Scientists discover a new method for building DNA-damaging molecules with anticancer potential

Scientists at Dublin City University (DCU, Ireland), in collaboration with research teams in Chimie ParisTech – PSL (France), Chalmers University of Technology, Sahlgrenska University Hospital, Gothenburg (Sweden), and the University of Limerick (Ireland), have developed a new chemical strategy for designing metal-based compounds capable of damaging cancer cell DNA, offering a potential new direction for future anticancer drug development. Led by Professor Andrew Kellett, this European consortium has created a series of molecules that cut DNA through a distinct chemical mechanism compared with existing chemotherapy drugs. The research focuses on early-stage compounds that could form the basis of future therapies, particularly in cancers that become resistant to treatment. The scientists used click chemistry—a fast and reliable method for assembling molecular components—to create a family of compounds known as Tri-Click ligands. When combined with copper ions, these ligands form artificial metallo-nucleases, metal-containing agents designed to cleave DNA. Drug resistance remains one of the biggest challenges in cancer treatment. Tumours can adapt by repairing specific forms of DNA damage or by blocking the activity of conventional drugs. A key advantage of this new chemical strategy is that the compounds damage DNA via pathways that differ from those targeted by many current cancer treatments. This means they may avoid some of the typical mechanisms that cancers use to become resistant. Professor Andrew Kellett, Professor of Inorganic and Medicinal Chemistry at DCU, said: "Click chemistry has transformed how we build complex molecules, but its potential as a platform to assemble DNA-damaging chemotherapeutics is underexplored. One of the major challenges in cancer treatment is drug resistance. By developing compounds that damage DNA in a different way, we aim to open up new possibilities for overcoming some of the limitations of existing therapies. While this research is still at an early stage, it provides a valuable platform for future drug development." Professor Gilles Gasser, Professor of Bioinorganic Chemistry at Chimie ParisTech, said: "This work is clear evidence of the potential of metal-based compounds for anticancer research, going beyond the currently heavily used platinum-based drugs. While still in its infancy, this study is a first step towards new solutions for cancer treatment. On a completely different note, this work is another demonstration of the power of collaboration between European scientists and institutions." Professor Damien Thompson, Director of SSPC, the Research Ireland Centre for Pharmaceuticals & Professor of Molecular Modelling at UL, said: "This work exemplifies the value of systematic, deep screening of molecular properties in the development of more effective medicines. Support from SSPC, the Research Ireland Centre for Pharmaceuticals, enabled strong collaboration between our experimental and modelling teams, and this new design strategy marks a key milestone in developing sustainable, well-tolerated anticancer drugs." Professor Fredrik Westerlund, Professor of Molecular Bioscience at Chalmers University of Technology, said: "This study truly highlights how combined expertise across Europe can lead to innovative results in a research field of paramount importance. Novel treatments to resistant cancer tumours are highly desired in the clinics, and the metal compounds developed in this study have many of the properties that are eagerly sought for." Introduction — Are GLP-1 Drugs Forever?Have you heard experts say that GLP-1 medications like Ozempic, Wegovy, or Zepbound are forever drugs — because once you stop, you regain all the weight?I'm not fully convinced that's true.I've found three promising approaches being developed that may help people stop medication without immediate weight regain.Each approach is completely different:One focuses on the stomachOne on the brainOne on structured lifestyle interventionI'm Dr. Gary Pepper. I've practiced endocrinology for over 30 years. This podcast is unsponsored, and I receive no financial compensation from the treatments discussed. This content is educational only and not a substitute for medical care from your physician.Chapter 1 — Resetting the Gut (DMR)The first approach is called Duodenal Mucosal Resurfacing (DMR).The theory is that while you're on GLP-1 medication, the part of the stomach that normally produces appetite-regulating hormones becomes dormant. After long enough, it may struggle to restart when medication stops.DMR removes the top layer of the stomach lining using a heated endoscope. This forces regeneration of fresh cells that produce appetite-controlling hormones.In a short three-month study, patients who underwent the procedure did not regain weight after stopping medication — some continued to lose.This sounds dramatic, but the technique is already FDA-approved for other stomach conditions. It is not yet approved for weight management, but it shows where research is heading.Chapter 2 — Retraining the Brain (rTMS)The second approach targets the brain instead of the gut.It's called Transcranial Magnetic Stimulation (rTMS) — already used for depression treatment.Magnetic coils stimulate appetite control centers in the brain. After just five weeks of treatment, participants lost as much weight over a year as those taking semaglutide for a full year.This is early research, but it suggests appetite control may eventually be trained neurologically rather than chemically.Chapter 3 — Exercise as Hormone RehabilitationThe third approach is the most practical and immediately available: structured exercise.Researchers at the University of Copenhagen studied people stopping GLP-1 medication after a year of use.Those who followed a structured program of resistance and cardio training regained almost no weight — averaging about five pounds over a year.Those returning to sedentary habits regained most of their weight.Exercise appeared to restore the body's natural GLP-1-like hormone production.This is not about willpower.It's biological rehabilitation.Closing — A Changing LandscapeThese three approaches show that weight regain after GLP-1 is not inevitable.The science is evolving quickly.I'll continue tracking new developments and sharing updates here.Thanks for listeninGary Pepper, M.D., an associate professor at a prominent medical school and endocrinologist with 40 years of experience, brings clarity to numerous topics within the field of metabolic health. At the turn of the twentieth century, education was undergoing rapid changes. Teachers sought creative ways to engage students in subjects that were often taught by rote memorization. Geography, in particular, was considered a cornerstone of a well-rounded education, yet it was sometimes reduced to reciting capitals and drawing borders. Heffron's work introduced a tactile and visual method known as “chalk modeling,” in which teachers could draw raised relief maps directly on the blackboard to show mountains, rivers, and valleys in a more dynamic way.
