They were believed to pull the chariot of Teššub, the Hurrian weather god. Šeri additionally could function as a deity mediating between petitioners and his master, but no individual role was ever assigned to Ḫurri. In addition to appearing in Hurrian offering lists and theophoric names, for example from Nuzi, Šeri and Ḫurri are also attested in Hittite and Mesopotamian sources. He has also been the Cognition Core Leader for the Dominantly Inherited Alzheimer Network (DIAN) studies for 11 years, and is Principal Investigator of ARC-DS, a multi-site study to develop a mobile data collection platform in Down syndrome-associated Alzheimer's disease. Sára Erzsebet Zsadányi is a PhD Student at the Sant Pau Memory Unit – Neuroimaging Core and Universitat Autonoma de Barcelona. Originally from New Zealand, Sára studied psychology and linguistics, but always felt really drawn to neuroscience and especially to Alzheimer's disease. Though highly variable in terms of age at symptom onset (AAO), hundreds of variants in PSEN1 cause autosomal dominant forms of AD (ADAD) with nearly complete penetrance. PSEN1 forms the catalytic core of the {gamma}-secretase complex and thereby directly mediates the production of longer, aggregation-prone A{beta} peptides relative to shorter, non-aggregating peptides. We hypothesized that the broad AAO and biomarker heterogeneity seen across ADAD would be predictable based on mutation-specific differences in the production of A{beta} species. Methods: A{beta}-37, 38, 40, 42, and 43 production was quantified from 162 unique PSEN1 variants expressed in HEK293 cells. Prediction of AAO was carried out in 107 variants with available AAO and then replicated in 55 variants represented across 190 PSEN1 mutation carriers who have detailed cognitive and biomarker data from the Dominantly Inherited Alzheimer's Network (DIAN). Results: Variations in A{beta} production across the 162 mutations examined in cell-based models were highly predictive of AAO. In those with corresponding in vivo data from the DIAN study, our cell-based {gamma}-secretase composite was strongly associated with biomarker and cognitive trajectories. Conclusions: These findings elucidate the critical link between {gamma}-secretase function, A{beta} production, and AD progression and offer mechanistic support for the amyloid hypothesis. The approach used here represents a powerful tool to account for heterogeneity in disease progression in ADAD clinical trials and to assess the pathogenicity of variants of unknown significance or with limited family history. Abstract: Multiple neurodegenerative disorders, including Parkinson,s disease (PD) and Alzheimer,s disease-associated dementia (ADAD), are linked with dopaminergic (DA) neuron death and a resulting reduction in dopamine levels in the brain. DA neuron degeneration and the risk of developing PD is connected to genetic mutations affiliated with lysosomal function and protein degradation. Accessible human cellular models for PD-relevant genetic mutations are needed to investigate mechanisms of DA cell death and define points of therapeutic intervention. Human induced pluripotent stem cell (iPSC)-derived midbrain DA neurons offer a developmentally and physiologically relevant in vitro model for investigating PD pathogenic mechanisms across genetic backgrounds. In this study, we generated DA neurons using iPSCs from two clinically diagnosed PD patients, one harboring an inherited GBAN370S mutation and the other a mutation in LRRK2G2019S and compared pathophysiology against DA neurons from genetically engineered SNCAA53T iPSCs and its isogenic apparently healthy normal (AHN) iPSCs. Our results present a novel phenotype for GBAN370S and LRRK2G2019S derived DA neurons, showing that they produced and released significantly more dopamine compared to the AHN and SNCAA53T mutant DA neurons. All mutant DA neurons developed deficient glucocerebrosidase (GCase) activity, increased mitochondrial stress, aberrant neuronal activity patterns, and increased -synuclein accumulation. Together these data suggest potentially divergent origins of PD pathogenesis in GBAN370S and LRRK2G2019S DA neurons. In addition, compound screening confirmed that GCase modulators can rescue enzyme activity and impact neural activity across all DA mutant neurons, to varying degrees. These data demonstrate unique in vitro phenotypes associated with PD and suggest a diversity of underlying mechanisms across different genetic backgrounds. This episode is an introduction to the epic Gilgamesh and in fact a prelude to a future edition where I tell you the story in one episode. The Epic of Gilgamesh was rediscovered in the Library of Ashurbanipal in 1849. After being translated in the early 1870s, it caused widespread controversy due to similarities between portions of it and the Hebrew Bible. Gilgamesh remained mostly obscure until the mid-20th century, but, since the late 20th century, he has become an increasingly prominent figure in modern culture. When the gods created Gilgamesh they gave him a perfect body. Shamash the glorious sun endowed him with beauty, Adad the god of the storm endowed him with courage, the great gods made his beauty perfect, surpassing all others, terrifying like a great wild bull. Today on the show we have three filmmakers taking on this topic straight on with the production of their short film "The Hard R."
