Los hombres no tenían pues acceso a los cuentos, a las leyendas y a los mitos. Los grandes reinos hacían todos los esfuerzos para poder contar con sus historias y cuentos, pero cada que una historia aparecía era almacenada por el dios Nyame y no había nada que pudieran ofrecer al dios para que les abriera el acceso a ellos. Al mismo tiempo había un dios araña que se llamaba Anansi que era muy cercano a los hombres y que preocupado por la falta de historias decide ofrecer un precio por las historias. Anansi decide contarle a su familia la difícil tarea que tiene por delante y su esposa Aso decide ayudarlo para que pueda llevar a cabo cada uno de sus retos. Primero fue la piton Onini: Para ello Anansi simulo que tenía una discusión con su esposa acerca del tamaño de Onini, diciendo Aso alegaba que Onini era más pequeño que una rama de bambú y Anansi decía que no era así. La serpiente escucha a Anasi y decide ayudar a Anansi en la discusión con su esposa. De esta manera le pregunta a Anansi como puede demostrarse. Anansi le dice que la mejor manera es que se enrolle alrededor de una rama que el había preparado y así probar su enorme longitud, La serpiente Onini gustosa se acerca a la rama y se enrolla. Obviamente era más grande que una rama de bambu, pero en ese momento Anansi se acerca a la rama y usando unas cuerdas preparadas previamente ata la serpiente Onini, y tomando la vara de bambu con la Piton amarrada a ella, se la lleva triunfante al dios Nyame. MAKING PRAYER WORK!This episode is a follow up to Episode 59 where I talked about prayer and the fact that it works! I like the quiet moments of conversation, of listening to the inner whispers, of acknowledging the communication centers including my gut for insight, knowingness, and pure unadulterated intelligence that flows to me. I am sharing my native, traditional prayer format — the libation..The most common or frequently used ritual of prayer in the Akan tradition is the pouring of libation. This is done on different occasions, including naming ceremonies, rites of passage (e.g., puberty, birthdays, weddings, graduations), funerals and mourning rites, and traditional festivals. The libation or prayer acknowledges Nana Nyame—God, Twereduampong—The Omnipotent God, Eno Asaase Yaa—Mother Earth, and the ancestors. It asserts faith and expresses gratitude for what has been accomplished by Nyame's grace. The prayers also make several declarations of wealth earned or to be earned for the clan; they affirm good health, protection from evil, and blessings for the young children born to expand the lineage. The closing remarks ask for more blessings for the specific occasion and for all future endeavors. A PrayerNana Nyame, Twereduampong, Bo Adee—wo nsa nie. God, Good Mother, My Great Ancestors, Benevolent Spirits and Good Messengers, Drink, drink, drink!"The prayer is representative of the format of libations, where all the Akan concepts about the Supreme Being, Mother Earth, Spirits, Deities, Ancestors, Directions, are addressed and thanked before any supplication. The prayer ends by acknowledging the presence of All, and always with an offering of a drink, usually clear liquid, like water or gin. The idea is to create a prayer in your own words, using a format that resonates with you, and express the desire you have and commit to having a conversation with the God in You! Busiswa, DJ Lag, Karen Nyame KG and Scratchclart talk about the impact of the pandemic on amapiano and gqom, what it was like getting back to the clubs when things opened up again, and the global rise in popularity of these genres. Busiswa is a singer and poet from South Africa. Her music career began when she featured on Sir Bubzin's track Syaphambana in 2011, and she's since gone on to feature on DJ Zinhle's global hit My Name Is, as well as Beyonce's My Power as part of The Lion King: The Gift soundtrack. She's released three studio albums, and her most recent single Lagos came out late last year. DJ Lag is a prolific DJ and record producer who is widely recognized as the king of gqom music, a genre of electronic dance music from Durban in South Africa. His bass-heavy, minimalist sound has pushed him into the upper ranks of the global electronic music scene, and last year he released his debut full-length album, Meeting with the King. Karen Nyame KG is a producer, DJ and broadcaster who has been one of dance music's true innovators of the last decade. She's been dubbed the Goddess of Rhythm, and is known for her energetic, polyrhythmic sound that is lighting up the clubs of London and beyond. Scratchclart is an electronic musician, DJ and record producer who is part of the fabric of grime music and UK funky. He's the head of the highly respected DVA Music label, and in recent years he's been building bridges between music scenes in the UK and South Africa, in particular with his DRMTRK series and his new EP Beyond Gqom & Grime.

Abstract: Mutations in the lysosomal membrane protein CLN3 cause Juvenile Neuronal Ceroid Lipofuscinosis (JNCL). Activation of the lysosomal ion channel TRPML1 has previously been shown to be beneficial in several neurodegenerative disease models. Here, we tested whether TRPML1 activation rescues disease-associated phenotypes in CLN3-deficient retinal pigment epithelial (ARPE-19 CLN3-KO) cells. ARPE-19 CLN3-KO cells accumulate LAMP1 positive organelles and show lysosomal storage of mitochondrial ATPase subunit C (SubC), globotriaosylceramide (Gb3), and glycerophosphodiesters (GPDs), whereas lysosomal bis(monoacylglycero)phosphate (BMP/LBPA) lipid levels were significantly decreased. Activation of TRPML1 reduced lysosomal storage of Gb3 and SubC but failed to restore BMP levels in CLN3-KO cells. TRPML1-mediated decrease of storage was TFEB-independent, and we identified TRPML1-mediated enhanced lysosomal exocytosis as a likely mechanism for clearing storage including GPDs. Astrid Nyame and Adrea Propp have lived parallel life experiences as Saskatchewan track and field athletes for many years. They excelled in high school and had successful university experiences. They transitioned to high-performance athlete training centres away from their home province. Though they both had great experiences, ultimately, they decided to return to Saskatchewan. They have created SaskWatch Athletics, which began as a social platform and recently expanded to a podcast, to give a moment in the sun to these prairie athletes that don't always get their due. Abstract: Lysosomes degrade macromolecules and recycle their nutrient content to support cell function and survival over a broad range of metabolic conditions. Yet, the machineries involved in lysosomal recycling of many essential nutrients remain to be discovered, with a notable example being choline, an essential metabolite liberated in large quantities within the lysosome via the degradation of choline-containing lipids. To identify critical lysosomal choline transport pathways, we engineered metabolic dependency on lysosome-derived choline in pancreatic cancer cells. We then exploited this dependency to perform an endolysosome-focused CRISPR-Cas9 negative selection screen for genes mediating lysosomal choline recycling. Our screen identified the orphan lysosomal transmembrane protein SPNS1, whose loss leads to neurodegeneration-like disease in animal models, as critical for cell survival under free choline limitation. We find that SPNS1 loss leads to massive accumulation of lysophosphatidylcholine (LPC) and lysophosphatidylethanolamine (LPE) within the lysosome. Mechanistically, we revealed that SPNS1 is required for the efflux of LPC species from the lysosome to enable their reesterification into choline-containing phospholipids in the cytosol. Using cell-based lipid uptake assays, we determine that SPNS1 functions as a proton gradient-dependent transporter of LPC. Collectively, our work defines a novel lysosomal phospholipid salvage pathway that is required for cell survival under conditions of choline limitation, and more broadly, provides a robust platform to deorphan lysosomal gene functions. It represents a central trafficking hub that sorts proteins and lipids to various destinations or for secretion from the cell. The Golgi has emerged as a docking platform for cellular signalling pathways including LRRK2 kinase whose deregulation leads to Parkinson disease. Golgi dysfunction is associated with a broad spectrum of diseases including cancer, neurodegeneration, and cardiovascular diseases. To allow the study of the Golgi at high resolution, we report a rapid immunoprecipitation technique (Golgi-IP) to isolate intact Golgi mini-stacks for subsequent analysis of their content. By fusing the Golgi resident protein TMEM115 to three tandem HA epitopes (GolgiTAG), we purified the Golgi using Golgi-IP with minimal contamination from other compartments. We then established an analysis pipeline using liquid chromatography coupled with mass spectrometry to characterize the human Golgi proteome, metabolome and lipidome. Subcellular proteomics confirmed known Golgi proteins and identified novel ones. Metabolite profiling established the first known human Golgi metabolome and revealed the selective enrichment of uridine-diphosphate (UDP) sugars and their derivatives, which is consistent with their roles in protein and lipid glycosylation. Furthermore, targeted metabolomics validated SLC35A2 as the subcellular transporter for UDP-hexose. Finally, lipidomics analysis showed that phospholipids including phosphatidylcholine, phosphatidylinositol and phosphatidylserine are the most abundant Golgi lipids and that glycosphingolipids are enriched in this compartment. Altogether, our work establishes a comprehensive molecular map of the human Golgi and provides a powerful method to study the Golgi with high precision in health and disease states. There's a real need to improve road safety in Ghana and Sub-Saharan Africa as a whole, says Eric Nyame-Baafi, a road safety consultant for the Bloomberg Initiative for Global Road Safety in Ghana. “A total of 650 people die on African roads every day. In this week's episode of Lunch & Learn with Dr. Berry, we bring you a special guest, Dr. Theodore Nyame, who is a board-certified plastic surgeon from Harvard Medical School. He also completed a research fellowship also at Harvard Medical School. Dr. Nyame comes to us from Ghana, West Africa. After coming to the realization that the conditions of Ghana leave many of the citizens there in need of reconstructive procedures, he selected cosmetic surgery as his medical specialty. During his undergraduate studies, Dr. Nyame attended Cornell University where he graduated top of his class before continuing on to Harvard Medical School. After receiving his Doctorate of Medicine, Dr. Nyame kicked off what has developed into an international tour of medical missions and speeches. He uses these opportunities to enhance the quality of information circulated through the industry. Today, Dr. Nyame will share his experiences from when he was a child until he became successful. From a very young age, Dr. Theo Nyame showed a level of desire and dedication towards providing positive outcomes for himself and those around him. The secret to Dr. Nyame's unconditional passion for results-oriented service to his patients is not a secret at all. “For me, walking out of a treatment knowing that I delivered the best work possible for my patients is the most important,” he explains.
