Mein heutiger Gast ist jemand, von dem viele Leute denken, sie würden ihn richtig gut kennen. Dabei ist er seit rund 30 Jahren vor allem als Kunstfigur in der Öffentlichkeit unterwegs und versteckt seine wahre Identität ziemlich gut. Der Mann, den wir als Atze Schröder kennen, steht für den Proll aus dem Ruhrgebiet mit Lockenmähne, getönter Sonnenbrille und großer Klappe. Ich erinnere mich noch, wie meine Schwester und ich als Schulkinder über seine Serie „Alles Atze“ auf RTL gekichert haben, obwohl wir wahrscheinlich nur jeden zweiten Witz überhaupt verstanden haben. Atze Schröder ist nicht nur im Fernsehen zu sehen, sondern füllt mit seinen Bühnenprogrammen riesige Hallen. Er hat sieben Mal den Deutschen Comedypreis gewonnen und inzwischen auch das Podcasten für sich entdeckt. Vielleicht kennt ihr ja „Betreutes Fühlen“, wo er immer dienstags mit Leon Windscheid über Psychologie spricht oder „Zärtliche Cousinen“, was eher so ein typischer Comedy-Laber-Podcast ist. Das Interessante in Atzes Fall ist – und das ist ja eh meine These übers Podcasten – dieses Medium lässt nicht so richtig zu, dass man da dauerhaft eine Kunstfigur aufrechterhält, weil man eben jede Woche so lange mit jemandem über Alltägliches redet. Und so hat sich in den vergangenen drei, vier Jahren auch Atze ein bisschen von anderen Seiten gezeigt und offenbart, wer der Mensch hinter der Rolle ist. Wusstet ihr zum Beispiel, dass er in seiner Freizeit super viel liest und Erstausgaben von Romanen sammelt? Dass er gerne mal die Pyrenäen durchwandern und auf einer einsamen Insel leben würde? Oder dass er vor seiner Comedy Karriere als Kaufmann in den Niederlanden gearbeitet hat? Abstract: Detection of repeating patterns within continuous sound streams is crucial for efficient auditory perception. Previous studies demonstrated a remarkable sensitivity of the human auditory system to periodic repetitions in randomly generated sounds. Automatic repetition detection was reflected in different EEG markers, including sustained activity, neural synchronisation, and event-related responses to pattern occurrences. The current study investigated how listeners attention and the temporal regularity of a sound modulate repetition perception, and how this influence is reflected in different EEG markers that were previously suggested to subserve dissociable functions. We reanalysed data of a previous study in which listeners were presented with random acoustic sequences with and without repetitions of a certain sound segment. Repeating patterns occurred either regularly or with a temporal jitter within the sequences, and participants' attention was directed either towards or away from the auditory stimulation. Across both regular and jittered sequences during both attention and in-attention, pattern repetitions led to increased sustained activity throughout the sequence, evoked a characteristic positivity-negativity complex in the event-related potential, and enhanced inter-trial phase coherence of low-frequency oscillatory activity time-locked to repeating pattern onsets. While regularity only had a minor (if any) influence, attention significantly strengthened pattern repetition perception, which was consistently reflected in all three EEG markers. These findings suggest that the detection of pattern repetitions within continuous sounds relies on a flexible mechanism that is robust against in-attention and temporal irregularity, both of which typically occur in naturalistic listening situations. Yet, attention to the auditory input can enhance processing of repeating patterns and improve repetition detection. Copy rights belong to original authors. Abstract: The development of novel drugs for Alzheimer's disease has proven difficult, with a high failure rate in clinical trials. Typically, transgenic mice displaying amyloid-{beta} peptide brain pathology are used to develop therapeutic options and to test their efficacy in preclinical studies. However, the properties of A{beta} in such mice have not been systematically compared to A{beta} from the patient brains. Here, we determined the structures of nine ex vivo A{beta} fibrils from six different mouse models by cryo-EM. We found novel A{beta} fibril structures in the APP/PS1, ARTE10, and tg-SwDI models, whereas the human familial type II fibril fold was found in the ARTE10, tg-APPSwe, and APP23 models. The tg-APPArcSwe mice showed an A{beta} fibril whose structure resembles the human sporadic type I fibril. These structural elucidations are key to the selection of adequate mouse models for the development of novel plaque-targeting therapeutics and PET imaging tracers. Abstract: The superficial layers of the superior colliculus (SC) are highly visual and receive direct input from the retina. Nonetheless, neural activity in the superficial SC (sSC) is modulated by locomotion and pupil-linked arousal. Here we show that visual responses of neurons in the sSC are additionally modulated by reward delivered prior to the visual stimulus. We trained mice to perform a visual detection task and recorded the activity of neurons in the SC using two-photon calcium imaging and electrophysiological recordings using high-density silicone probes (Neuropixels). Neurons across all layers of the SC responded to various task events, including reward delivery. However, responses to events like licking or movements did not explain the visual response modulation by reward. Electrophysiological recordings showed that most of the reward modulation occurred in the superficial rather than the deeper layers of the SC. Neurons also exhibited modulation by pupil-linked arousal, which was independent of the reward modulation. Performance of a population decoder to detect visual stimuli improved significantly by reward modulation but not by pupil-linked arousal modulation. Our results indicate that behavioural factors other than locomotion and arousal modulate visual activity in the SC. Copy rights belong to original authors. Abstract: Alzheimer's disease (AD) is a multifactorial disorder driven by abnormal amyloid {beta}-peptide (A{beta}) levels. To identify new druggable pathways involved in the A{beta} cascade we here investigated the AD pathophysiological role of the presenilin-like intramembrane protease signal peptide peptidase-like 2b (SPPL2b). A{beta}42 induced a biphasic modulation of SPPL2b expression in human cell lines and ex vivo mouse brain slices. In addition, SPPL2b was elevated in AppNL-G-F knock-in AD mice as well as in human AD samples. Early high neuronal expression of SPPL2b was followed by a downregulation in late AD pathology in both AppNL-G-F mice and Braak stage V AD brains. Importantly, SPPL2b overexpression or its genetic deletion significantly increased or reduced APP cleavage and A{beta} production, respectively. Thus, our results strongly support the involvement of SPPL2b in AD pathology. The early A{beta}-induced SPPL2b upregulation may enhance A{beta} production in a vicious cycle further aggravating the A{beta} pathology suggesting SPPL2b as a potential anti-A{beta} drug target. Abstract: For colour vision, retinal circuits must separate information about intensity and wavelength. This requires circuit level comparison of at least two spectrally distinct photoreceptors. However, many vertebrates use four or more, and in those cases the nature and implementation of this computation remains poorly understood. Here, we establish the complete circuit architecture and function of outer retinal circuits underlying colour processing in the tetrachromatic larval zebrafish. Our findings reveal that the specific spectral tunings of the four cone types near optimally rotate the encoding of natural daylight in a principal component analysis (PCA)-like manner to yield one primary achromatic axis, two colour opponent axes as well as a secondary UV-achromatic axis for prey capture. We note that fruit flies - the only other tetrachromat species where comparable circuit-level information is available - use essentially the same strategy to extract spectral information from their relatively blue-shifted terrestrial visual world. Together, our results suggest that rotating colour space into achromatic and chromatic axes at the eye's first synapse may be a fundamental principle of colour vision when using more than two spectrally well separated photoreceptor types. Abstract: Chronic imaging of neuronal networks in vitro has provided fundamental insights into mechanisms underlying neuronal function. Existing labeling and optical imaging methods, however, cannot be used for continuous and long-term recordings of the dynamics and evolution of neuronal networks, as fluorescence indicators can cause phototoxicity. Here, we introduce a versatile platform for label-free, comprehensive and detailed electrophysiological live-cell imaging of various neurogenic cells and tissues over extended times. We report on a novel dual-mode high-density microelectrode array, which can simultaneously record in i) full-frame mode with 19,584 recording sites and ii) high-signal-to-noise mode with 246 channels. We set out to demonstrate the capabilities of this platform with recordings from primary and iPSC-derived neuronal cultures and tissue preparations over several weeks, providing detailed morpho-electrical phenotypic parameters at subcellular, cellular and network level.
