Abstract: Mutations in the SNCA gene cause autosomal dominant Parkinsons disease (PD), with progressive loss of dopaminergic neurons in the substantia nigra, and accumulation of aggregates of alpha-synuclein. However, the sequence of molecular events that proceed from the SNCA mutation during development, to its end stage pathology is unknown. Utilising human induced pluripotent stem cells (hiPSCs) with SNCA mutations, we resolved the temporal sequence of pathophysiological events that occur during neuronal differentiation in order to discover the early, and likely causative, events in synucleinopathies. We adapted a small molecule-based protocol that generates highly enriched midbrain dopaminergic (mDA) neurons ( greater than 80%). We characterised their molecular identity using single-cell RNA sequencing and their functional identity through the synthesis and secretion of dopamine, the ability to generate action potentials, and form functional synapses and networks. RNA velocity analyses confirmed the developmental transcriptomic trajectory of midbrain neural precursors into mDA neurons using our approach, and identified key driver genes in mDA neuronal development. To characterise the synucleinopathy, we adopted super-resolution methods to determine the number, size and structure of aggregates in SNCA-mutant mDA neurons. At one week of differentiation, prior to maturation to mDA neurons of molecular and functional identity, we demonstrate the formation of small aggregates; specifically, beta-sheet rich oligomeric aggregates, in SNCA-mutant midbrain immature neurons. The aggregation progresses over time to accumulate phosphorylated aggregates, and later fibrillar aggregates. When the midbrain neurons were functional, we observed evidence of impaired physiological calcium signalling, with raised basal calcium, and impairments in cytosolic and mitochondrial calcium efflux. Once midbrain identity fully developed, SNCA-mutant neurons exhibited bioenergetic impairments, mitochondrial dysfunction and oxidative stress. During the maturation of mDA neurons, upregulation of mitophagy and autophagy occured, and ultimately these multiple cellular stresses lead to an increase in cell death by six weeks post-differentiation. Our differentiation paradigm generates an efficient model for studying disease mechanisms in PD, and highlights that protein misfolding to generate intraneuronal oligomers is one of the earliest critical events driving disease in human neurons, rather than a late-stage hallmark of the disease. Submitted photo | YourClassical Anat Spiegel Spiegel is a Dutch-Israeli composer who now makes her home in the Twin Cities. Rimon executive producer David Harris saw her work as a Cedar Commissions fellow and commissioned Thursday’s performance. “I would describe her music and her performances as bold, theatrical, hugely imaginative — and always surprising,” Wernick said. The Zoom event is entitled “I Once Was a Boy: on the Edge with poet Anna Margolin.” Spiegel’s song series puts music to poems written in Yiddish by Anna Margolin. The evening includes a conversation between Spiegel and vocalist Sarah Larsson about the Yiddish language, poetry, and music. The event Thursday at 7 p.m. is free, but registration is required. Landscape painter Joshua Cunningham recently made a trip to Duluth, Minn., to get a sneak peak at the Great Lakes Academy of Fine Arts’ first student show. The school is a four-year program that teaches a classical style of art. The first graduating class was in 2020, but the pandemic prevented the academy from putting on a year-end showcase. This year’s show includes work by the first two graduating classes as well as current students. Cunningham was intrigued by both the showcase and its location; the school is housed in a decommissioned Catholic church, with pews turned into easels surrounded by original stonework and wooden scaffolding. “It's really neat because beauty endures beyond its original purpose. And so for the students to be practicing and learning in a space about beauty for a bigger thing,” Cunningham said. As for the artwork, Cunningham said “some of those drawings and paintings will be the fruit of weeks of observation. Just think about the last time you spent weeks looking at and studying something.” The student showcase runs Friday from 5 p.m. to 9 p.m. and Saturday from noon to 5 p.m. Looking ahead to next weekend, visual artist Heather Lou in St. Paul is planning to attend the New Eyes Festival 2021: (Un)Scene. The annual festival of short plays from Theater Mu and the Playwrights’ Center will feature a series of 10-minute plays, presented virtually. This year’s playwrights are Carla Ching, Aditi Brennan Kapil, Saymoukda Duangphouxay Vongsay, Isabella Dawis, and Lisa Marie Rollins. This year, the five playwrights respond in some way to growing anti-Asian violence in the country. Lou also appreciates that the plays explore “the ways our community can be celebrated and put in the limelight.” Friday, May 21 at 7 p.m., you can hear from the playwrights and producer Katie Hae Leo during a “Mu-tini Hour” on Facebook live. What company conduct was rewarded? II. J&F Investments DOJ Perspective How do you evaluate something so massive, yet largely resolved by the time the DOJ takes the lead? How is it to work with Brazilian prosecutors? Does each large international investigation have its own character and cadence? Company Perspective This case had difficult corporate governance issues. Your M&A protocol needs to be in place but the risks from each transaction are bespoke. In many ways, the case was a testament to deep pockets. Pay attention to what is said in the resolution documents. III. Goldman Sachs DOJ Perspective Obviously as the biggest FCPA prosecution ever, how does the Dept. work up such a case. Are there multiple prosecutors working up different aspects of the case? How do you coordinate both internationally and domestically with all the other regulatory bodies? What is the one pie concept and why is it so important? Company Perspective Why was there no monitor? Compliance needs to be persistent and persistently right. Why are compliance red flags also commercial red flags? Will clawbacks become the norm? The country's top bureaucrat, Michael Wernick, is leaving his job, telling Prime Minister Justin Trudeau in an open letter that recent events show him there is no path for a "relationship of mutual trust" with the opposition parties. Wernick has been clerk of the Privy Council since 2016, shortly after the Liberals assumed office. But he has come under fire in the past few weeks after being accused by former attorney general Jody Wilson-Raybould of being among a number of officials, including Trudeau and his senior staff, who pressured her to head off criminal charges for SNC-Lavalin. More testimony today at the Commons Justice Committee, on the SNC-Lavalin scandal. Michael Wernick, clerk of the Privy Council, says he is unapologetic for raising alarms at his first appearance before the committee, where he talked about the heated political rhetoric potentially leading to violence and foreign interference in this year's federal election. Wernick said he is deeply worried and that the country needs a debate about foreign interference in elections.
