Abstract: The cellular actin cortex provides crucial mechanical support and plays critical roles in numerous functions, including cell division and migration. The proteins of the ERM family, ezrin, radixin, and moesin, are central to these processes by linking the plasma membrane to the actin cytoskeleton. To investigate the individual contributions of these three proteins to leukocyte migration, we generated single and triple ERM knock-out macrophages. Surprisingly, we found that even in the absence of ERMs, macrophages can still form the different actin structures promoting cell migration, such as filopodia, lamellipodia, podosomes, and ruffles. Furthermore we discovered that, unlike every other cell type previously investigated, the single or triple knock-out of ERMs does not affect macrophage migration in a large diversity of contexts. Finally, we demonstrated that the loss of ERMs in macrophages does not affect the mechanics of their actin cortex. These findings challenge the notion that ERMs are universally essential for cortex mechanics and cell migration and support the notion that the macrophage cortex may have diverged from that of other cells to allow for their adaptive cortical plasticity. A., Kim, J. H., Sia, G. M. Abstract: Microglia play a pivotal role in mediating the neurological effects of chronic stress, and is implicated in stress-associated diseases such as depression. However, the biological states of microglia in the stressed brain remains unclear. Here, using single cell RNA sequencing of mouse cortical microglia, we identify a stress-associated microglia state marked by high expression of apolipoprotein E (ApoE). In the prefrontal cortex, ApoEhigh microglia are localized to layer 2/3, and are caused by stress-induced local complement activation in the upper cortical layers. In mice lacking complement component C3, chronic stress does not induce the ApoEhigh microglial state, and these mice are protected from stress-induced synapse loss, anhedonia, and working memory deficits. Our data show that spatially-restricted complement and microglia activation can result in regional vulnerability of synapses in brain disease, and highlight the role of immune processes in causing region-specific circuit defects that are associated with many psychiatric diseases. My special guest today is my beautiful and fantastic friend, Lyndsay Carreno (@LyndsCarreno). Lyndsay is a world traveler, political activist, historian, music festival veteran, and proud to say one of my best friends in the whole world. The first time I met Lyndsay was at a concert in Atlanta, and from the moment I met her I knew I wanted to be her friend. Its so crazy looking back on it because now I know her family, we've traveled all around the world together, gone to countless music festivals and concerts together, and she has introduced me to some really awesome people. It's weird to think how much different my life might have been if I had never met her. Lyndsay moved to Washington, DC six months ago, and didn't know a soul. Just straight hustled from the ground up living in some ridiculous housing environments to now having a ballin' apartment with her roommate, Dana, who makes some tasteful cameos in this episode. She has always wanted to move out there, and I am super duper proud of her for conquering her fears and just going for it. She was also sick the day that we recorded this, and she still totally killed it. I was kinda afraid we wouldn't be able to record when she told me that she got sent home from work since she was sick, and she still rallied through. That's just the kind of person she is.  On this episode we covered a lot of topics. She shared her stories from her childhood growing up in England, the stories of the impact her dad made on her life, her dad passing away last year, and at the end we talked some good ol' american politics. Lyndsay is such a strong woman and I really want to thank her for being so vulnerable and sharing so much with us. Death is never a fun subject, but I'm really glad we were able to talk about it and hopefully listening to this can help some other people who may be dealing with loss right now, too. Make sure to follow her on Instagram @LyndsCarreno   If you would like to support the podcast, please rate 5 stars and review on iTunes. Music by Calvin Kraakevik
