It's Miriam Shor, y'all! You Might Know Her From Younger, Hedwig and the Angry Inch, Magic Hour, GCB, Shortbus, Swingtown, Maestro, American Fiction, and Guardians of the Galaxy Vol 3. Miriam gave us all the scoop on grounding the zaniness of Diana Trout on Darren Star's Younger, leading the indie comedy Magic Hour, and appearing in "brilliant but canceled" series like GCB and Swingtown. All that, plus Miriam talked to us about being one of John Cameron Mitchell's "players," appearing in Shortbus and, of course, originating the role of Yitzhak in the Hedwig universe; popping up in prestige Oscar films; her musical theatre roots; New York City history; and dying by way of a paintbrush in The Americans. We just LOVED Miriam.

Fintan's news is bigger though: a new sponsor has joined the podcast family in the shape of Optimove, and after an in-depth product demo the chaps are bedazzled. Bedazzled!  The gents discuss the product, and Optimove's presence at ICE, and Amazon gift vouchers, and more [0:00 - 20:14]. Oh, and our something different is, we've done video this time. It might not be live when this goes live, but you can see the channel here. Then the week's very special guest is the wonderful Aideen Shortt, advisor to Curacao's Minister of Finance and who has been at the heart of the island's regulatory overhaul. Aideen has had a long and impressive career in gambling, with many big-name companies and quality job titles on her resumé. Aideen discusses the changes to Curacao's gambling licensing regime, clarifying that the LOK has not been rejected and explains the parliamentary process it is going through. She emphasises the need for modernization and the economic, social, and reputational benefits it will bring to Curacao, discusses the timeline and process for existing and new licensees, and explains the flexibility and transparency of the GCB portal. Rather excitingly, we talk about the draft crypto legislation and the involvement of experts in shaping it, the game-changing potential of Curacao and the importance of forward-looking legislation.  At ICE, Curacao's Gaming Control Board will be at N9-260 so stop by and say hello, tell them we sent you. A provocative defense of a forgotten Chinese approach to identity and difference. Historically, the Western encounter with difference has been catastrophic: the extermination and displacement of aboriginal populations, the transatlantic slave trade, and colonialism. China, however, took a different historical path. In Chinese Cosmopolitanism: The History and Philosophy of an Idea (Princeton UP, 2023), Shuchen Xiang argues that the Chinese cultural tradition was, from its formative beginnings and throughout its imperial history, a cosmopolitan melting pot that synthesized the different cultures that came into its orbit. Unlike the West, which cast its collisions with different cultures in Manichean terms of the ontologically irreconcilable difference between civilization and barbarism, China was a dynamic identity created out of difference.  The reasons for this, Xiang argues, are philosophical: Chinese philosophy has the conceptual resources for providing alternative ways to understand pluralism. Xiang explains that "Chinese" identity is not what the West understands as a racial identity; it is not a group of people related by common descent or heredity but rather a hybrid of coalescing cultures. To use the Western discourse of race to frame the Chinese view of non-Chinese, she argues, is a category error. Xiang shows that China was both internally cosmopolitan, embracing distinct peoples into a common identity, and externally cosmopolitan, having knowledge of faraway lands without an ideological need to subjugate them. Contrasting the Chinese understanding of efficacy--described as "harmony"--with the Western understanding of order, she argues that the Chinese sought to gain influence over others by having them spontaneously accept the virtue of one's position. These ideas from Chinese philosophy, she contends, offer a new way to understand today's multipolar world and can make a valuable contribution to contemporary discussions in the critical philosophy of race. For readers interested in how GCB and the Greek philosophical justification of GCB, domination, and destruction of barbarians still inform productions and consumptions of racist ideology as embodied in The Turner Diaries, see for example, here, here, and here.  Readers interested in the Vāda project that employs Indian epistemology to evaluate contemporary political claims, see here.  Jessica Zu is an intellectual historian and a scholar of Buddhist studies.

Let the mixture sit for 20-25 minutes, then strain through a fine mesh sieve. Use a spoon to press as much juice out of the berries as possible. Add the bourbon, lemon juice, Chambord and berry agave mixture to a large pitcher. Refrigerate for at least 4 hours.  Add sparkling water/club soda just before serving! You can either add it straight to the pitcher, or top off each individual drink.  Optional: garnish with fresh berries and mint. (TDCA), Vall d'Hebron Hospital Universitari, Universitat Autònoma de Barcelona, University of Texas MD Anderson Cancer Center, and Labcorp Drug Development report an updated analysis from a phase I study of the spleen tyrosine kinase (SYK) and FMS-like tyrosine kinase 3 inhibitor mivavotinib. They present data for the overall cohort of lymphoma patients and the subgroup of patients with diffuse large B-cell lymphoma (DLBCL), including an expanded cohort not included in the initial report. “Mivavotinib (TAK-659/CB-659) is an investigational, oral, reversible, potent dual inhibitor of spleen tyrosine kinase (SYK) and FMS-like tyrosine kinase 3 (FLT3) [18]. SYK is an essential component of the B-cell receptor signaling pathway; abnormal SYK signaling has been implicated in the pathogenesis of DLBCL and several other B-cell malignancies.” Patients with relapsed/refractory lymphoma for which no standard treatment was available received mivavotinib 60–120 mg once daily in 28-day cycles until disease progression/unacceptable toxicity. A total of 124 patients with lymphoma, including 89 with DLBCL, were enrolled. Overall response rates (ORR) in response-evaluable patients were 45% (43/95) and 38% (26/69), respectively. Median duration of response was 28.1 months overall and not reached in DLBCL responders. In subgroups with DLBCL of germinal center B-cell (GCB) and non-GCB origin, ORR was 28% (11/40) and 58% (7/12), respectively. Median progression free survival was 2.0 and 1.6 months in the lymphoma and DLBCL cohorts, respectively. Grade ≥3 treatment-emergent adverse events occurred in 96% of all lymphoma patients, many of which were limited to asymptomatic laboratory abnormalities; the most common were increased amylase (29%), neutropenia (27%), and hypophosphatemia (26%). “These findings support SYK as a potential therapeutic target for the treatment of patients with B-cell lymphomas, including DLBCL.” DOI: https://doi.org/10.18632/oncotarget.28352 Correspondence to: Leo I. A new research paper was published in Oncotarget's Volume 13 on November 17, 2022, entitled, “Mutation analysis performed on tumor biopsies from patients with newly-diagnosed germinal center aggressive B cell lymphomas.” Comprehensive genomic analyses of tumor biopsies from patients with newly-diagnosed germinal center B cell (GCB) diffuse large B cell/high grade B cell lymphoma (DLBCL/HGBL) have identified molecular subtypes predictive of inferior survival, which are characterized by somatic mutations that can be detected through clinical laboratory mutation analysis (CLMA). To determine the frequency and predictive value of individual genetic mutations associated with these experimentally-defined poor-risk subgroups, researchers Daniel J. Landsburg, Jennifer J.D. Morrissette, Stephen J. Schuster, Sunita D. Nasta, James N. Gerson, Stefan K. Barta, Jakub Svoboda, Elise A. Chong, and Megan S. Lim from the University of Pennsylvania reviewed the findings from CLMA performed on tumors from patients with newly-diagnosed GCB DLBCL/HGBL who were previously treated at the University of Pennsylvania. “Thus, we sought to analyze the results of CLMA performed at our institution on tumors from patients with newly-diagnosed GCB DLBCL/HGBL previously-treated with first line immunochemotherapy to determine the frequency and predictive value of the presence of individual genetic mutations associated with these experimentally-defined poor-risk subgroups.” CLMA was successfully performed on 58/59 patient tumor biopsies with a median turnaround time of 16 days, and 51 on which CLMA was routinely performed with adequate clinical follow-up were analyzed. Patients whose tumors demonstrated CREBBP mutation experienced a lower estimated rate of 2-year disease free survival (DFS) as compared to those whose tumors did not (45% [95% CI 18–68%] vs. 67% [95% CI 44–83%], P = 0.045). The researchers note that CREBBP mutations may be frequent and predict for inferior DFS in patients with newly-diagnosed GCB DLBCL/HGBL. Furthermore, CLMA may be practically-applied to translate experimental findings into those with more direct application to risk stratification and clinical trial design in subsets of patients with DLBCL/HGBL. “In conclusion, CLMA performed on tumor biopsies from patients with newly-diagnosed GCB DLBCL/HGBL revealed frequent mutations in CREBBP which were predicted to result in loss of function as well as a significantly lower rate of estimated DFS at 2 years.” DOI: https://doi.org/10.18632/oncotarget.28309 Correspondence to: Daniel J.
