Abstract: The updating of contextual memories is essential for survival in a changing environment. Accumulating data indicate that the dorsal CA1 area (dCA1) contributes to this process. However, the cellular and molecular mechanisms of contextual fear memory updating remain poorly understood. Postsynaptic density protein 95 (PSD-95) regulates the structure and function of glutamatergic synapses. Here, using dCA1-targeted genetic manipulations in vivo, combined with ex vivo 3D electron microscopy and electrophysiology, we identify a novel, synaptic mechanism that is induced during attenuation of contextual fear memories and involves phosphorylation of PSD-95 at Serine 73 in dCA1. Our data provide the proof that PSD-95-dependent synaptic plasticity in dCA1 is required for updating of contextual fear memory. Copy rights belong to original authors. Including rob a bank, hold eight hostages for 14 hours, and get his friend killed in the process. Liz needed gender confirmation surgery, and like today, it was expensive. John saw it as a way to save her life and so, in the name of love, he attempted to rob the Chase bank in the Gravesend district of Brooklyn. Later, a movie, Dog Day Afternoon starring Al Paccino would go on to fund Liz's much needed surgeries and John would claim himself a hero, a real Robin Hood. Book Vs Movie: “Dog Day Afternoon” The Classic Hollywood 1974 film Vs “The Boys in the Bank” from 1972 article from Life Magazine On August 22, 1972, John Wojtowicz and two accomplices entered a bank In Brooklyn at 450 Avenue P (in Gravesend) and started a robbery. One left early, (20-year-old Bobby Westeberg) while Wojtowicz and 18-year-old Sal Naturale proceeded to steal almost $200,000 in cash and traveler's checks before being surrounded by the police. What happened for the next 14 hours was a standoff and media circus for the ages. Turns out Wojtowicz said he needed the money for a “sex change” operation (as it was known at the time) for his partner Ernest Aron. The police brought Aron to the scene straight from the hospital where they had attempted suicide the week before. Another of Wojtowicz lover was brought out and he kissed him on the mouth in front of the news camera which hundreds of locals who made up the crowd. He also threw money into the air and ordered pizza for the bank hostages held inside. Eventually, the robbers and many of the bank hostages were taken to JFK airport where Naturale was shot in the chest and killed immediately. Wojtowicz was arrested and eventually spent a few years in jail for his crimes. He married again while in prison and had a tough time holding down a job because of his past. In the end, he was reduced to trying to make some money by posing in front of the bank for autographs. He died of cancer in 2006 at the age of 60. Aron eventually had gender reassignment surgery and died from the complications of AIDS in 1987. She took the name, Elizabeth Eden. Writers P.F. Kluge and Thomas Moore wrote about the crime and hostage situation on September 22, 1972, for Life magazine and the title of the article was “The Boys in the Bank” that was bought by Warner Brothers pictures and developed into the film Dog Day Afternoon. The 1975 film directed by Sidney Lumet and screenplay by Frank Pierson changes a few of the major details. The lead crook is “Sonny Wortzik” played by Al Pacino and his main cohort is Sal Naturile played by 40-year-old acting legend John Cazale. Filmed in Winsdor Terrace, Brooklyn (near Margo D's home!) the movie would become a huge moneymaker and nominated for all of the major categories at the Academy Awards in 1976 (and losing most to One Flew Over the Cuckoos Nest which was the movie to beat that season.) The film is a modern classic and entered in 2009 to the Library of Congress and in the National Film Registry. You observe a phenomenon, and come up with an explanation for it. That's true for scientists, but also for literally every person. (Why won't my car start? I bet it's out of gas.) But there are literally an infinite number of possible explanations for every phenomenon we observe. How do we invent ones we think are promising, and then decide between them once invented? Simon DeDeo (in collaboration with Zachary Wojtowicz) has proposed a way to connect explanatory values (“simplicity,” “fitting the data,” etc) to specific mathematical expressions in Bayesian reasoning. Abstract: The ability to update and extinguish fearful memories is essential for survival. 30 Accumulating data indicate that the dorsal CA1 area (dCA1) contributes to this process. 31 However, the cellular and molecular basis of fear memory updating remains poorly 32 understood. Postsynaptic density protein 95 (PSD-95) regulates the structure and function of 33 glutamatergic synapses. Here, we investigated the role of dCA1 PSD-95-driven synaptic 34 plasticity in contextual fear extinction. Using dCA1-targeted genetic manipulations in vivo 35 combined with PSD-95 immunostaining and 3D electron microscopy ex vivo, we demonstrate 36 that phosphorylation of PSD-95 at serine 73 PSD-95(S73) is necessary for contextual fear 37 extinction-induced expression of PSD-95 and remodeling of glutamatergic synapses. 38 Surprisingly, PSD-95 phosphorylation is not necessary for fear memory formation or early 39 extinction but is required for updating a partly extinguished fear memory, affecting its 40 persistence. Using a chemogenetic manipulation, we confirm that updating of the partly 41 extinguished fear requires PSD-95 expression and dCA1 activity during a prior extinction 42 session. Overall, our data indicate that dCA1 synapses are remodeled upon the extinction of 43 contextual fear memories; this process relies on PSD-95(S73) phosphorylation and enables 44 future updating of a partly extinguished contextual fear memory. Abstract: Structural neural changes following concussion are often not captured by standard imaging techniques. However, there is growing evidence that damage to white matter tracts and change in functional network connectivity may be observed following concussive injury. We investigated behavioural performance on a cognitive-motor integration (CMI) task in conjunction with alterations in resting state functional connectivity (rs-FC) in brain networks in a population of 30 female varsity athletes, with 16 having a previous history of concussion. Behavioural performance on accuracy, timing, and trajectory measures of a CMI task were assessed between the concussion history group and the control group. Rs-FC within the nodes of the Default Mode Network (DMN), Dorsal Attention Network (DAN), the Frontoparietal Network, and the Anterior Cerebellar Lobule Network, was assessed against performance scores on accuracy, timing, and trajectory measures. Main findings indicate no difference in behavioural performance between those with concussion history and those without, in contrast to previous findings in a group of primarily male varsity athletes. In addition, no difference in rs-FC was noted in correlation with behavioural performance scores on either accuracy, timing, or trajectory. These findings may suggest sex-related differences in performance on a CMI task, and a resiliency in both functional network connectivity and visuomotor skilled performance in varsity female athletes. The working title for this one was: “Three Gays Rob a Bank”. And we have an idea for the sitcom version. In August 1972, John Wojtowicz attempted to rob a Brooklyn bank to finance his lover's sex-reassignment surgery. The act resulted in a fourteen-hour hostage situation that was broadcast on TV. Three years later Al Pacino portrayed his crime in DOG DAY AFTERNOON. Directors Allison Berg and Frank Keraudren began filming THE DOG in 2002 and continued for ten years. Drawing upon extraordinary archival footage, the film shuffles between the 1970s and the 2000s. We gain a historic perspective on New York's gay liberation movement, in which Wojtowicz played an active role. In later footage, he remains a subversive force, backed by the unconditional love of his mother Terry, whose wit and charm infuse the film.
