Human decision-making – even when a lot of money is at stake, is not always rational. A lot of the time it's based on intangible factors that influence people in ways that can't be explained or justified by data or analysis – this is after all, the entire basis for the field of behavioural economics, and it's not new information. So why, in an industry like venture capital, are people still led oftentimes more by their gut than their head? The answer, of course, is human nature. But in an industry that looks for outliers, that tries to go against the grain, why is it not more of a priority to minimise the amount of returns left on the table by counteracting well-known flaws in our thinking? My guest today is Sharon Vosmek, CEO and managing partner of VC firm Astia, which has, at the core of its thesis, diversity. Not just diversity for its own sake, though Vosmek argues that that would be good enough – but rather as a tangible, strategic way to cast the net as widely as possible, and to eliminate the biases that ultimately end up hurting your return profile. Astia recently released a white paper on what it calls the Sift – its method of sourcing and screening companies for investment, in a way that tries to filter out the pitfalls that people fall into because of their biases or hunches. Things like not starting the process with introductions, but rather an in-depth assessment of the business before face-to-face meetings, or drawing on the experience of a large diverse group of external advisors to assess each company – thereby also spreading the biases across a wider set of experiences. At a time when diversity and inclusion initiatives are coming under attack, Vosmek defends this not as some gimmick, but as a way to demonstrably bring in more money – indeed, Astia's data has shown a far lower failure rate for startups that have made it through the Sift, than the average, and a higher rate not just of survival, but of successful exits, than failures. We also talk about the biases that still exist and how they may manifest themselves, the potential impact that the perception of being an “inclusive” fund may have on fundraising, and the wider impact of a current climate that is hostile to inclusion initiatives. Left Internal Mammary Artery (LIMA) to Left Anterior Descending (LAD) artery anastomosis is the cornerstone of Coronary Artery Bypass Graft (CABG) surgery. Anastomosis of the LIMA to the Great Cardiac Vein (GCV) is a known but rare complication of the surgery. Currently there are no clear guidelines in regard to further management. We report a case of a LIMA to GCV anastomosis managed with a drug eluting stent (DES) to the mid LAD after ruling out a significant left to right heart shunt. The patient determines when the angina is no longer stable angina.The placebo effect of our interventions should not be discounted.LIMA to GCV anastomosis creates a left to right cardiac shunt. A Qp:Qs greater than 1.5 signifies a significant shunt.Increasing the pressure in the coronary sinus may actually be beneficial to the patient.LIMA-LAD is remains the most efficacious and long lasting graft but why not other arterial grafts? Notes - Coronary Artery Bypass Grafting: An Iatrogenic Left to Right Cardiac Shunt Iatrogenic anastomosis of the LIMA to the GCV is a rare but noted complication of CABG surgery. Review of the literature has reported under 40 such cases of arteriovenous fistula formation in the coronary system. Detection of the anastomosis generally stems from recurrent angina which can be attributed to unresolved ischemia or coronary steal syndrome but also can be detected with new heart failure (namely right sided heart failure due to left to right shunting). Diagnosis is usually made with coronary angiography, but CT coronary angiography has also been reported. Due to the rarity of this complication, no clear guidelines are in place directing the management leaving it to the discretion of the various Heart Teams. Evaluating for signs of heart failure and/or ischemia, and measuring the Qp:Qs have been the most common signs directing management. Various options are available for closing the fistula and include coil or balloon embolization, vascular plugs, venous ligation or a covered stent. Redoing the surgery is also an option. Spontaneous closure of the fistula has also been reported. Lastly, if redo surgery is not performed then regardless of fistula closure, coronary intervention for the native diseased artery may be pursued to relieve symptoms. Green Card Voices, the Podcast Season 1 host Mahlet aka MK, Season 2 #LoveYourAsianNeighbors host and co-manager Tri, and Beyond Allyship podcast manager and co-host Asha Thanki join each other after the end of the season 3 Beyond Allyship finale in October 2020, barely a week before the momentous 59th United States presidential election on Tues Nov 3rd, 2020. Tri has a lot of questions for the hosts, including: Mahlet’s thoughts, as the host who was part of the start of the overall life of the podcast, on what the unplanned trajectory, as impacted by worldwide pandemic and the U.S national social uprisings, of the podcast series means to her Tri’s thought process in the conception and shift to season 2 of GCV the Podcast, titled the #LoveYourAsianNeighbors series. As GCV’s then-social media intern with an amateur enthusiasm for podcasts as a long-time podcast consumer, Tri took on the task of shifting the format of the podcast to best demonstrate GCV’s role as a storytelling platform during a global pandemic whose circumstances also amplified anti-Chinese and Chinese-adjacent attitudes and actions in countries such as the U.S. Asha’s circumstances and task that became creating the season 3 series, Beyond Allyship, of stepping into the podcast manager role in 2020, in the wake of George Floyd’s murder by Minneapolis police and the consequent uprisings that began in Minnesota and has since spread to many sites of allied protests worldwide, and shifting the podcast’s goals, yet again, to address the social injustices against peoples grouped and distinguished as “Black” and African under the banner of a Minneapolis-based non-profit that primarily seres as a platform for immigrant and refugee first-person storytelling Asha sharing how GCV the Podcast is set up well to reach listeners in many different places as it concerns their understanding of the political, historical, and cultural moment we’re living by facilitating a platform for guests to demonstrate what work they have already been doing, rather than the hosts guiding the conversations outside of the immediate experiences of the guests Tri’s comment on how the Green Card Voices became a veritable “time capsule” for three stages of GCV’s varied alignment of commitments to immigrant and refugee first-person storytelling first on its own terms, then in emphasized relationship to Asian U.S diasporic peoples during a global pandemic, and in most recent memory in relationship with often non-immigrant Black peoples and recent-diasporic African peoples Tri asks Mahlet, Asha, and is asked himself: “How are you? In recent years data sets have become increasingly more complex requiring more flexible instruments for their analysis. Such a flexible instrument is regression analysis based on a structured additive predictor which allows an appropriate modelling for different types of information, e.g.~by using smooth functions for spatial information, nonlinear functions for continuous covariates or by using effects for the modelling of cluster--specific heterogeneity. In this thesis, we review many important effects. Moreover, we place an emphasis on interaction terms and introduce a possibility for the simple modelling of a complex interaction between two continuous covariates. Mainly, this thesis is concerned with the topic of variable and smoothing parameter selection within structured additive regression models. For this purpose, we introduce an efficient algorithm that simultaneously selects relevant covariates and the degree of smoothness for their effects. This algorithm is even capable of handling complex situations with many covariates and observations. Thereby, the validation of different models is based on goodness of fit criteria, like e.g.~AIC, BIC or GCV. The methodological development was strongly motivated by case studies from different areas. As examples, we analyse two different data sets regarding determinants of undernutrition in India and of rate making for insurance companies. Furthermore, we examine the performance or our selection approach in several extensive simulation studies.

Neue Therapieansätze zur Krebsbehandlung aus dem Bereich der Gen- und Immuntherapie haben als Ergänzung zu den konventionellen Behandlungsmethoden besondere Bedeutung erlangt. Zu ihnen zählen u.a. die Suizidgentherapie und die Verwendung Zytokin-Gen-modifizierter Tumorzellvakzine. Beide Strategien wurden in der vorliegenden Arbeit herangezogen, um eine Immunantwort gegen das murine B-Zell-Lymphom A20 zu erzeugen. 1. Versuche mit HSV-tk-Gen-transduzierten Lymphomzellen. Das Prinzip der Suizidgentherapie mit dem HSV-tk/GCV-System beruht auf der Transduktion des HSV-tk-Gens in Tumorzellen, die dadurch gezielt durch GCV abgetötet werden können. Dabei werden auch nicht-transduzierte, angrenzende Zellen miterfasst („Bystander“-Effekt). Ganciclovirsensible, HSV-tk-Gen-transduzierte A20-Zellen (A20-TK) zeigten in vitro einen eingeschränkten „Bystander“-Effekt. In vivo konnten A20-TK-Zellen nur bei jeder dritten Maus vollständig durch GCV eliminiert werden, wobei sich jedoch bei der Hälfte der überlebenden Tiere eine dauerhafte, systemische Antitumorantwort induzieren ließ. 2. Versuche mit HSV-tk-Gen-transduzierten Triomzellen. Die Triomzelle BiV, hervorgegangen aus der Fusion des Lymphoms A20 mit der Hybridomzellinie 2.4G2, dirigiert Tumorantigene über die Expression eines anti-Fc-Rezeptor-Antikörpers an APC und führt auf diese Weise zur Tumorabstoßung. Trotz Xenogenität sind BiV-Zellen jedoch bis zu 11 Wochen nach Immunisierung in vivo nachweisbar. Auch HSV-tk-Gen-transduzierte BiV-Zellen (BiV-TK) konnten trotz GCV-Verabreichung vier Wochen post iniectionem in den Milzen immunisierter Mäuse nachgewiesen werden, allerdings in niedriger Frequenz. Obwohl nicht alle BiV-TK-Zellen durch GCV in vivo eliminiert werden, hatte eine frühzeitige Rückholung der Zellen einen verminderten Antitumoreffekt zur Folge. Im Gegensatz dazu beeinflusste eine späte GCV-Gabe die Tumorabstoßung nicht. Dies deutet darauf hin, dass für einen potenten Tumorschutz die langdauernde Verfügbarkeit von Vakzinezellen im Sinne einer persistierenden Antigenquelle erforderlich sein könnte. 3. Versuche mit GM-CSF-Gen-transfizierten Triomzellen. GM-CSF-Gen-transfizierte Triomzellen (BiV-BNG) schützten in Präimmunisierungsversuchen effektiver vor Tumorwachstum als Wildtyp-BiV-Zellen, bei therapeutischer Applikation wurde der Antitumoreffekt jedoch überaschenderweise aufgehoben. Durch Blockierung der Triomzell-vermittelten Tumorantigen-Heranführung an APC durch BiVneg-BNG bzw. den Antikörper 2.4G2 konnte die ursprüngliche Antitumorantwort vollständig bzw. teilweise wiederhergestellt werden. Eine mögliche Erklärung hierfür wäre eine hemmende Interaktion zwischen intaktem BiV-Protein und GM-CSF, resultierend in einer Hemmung der APC. Offenbar beschränkt sich die Hemmung nur auf die betroffene APC, denn BiV-BNG hob den Therapieeffekt von A20-BNG bzw. BiV bei simultaner Applikation nicht auf. Die Applikationsroute als mögliche Ursache der Wirkungslosigkeit von BiV-BNG im Therapieansatz wurde durch Änderung des Präimmunisierungsprotokolls ausgeschlossen. Da IL-4-defiziente Mäuse bei therapeutischer BiV-BNG-Gabe eine bessere Überlebensrate erreichten als Wildtypmäuse, könnte der aufgehobene Therapieeffekt von BiV-BNG mit einer Verschiebung des Th1/Th2-Verhältnisses zu Gunsten einer Th2-Antwort zusammenhängen.
