We're about to go into a place that many people have never contemplated. Thank goodness, because we're going to go behind bars and into what really creates prisoners. Let's start with what's the definition of a prisoner? What's the definition of a jail? What if not all jails have bars? Rocky is a 35 year old successful Indian businessman born in New York City and living in Los Angeles. Rocky now owns and runs a showroom in a high traffic area of downtown near LA Live. Rocky also recently upgraded to Beverly Hills. Rocky is a felon as well as a Philosophy Graduate (both) from the state of Alabama. He spent “high school” life in 9 different facilities, schools, and boot camps across the world, some of which are now closed due to severe child abuse, rape, and torture. Yes, Rocky almost died at least 7 times inside the facilities that were supposed to keep him safe. Half of Rocky's face and jaw are fake from a murder attempt. He's been shot at, stabbed, kidnapped, and kicked in the face. Rocky has had 20 major surgeries. Rocky's “been hurt, hurt others, and seen hurt up close.” Rocky's also been loved, loved others, and seen love up close. Rocky now has amazing friends and family in almost every continent in the world. His business allows him to travel freely and meet amazing souls across our planet. Rocky speaks 4 languages and can hold a conversation with any man, woman, or child in the world. Rocky Singh Kandola is a WWASP survivor....twice. If you don't know anything about the World Wide Association of Specialty Programs (WWASP), it was a horrendous way to try and modify behavior of young adolescent boys and girls. Torture and emotional/physical/sexual abuse were endured day after day usually for months at a time. Children were abruptly removed from their beds in the middle of the night, as if it were a kid napping, and taken far away from their home to one of the WWASP camps. If enduring two tours of WWASP wasn't bad enough for this young man, he also had a destructive past with drug and alcohol.Rocky graciously takes us into his past experiences before discussing his amazing career as an entrepreneur and owning his own hair extension business called Hair Maiden India.

Abstract: The combination of phase separation and disorder-to-order transitions can give rise to ordered, semi-crystalline fibrillar assemblies that underlie prion phenomena namely, the non-Mendelian transfer of information across cells. Recently, a method known as Distributed Amphifluoric Foerster Resonance Energy Transfer (DAmFRET) was developed to study the convolution of phase separation and disorder-to-order transitions in live cells. In this assay, a protein of interest is expressed to a broad range of concentrations and the acquisition of local density and order, measured by changes in FRET, is used to map phase transitions for different proteins. The high-throughput nature of this assay affords the promise of uncovering sequence-to-phase behavior relationships in live cells. Here, we report the development of a supervised method to obtain automated and accurate classifications of phase transitions quantified using the DAmFRET assay. Systems that we classify as undergoing two-state discontinuous transitions are consistent with prion-like behaviors, although the converse is not always true. We uncover well-established and surprising new sequence features that contribute to two-state phase behavior of prion-like domains. Additionally, our method enables quantitative, comparative assessments of sequence-specific driving forces for phase transitions in live cells. Finally, we demonstrate that a modest augmentation of DAmFRET measurements, specifically time-dependent protein expression profiles, can allow one to apply classical nucleation theory to extract sequence-specific lower bounds on the probability of nucleating ordered assemblies. Taken together, our approaches lead to a useful analysis pipeline that enables the extraction of mechanistic inferences regarding phase transitions in live cells. Rocky Singh Kandola, is an 31-year-old successful Indian businessman born in New York City and living in Los Angeles. He owns and run a showroom in a high traffic area of downtown near LA Live. He is a felon as well as a Philosophy Graduate (both) from the state of Alabama. He spent his “high school” life in 9 different facilities and boot camps across the world, some of which are now closed due to severe child abuse, rape, and torture. He has almost died at least eight times. Half of his face and jaw are fake from an attempted murder on his life. He's been shot at, stabbed, kidnapped, and kicked in the face. He had over 20 major surgeries in his adult life. He's has been hurt, hurt, and seen hurt up close. He has also loved, have seen love, and been in love. He has amazing friends and family in almost every continent in the world. His business allows him to travel freely and meet these fantastic souls across our planet. He can speak four languages and hold a conversation with any man, woman, or child in the word. The "troubled teen industry" is a mostly unregulated collection of for-profit programs that claim to rehabilitate out-of-control youth. Between 50,000 and 100,000 adolescents currently spend at least part of the year in these facilities. The hopes are that their enrollment – or confinement, depending on the arrangement or their perspective – can prevent these relatively privileged kids from joining the 48,000 youth caught in the U.S. juvenile justice system. Some are boot camps or ranches or wilderness therapy programs. Some are military-style academies that focus on sparse surroundings, lots of physical activities and punishments. There are also a variety of residential treatment programs that aim to change deviant behaviors like poor school performance, self-harming, drug abuse, insubordination and more. Welcome to the very first episode of the Mother’s Quest podcast! I am so honored to bring you this conversation with Navjit Kandola. Navjit is a spiritual adviser, educator, speaker, filmmaker, full-time mom, and the director of the Nizhoni School for Global Consciousness. Navjit works with clients to heal subconscious wounds, release anxiety and strain, identify habits that spiral into negative behaviors, and unblock stagnant energy in relationships and projects. It felt right to have my first conversation on the podcast with Navjit because she was one of the first people I reached out to after I decided to launch Mother’s Quest. I found her through Danielle LaPorte’s website. Danielle, another profoundly spiritual mother, who is also an entrepreneur, author and poet, is a long-time friend of Navjit’s and refers to Navjit as a masterful soul sister who is lucid as a diamond. I determined I needed some of that guidance and reached out to Navjit for a phone session. I proceeded to tell her about my desire to embark on my own hero’s journey and all my hopes and dreams for Mother’s Quest. She helped me think about the metaphor I was choosing and encouraged me to make it my own...to hold a vision for a journey that could include more connection and ease than the typical one found in epic stories. Those words stayed with me and have been a guide as I have gone through the ups and downs of birthing my new venture and laying a foundation for the Mother’s Quest community. More recently, I reached out to Navjit again, distraught with fear and sadness in the wake of the election and consumed by self-doubt in the days leading up to this podcast launch. I got to experience firsthand how Navjit’s healing presence could help me tap into the wisdom of my inner child, a term she refers to in this interview. It was a powerful and transformative experience and one that I wrote about the next day on a blog post that I will include in the show notes. I share all this because this first conversation has a different tone than many of the episodes that follow. It is a deep dive into spirituality and consciousness. And, if these are topics you don’t typically gravitate to, I encourage you to stay with it. Navjit talks about being grateful for those magical moments when we’re present and connected with ourselves, our children, and with nature.
