I first learned about cryonics when I read Eliezer and Robin's posts about it on Overcoming Bias years ago. I got cryopilled. Somewhat amazingly to me, I'm now a researcher in this field. So I thought this community might be interested to know that I was one of several co-authors on a paper just published in Frontiers in Medical Technology, titled "Structural brain preservation: a potential bridge to future medical technologies". In this paper, we propose reframing cryonics as a type of structural brain preservation, focusing on maintaining the brain's physical structure that encodes memories and personality, rather than making the focus about low-temperature storage per se. We explore what brain structures likely need to be preserved to retain long-term memories and other valued aspects of personal identity. We then review different methods of brain preservation, including cryopreservation, aldehyde-stabilized cryopreservation, fluid preservation, and fixation followed by polymer embedding. The paper also discusses the two most commonly discussed potential future revival technologies, i.e. molecular nanotechnology and whole brain emulation. We argue that this structural preservation framing may be more technically grounded and agreeable to mainstream researchers than some of the traditional ways that cryonics has been discussed. As a personal reflection here, I want to briefly discuss the idea of fluid preservation, which is one topic discussed in our review paper. I remember first reading about this idea in approximately 2017 on a cryonics mailing list. Even though I was already sold on the idea of aldehyde-stabilized cryopreservation -- using fixation as a short-term bridge to cryoprotection and cryopreservation, I remember thinking that the idea of simply leaving the brain in fixative solution for the long-term was bizarre and outlandish. Around 2020-2022, I spent a good amount of time researching different options for higher temperature (and thus lower cost) brain preservation. Mostly I was looking into different methods for embedding fixed brain tissue in polymers, such as paraffin, epoxy, acrylates, or silicon. I also studied the options of dehydrated preservation and preserving the fixed brain in the fluid state, which I was mostly doing for the sake of completeness. To be clear, I certainly don't want to make it seem like this was a lone wolf effort or anything. I was talking about the ideas with friends and it was also in the zeitgeist. For example, John Smart wrote a blog post in 2020 about this, titled "Do we need a noncryogenic brain preservation prize?" (There still is no such prize.) In 2022, I was reading various papers on brain preservation (as one does), when I came across Rosoklija 2013. If I recall correctly, I had already seen this paper but was re-reading it with a different eye. They studied human and monkey brain tissue that had been preserved in formalin for periods ranging from 15 months to 55 years, using the Golgi-Kopsch silver staining method to visualize neuronal structures. They reported that even after 50 years of formalin fixation at room temperature, the method yielded excellent results. In particular, they had this figure: That's a picture showing well-impregnated neurons with preserved dendritic spines. Looking at this picture was a viewquake for me. I thought, if fluid preservation can preserve the structure of the 1-5% of cells that are stained by the Golgi-Kopsch method, why not other cells? And if it can work in this one part of the brain, why not the whole brain? And if it can do it for 50 years, why not 100 or 150? This is exploratory investigation of a new-ish hypothesis, it is not intended to be a comprehensive review of the field or even a a full investigation of the hypothesis. I've always been skeptical of the seed-oil theory of obesity. Perhaps this is bad rationality on my part, but I've tended to retreat to the sniff test on issues as charged and confusing as diet. My response to the general seed-oil theory was basically "Really? Seeds and nuts? The things you just find growing on plants, and that our ancestors surely ate loads of?" But a twitter thread recently made me take another look at it, and since I have a lot of chemistry experience I thought I'd take a look. The PUFA Breakdown Theory It goes like this: PUFAs from nuts and seeds are fine. Deep-frying using PUFAs causes them to break down in a way other fatty acids do not, and these breakdown products are the problem. Most of a fatty acid is the "tail". This consists of hydrogen atoms decorating a backbone of carbon atoms. Each carbon atom can make up to four bonds, of which two have to be to other carbons (except the end carbon which only bonds to one carbon) leaving space for two hydrogens. When a chain has the maximum number of hydrogen atoms, we say it's "saturated". These tails have the general formula CnH2n+1: For a carbon which is saturated (i.e. has four single bonds) the bonds are arranged like the corners of a tetrahedron, and rotation around single bonds is permitted, meaning the overall assembly is like a floppy chain. Instead, we can have two adjacent carbons form a double bond, each forming one bond to hydrogen, two bonds to the adjacent carbon, and one to a different carbon: Unlike single bonds, double bonds are rigid, and if a carbon atom has a double bond, the three remaining bonds fall in a plane. This means there are two ways in which the rest of the chain can be laid out. If the carbons form a zig-zag S shape, this is a trans double bond. If they form a curved C shape, we have a cis double bond. The health dangers of trans-fatty acids have been known for a long while. They don't occur in nature (which is probably why they're so bad for us). Cis-fatty acids are very common though, especially in vegetable and, yes, seed oils. Of course there's no reason why we should stop at one double bond, we can just as easily have multiple. This gets us to the name poly-unsaturated fatty acids (PUFAs). I'll compare stearic acid (SA) oleic acid (OA) and linoleic acid (LA) for clarity: Linoleic acid is the one that seed oil enthusiasts are most interested in. We can go even further and look at α-linoleic acid, which has even more double bonds, but I think LA makes the point just fine. Three fatty acids, usually identical ones, attach to one glycerol molecule to form a triglyceride. Isomerization As I mentioned earlier, double bonds are rigid, so if you have a cis double bond, it stays that way. Mostly. In chemistry a reaction is never impossible, the components are just insufficiently hot. If we heat up a cis-fatty acid to a sufficient temperature, the molecules will be able to access enough energy to flip. The rate of reactions generally scales with temperature according to the Arrhenius equation: v=Aexp(EakBT) Where A is a general constant determining the speed, Ea is the "activation energy" of the reaction, T is temperature, and kB is a Boltzmann's constant which makes the units work out. The studies created much of the evidentiary foundation now on the academic record about the still very young field of pediatric gender medicine today. The publicity surrounding his research in turn served as the impetus for the governmental inquiry into that country's only pediatric gender clinic, culminating in the issuance of an order shuttering the Tavistock Gender Identity Development Service in July of 2022. Biggs' studies revealed, among other things, that a longitudinal study launched by the Tavistock into the effects of puberty blockers on gender dysphoric youth had resulted in increasing levels of depression and suicidality in those whose psychic distress the drugs were ostensibly being used to relieve. The service hid the results that proved that the ongoing practice was harmful on balance and continued undeterred by evidence with the dramatic expansion of the practice outside of the research context that was its initial pretext to be permitted at all. In other words, Biggs had proven that the Tavistock knew it was doing harm when it practiced “gender affirming care.” Thank you for reading Year Zero. This post is public so feel free to share it.Biggs began his investigations into gender ideology soon after being admonished by an American student in a graduate seminar he led that featured discussion of a Guardian article on the rising number of transgender identified youth. “Things were said in that discussion that should not have been have been said,” Biggs recalled the student warning him. “That was first my encounter with this kind of student woke activism, saying that we shouldn't discuss things… As somebody whose job is to look into things and look into truth and look into reality, it's my job to probe that. If you tell me I can't look into something, that's when I want to look into it.” It did not take long for Biggs to discover what was being hidden behind the demands not to look — a vast complex of spurious dogma and corrupted research that did not survive reasoned scrutiny. He has swiftly set about dismantling this corpus in dryly understated but ruthlessly unsparing prose. I met with Biggs at the Genspect: the Bigger Picture conference in Killarney, Ireland in April, where he gave a presentation on his puberty blocker research. I had a wide-ranging conversation with him about the trajectory that brought him into the gender issue and the wider sea change in attitudes enabling the ongoing takeover of academia by acolytes of the ongoing non-electoral politics of institutional capture that we refer to at Year Zero as the Successor Ideology.As always, a full transcript will be made available to paid subscribers. A novel lethal infectious neurological disease emerged in American deer a few decades ago. Since then, it's spread rapidly across the continent. In areas where the disease is found, it can be very common in the deer there. Chronic wasting disease isn't caused by a bacteria, virus, protist, or worm – it's a prion, which is a little misshapen version of a protein that occurs naturally in the nervous systems of deer. Chemically, the prion is made of exactly the same stuff as its regular counterpart – it's a string of the same amino acids in the same order, just shaped a little differently. Both the prion and its regular version (PrP) are monomers, single units that naturally stack on top of each other or very similar proteins. The prion's trick is that as other PrP moves to stack atop it, the prion reshapes them – just a little – so that they also become prions. These chains of prions are quite stable, and, over time, they form long, persistent clusters in the tissue of their victims. We know of only a few prion diseases in humans. They're caused by random chance misfolds, a genetic predisposition for PrP to misfold into a prion, accidental cross-contamination via medical supplies, or, rarely, from the consumption of prion-infected meat. Every known animal prions is a misfold of the same specific protein, PrP. PrP is expressed in the nervous system, particularly in the brain – so infections cause neurological symptoms and physical changes to the structure of the brain. Prion diseases are slow to develop (up to decades), incurable, and always fatal. There are two known infectious prion diseases in people. One is kuru, which caused an epidemic among tribes who practiced funerary cannibalism in Papua New Guinea. The other is mad cow disease, also known as bovine spongiform encephalopathy (BSE) AKA Variant Creutzfeldt-Jakob disease, which was first seen in humans in 1996 in the UK, and comes from cows. Chronic wasting disease (CWD). Is, like every other animal prion disease, a misfold of PrP. PrP is quite similar in both humans and deer. Is found in multiple deer species which are commonly eaten by humans. Can be carried in deer asymptomatically. But it doesn't seem to infect people. Is it ever going to? If a newly-emerged virus were sweeping across the US and killing deer, which could be spread through consuming infected meat, I would think “oh NO.” I'd need to see very good evidence to stop sounding the alarm. Now, the fact that it's been a few decades, and it hasn't spread to humans yet, is definitely some kind of evidence about safety. But are we humans basically safe from it, or are we living on borrowed time? If you live in an area where CWD has been detected, should you eat the deer? Sidenote: Usually, you'll see “BSE” used for the disease in cows and “VCJ” for the disease in humans. But they're caused by the same agent and this essay is operating under a zoonotic One Health kind of stance, so I'm just calling the disease BSE here. (As well as the prion that causes it, when I can get away with it.) In short The current version of CWD is not infectious to people. We checked. BSE showed that prions can spill over, and there's no reason a new CWD variant will never do the same. The more cases there are, the more likely it is to spill over. That said, BSE did not spill over very effectively. It was always incredibly rare in humans. It's an awful disease to get, but the chance of getting it is tiny. Prions in general have a harder time spilling over between species than viruses do. CWD might behave somewhat differently but probably will stay hampered by the species barrier. In some health and fitness circles, where ascorbic acid is considered a "synthetic form of vitamin C," influencers and some nutritionists warn their followers of the dangers of ascorbic acid. But is it really as bad as they claim, or like many things in nutrition, are they misinformed? I'll try to set the record straight, covering many of the reasons some people warn against using ascorbic acid, and also covering its benefits and why I use and recommend it. If you have thoughts or questions I don't cover, share them in the comments area. What is Ascorbic Acid? You may have heard the term "ascorbic acid," but what exactly is it? Ascorbic acid is a water-soluble vitamin, commonly known as vitamin C. Chemically speaking, it is an organic compound with the molecular formula C₆H₈O₆. This essential nutrient plays a crucial role in the functioning of our bodies. It acts as an antioxidant, protecting cells from harmful free radicals, which are unstable molecules that can cause damage. Ascorbic acid also plays a vital role in collagen synthesis, a protein that supports the structure and health of our skin, bones, and connective tissues. One study published in the Journal of the American College of Nutrition found that adequate vitamin C intake is associated with a reduced risk of chronic diseases, such as cardiovascular disease and certain types of cancer.Johnston, C. S., et al. (2014). Vitamin C supplementation slightly improves physical activity levels and reduces cold incidence in men with marginal vitamin C status: A randomized controlled trial. Journal of the American College of Nutrition, 33(5), 417-424. Another study published in Nutrients emphasized the role of ascorbic acid in enhancing immune function and supporting the body's defense against pathogens.Carr, A. C., et al. (2017). Vitamin C and immune function. Nutrients, 9(11), 1211. What is the difference between ascorbic acid and vitamin C? The terms "ascorbic acid" and "vitamin C" are often used interchangeably, but they're not the same. All ascorbic acid is vitamin C, but not all vitamin C is ascorbic acid. Ascorbic acid is a specific form of vitamin C. Chemically, ascorbic acid is the purest form of vitamin C, with the molecular formula C₆H₈O₆ I mentioned above. It is the active and most well-known component of vitamin C. However, vitamin C is a broader term that encompasses not only ascorbic acid but also other related compounds with vitamin C activity. These compounds include dehydroascorbic acid, ascorbyl palmitate, and various mineral ascorbates, which are salts of ascorbic acid. While ascorbic acid is the most common and readily available form of vitamin C, "vitamin C" describes the presence and activity of all the compounds. They contribute to the overall vitamin C activity and have similar functions and benefits as ascorbic acid itself. Different forms of vitamin C can be found in a variety of food sources.
