Dr. Young-Hui Chang is a Professor of Biological Sciences at Georgia Institute of Technology where he directs research in the Comparative Neuromechanics Laboratory. Research in Young-Hui's lab aims to examine how the control of movement by the nervous system is influenced by mechanics and physics during locomotion. He is interested in broad mechanisms for behaviors like walking, running, and hopping that apply within and across species. Young-Hui likes to spend his free time with his family. He, his wife, and his two boys enjoy exploring the outdoors, hiking, and camping together. Though Young-Hui was not always a particularly outdoorsy person, enrolling his sons in the Scouts program has provided an avenue for him and his family to learn more and get outside. Young-Hui received his B.S. in Mechanical Engineering and his M.S. in Animal Physiology from Cornell University. Next, he conducted his doctoral studies at the University of California, Berkeley, earning his PhD in Integrative Biology in 2000. Prior to joining the faculty at Georgia Tech, Young-Hui was a postdoctoral researcher at Emory University. While at Emory, he was awarded the Association of Korean Neuroscientists President Outstanding Research Hanwha Award. Young-Hui has also been awarded an NSF CAREER Award. In this interview, he discusses his experiences in life and science.

J., Ullian, E. M., Kampmann, M. Abstract: The sheer complexity of the brain has complicated our ability to understand its cellular mechanisms in health and disease. Genome-wide association studies have uncovered genetic variants associated with specific neurological phenotypes and diseases. In addition, single-cell transcriptomics have provided molecular descriptions of specific brain cell types and the changes they undergo during disease. Although these approaches provide a giant leap forward towards understanding how genetic variation can lead to functional changes in the brain, they do not establish molecular mechanisms. To address this need, we developed a 3D co-culture system termed iAssembloids (induced multi-lineage assembloids) that enables the rapid generation of homogenous neuron-glia spheroids. We characterize these iAssembloids with immunohistochemistry and single-cell transcriptomics and combine them with large-scale CRISPRi-based screens. In our first application, we ask how glial and neuronal cells interact to control neuronal death and survival. Our CRISPRi-based screens identified that GSK3{beta} inhibits the protective NRF2-mediated oxidative stress response in the presence of reactive oxygen species elicited by high neuronal activity, which was not previously found in 2D monoculture neuron screens. We also apply the platform to investigate the role of APOE-{epsilon}4, a risk variant for Alzheimer's Disease, in its effect on neuronal survival. This platform expands the toolbox for the unbiased identification of mechanisms of cell-cell interactions in brain health and disease. Copy rights belong to original authors. Listen to a blog summary of a research paper published by Aging in Volume 14, Issue 24, entitled, "12-year evolution of multimorbidity patterns among older adults based on Hidden Markov Models." _______________________________________ Multimorbidity is a term that refers to living with two or more chronic diseases at the same time, and the prevalence of this phenomenon increases with age. In addition, humans tend to evolve and transition into distinct patterns of multimorbidity. These still ill-defined patterns of multimorbidity may offer a window of opportunity for researchers. Since the aging population continues to grow in many parts of the world, researchers are motivated to better understand these patterns and how they evolve and transition over time in order to develop interventions and therapeutics for healthier aging. However, this is a challenging task for several reasons. “Multimorbidity is associated with a higher risk of polypharmacy and decreased quality of life, and challenges the decision-making of clinicians that lack effective guidelines for the management and treatment of patients with cohexisting complex diseases [4].” While researchers have investigated multimorbidity, not all studies are created equal—rendering meta-analyses largely incongruent (thus far). One reason the evolution of multimorbidity patterns is so challenging to study is because most study designs are not powered to account for the dynamic nature of multimorbidity in old age. Another reason is that various studies use different lists of diseases. (Some studies include ten conditions or less and others include 200+ conditions.) Finally, most statistical methods used to organize data are not able to properly handle the complexity of multimorbidity. “Exploring how multimorbidity patterns evolve throughout people's lives and the time subjects remain within specific patterns is still an under-researched area [7, 8]. The trailer, and first twenty minutes, suggested that the show would be both heavy-handed (a butterfly in a jar to symbolize a hot mom being smothered by the confines of domesticity? groundbreaking) and bizarrely menacing in tone. If we're going to watch a campy, graphically sexual romance, we prefer a bit more spark and frothy fun and a bit less foreboding voiceover. But then, well, everyone started watching it. A lingering moment of male full-frontal fueled a think-piece cycle. Reviews were, uh, less than kind. Against our own initial instincts, we grew intrigued. And so, at last, we binged. “Sex/Life” tells the story of Billie, a ragingly hot and miserable Greenwich stay-at-home mom. She has a preschooler and a newborn, a sculpted Greek god of a husband who is also a successful hedge funder and living saint, a long-abandoned career as a psychologist and PhD candidate, and a throbbing need for sexual adventure and fulfillment that has begun to explode messily all over her life. In a melodramatic voiceover, taken from the pages of her Word doc journal, Billie recalls the explosive sex she used to have with her bad-boy ex, Brad, who broke her heart, and yearns for a rekindling of that part of herself. And then, oops, her husband reads the diary — AND she runs into Brad, who she learns has been hooking up with her best friend, Sasha, but still has feelings for her. Who will she choose? What will her husband do? The drama unfolds from here, but all the plot is crammed into a handful of minutes in between endless explicit sex scenes, which manage to feel more grim than sensual.In this discussion, we cover the first four episodes of “Sex/Life”: getting to know the paper-flat characters, unpacking the themes, and breaking down the credulity-stretching plot turns. Abstract: The Src-like adaptor proteins (SLAP/SLAP2) bind to CBL E3 ubiquitin ligase to down regulate antigen, cytokine, and tyrosine kinase receptor signaling. In contrast to phospho-tyrosine dependent binding of CBL substrates through its tyrosine kinase binding domain (TKBD), CBL TKBD associates with the C-terminal tail of SLAP2 in a phospho-independent manner. To understand the distinct nature of this interaction, a purification protocol for SLAP2 in complex with CBL TKBD was established and the complex crystallized. However, determination of the complex crystal structure was hindered by apparent SLAP2 degradation during the crystallization process, such that only CBL TKBD residues could be modeled initially. Close examination of the CBL TKBD structure revealed a unique dimer interface that included two short segments of electron density of unknown origin. To elucidate which residues of SLAP2 to model in this unassigned density, a co-expression system was generated to test SLAP2 deletion mutants and define the minimal SLAP2 binding region. As well, SLAP2 degradation products were analyzed by mass spectrometry. Model building and map generation features of the Phenix software package were employed, leading to successful modeling of the C-terminal tail of SLAP2 in the unassigned electron density segments. E., Povlishock, J. T., Rowe, R. K. Abstract: Diffuse brain injury is better described as multi-focal, where pathology can be found adjacent to seemingly uninjured neural tissue. In experimental diffuse brain injury, pathology and pathophysiology have been reported far more lateral than predicted by the impact site. Finite element biomechanical models of diffuse brain injury predict regions of maximum stress and strain. However, the application of a skull with uniform thickness may mask the pathophysiology due to varying thickness of human and animal skulls. Force applied to the intact skull would diffuse the forces, whereas forces applied through an open skull are distributed along paths of least resistance within and then exiting the skull. We hypothesized that the local thickening of the rodent skull at the temporal ridges serves to focus the intracranial mechanical forces experienced during brain injury and generate predictable pathology in underlying cortical tissue. We demonstrated local thickening of the skull at the temporal ridges using contour analysis of coronal skull sections and oblique sectioning on MRI. After diffuse brain injury induced by midline fluid percussion injury (mFPI), pathological foci along the anterior-posterior length of cortex under the temporal ridges were evident acutely (1, 2, 7 days) and chronically (28 days) post-injury by deposition of argyophilic reaction product. Area CA3 of the hippocampus and lateral nuclei of the thalamus showed pathological change, suggesting that mechanical forces to or from the temporal ridges shear subcortical regions. A proposed model of mFPI biomechanics suggests that injury force vectors reflect off the skull base and radiate toward the temporal ridge due to the material properties of the skull based on thickness, thereby injuring ventral thalamus, dorsolateral hippocampus, and sensorimotor cortex. Surgically thinning the temporal ridge prior to injury reduced the injury-induced inflammation in sensorimotor cortex. These data build evidence for the temporal ridges of the rodent skull to contribute to the observed pathology, whether by focusing extracranial forces to enter the cranium or intracranial forces to escape the cranium. Abstract: Setting in of reproductive senescence (RS) gives rise to several changes, making aged individuals susceptible to neurodegenerative diseases, cardiovascular, and bone disorders amongst others. The present study deciphers the association of reproductive senescence, presence/absence of the sex hormone estradiol with age-associated neurodegenerative diseases. We employed RNAi induced silencing of a subset of 22 genes that are known to delay RS, followed by studies on alpha-Synuclein aggregation and associated effects in the transgenic C. elegans. These studies led us to functional characterisation of the Na+/H+ exchanger, expressed exclusively in the gut. We found that RNAi of nhx-2 ameliorates the effects associated with alpha-Synuclein aggregation via mimicking dietary restriction as it alters food absorption from the gut. Our studies further elucidated that such effects are Sir-2.1 driven as nhx-2 RNAi did not delay reproductive senescence when sir-2.1 was silenced concurrently. As estradiol plays a central role in both reproductive health as well as neuronal health, we performed structural binding analysis that demonstrated the binding potential of the estradiol receptor NHR-14 with nhx-2 gene. Hence, we treated the worms with estradiol and observed that the transcription levels of nhx-2 were elevated above the endogenous level. To unravel the underlying molecular mechanism of induction we performed ChIP analysis and it revealed that estradiol treatment gives rise to enhanced NHX-2 levels through inducing the promoter-specific histone H3 acetylation (H3K9) and lysine methylation (H3K4me3).
