While many of us may have been taught that single genes produce single proteins that have single functions, protein function is far more dynamic than that. This representation of the central dogma is, however, very much a simplification of its original formulation by Francis Crick and over-applying it can lead us down spurious paths and faulty conclusions. For our final episode of our 3-part series on the Central Dogma, we discuss (one of) the final products of DNA and RNA: proteins! We cover everything from where proteins come from to some of their roles as molecular machines. Along the way, we touch on topics such as how eating protein helps you make muscles to "the protein fold that kills." Ashley's Biggest Takeaways The U.S. Centers for Disease Control and Prevention (CDC)'s Morbidity and Mortality Weekly Report first reported on a cluster of unusual infections in June of 1981, which would become known as AIDS. Evidence suggested that the disease was sexually transmitted and could be transferred via contaminated blood supply and products, as well as contaminated needles, and could be passed from mother to child. All hemophiliacs of this generation acquired AIDS (15,000 in the U.S. alone). The fact that the microbe was small enough to evade filters used to screen the clotting factor given to hemophiliacs indicated that the etiologic agent was a virus. AIDS patients had low counts of T-lymphocytes called CD4 cells. By 1993, the most likely virus candidates included, a relative of hepatitis B virus, some kind of herpes virus or a retrovirus. Howard Temin discovered reverse transcriptase, working with Rous sarcoma in the 50s and 60s. His work upset the Central Dogma of Genetics, and at first people not only did not believe him, but also ridiculed him for this claim. Research conducted by David Baltimore validated Temin's work, and Temin, Baltimore and Renato Dulbecco shared the Nobel Prize for the discovery in 1975. Robert Gallo of the U.S. National Institute of Health (NIH), discovered the first example of a human retrovirus—human T-cell lymphotropic virus (HTLV-1). Françoise Barré-Sinoussi worked on murine retroviruses in a laboratory unit run by Luc Montagnier, where she became very good at isolating retroviruses from culture. In 1982, doctors gave lab Montagnier's lab a sample taken from a with generalized adenopathy, a syndrome that was a precursor to AIDS. Barré-Sinoussi began to detect evidence of reverse transcriptase in cell culture 2 days after the samples were brought to her lab. Barré-Sinoussi and Luc Montagnier were recognized for the discovery of HIV with the 2008 Nobel Prize in Physiology or Medicine. In this episode, I discuss the Central Dogma of Molecular Biology, which is the observation that all living things have DNA, RNA and Protein, and that DNA codes for RNA, and RNA codes for Protein. This "DNA -> RNA -> Protein" system undergirds all living things on Earth. The question is this: Could this system come about by random chance in the prebiotic soup of the early Earth? Assuming there were plenty of DNA nucleotides floating around, could they come together to form a small, 300 nucleotide, meaningful piece of DNA? Also, if all the necessary amino acids were present, could they slam together and form a meaningful/functional protein in a reasonable amount of time? Let's explore some of the probability calculations provided by Dr. Stephen C. Meyer, a former geophysicist and philosopher of science, who has looked at this system extensively. He is allied with them through the aspect of Nebelun and his inclusion in the Gnomish pantheon. We have Dr. Bruce Lipton on the show today! He wrote The Biology of Belief, and he is here to talk about how our consciousness creates our life experiences. Ultimately, we manifest our beliefs. Our lives and our own health may be a manifestation of our belief system. Dr. Lipton is a stem cell biologist and is an internationally recognized authority in bridging science and spirit. A cell biologist by training, he taught at the University of Wisconsin’s School of Medicine, and later performed pioneering studies at Stanford University. Biology's Central Dogma has been that life is controlled by genes and it's only in the past 50 years – as we have increased knowledge of the molecular mechanisms underlying the regulation of gene expression – that we know that our behaviors and environment can cause changes that affect the way in which our genes work. If the possession of the gene doesn't necessarily mean that you get the disease, but a life out of harmony can activate the gene that we don't want to activate… what are you going to do about it? The power is in your hands!

Segment 1: Introduction to Population Genetics: Central Dogma, Allele Frequency Equation and the Gene PoolIntroduce the episode topic Include definitions and vocabulary Will be discussing the topic of population genetics which is the study of genetic variation within a population and looking into changes in the frequencies of genes and alleles in populations over time Natural selection is one of the most influential factors that can affect a population's genetic composition Central dogma of biology is when the instructions contained in DNA are converted into a functional product, a phenotype DNA, contains the genes that determine who you are, and proteins determine the structure and function of all your cells It describes the two-step process, transcription and translation, of how information in genes flow into proteins, creating a string of amino acids called polypeptides The DNA has the information which is used by the RNA to make the proteins The Allele Frequency Equation: an allele is a version of a gene and a heritable unit that controls a particular feature of an organism The allele frequency refers to how often a particular allele appears in a population An equation called the Hardy-Weinberg equation is used to calculate the genetic variation in a population: p^2 + 2pq + q^2 p^2 and q^2 are the allele frequencies of the homozygous recessive and homozygous dominant, and 2pq is the allele frequency of the heterozygous genotypes To get p and q individually, you calculate actual/total # of alleles With this knowledge, you are able to calculate the total allele frequencies using the equation p + q = 1 The gene pool is calculated using the equation just mentioned, p + q = 1 since it is the sum of both allele frequencies A gene pool is the collection of different genes within an interbreeding population, and refers to its genetic diversity The larger the gene pool, the greater genetic diversity, and the better a population is able to withstand environmental challenges Segment 2: Examples of Population GeneticsHave a natural transition into an example… no need to say “segment 2” Provide detailed example(s) of your topic This first example is a depiction of the central dogma and the different processes at work from Khan Academy DNA directs the construction of the chain of amino acids through transcription, which is when the DNA sequence of a gene is copied to make an RNA molecule During the second process, translation, mRNA is decoded to specify the amino acids of the polypeptide chain Overall, information flows from DNA to RNA to a protein, and this directional flow is why it is the central dogma of molecular biology An example using pea plants demonstrates how to calculate the allele frequency of a population using the total number of alleles and fractions There are 9 pea plants, meaning 18 total alleles 6 of them are homozygous dominant (WW), 1 is heterozygous (Ww), and 2 are homozygous recessive (ww) To calculate p and q, set up fractions and convert them into percentages There are 13 copies of the W allele and 5 copies of the w allele, so the allele frequencies for each are 72% and 28% respectively If you notice, they add up to 1, or 100% To show a gene pool, here is a picture of butterflies of 3 different colors: orange, white, brown You can see the diversity in the population through the different allele combinations: AA (brown), Aa(orange), aa (white) Segment 3: Digging Deeper into Population GeneticsHow does this topic fit into the greater picture of evolution? I recently saw an estimate that if you took all the novel coronaviruses in the world (the actual viruses, not patients), you could fit them into a bucket no more than a couple of liters in volume. A huge impact has been wrought by a very small amount of stuff. The world of viruses is vast and complicated, and we’re still learning some of its basic features. Today’s guest David Baltimore won the Nobel Prize in Physiology or Medicine for the discovery that genetic information in viruses could flow from RNA to DNA, establishing an exception to the Central Dogma of Biology. He is the author of the Baltimore Classification scheme for viruses, and has done important research in the role of viruses in diseases from AIDS to cancer. We talk about what viruses are, how they work, and the status of the novel coronavirus we are currently battling. He is currently the Robert Andrews Millikan Professor of Biology at Caltech. At age 37 he was awarded the Nobel Prize, which he shared with Howard Temin and Renato Dulbecco. The mRNA transcript is named after the Latin word for writing (1:50). RNA Polymerase will synthesize mRNA in the 5’-3’ direction, just like DNA polymerase. Do you know Chargaff’s rules? (3:22). The mRNA transcript will need to go through modification as it exits the nuclear pore (3:40). Translation involves mRNA, rRNA and tRNA to form the polypeptide (4:50).
