Abstract: Aberrantly accumulated metabolites such as fumarate elicit intra- and inter-cellular prooncogenic cascades, yet current methods to measure them require sample perturbation or disruption and lack spatio-temporal resolution, limiting our ability to fully characterize their function and distribution in cells and within a tissue. Raman spectroscopy (RS) is a powerful bio-analytical tool that directly characterizes the chemical composition of a sample based solely on the optical fingerprint of vibrational modes. Here, we show for the first time that RS can directly detect fumarate in living cells in vivo and animal tissues ex vivo. Using the observed linear relationship between Raman scattered intensity and fumarate concentration, we demonstrate that RS can distinguish between Fumarate hydratase (Fh1)-deficient and Fh1-proficient cells based on their fumarate concentration. Moreover, RS reveals the spatial compartmentalization of fumarate within cellular organelles: consistent with disruptive methods, in Fh1-deficient cells we observe the highest fumarate concentration (37 +/- 19 mM) in the mitochondria, where the TCA cycle operates, followed by the cytoplasm (24 +/- 13 mM) and then the nucleus (9 +/- 6 mM). Finally, we apply RS to tissues from an inducible mouse model of FH loss in the kidney, demonstrating that RS can accurately classify FH status in these tissues. These results suggest that RS could be adopted as a valuable tool for small molecule metabolic imaging, enabling in situ dynamic evaluation of fumarate compartmentalization. Abstract: Cell culture, the workhorse of biomedical research, is generally considered to be hyperoxic. However, oxygen consumption by cells is underappreciated. High cellular respiration rates can rapidly deplete oxygen, resulting in local hypoxia. Increasing pericellular oxygen levels rewired the metabolism of multiple post-mitotic cell-lines, both in monolayer and organoid culture. Under standard conditions, cultured adipocytes are hypoxic and highly glycolytic. Increased oxygen availability diverted glucose flux toward mitochondria and increased lipogenesis from glucose-derived carbon. These metabolic changes were coupled to thousands of gene expression changes, and rendered adipocytes more sensitive to insulin and lipolytic stimuli. Importantly, pathway analyses revealed increasing oxygen tension made in vitro adipocytes more similar to in vivo adipose tissue. hPSC-derived hepatocytes and cardiac organoids were also functionally enhanced by increased oxygen. Our findings suggest that oxygen is limiting in many standard cell culture systems, and highlight how controlling oxygen availability can improve translatability of cell models. Long gone are the days of popping pimples in secret or attempting DIY acne solutions, like toothpaste or liquid Advil, as an influx of acne-positivity brands have entered the market. In turn, the stigma surrounding acne is slowly being erased. Peace Out Skincare, a brand dedicated to acne and aging products, launched in 2017 “to market effective products that deliver on the promises,” said Enrico Frezza, Peace Out Skincare founder and CEO, on the latest Glossy Beauty Podcast. Frezza struggled with acne as both a teenager and adult. As a beauty outsider (Frezza's background is in cybersecurity), he hoped to “build a strong, accurate, positive community where people can talk openly about the mental struggles of acne and educate one another.” Notably, among the brand's repertoire are the Peace Out Acne Dots. Despite the small size of the patches, the acne healing dots contain a complex ingredient profile of salicylic acid, retinol, aloe vera and calcium. Nowadays they're considered as much an acne treatment as a fashion statement. Peace Out has also released products that address wrinkles, dark spots and puffy eyes, as well as topical products like its acne and retinol serums. And while the brand started as a Sephora exclusive, Peace Out looks forward to its partnership with Kohl's through the retailer's shop-in-shop to reach the everyday customer through its “affordable pricing,” he said. Peace Out products start at $5 and range up to $34. Additionally, the brand found success on its DTC website, as online shopping became the new normal as a result of the pandemic. Apart from the success found on its website, Peace Out's digital presence has manifested into an acne docuseries, "Acne Champs." The brand can also be found on Facebook, with a focus on anti-aging, as well as TikTok, where the brand reaches its Gen-Z audience through acne education and fun videos, he said. “Instagram is a balance between the two,” targeting the millennial consumer.

Enrico Frezza went from testing skincare products in his kitchen (and sometimes burning layers of skin off) to building the number one acne brand in Sephora North America. As the founder and CEO of Peace Out Skincare, Enrico is challenging the clinical, unappealing acne industry with unique, effective products and a brand centered around “acne positivity.”Initially, Enrico's goal was to find a solution to his own acne problems. After trying every acne spot treatment in the drug stores, he set out to make his own product by mixing hydrocolloid wound care dressing with salicylic acid. He left it on overnight and when he woke up his pimples were gone, but a layer of his skin was gone, too. It wasn't great, but it was a start and Enrico knew he had found a solution. He approached 65 different labs that all told him it wasn't possible to mix active ingredients with hydrocolloid dressing. Undeterred, Enrico (who worked in cybersecurity and had zero scientific background) researched patents and found one with a mix of ingredients that gave him hope his product was possible to produce. He met with the business that held the patent and they started on the R&D process. Together, Enrico and his husband, Junior, who has 20 years of experience in consumer marketing, built the Peace Out brand — as in "peace out" to your skin imperfections. After seeing how clinical, doctor-driven, and sometimes scary acne marketing was, Enrico wanted to build an upbeat, engaging, and simple brand while bringing breakthrough technology to the industry. From the start, their goal was to launch with Sephora. Among all the proteins expressed by the virus, RNA helicase is a fundamental protein for viral replication, and it is highly conserved among the coronaviridae family. To date, there is no high-resolution structure of helicase bound with ATP and RNA. We present here structural insights and molecular dynamics (MD) simulation results of the SARS-CoV-2 RNA helicase both in its apo form and in complex with its natural substrates. Our structural information of the catalytically competent helicase complex provides valuable insights for the mechanism and function of this enzyme at the atomic level, a key to develop specific inhibitors for this potential COVID-19 drug target. This Thanksgiving, try asking a college-aged relative whether they know it used to be illegal to own a phone. Today's smart-phone-owning young people do not remember the bygone era (pre-90s) when you had to rent it from the phone utility, Bell Labs.
