For today’s installment of our 40 Days Meditation series, our brother Martin Steinbereithner provides commentary on Num 12:16-13:33. Listen below, download here, or search for Words from the Brothers on your favourite podcasting app. After that the people set out from Hazeroth, and camped in the wilderness of Paran. The Lord spoke to Moses, saying, “Send men to spy out the land of Canaan, which I am giving to the people of Israel. From each tribe of their fathers you shall send a man, every one a chief among them.” So Moses sent them from the wilderness of Paran, according to the command of the Lord, all of them men who were heads of the people of Israel. And these were their names: From the tribe of Reuben, Shammua the son of Zaccur; from the tribe of Simeon, Shaphat the son of Hori; from the tribe of Judah, Caleb the son of Jephunneh; from the tribe of Issachar, Igal the son of Joseph; from the tribe of Ephraim, Hoshea the son of Nun; from the tribe of Benjamin, Palti the son of Raphu; from the tribe of Zebulun, Gaddiel the son of Sodi; from the tribe of Joseph (that is, from the tribe of Manasseh), Gaddi the son of Susi; from the tribe of Dan, Ammiel the son of Gemalli; from the tribe of Asher, Sethur the son of Michael; from the tribe of Naphtali, Nahbi the son of Vophsi; from the tribe of Gad, Geuel the son of Machi. These were the names of the men whom Moses sent to spy out the land. And Moses called Hoshea the son of Nun Joshua. Moses sent them to spy out the land of Canaan and said to them, “Go up into the Negeb and go up into the hill country, and see what the land is, and whether the people who dwell in it are strong or weak, whether they are few or many, and whether the land that they dwell in is good or bad, and whether the cities that they dwell in are camps or strongholds, and whether the land is rich or poor, and whether there are trees in it or not. Be of good courage and bring some of the fruit of the land.” Now the time was the season of the first ripe grapes. So they went up and spied out the land from the wilderness of Zin to Rehob, near Lebo-hamath. They went up into the Negeb and came to Hebron. Ahiman, Sheshai, and Talmai, the descendants of Anak, were there. (Hebron was built seven years before Zoan in Egypt.) And they came to the Valley of Eshcol and cut down from there a branch with a single cluster of grapes, and they carried it on a pole between two of them; they also brought some pomegranates and figs. That place was called the Valley of Eshcol, because of the cluster that the people of Israel cut down from there. At the end of forty days they returned from spying out the land. And they came to Moses and Aaron and to all the congregation of the people of Israel in the wilderness of Paran, at Kadesh. They brought back word to them and to all the congregation, and showed them the fruit of the land. And they told him, “We came to the land to which you sent us. It flows with milk and honey, and this is its fruit. However, the people who dwell in the land are strong, and the cities are fortified and very large. And besides, we saw the descendants of Anak there. The Amalekites dwell in the land of the Negeb. The Hittites, the Jebusites, and the Amorites dwell in the hill country. And the Canaanites dwell by the sea, and along the Jordan.” But Caleb quieted the people before Moses and said, “Let us go up at once and occupy it, for we are well able to overcome it.” Then the men who had gone up with him said, “We are not able to go up against the people, for they are stronger than we are.” So they brought to the people of Israel a bad report of the land that they had spied out, saying, “The land, through which we have gone to spy it out, is a land that devours its inhabitants, and all the people that we saw in it are of great height. And there we saw the Nephilim (the sons of Anak, who come from the Nephilim), and we seemed to ourselves like grasshoppers, and so we seemed to them.” Num 12:16-13:33

On this episode of Translating Proteomics, hosts Parag Mallick and Andreas Huhmer of Nautilus Biotechnology discuss the challenges and opportunities of plasma proteomics. Their conversation focuses on:· Why blood plasma may be a good source of protein biomarkers· Current methodologies and pitfalls in plasma proteomics· The path forward for plasma proteomicsWhat is Plasma Proteomics? For those who are new to this topic, plasma is the liquid portion of the blood distinct from fractions containing red and white blood cells. Given the relatively non-invasive ways physicians can collect patient plasma, and the blood's intimate association with tissues throughout the body, plasma is potentially an excellent source of protein biomarkers. Yet, it is quite difficult to measure the levels of all plasma proteins because their concentrations span over 12 orders of magnitude. This episode features an in-depth discussion of the ways plasma proteomics efforts have and have not lived up to the promise of biomarker discovery and what we can do to advance plasma biomarker discovery efforts in the future.Chapters00:00 – 01:01 – Intro01:02 – 4:55 – What is the promise of plasma proteomics?04:55 – 07:23 – Is the plasma proteome really the best source of biomarkers?07:23 – 10:16 – How do proteins get into the blood and what are the implications for biomarker discovery?10:16 – 13:59 – Is it clear that proteins are the best candidates for blood biomarkers?13:59 – 19:57 – Advances in and the future of comprehensive plasma proteomics19:57 – 22:31 – Pros and cons of fractionating the plasma proteome to discover biomarkers22:31 – 28:14 – Progress in identifying multiomic plasma biomarkers and the path forward28:14 – End – OutroResourcesNano-omics: nanotechnology-based multidimensional harvesting of the blood-circulating cancerome (Gardner et al. 2022)o Review from focused on the development multiomics liquid biopsiesMulticompartment modeling of protein shedding kinetics during vascularized tumor growth (Machiraju et al. 2020)o Work from Parag's Lab investigating tumor protein sheddingSimulation of the Protein-Shedding Kinetics of a Fully Vascularized Tumor (Frieboes et al. 2015)o Tumor protein shedding work from Parag's LabMathematical model identifies blood biomarker-based early cancer detection strategies and limitations (Hori and Gambhir et al. 2011)o Study modeling how much protein could be shed and detected from different size tumorsThe human plasma proteome: history, character, and diagnostic prospects (Anderson and Anderson 2002)o Review discussing...

Better late than never, right? Nintendo has unveiled its latest accessory to the Switch: a charging stand for your Joy-Con. This would've been great...maybe at launch? And it's not like the Switch was in desperate need for a charging stand—Hori makes a pretty good one AND it's an officially licensed Nintendo product. The kicker? The Hori stand charges four Joy-Con at once, while Nintendo's only does two. In other news, Sony may have accidentally leaked the latest Helldivers 2 Warbond themselves, and Splitgate, a cult favorite from the late-PS4 era, is getting a sequel.

Big Idea: Jesus always delivers on his promises even when we don't deserve it. In the Beginning: Jacob Genesis 36:1-43 I. You will be fruitful and multiply. 1-8 These are the family records of Esau (that is, Edom). Esau took his wives from the Canaanite women: Adah daughter of Elon the Hethite, Oholibamah daughter of Anah and granddaughter of Zibeon the Hivite, and Basemath daughter of Ishmael and sister of Nebaioth. Adah bore Eliphaz to Esau, Basemath bore Reuel, and Oholibamah bore Jeush, Jalam, and Korah. These were Esau's sons, who were born to him in the land of Canaan. Esau took his wives, sons, daughters, and all the people of his household, as well as his herds, all his livestock, and all the property he had acquired in Canaan; he went to a land away from his brother Jacob. For their possessions were too many for them to live together, and because of their herds, the land where they stayed could not support them. So Esau (that is, Edom) lived in the mountains of Seir. II. You will father many nations. 9-19 These are the family records of Esau, father of the Edomites in the mountains of Seir. These are the names of Esau's sons: Eliphaz son of Esau's wife Adah, and Reuel son of Esau's wife Basemath. The sons of Eliphaz were Teman, Omar, Zepho, Gatam, and Kenaz. These are the sons of Esau's wife Basemath. These are the sons of Esau's wife Oholibamah daughter of Anah and granddaughter of Zibeon: She bore Jeush, Jalam, and Korah to Edom. These are the chiefs among Esau's sons: the sons of Eliphaz, Esau's firstborn: chief Teman, chief Omar, chief Zepho, chief Kenaz, chief Korah, chief Gatam, and chief Amalek. These are the chiefs descended from Eliphaz in the land of Edom. These are the sons of Adah. These are the sons of Reuel, Esau's son: chief Nahath, chief Zerah, chief Shammah, and chief Mizzah. These are the chiefs descended from Reuel in the land of Edom. These are the sons of Esau's wife Basemath. These are the sons of Esau's wife Oholibamah: chief Jeush, chief Jalam, and chief Korah. These are the chiefs descended from Esau's wife Oholibamah daughter of Anah. These are the sons of Esau (that is, Edom), and these are their chiefs. III. Seir, who? 20-30 These are the sons of Seir the Horite, the inhabitants of the land: Lotan, Shobal, Zibeon, Anah, Dishon, Ezer, and Dishan. These are the chiefs among the Horites, the sons of Seir, in the land of Edom. The sons of Lotan were Hori and Heman. Timna was Lotan's sister. These are Shobal's sons: Alvan, Manahath, Ebal, Shepho, and Onam. These are Zibeon's sons: Aiah and Anah. This was the Anah who found the hot springs in the wilderness while he was pasturing the donkeys of his father Zibeon. These are the children of Anah: Dishon and Oholibamah daughter of Anah. These are the chiefs among the Horites, clan by clan, in the land of Seir. IV. Kings (and chiefs) will come from you. 31-43 These are the kings who reigned in the land of Edom before any king reigned over the Israelites: Bela son of Beor reigned in Edom; the name of his city was Dinhabah. When Bela died, Jobab son of Zerah from Bozrah reigned in his place. When Jobab died, Husham from the land of the Temanites reigned in his place. When Husham died, Hadad son of Bedad reigned in his place. He defeated Midian in the field of Moab; the name of his city was Avith. When Hadad died, Samlah from Masrekah reigned in his place. If you enjoy this show and would like to help me spread the word about it, or support it financially, you can find out more at nuancespod.com/support GUEST BIO Prof. Sachi Schmidt-Hori is interested in investigating how gender, sexuality, corporeality, and power are represented and negotiated in pre-seventeenth-century Japanese narratives and illustrations. Her first book, Tales of Idolized Boys: Male-Male Love in Medieval Japanese Narratives (University of Hawai`i Press, 2021) is on medieval chigo monogatari (Buddhist acolyte tales), which often depict romantic relationships between Buddhist priests and adolescent boys. These tales challenge a host of normative and moral standards we (academics, especially) internalize, including such ideas as "sexual orientation," "transgenerational sex," and "sexual agency." Her current project is on the literary representations of "milk kinships" in pre-seventeenth Japanese tales, in cluding the Tale of Ochikubo, the Tale of Genji, the Tales of the Heike. She is also the director of the Springboard Japan Project, a multi-purpose open-access forum for Japan Studies. Web MENTIONED YOASOBI (J-Pop duo) The Tale of Ochikubo (Ochikubo Monogatari) The Tale of Genji (Genji Monogatari) Tales of Idolized Boys: Male-Male Love in Medieval Japanese Buddhist Narratives TAKEAWAYS The equation of your career/life choices is for you to solve. It doesn't have to make sense to anyone else if it makes sense for you. You can seek out spaces where being you is a plus instead of a minus. Don't settle. If you're not a white man, emulating white men probably won't help. Be goofy :-) Language shapes our discussions. Identity is seen as static, whereas actions are choices that we could change in the future, not immutable character traits. How integrity is perceived is one of many things that can vary widely outside the West. In Japan, parenting is about taming the child's ego. In the U.S., parenting is about nurturing the child's ego. Study Passage: Genesis 35:16-37:116 Then they journeyed from Bethel. When they were still some distance from Ephrath, Rachel went into labor, and she had hard labor. 17 And when her labor was at its hardest, the midwife said to her, “Do not fear, for you have another son.” 18 And as her soul was departing (for she was dying), she called his name Ben-oni; but his father called him Benjamin. 19 So Rachel died, and she was buried on the way to Ephrath (that is, Bethlehem), 20 and Jacob set up a pillar over her tomb. It is the pillar of Rachel's tomb, which is there to this day. 21 Israel journeyed on and pitched his tent beyond the tower of Eder. 22 While Israel lived in that land, Reuben went and lay with Bilhah his father's concubine. And Israel heard of it. Now the sons of Jacob were twelve. 23 The sons of Leah: Reuben (Jacob's firstborn), Simeon, Levi, Judah, Issachar, and Zebulun. 24 The sons of Rachel: Joseph and Benjamin. 25 The sons of Bilhah, Rachel's servant: Dan and Naphtali. 26 The sons of Zilpah, Leah's servant: Gad and Asher. These were the sons of Jacob who were born to him in Paddan-aram. 27 And Jacob came to his father Isaac at Mamre, or Kiriath-arba (that is, Hebron), where Abraham and Isaac had sojourned. 28 Now the days of Isaac were 180 years. 29 And Isaac breathed his last, and he died and was gathered to his people, old and full of days. And his sons Esau and Jacob buried him. 36 These are the generations of Esau (that is, Edom). 2 Esau took his wives from the Canaanites: Adah the daughter of Elon the Hittite, Oholibamah the daughter of Anah the daughter of Zibeon the Hivite, 3 and Basemath, Ishmael's daughter, the sister of Nebaioth. 4 And Adah bore to Esau, Eliphaz; Basemath bore Reuel; 5 and Oholibamah bore Jeush, Jalam, and Korah. These are the sons of Esau who were born to him in the land of Canaan. 6 Then Esau took his wives, his sons, his daughters, and all the members of his household, his livestock, all his beasts, and all his property that he had acquired in the land of Canaan.
