I'm Stacey Simms and every other Friday I bring you a short episode with the top diabetes stories and headlines happening now. XX   The understanding of type 1 has changed a lot in the last 10-15 years, specifically with the definition of stages of the disease. Now some researchers suggest the current 2015 model is ready for another update. University of Florida Health diabetes researchers and collaborators last week issued a call to action in The Lancet Diabetes & Endocrinology, urging the development of a more precise method of calculating type 1 diabetes risk. Under the current staging system, this is Stage 1 of the disease, when a patient does not yet have elevated blood sugar but has two or more of these autoantibodies in the blood. Stage 2 is diagnosed when blood sugar begins to rise, indicating a dysfunctional pancreas in a patient who does not yet have clinical type 1 diabetes. That comes in Stage 3 with high blood sugar and, eventually, the classic symptoms of the disease, including frequent urination, weight loss and fatigue. Jacobsen, however, said the staging system requires more precision. This is especially important as the current staging model determines if a patient is eligible to receive a drug therapy called teplizumab, the only Food and Drug Administration-approved medication that can delay the onset of type 1 diabetes by an average of two years. The staging system, however, does not include a patient's age and these researchers say additional markers that would be important to consider include a greater array of abnormal metabolic measures beyond blood sugar and the presence of more autoantibodies.   https://www.gainesville.com/story/news/healthcare/2024/12/04/uf-health-scientists-propose-update-to-type-1-diabetes-staging-system/76734459007/ XX A product called InsuJet gets approval in Canada. Made by NuGen Medical Devices, InsuJet™ is a needle-free injection system. It's already approved for sale in 42 countries – not yet the US. Their website says InsuJet's jet-injected fluid penetrates the skin through a small orifice of a special nozzle, creating a thin, high pressure stream. This way, insulin doesn't remain around the place of injection, like it does with a needle. Instead, with InsuJet, the insulin is deposited into the subcutaneous tissue where it's easily absorbed and acts faster– saving you money, time, and discomfort.   https://finance.yahoo.com/news/nugen-announces-canada-wide-launch-221500674.html XX Individuals diagnosed with type 2 diabetes before the age of 50 may be at a higher risk of developing dementia later in life. A study led by researchers at NYU Rory Meyers College of Nursing, says the risk is notably greater among adults with obesity. Type 2 diabetes is a known risk factor for dementia. Although the underlying mechanisms are not fully understood, scientists think that some of the hallmarks of diabetes—such as high blood sugar, insulin resistance, and inflammation—may encourage the development of dementia in the brain.     The researchers note that this greater understanding of the connection between diabetes onset, obesity, and dementia may help inform targeted interventions to prevent dementia. https://scitechdaily.com/diagnosed-young-with-diabetes-your-dementia-risk-may-be-higher/ XX Embecta is out of the insulin patch pump business – just months after getting the ok from the FDA. Embecta received FDA clearance for its insulin patch pump in September. The device is indicated for people with Type 1 or Type 2 diabetes and includes a 300-unit insulin reservoir to support people with higher daily insulin needs. However, the version Embecta received clearance for was an open-loop patch pump, where users manually control insulin delivery, while the firm's goal was to develop a closed-loop version, which automatically delivers insulin when needed. The CEO also expects competition in closed-loop devices for people with Type 2 diabetes to intensify, with Insulet receiving FDA clearance for its Omnipod 5 patch pump for Type 2 diabetes in August. https://www.medtechdive.com/news/embecta-discontinue-patch-pump/734059/ XX Sanofi invests in a company developing a new drug to prevent hypoglycemia. We reported on Zucara Therapeutics back in 2022 and they've been moving ahead with ZT-01. ZT-01 is a medication being developed to help prevent dangerous low blood sugar (hypoglycemia) by fixing how the body normally fights it. In people without diabetes, a hormone called glucagon is released to raise blood sugar when it gets too low. But in people with insulin-dependent diabetes, like type 1 diabetes or some forms of type 2 diabetes, this process doesn't work well because another hormone, somatostatin (SST), blocks glucagon from being released. ZT-01 works by blocking the effects of somatostatin, allowing the body to release more glucagon when it's needed. They published their findings Dec. 2 in the journal Diabetologia. The new research followed on data from other studies that had suggested that people with especially low levels of vitamin D might face a higher risk for diabetes. That was true for people who were already prediabetic, especially. To find out, researchers led by Jirki Vyrtanen, from the University of Eastern Finland, randomly assigned nearly 2,300 people aged 60 or older to receive either placebo pills or 40 or 80 micrograms of vitamin D3 supplements per day, for five years.  At the end of the five years, "105 participants developed type 2 diabetes: 38 in the placebo group, 31 in the group receiving 40 micrograms of vitamin D3 per day, and 36 in the group receiving 80 micrograms of vitamin D3 per day," according to a university news release. In other words, there were no significant differences in the rate at which people developed type 2 diabetes, regardless of how much supplemental vitamin D they took. Vytanen's team also saw no benefit from high-dose supplements on a person's blood sugar or insulin levels, or measures of overweight/obesity. Carl Ichan - Hand Slap - Carl Icahn and his publicly traded company Icahn Enterprises settled with the SEC over allegedly failing to disclose billions worth in stock-backed borrowing. - Icahn and IELP will pay a combined $2 million in fines, without admitting or denying wrongdoing, over the failure to disclose as much as $5 billion in margin loans that were backed by Icahn's stake in the company. - Icahn, a well-known activist investor, had been facing pressure from another activist short seller in 2023. (Hindenburg which is still short the shares) Circle K 11? - Canada's Alimentation Couche-Tard (Circle K) has sounded out Japan's Seven & i (7-Eleven) about a potential takeover, the two companies said on Monday, making the 7-Eleven owner the largest-ever Japanese target of a foreign buyout. - While the value of the offer has not been disclosed, the bid is the latest example of the growing interest in Japanese companies by Western investors, who have been drawn by the country's push for better governance. - News of the deal sent shares of Seven & i surging by almost 23% in Tokyo Chicken and the Egg - Chicken prices are going up - Perdue Foods is recalling more than 167,000 pounds of frozen chicken nuggets and tenders after some customers reported finding metal wire embedded in the products. - According to Perdue and the U.S. Agriculture Department's Food Safety and Inspection Service, the recall covers select lots of three product...

Our big story this week is from AMD. They're opening up their wallet to the tune of nearly $5 billion to buy ZT Systems. The two companies have had a preexisting partnership, with ZT having collaborated on the EPYC processor lines. ZT's biggest customers are AWS and Azure, as ZT specializes in hyperscale AI systems that are bought by the rack. This move follows a very recent acquisition of Silo AI, which we covered on the Rundown, as well as their last big acquisition of Xilinx. AMD wil met een overname ter waarde van bijna vijf miljard dollar een slag slaan in de strijd tegen Nvidia. Joe van Burik vertelt erover in deze Tech Update. AMD, oftewel Advanced Micro Devices, wil voor omgerekend 4,4 miljard euro ZT Systems overnemen, dat zich specialiseert in dataclusters voor bedrijven. Dat betekent dat bedrijven die AI-systemen ontwikkelen en uitrollen, dat zo sneller kunnen doen. I'm Stacey Simms and these are the top diabetes stories and headlines happening now XX In the news is brought to you by the T1D Exchange, make your voice heard and drive research that matters XX Buckle up there is a LOT going on XX Medtronic's Minimed 780G AID system gets FDA approval. This is for people aged 7 years and older who have type 1 diabetes. Medtronic will begin taking preorders on May 15, 2023. Users of the current MiniMed 770G will be eligible for free remote software upgrades. The 780G is currently available in 105 countries. It has been available in Europe since 2020 and in the UK since 2021. The biggest difference from other systems is that it can automatically administer bolus correction insulin doses every 5 minutes. It will also automatically adjust basal insulin The insulin pump's infusion set can be worn for 7 days, rather than 3 days as with the older system, and the glucose target level can be set as low as 100 mg/dL. In the pivotal US trial, overall, patients who used the 780G spent 75% of the time in ideal glucose range (70-180 mg/dL) and 1.8% of the time below that range. Overnight, the figures were 82% and 1.5%, respectively. With the glucose target set at 100 mg/dL and active insulin time set to 2 hours, patients spent 78.8% of time in range without increased hyperglycemia. https://www.medscape.com/viewarticle/991091 XX U.S. Food and Drug Administration (FDA) lifted Medtronic's warning letter sent in December 2021. The resolution of the warning letter follows ongoing remediation actions from the company and proactive actions to continue to strengthen its quality systems. Insulet says “Omnipod GO was designed to serve the more than three million people using basal insulin or transitioning to insulin therapy to treat their type 2 diabetes” Omnipod GO is a standalone system that provides a fixed rate of continuous rapid-acting insulin for 72 hours. It's tubeless and waterproof just like a regular Omnipod but it's offered in seven different pre-programmed daily rates, ranging from 10 to 40 units per day, and operates without the need for a handheld device to control the Pod. It has been cleared for use with the following U-100 insulins: NovoLog®, Fiasp®, Humalog®, Admelog®, and Lyumjev®. The product was developed to serve people with type 2 diabetes earlier in their treatment journey by starting them on Pod therapy for their insulin delivery, rather than daily injections. If a patient becomes insulin-intensive, meaning they require both basal and bolus insulin, the transition to another Omnipod product would be seamless. Positive results could further extend the company's reach across the global diabetes community, she added. The company also intends to utilize study data to submit for an integrated CGM (iCGM) designation in 2023. Senseonics holds an FDA investigational device exemption (IDE) to expand the trial to pediatric patients. Amphastar Pharmaceuticals is buying the Glucagon nasal spray from Lilly in a deal worth up to $1.08 billion dollars. Launched in 2019, Baqsimi saw worldwide sales worth $139.3 million in 2022, the companies said. You may remember that Lilly had acquired Baqsimi from Locemia Solutions in 2015. If approved, the drug would be the first therapy to specifically address low blood sugar at night. We've talked about this.. Zucara Therapeutics is testing an oral medication currently known as ZT-01 will attempt to restore the body's natural ability to respond to low blood sugar levels. Researchers will evaluate the ability of the drug to blunt hypoglycemia caused by insulin in type 1 diabetes. This phase 2 clinical trial follows positive results from the company's earlier phase 1 study which included 18 participants with type 1 diabetes who received the treatment. Of these 18, 16 (89%) had a meaningful increase in glucagon production after being given ZT-01, with no serious health events during the trial. https://diatribe.org/drug-prevent-nighttime-lows-approved-enter-clinical-trials XX The T1D Exchange Registry is a research study conducted online over time, designed to foster innovation and improve the lives of people with T1D. The registry aims to improve knowledge of T1D, accelerate the discovery and development of new treatments and technologies, and generate evidence to support policy or insurance changes that help the T1D community. By sharing opinions, experiences and data, patients can help advance meaningful T1D treatment, care and policy. XX Australian scientists have designed a new way to swallow insulin, in a pill form. The design also has potential uses for delivering other protein drugs, such as antibiotics and cancer treatments. Insulin is made of smaller versions of proteins called peptides. Previous attempts to develop an orally administered insulin have found that the severe pH levels in the gastrointestinal tract degrade the peptides, causing the drug to lose its function. To get around this problem, the scientists encapsulated the insulin in a lipid-based nanomaterial placed inside an enteric capsule. In animal studies, long acting insulin was absorbed better than fast acting.
