Abstract: Alzheimer's disease (AD) is a multifactorial disorder driven by abnormal amyloid {beta}-peptide (A{beta}) levels. To identify new druggable pathways involved in the A{beta} cascade we here investigated the AD pathophysiological role of the presenilin-like intramembrane protease signal peptide peptidase-like 2b (SPPL2b). A{beta}42 induced a biphasic modulation of SPPL2b expression in human cell lines and ex vivo mouse brain slices. In addition, SPPL2b was elevated in AppNL-G-F knock-in AD mice as well as in human AD samples. Early high neuronal expression of SPPL2b was followed by a downregulation in late AD pathology in both AppNL-G-F mice and Braak stage V AD brains. Importantly, SPPL2b overexpression or its genetic deletion significantly increased or reduced APP cleavage and A{beta} production, respectively. Thus, our results strongly support the involvement of SPPL2b in AD pathology. The early A{beta}-induced SPPL2b upregulation may enhance A{beta} production in a vicious cycle further aggravating the A{beta} pathology suggesting SPPL2b as a potential anti-A{beta} drug target. Report reçoit deux marins, qui, en 2023, disputeront ensemble The Ocean Race à bord de l'Imoca Biotherm, dont la mise à l'eau est prévue mercredi 31 août en fin de journée à Lorient : son skipper, Paul Meilhat, et Anthony Marchand, qui fera partie de l'équipage dévoilé la semaine dernière. Avant de parler de cette actualité, les deux marins, qui ont fait leurs armes sur le circuit Figaro Beneteau, évoquent la 53e édition en cours, Anthony Marchand estimant que Tom Laperche, le plus rapide de la flotte, mériterait de l'emporter au vu de sa régularité sur le support depuis maintenant deux ans. Celui qui a participé à dix reprises à la Solitaire (deux podiums) confie qu'il reviendra un jour, mais seulement s'il peut disputer tout le programme d'avant-saison, indispensable selon lui pour performer. Paul Meilhat raconte ensuite comment il se sent à la veille de mettre à l'eau son Imoca Biotherm, une première construction pour le vainqueur de la Route du Rhum qui, pour des questions de timing et de coût mais aussi de design, a choisi un sistership de l'actuel LinkedOut de Thomas Ruyant. En version “upgradée”, cependant, avec “une étrave de dernière génération, des foils, des systèmes de safrans et une casquette différents.” Débuté en décembre, le chantier a duré jusqu'en août chez Persico, en Italie, où le skipper et sa petite équipe (qui compte notamment Baptiste Chardon et Marc Liardet) ont passé tout l'été, d'où une certaine fatigue, mais “beaucoup d'enthousiasme au moment de mettre à l'eau.” Les deux marins donnent par ailleurs leur avis sur les nouveaux 60 pieds sortis de chantier cet été, entre “faux bateaux neufs”, selon l'expression de Paul Meilhat, car construits dans les moules d'Imoca existants, et bateaux vraiment différents, comme Malizia et Charal qui ont fait l'objet de “beaucoup de développement.” Le skipper de Biotherm confie ensuite pourquoi il a absolument voulu participer à The Ocean Race, un désir qui remonte à ses années Figaro, et, selon lui, “la meilleure manière de préparer le Vendée Globe.” L'occasion pour Anthony Marchand de raconter les souvenirs de sa participation avec l'équipage espagnol de Mapfre en 2014-2015, “une expérience hyper enrichissante qui m'avait permis de progresser.” Paul Meilhat explique ensuite son choix d'équipage pour The Ocean Race, entre nécessité d'avoir des marins aguerris sur ce type de support (Anthony Marchand, Damien Seguin) et désir de s'ouvrir à d'autres expériences - “Je ne veux pas que ce soit une balade autour du monde entre Bretons” - d'où la présence de la Portugaise Mariana Lobato, de l'Italien Giulio Bertelli - rencontré chez Persico - et d'Amélie Grassi. Il finit par détailler son planning d'ici le départ de The Ocean Race - tests de jauge, premières navigations, Défi Azimut, Route du Rhum, retour vers Alicante, chantier. Deuxième invitée d'Un été de navigatrices, une certaine Marie Riou, avec un épisode d'Into The Wind (le numéro 22) enregistré chez elle en presqu'île de Crozon, à l'automne 2020. Depuis, celle qui fut, entre autres, vainqueure inoubliable de la Volvo Ocean Race 2017-2018 à bord de Dongfend Race Team, a poursuivi sa route, goûtant à l'Imoca à bord de Corum pour The Ocean Race Europe en 2021, au Class40 avec Amélie Grassi sur La Boulangère Bio lors de la dernière Transat Jacques Vabre. Et même, en juin dernier, à un titre de champion du monde militaire avec Jean-Baptiste Bernaz en J80 alors qu'elle attendait un heureux évènement... Un épisode à retrouver sur notre site et sur toutes les plateformes d'écoute. __ Les femmes qui ont gagné quatre titres de champions du monde en voile légère tout en comptant une victoire dans la Volvo Ocean Race ne sont pas légion sur la planète voile. La Française Marie Riou, 39 ans, appartient à ce cercle très fermé. La native de Plougastel, au bord de la rade de Brest, tombée dans la voile toute petite dans le sillage d'un père et de deux frères passionnés, a connu un début de carrière fulgurant, devenant championne du monde et d'Europe de 420 en 1999 (avec Anne-Claire Le Berre). L'enchaînement s'avère ensuite plus difficile mais après quelques années de recul, la voilà de retour par le match-racing où les titres s'accumulent et l'emmènent aux JO de Londres en 2012 en compagnie de Claire Leroy, qui règne alors sur la discipline. Puis c'est la rencontre - les retrouvailles à - avec Billy Besson et l'enchaînement incroyable de quatre titres de champions du monde de Nacra 17 en 4 ans - ainsi qu'un titre de marins de l'année en 2015 - qui font d'eux les grandissimes favoris des jeux de Rio 2016. Après la blessure de Billy et l'échec de Rio, la voilà qui rebondit et embarque en 2017 sur Dongfeng pour une inoubliable campagne qui voit le projet franco-chinois remporter la Volvo Ocean Race. Et Marie Riou d'être désignée Rolex Sailor of The Year en 2018. Puis c'est l'aventure SailGP - seule femme de la flotte sur ces catas à foils qui se baladent à 50 noeuds -, du Diam24, une nouvelle campagne olympique avec Benjamin Schartz vers Paris 2024... Au fil de deux heures de discussion, on découvre la trajectoire d'une femme directe et authentique, naviguant au gré des rencontres. Tranquillement, discrètement, Marie Riou se bâtit brique par brique un palmarès incroyable sur tous les supports. Un parcours éclectique doré sur tranche, sans doute loin d'être fini. Both music and the psyche work through similar mechanisms such as repetition, imitation, variation (transformation), intimacy and the work of mourning, of the negative and of nostalgia. Silence and absence, as well as time and temporality, are key elements both in music and in symbolization and subjectivation processes. Viewing music as a primal and inherent characteristic of psychic structure and functioning allows both a better understanding of somato-psychic development and a musically informed psychoanalytic listening. Ludovica Grassi is a child neuro-psychiatrist, a full member of the Italian Society of Psychoanalysis, a qualified expert in child and adolescent psychoanalysis, and a psychoanalyst of couples and families. She is currently Treasurer of the Italian Psychoanalytic Society. Her main scholarly interests are infant psychoanalysis, ethno-psychoanalysis, and the role of music in psychic development. She has written on these topics as well as on individual, couple and family psychoanalysis. Her book The Sound of the Unconscious: Psychoanalysis as Music was published by Routledge in June 2021.
