Hey gang! This week on the show the history segment is chock full of WrestleMania goodness! So we cover that before getting into The Smackdown Symposium which took place hot off the heels of the classic WrestleMania 17 from Houston. There is a big time change in the landscape of the WWF in 2001 as SCSA has aligned himself with the dastardly Vince McMahon and HHH. Tonight's special guest is John Anderson from River Ridge, Louisiana, a child abuse survivor, activist, volunteer and member of the NAASCA family. "My abuse started at the age of nine at St. Matthew in River Ridge, Louisiana," he explains. "It continued for three weeks in class in front of other students. I was threatened and scared. My friends saw Brignac abusing me with their own eyes. So did many other students," he says.  "It took the help of one of the witnesses to tell my parents. I couldn't." John goes on, "I started breaking down. My life would never be the same. 6 different schools later I barely graduated. My mother was in her third trimester with my brother while trying to convict George Brignac. We had him arrested and with a bench trial Brignac was acquitted for the reasons of conspiring to expose his celibacy. I had two witnesses. Brignac managed to get 26 victims after me. I was the first." Today he's getting better. "I suffer extensive damage physically and mentally. C-PTSD, fibromyalgia, permanent nerve damage, depression and anxiety. I have been going to doctors since I was 10. I use my experience of 45 years to help any and all victims." With little support from the baby's father, a pill habit to feed, Jillian soon found herself in a cycle of addiction, psychological, and physical abuse. In the years following, she went from one bad relationship to the next. Eventually, she found her saving grace in her Catholic faith, her husband Lonnie and her children, and her determination to stay clean. 'My Ugly Truth: Life Beyond Abuse' is a riveting story, about not just the social and emotional realities of addiction, but also the monetary and opportunity costs of single parenthood and the secret lives of abusers and the abused who live to tell their stories. Jillian is an entrepreneur, a two-time published author (the other book is a cook book, 'Spicy, Cajon Love'), an activist and advocate, an inventor, outdoorswoman, wife and mother of three. She is a co-owner of NOLA Prestige Electric, a women/minority/disabled veteran-based contracting company. Jillian was recently appointed to the Board of Directors of SCSA, and serves on a committee where she and her colleagues recently triumphed, passing Bill 492, allowing victims of Childhood Sexual Abuse to have their day in court. Tonight's special guest is Richard Windmann from Fort Worth, Texas, who was sexually abused in Boy Scouts by four leaders who passed him around, by a sex crimes detective who worked on his case who was later jailed himself, and later by the janitor and a priest at a Jesuit high school. Richard's childhood was filled with sexual abuse. "I was a typical mid-city kid." Unbeknownst to his mother the boy scout troop he joined was started by pedophiles and all four of his Boy Scout leaders raped him. Men would fly in from out of town to have sex with me. It was two years before detectives would find this pedophile ring. Then he contacted two police officers and one told him not to call back. "As a kid I did not know to call the District Attorney," he explains. "Then the sex crimes detective also raped me and scarred me by showing me a picture of a dead teen. I was then called in to a court case against this sex crimes detective. The court found that my testimony was believed over the detective's testimony." Richard stays busy helping others. "I find my healing through helping others find their voice," he says. "I am the president of a group which helps adult survivors of child abuse try to get justice and tell their story. The name of the organization is 'Survivors of Childhood Sex Abuse'." SCSA is an advocacy and support group for victims and survivors of childhood sex abuse. They offer resources and support free of charge to victims and survivors. The mission of Survivors of Childhood Sex Abuse is to empower victims to become survivors and thrive. "Through understanding, empathy, education and advocacy, we support you and your loved ones in your healing journey," he says. "We will listen to you, believe you, and we support you."

Go to the Caribbean, go to Africa." Watch the Video  On Aug. 14 1862, Abraham Lincoln hosted a “Deputation of Free Negroes” at the White House, led by the Rev. Joseph Mitchell, commissioner of emigration for the Interior Department. It was the first time African Americans had been invited to the White House on a policy matter. The five men were there to discuss a scheme that even a contemporary described as a “simply absurd” piece of “charlatanism”: resettling emancipated slaves on a 10,000-acre parcel of land in present-day Panama.   Edward M. Thomas, John F. Cook, Cornelius C. Clark, John T. Costin, and Benjamin McCoy—met with Lincoln in August 1862 to debate his proposal for a black colony in Central America Cook, Costin, and Clark were members of the Social Civil and Statistical Association, A black city-based organization. The SCSA had sought to "banish several emigration promoters from Washington" weeks before the delegation met with Lincoln. Many of the SCSA's members were connected with the prestigious Fifteenth Street Presbyterian Church.

The aim of this study was to examine if the assessments of sperm quality can be better objectified and standardized by using flow cytometric examinations. For this investigation temporary deterioration of sperm quality was induced by a local heating of testes in bulls. The scrotal surface temperature of 4 bulls was increased during the local heating of testis by enclosing the entire scrotum for 48 hours by 6.4°C to 7.1°C. Semen was collected 3 times a week from Day -7 to Day 61 and once a week from Day 68 to Day 83 (Day 0 = the day of scrotal insulation). Each ejaculate was conventionally examined (numbers of total spermatozoa, sperm concentration, morphology and proportion of vital spermatozoa after staining with bromphenol blue nigrosin). The percentage of motile spermatozoa and progressive motile spermatozoa of motile sperm were determined with a computer-assisted motility analysis. Using flow cytometry the proportions of sperm with the following parameters were examined and analyzed: intact plasma membranes after staining with SYBR®14/PI, high mitochondrial membrane potentials after staining with JC1, damaged acrosome status after staining with FITC-PNA/SYTO®17/PI, and sperm with defective chromatin structure or rather with high DNA fragmentation index (DFI) by SCSA. The deterioration of sperm quality after elevating testicular temperature was correspondent to a large extent to the typical changes of the conventional sperm parameters, which result is in agreement with previous similar studies. After testicular hyperthermia (Day 0) changes of sperm quality occurred in the following sequence. There was a notable increase in secondary sperm abnormalities as well as a decrease in sperm motility on Day 9 after testicular hyperthermia. The proportions of vital spermatozoa after staining with bromphenol blue nigrosin, total sperm count and sperm concentration were decreased on Day 12 for the first time. At the same time the primary sperm abnormalities began to increase. The primary abnormalities most frequently encountered were morphological head defects of spermatozoa. Concurrent with the changes specified above the following alterations of sperm parameters were observed with flow cytometry. Beginning on Day 7 after testicular hyperthermia sperm with defective acrosome status increased. From Day 9 on sperm with intact plasma membranes and sperm with high mitochondrial membrane potentials began to decrease. On Day 12 after testicular hyperthermia the proportion of sperm with defective chromatin structure (spermatozoa with high DFI) started to increase significantly. The relationships between the proportions of defective sperm chromatin structure assessed with the SCSATM test and the proportions of sperm head defects were highly significant (r = 0.81; P < 0.0001). The proportion of sperm with a high mitochondrial membrane potential correlated positively to the sperm motility (r = 0.83; P < 0.0001). Significant correlations between the viability assessed by light microscopy and the percentages of spermatozoa with intact plasma membrane obtained by flow cytometry (r = 0.77; P < 0.0001) occurred. The increase of the proportion of the sperm with defective chromatin structure (spermatozoa with high DFI) was more clearly pronounced than that of the morphological abnormal sperm heads. This result indicates that heat stress had led to chromosome defects not only in morphologically abnormal but also in normally appearing sperm cells. In addition, morphological sperm abnormalities, especially sperm head defects, may be indicative of chromosome abnormalities also in normally appearing sperm cells of an ejaculate. This study shows that flow cytometric assessments of the ejaculate is a reliable and objective method in semen investigation. It provides additional important information about the sperm quality. In further studies, it has to be clarified, whether the prospective fertilization capacity from an ejaculate and/or the fertility of a bull can be predicted with this method more reliable than with the conventional sperm evaluation.

The aim of these examinations was to check various flow cytometric methods to establish whether they are suitable objective methods for assessing the quality of cryopreserved sperm from stallions. The plasma membrane integrity, the mitochondrial membrane potential, the acrosomal status and the integrity of the chromatin structure in cryopreserved sperm from stallions were assessed. For this purpose various fluorescent stainings were carried out both immediately after the semen samples were thawed and after a three-hour period of incubation at 37 C with SYBR® 14, JC-1 and SYTO® 17 / propidium iodide / FITC-PNA and a sperm chromatin structure analysis (SCSA™) and were then evaluated. The sperm quality parameters obtained by flow cytometry showed good reproducibility. The results of measurements of three different straws of an ejaculate which were carried out independently of each other did not differ significantly. The intra-class correlation coefficients were between 0.88 and 0.98. The flow cytometric tests evaluated in this examination were therefore considered to be reliable. In order to demonstrate the correlations between the routine examination methods for assessing the sperm quality and the analysis processes evaluated in this study, the viability and the morphology of the sperms after thawing were assessed by light microscopy using bromide phenol blue nigrosine smears. A computer controlled motility analysis was also carried out. Moderate correlations (0.60 > r > 0.58; p < 0.0001) were obtained between the viability assessed by light microscopy and the percentage of vital sperms obtained by flow cytometry. The proportion of progressively forward moving sperms correlated positively (r = 0.50; p < 0.01) to the proportion of sperms with a high mitochondrial membrane potential. The relationships between the results of the SCSA™ test and the proportions of both primary and secondary sperm anomalies were weakly pronounced (r = 0.24; p < 0.01). In the flow cytometric assessment of the viability, the mitochondrial membrane potential and the integrity of the acrosome it was shown that the ejaculate-related variations were approximately the same as the variations between the stallions. Regarding the results of the sperm chromatin structure the individual variations (88 - 91%) were considerably greater than those dependent on the ejaculate (9 – 12%). It was also shown that the results of the flow cytometric sperm quality assessment did not vary significantly in stallions which had a period (four months) of sexual rest before providing ejaculate compared to stallions which ejaculates were collected with a regular frequency. In terms of the animals’ age there was only a weak relationship (r = 0.43; p < 0.01) with integrity of the sperm chromatin structure. The results of the flow cytometric methods for assessing the viability (SYBR® 14/PI, JC-1 and SYTO® 17/FITC-PNA/PI combination staining) were comparable (r > 0.96; p < 0.0001). The latter is the only one of these tests which provides additional information about the integrity of the acrosome of the sperms. The combination of this triple staining with the sperm chromatin structure analysis (SCSA™) also provides further important information about the sperm quality. To check the relationships between fertility of the stallions and the various sperm quality parameters obtained by flow cytometry, only seasonal pregnancy rates from fresh semen insemination were available. In terms of the viability and the acrosomal status of the sperms after thawing, these showed no correlations (p > 0.05) with the fertility of the stallions. On the other hand, a negative relationship was established between the integrity of the chromatin structure and the fertility of the stallions (r = - 0.51 / - 0.59). This study shows that the flow cytometric assessment of the quality of cryopreserved sperm from stallions represents a reliable and objective method. In addition to the routine assessment of sperm quality, it provides important additional information, for example about the intactness of the acrosome and the integrity of the sperm chromatin structure.
