tell. When she was first diagnosed with breast cancer, her jaw had hit the floor. She was a health coach! She drank green smoothies, meditated, and did yoga, so how could someone so health-conscious get cancer? Well, it made her re-evaluate her life and to seek pleasure and satisfaction in every moment. This mindset shift turned out to be one of the happiest times of her life. As she shared her story with women across the globe, Christine realized that there was so much more to being healthy than green smoothies.   Key Insights & Aha Moments: *Please share this podcast! *Who is Christine? She's a mom, wife to her high school sweetheart, and a cancer survivor. *It was such a shock to Christine when she found out she had breast cancer because she was the healthiest girl in the room. *Christine used a combination of both traditional and alternative medicine. *Christine's book, The Healthy Girl's Guide to Breast Cancer, was written two years ago. *What has happened to Christine and her book since then? *Christine did have some fears that she was going to be burned at the stake for her alternative choice in medicine, for not putting her children first. *What does Christine's business look like today? *The products you put on your body can be toxic to you. Christine found out after her diagnosis that the lotion she was using for the last 20 years had harmful ingredients in it. *A reader, who was also a makeup artist working for a cancer-related non-profit, felt sick that she was using these harmful makeup products on women fighting cancer. She asked Christine for her help to create healthy, non-toxic products. *Fast forward to today, Christine is about to launch her makeup product line, Life On Mars. *What's Christine putting on her skin nowadays? Coconut oil! *Despite being in crunch mode to release the Life On Mars website, Christine still tells her team to practice self-care and to close that laptop by 4pm. *What's the story behind the Life On Mars name? *What advice does Christine have for those struggling to get their business online? *It's important to not get sucked down a rabbit hole when you see other people succeeding. Stop! There's room enough for all of us. *What has Christine learned about business? Self-care has to come first. *Best piece of life advice? Have fun! Flip those stories in your head. *Look out for Christine's audio book! *Be sure to stay till the end where I share a story on how I recovered from a really tough injury.   Maven Moment: Christine's message about sharing your story and to not make it bigger than it is reminded me of a horrible accident that happened to me when I was ice skating. I fell so perfectly that I pretty much broke everything. I know, ouch, but looking back, that accident made me a better healer. Why? Just due to the nature of the injury, I knew I had to use both traditional and alternative medicine to help heal me. When I came back, the doctors said I had made a full recovery, which was rare for the type of injury I had sustained. The entire experienced soften me and made me appreciate the power of both forms of medicine. It also made me more compassionate towards what my patients were going through too. The reason why I bring this story up is because there's so much power to your story and what you tell yourself. You have a story like this as well. You had to overcome things in your past. Our souls need rest too; and without rest, we cannot find the resources we need to move life forward. However, finding rest for our souls doesn’t just happen. We need to intentionally set aside time to reflect and seek God’s transformational and restorative power. On August 30, 2015, Pastor Chad gives us three exercises to examine our souls, so we can find rest.

Happy New Year! 2012 had many ups and downs, and that counts in comedy podcasting, too. Some podcasts went off the rails — such as Affirmation Nation and The Gentlemens' Club — never to be heard from again. While at the same time, milestones were passed for others. What did WTF, Nerdist, and Never Not Funny have in common? They all dropped their 300th episode this year! Congrats! And You Made It Weird, Who Charted?, Walking the Room, The Long Shot, and Sklarbo Country all turned the corner on their episode 100 in 2012. And the granddaddy of the comedy podcast ilk, Tom Scharpling's The Best Show On WFMU, actually rang up its 500th episode! I came up with the idea of doing a Top 10 Comedy Podcasts of 2012 but, unfortunately, the concept hit me the day before I was going to record Epi45. I didn't have the time or even a methodology in mind to pull it off, so I did the next best thing. I borrowed a list from the AV Club. They had their panel of reviewers dole out 50 points a piece amongst the podcasts out there in the universe, did careful calculations and considerations before posting their list. I swear we'll do something like that…next year. In the meantime, here's how the AV Club's list shook out: 10. Tompkast5.   (tie) Sklarbro Country / The Best Show on WFMU4.   You Made It Weird3.   Savage Love2.   Comedy Bang Bang1.   WTF This list was meant to be from ALL realms of podcasts but, as you can see, the majority are comedy-based in nature. Three of them have never been clipped on Succotash before, so we make up for that in this episode, with the exception of Savage Love, hosted by Dan Savage out of Seattle, Washington. The reason being that it is not a comedy p'cast but, instead, a very good and somewhat edgy relationship and sexual advice show. (It's currently #1 on Stitcher's Lifestyle & Health Top 100 list.) POD MAFIA IRREGULARS Last episode was our Succotash Christmas Card episode to those podcasts in the Pod Mafia collective of internet shows. But there are also a group of shows that have been very supportive of not just our podcast but a number of the Pod Mafia shows as well, so I'm referring to them as the Pod Mafia Irregulars. And this episode we pay tribute to them by clipping some recent editions of The Brit & The American, The Clutch & Wiggle Experience, The Random Variety Show, Toggle The Switch and Bonn And Obo. CLIPS Pop Culture Happy HourNPR's entertainment and pop culture round-table podcast features Linda Holmes, Trey Graham, Stephen Thompson & Glen Weldon having spirited discussions of movies, books, television, nostalgia, and anything else that might be making them happy. NerdistCongrats to Chris Hardwick, Jonah Ray and Matt Myra! Their Nerdist podcast hit Epi300 this past week or so. And to celebrate the moment, they returned to whence they began, doing the podcast in the office of Thomas Lennon’s house. Lennon was their very first guest and (for my money) he is one of the funniest guests in podcasting, ranking up there with the likes of Paul F. Tompkins. The Best Show on WFMUThe Best Show on WFMU with Tom Scharpling is a combination music/call-in/comedy radio show originating from New Jersey-based radio station WFMU. The Best Show first aired on October 10, 2000 and, as mentioned up top, they've aired/dropped over 500 episodes! The Brit & The AmericanThe Brit and The American – namely Alba Lewis and Tom Moore – have translocated to Peru for the next 6 months but continue to pump out the podcasts on a regular basis. We feature a taste from a recent episode, which I think is PRE-Peru. The Clutch & Wiggle ExperienceYour hosts Rob and Craig (along with K-Love) take you on a verbal whirlwind as they spin, annihilate and humiliate everything and anything in the world today. Straight from the basement on a real air hockey table, they bring comedy and "justice" the way no one else sees the world. (Their CWERadio.com also rebroadcasts episodes of Succotash each week!  Random Variety Show PodcastCharley Miller is a good friend of Succotash and we’d be remiss if we didn’t include him in this belated Christmas card to the Pod Mafia Irregulars. In this clip Groucho Marx, who handles most of the heavy lifting when it comes to the interviews on  RVSP, talks to Libby Marie and Lishia from the Two Funny Girls podcast. Toggle the SwitchCross-Pacific podcast Toggle The Switch features Susan Wygmans from America and Carl McAteer from Australia. Susan started out as part of the Cuddlah Army, a total fan of Walking The Room with Greg Behrendt and Dave Anthony, and now she finds herself with listeners of her own. We have a snippet of their show talking about “humpy time” and, right after that, we’ve got a little holiday message from the pair that was delayed due to a snag in our Tweetsack. BonnAndOboHere’s a show that’s definitely Pod Mafia worthy, not to mention they are requesting some guest vocal input from our very own Bill Haywatt. I’ve sent out a request to Bill’s favorite watering holes and, if we can get him upright long enough to record those lines, we’ll get them to you. In this clip, they bring in a very special guest. Epic Rant RadioAs part of Epic Rant Radio with Andre and Eric, they have a feature called “Rounds”, where the two guys go at it over certain subjects. In this clip, it’s the merits of watching old Sex And The City versus Glee. King MeanyA few episodes ago, I talked about a new sketch group out of England called King Meany. Liam Beirne, Andy Goddard, Shem Jeffs, Natalie Feehan, and Lizzie Milton were getting ready to start podcasting and now…they have! IllusionoidOne of my personal favorite podcasts is out of Canada. Illusionoid, with Paul Bates, Lee Smart, and Nug Nahrgang. This was their holiday offering, The Christmas Ninjas, and we play the whole thing. In the coming year, I'm hoping to have more interesting interviews with podcasters, comedians and interesting people from the world of entertainment. I also hope to keep up a regular release schedule, which should be welcome news for those of you you enjoy a helping of Succotash you can depend on.
