Matteus 6,33-34

Matthiesen, who was forced to hide his love for artist Russell Cheney during a time before homosexual love and marriage were accepted. The sister of Jimmy's first serious boyfriend shows up at a book signing for Jimmy's new novel and hands him a letter that sends him spinning back to memories of the first man he ever loved. James describes his sexual awakening and recalls haunting moments with Gregory, whose self-destructive personality was part of Jimmy's impetus for writing American Scholar. Horrigan's novel, which weaves in the study of Queer Theory, Jimmy's sexual awakening, and fears of the AIDS virus then sweeping across the globe. Horrigan whips back and forth from that difficult time to 2016, when his now middle-aged protagonist is now a professor and published author, but political polarization following the presidential election has inspired new fears throughout the gay community. Born and raised in Reading, Pennsylvania, Patrick E. Horrigan received his BA from The Catholic University of America and his PhD from Columbia University. He is the author of the novel Pennsylvania Station (Lethe Press; Indie Book Award finalist for best LGBTQ2 fiction) and the novel Portraits at an Exhibition (Lethe Press; winner of the Dana Award for fiction as well as the Mary Lynn Kotz Art-in-Literature Award, sponsored by the Library of Virginia and the Virginia Museum of Fine Arts). His other works include the memoir Widescreen Dreams: Growing up Gay at the Movies (University of Wisconsin Press), the play Messages for Gary: A Drama in Voicemail, and (with Eduardo Leanez) the solo show “You Are Confused”! He has written artists' catalogue essays for Thion's LIMI-TATE: DRAWINGS OF LIFE AND DREAMS (cueB Gallery, London) and Ernesto Pujol's LOSS OF FAITH (Galeria Ramis Barquet, New York). His essay “The Inner Life of Ordinary People” appears in Anthony Enns' and Christopher R. Smit's “Screening Disability: Essays on Cinema and Disability” (University Press of America). Horrigan and Eduardo Leanez are the hosts of “Actors with Accents”, a recurring variety show in Manhattan. Winner of Long Island University's David Newton Award for Excellence in Teaching, he taught literature for twenty-five years at LIU Brooklyn. Today, I dive into the complex and often nebulous world of data observability with a leading expert in the field, Taggart Matthiesen, the Chief Product Officer at LogicMonitor. With an impressive career trajectory that includes pivotal roles at Lyft, Twitter, and Salesforce, Taggart offers an insider's view on the challenges and opportunities of harnessing data for actionable business intelligence. The discussion opens with Taggart sharing his unique perspective on how data has evolved to become the heartbeat of every business. In a world saturated with information, how do enterprises sift through the 'noise' to identify that elusive 1% of data which can drive decision-making and business outcomes? Taggart elucidates on the growing potential in the data observability space, shedding light on why this has become indispensable for businesses of all scales. As AI continues to infiltrate every aspect of our lives, questions around data responsibility become increasingly pertinent. We explore the readiness—or the lack thereof—of using AI to contextualize data. They discuss the ethical and practical implications, offering a balanced view on the technological advancements and the cautionary tales that serve as important guideposts for AI adoption in data observability. The episode then shifts to explore LogicMonitor's AI ops tool, DEXTA, as a case study. Taggart recounts how this tool provided immediate value by pinpointing the cause of a network outage, thereby showcasing the utility of intelligent data observability solutions. What are the design principles that inform such solutions? And what safeguards does LogicMonitor put in place to ensure data integrity and security? If you are a business leader, a product manager in SaaS, or someone simply interested in the frontier of data science and AI. In that case, this episode offers a holistic view on where data observability is headed, why it matters, and how to leverage it responsibly for business success. Prepare for an insightful journey through the world of data, AI, and business innovation.

Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.06.29.547007v1?rss=1 Authors: Anders, J., König, C., Lender, C., Hellhund, A., Nehls, S., Shalabi, I., Honecker, B., Lorenzen, S., Meyer, M., Matthiesen, J., Cadar, D., Roeder, T., Metwally, N. G., Lotter, H., Bruchhaus, I. Abstract: Recently, two genes involved in pathogenicity in a mouse model of amoebic liver abscess were identified based on their differential expression between non-pathogenic (A1np) and pathogenic (B2p) clones of the Entamoeba histolytica isolate HM:1-IMSS. While overexpression of a gene encoding the metallopeptidase EhMP8-2 decreases the virulence of the pathogenic clone B2p, overexpression of the gene ehi_127670 (ehhp127), encoding a hypothetical protein, increases the virulence of the non-pathogenic clone A1np, while silencing this gene in B2p decreases virulence. To understand the role of both molecules in determining the pathogenicity of E. histolytica, silencing and overexpression transfectants were characterized in detail. Silencing of ehmp8-2, of the homologous gene ehmp8-1, or of both together in A1np trophozoites significantly altered the transcript levels of 60-350 genes. This strong change in the expression profile caused by the silencing of ehmp8-1 and/or ehmp8-2 implies that these peptidases regulate expression of numerous genes. Consequently, numerous phenotypic characteristics including cytopathic, hemolytic and cysteine peptidase activity were changed in response to their silencing. Silencing of ehhp127 in B2p trophozoites did not affect other genes, whereas overexpression in A1np trophozoites results in an altered expression of approximately 140 genes. EhHP127 appears to be important for trophozoite movement, as silencing negatively affects and overexpression positively affects trophozoite motility. Interestingly, the specific silencing of ehhp127 also impairs cytopathic activity, cysteine peptidase and hemolytic activity. All three molecules of interest, namely EhMP8-1, EhMP8-2, and EhHP127 can be detected in amoeba vesicles. Our results clearly show that the proteins studied here influence the pathogenicity of amoebae in different ways and use entirely different mechanisms to do so. Author summary: The human pathogen Entamoeba histolytica can live asymptomatically in the intestine or become invasive and cause fatal liver abscesses. Approximately 15,000 people die each year as a result of an amoebic infection. Recently, two clones with different pathogenicity (A1np: non-pathogenic; B2p: pathogenic) derived from the E. histolytica isolate HM:1-IMSS were compared at the transcriptome level. Two highly differentially expressed genes (ehhp127 encoding a hypothetical protein and ehmp82 encoding a metallopeptidase) were identified. Analysis of E. histolytica transfectants showed that silencing of ehhp127 and overexpression of ehmp8-2 in B2p trophozoites reduced amoebic liver abscess formation in the mouse model. In this study, we characterized E. histolytica silencing and overexpression transfectants of ehmp8-2, as well as of the homologous gene ehmp8-1 and of ehhp127. It was shown that the altered expression of the metallopeptidase genes has a strong influence on the expression of a large number of genes and that the phenotype is strongly altered as a result. Silencing of ehhp127 does not affect the overall expression profile. However, specific silencing has a negative effect on motility, cysteine peptidase, hemolytic and cytopathic activity. All three molecules were shown to be localized in trophozoite vesicles. Copy rights belong to original authors. Visit the link for more info Podcast created by Paper Player, LLC

Emma hosts Sara Matthiesen, Professor of History at George Washington University, to discuss her recent book Reproduction Reconceived: Family Making and the Limits of Choice After Roe V. Wade, and how reproductive rights and the concept of choice have been manipulated and under attack in the age of Roe. Professor Matthiesen and Emma begin by exploring the large-scale questions of the book, from how Roe v. Wade completely changed the conversation around bodily autonomy to one on the idea of choice, to how the treatment of family plays into this. Sara Matthiesen then gets into this idea of “choice,” how this euphemism for abortion came to be, and the importance of recognizing the preexisting conditions that are necessary to choose to start a family, as she contextualizes the Roe decision at the beginning of the neoliberal era of austerity, seeing Nixon's veto of universal childcare actually as a signal of the conservative fear of more women in the workforce and the downfall of the nuclear family, and an announcement of the state's withdrawal from bolstering the project of the family writ large. Next, she and Emma tie in the role of the sexual revolution and red scares to the conservative preservation of “family values” (straight, cis, white, middle class) and the complete destruction of the welfare state, especially for poorer Black and brown mothers, discussing what additional labors are forced onto people in order to reproduce and maintain their family. They then dive into the role mass incarceration played in this project against non-perfectly-nuclear families, particularly in the neglect incarcerated individuals face at the hands of the state, from healthcare neglect resulting in countless severe illnesses and injuries alongside incredibly poor pregnancy care, to the active work against allowing communication between incarcerated individuals and their families, especially when it comes to court-ordered hearings. They wrap up the interview by touching on the role of queer parenting in this conversation, walking through the state's lack of recognition of queer parenting as valid, and its insistence on the existence of a “male” and “female” provider as perhaps its only real involvement in the family. Emma also dives into Putin's statement that he was really trying to HELP Ukrainians this whole time, as the Pentagon ramps up their weapon support, before she moves to Eric Adams continued scapegoating of the homeless in the face of a terror attack by someone who routinely raged at houseless (and other minorities) on social media, all while the funding for the police spikes and countless supposedly “affordable” housing apartments are kept empty. And in the Fun Half: Emma takes a call with Ren from Oregon as they cover the rapid deterioration of trans rights in the US, with states like Alabama and Mississippi moving forward with bills for the forcible detransition of teenagers, and the complete lack of urgency from liberals on this matter, then, the Fox & Friends hosts reveal that the violent and insane people responsible for yesterday's terror attack in Brooklyn are the homeless people that angered the shooter. Stephen from Pittsburg gives some updates on PA unionization, DeSantis continues to live out his dreams of government in the 80s, and the ACA somehow makes a return to town halls. Hannah from Southern California gives an open and honest discussion about the need for African American and Queer studies in American high schools, Greg Kelly tells us not to worry because “slavery was just a phase, mom!” Alex from NY calls in for some more discussion on parents' relationship to schooling, plus, your calls and IMs!   Check out Sara's book here: https://www.ucpress.edu/book/9780520298217/reproduction-reconceived   Purchase tickets for the live show in Boston on May 15th HERE:   https://majorityreportradio.com/live-show-schedule   Become a member at JoinTheMajorityReport.com: https://fans.fm/majority/join Subscribe to the AMQuickie newsletter here:  https://madmimi.com/signups/170390/join Join the Majority Report Discord! http://majoritydiscord.com/ Get all your MR merch at our store: https://shop.majorityreportradio.com/ Check out today's sponsors: MySolarNerd.com: There are a lot of homeowners that aren't aware of the solar options currently available. We tend to associate Roe v. Wade, the landmark Supreme Court decision that decriminalized abortion in 1973, with the choice not to have children. But Roe was equally transformative for Americans' understanding of family--having and raising children also came to be thought of as a choice. In Reproduction Reconceived: Family Making and the Limits of Choice after Roe v. Wade (University of California Press, 2021), Sara Matthiesen highlights the distance between this idea of choice and worsening forms of inequality that have forced far too many to work harder simply to create and maintain a family. In this new and timely work, Matthiesen shows how the effects of incarceration, for-profit and racist healthcare, disease, and poverty have been worsened by state neglect. At its core, Reproduction Reconceived is an urgent historical account: of the myriad labors that families have been made to perform simply to survive, and of the inevitable costs that pile up when family making is seen as a private responsibility rather than a public good.
