Today's podcast: A majority of Canada's premiers calling on the Justin Trudeau federal government to forego the scheduled for April 1 increase in the federal carbon tax. Trudeau insists premiers are lying about the tax when they insist it will only add to the financial burdens Canadian families are experiencing. The parliamentary budget officer told us for a second or third time on air on February 25 the carbon tax Canadian middle class families will pay will not be made revenue neutral by federal government rebates. The tax appears integral to the rapidly diminishing national support for the Trudeau government among voters. Guest: Saskatchewan premier Scott Moe. The premier assured us on air as long ago as January that Saskatchewan will neither collect nor remit the carbon tax to the federal government. Premier Moe on Wednesday appeared before the parliamentary Government Operations and Estimates committee. Professor Sylvain Charlebois (Dalhousie university Agri Foods lab director) posted that according to Statscan the average Canadian is spending less than $250.00 monthly on groceries, a 19% decrease since 2021. According to Dalhousie's Canada Food Price Report the average person should spend $339 per month on healthy food. Born in London to Greek Cypriot parents Soulla Christodoulou spent much of her childhood living carefree days full of family, school and friends. She was the first in her family to go to university and studied BA Hotel & Catering Management at Portsmouth University. Years later, after having a family of her own she studied again at Middlesex University and has a PGCE in Business Studies and an MA in Education. Soulla is a Women's Fiction author and wrote her first novel Broken Pieces of Tomorrow over a few months while working full time in secondary education and is a mother of three boys. She has since published a historical fiction novel set in 1950s Cyprus and England, The Summer Will Come, and a short story, Unlocked which was inspired by her travels to India in February 2018. The Summer Will Come has been earmarked as a Book to Movie project and currently the Greek translation is being pitched to Greek publishers as well as a new edition of the book being released with Kingsley Publishers. She is a compassionate and empathetic supporter of young people. Her passion for teaching continues through private tuition of English Language and she also supports writers with mentoring and book editing and has recently set up a social media campaign promotion service for new authors. Abstract: Background: Mutations in genes encoding nuclear pore proteins (NUPs) cause steroid-resistant nephrotic syndrome (SRNS) and focal and segmental glomerulosclerosis (FSGS). The mechanisms of how NUP deficiency may cause podocyte dysfunction and failure of the kidney filtration barrier are elusive. The tightly controlled activity of the transcriptional effectors of the evolutionarily conserved Hippo pathway YAP and TAZ is essential for podocyte homeostasis. Here we analyze the role of NUPs in controlling YAP/TAZ nuclear import and activity in podocytes. Methods: We used quantitative label-free mass spectrometry to characterize the YAP/TAZ interactomes in podocytes, particularly identifying NUP interactions. Moreover, we specifically studied NUP205 in controlling YAP/TAZ nuclear import and YAP/TAZ-dependent target gene expression. Results: Here we identify the disease-causing nuclear pore proteins NUP107, NUP133, NUP205, and XPO5 as components of YAP and TAZ protein complexes in podocytes. We demonstrate that NUP205 is essential for YAP/TAZ nuclear import. The nuclear interaction of YAP/TAZ with TEAD1 and their transcriptional activity were dependent on NUP205 expression. Furthermore, we identify a feedback regulatory mechanism that controls YAP activity depending on TAZ-mediated NUP205 expression. Conclusion: This study links the disease protein NUP205 with the activity of the transcriptional regulators and Hippo effectors YAP and TAZ and suggests a pathogenic role of YAP/TAZ-deregulation in podocytes in patients with NUP205 mutations. Zachy shares how she sought Yoga in 2013 after suffering a horrific knee injury during a competitive volleyball match. Although she helped bring her team to the championship, it came at a cost; a great one however, which would allow her to find her passion and purpose. Abstract: Tauopathies such as Alzheimers disease are characterized by the aggregation and increased phosphorylation of the microtubule-associated protein tau. The pathological changes in tau are closely linked to neurodegeneration, making tau a prime candidate for intervention. However, the multiple facets of tau function and the lack of cellular tauopathy models that could support mechanism-based drug development hampers progress. Here we report the development of a live-cell imaging approach to quantitatively monitor pathological changes of human tau as it interacts with axonal microtubules. We show that a full-length aggregation-prone tau construct exhibits reduced interaction with microtubules as it increasingly aggregates. Through chemoinformatic analyses, we identified 2-phenyloxazole (PHOX) derivatives as putative polypharmacological small molecules that inhibit tau aggregation and modulate tau phosphorylation. We found that PHOX15 restores the physiological microtubule interaction of aggregation-prone tau in neurons and inhibits the first phase of tau aggregation in vitro. Furthermore, we report that PHOX15 inhibits the tau kinases GSK3beta and Cdk5, alters the kinome activity of model neurons, and reduces tau phosphorylation at disease-relevant sites. Molecular dynamics simulations highlight cryptic channel-like pockets crossing tau protofilaments and indicate that the binding of PHOX15 in one of the channels reduces the protofilaments ability to adopt a PHF-like conformation. The data show that our imaging approach provides a useful tool for identifying compounds that modulate tau-microtubule interaction in axons. We demonstrate that a polypharmacological approach to simultaneously treat tau aggregation and tau phosphorylation is able to restore physiological microtubule regulation, identifying PHOX15 as a promising drug candidate to counteract tau-induced neurodegeneration. Abstract: NMR measurements of cross-correlated nuclear spin relaxation provide powerful probes of polypeptide dynamics and rotational diffusion, free from contributions due to chemical exchange or interactions with external spins. Here, we report on the development of a sensitivity-optimized pulse sequence for the measurement of cross-correlated relaxation in methyl spin systems by analysis of the differential relaxation of transitions within the 13C multiplet. We describe the application of optimal design theory to implement a real-time 'on-the-fly' adaptive sampling scheme that maximizes the accuracy of the measured rate constants. The increase in sensitivity obtained using this approach enables, for the first time, quantitative measurements of rotational diffusion within folded states of translationally-arrested ribosome-nascent chain complexes of the FLN5 filamin domain, and can be used to place strong limits on interactions between the domain and the ribosome surface. Our fourth EUPL book club meeting spotlighted travel. The book club read three EUPL winning novels: Death drives an Audi (Døden kører Audi) by Kristian Bang Foss (Denmark, 2013), The son (Sin) by Andrej Nikolaidis (Montenegro, 2011), and The day the river froze (Τη μέρα που πάγωσε ο ποταμός) by Stavros Christodoulou (Cyprus, 2020). This event was organised in partnership with Lit Cities - an exciting digital initiative compiling a database of novels that unveil the magic of urban tales and equip the reader with a feeling for a city‘s inhabitants, their habits and idiosyncrasies. Lit Cities is the cultural hub for the world of literature and the poetic dimension of travelling. For our discussion on the theme, we were joined by Stavros Christodoulou, who spoke about his EUPL winning novel, upcoming new book and his writing career, as well as the cities and regions his works are based in. Four years of hard work and dedicated training saw Yiannis qualify to represent GB successfully ending a bronze medal. Success breeds success and once he got the taste for it Yiannis wanted more. After several national Aquathon championships and a Gold at the 2019 European Aquathon Championships, Yiannis is tiring his hand at Triathlon. Born in London to Greek Cypriot parents Soulla Christodoulou spent much of her childhood living carefree days full of family, school and friends. This PEP Talk episode features Dr. Joanna Christodoulou, who shares her research findings on the brain basis of reading. Dr. Christodoulou is the Director of the Brain, Education, and Mind (BEAM) Lab of the MGH Institute of Health Professions, where she integrates roles as developmental cognitive neuroscientist, clinician, and Associate Professor. She talks about her program of research, conducted at MIT in the Gabrieli Lab and at MGH Institute of Health Professions, which focuses on brain and behavior correlates of development, difficulties, and intervention effects for reading. Some of the topics she covers include the neural routes of reading, differences in neural activity associated with varying language systems, and instructional strategies that have demonstrated increased activation in specific brain regions. Dr. Christodoulou's award-winning research has been supported by organizations including the Spencer Foundation; the Fulbright Foundation; the Mind, Brain, and Behavior Initiative at Harvard; and the National Institutes of Health.

Emma, Tom, Judith and Sharne bring us a meaty 57 minutes in which they review a fairly controversial publication: Daisy Christodoulou's 'Seven Myths about Education' - a book that ruffled a fair few feathers on its publication in 2014, propelled its author to instant fame as a darling of the traditionalist wing of the education world, and had commentators even tipping her as the next head of Ofsted. The book itself aims to set out seven 'myths' which are apparently dearly-held by 'progressives', and to demolish them with evidence and science. Focusing mainly on the first two myths for reasons of time ('facts prevent understanding' and 'teacher-led instruction is passive'), our intrepid podcast quartet interrogate Christodoulou's work and try to get to the bottom of whether this really is 'the most important book of the decade on teaching' (Dylan Wiliam). Sharne then provides us with her wellbeing tip, which involves getting your trainers on and going for a walk, a shoutout to a student who changed careers to become a teacher and press-ganged her giant-vegetable-growing friend into participating in a memorable lesson, and looks at how to grab pupils with a memorable 'hook'.
