Mission encre noire Tome 37 Chapitre 403. J'ai couru, j'ai cru me perdre. I'm beautiful in my way /'Cause God Makes no mistakes / I'm on the right track, baby / I was born this way / Don't hide yourself in regret / Just love yourself and you're set (...) Express Yourself / So you can respect yourself ».

W., Landman, B. A., Gore, J. C. Abstract: Recent studies have revealed the production of time-locked blood oxygenation-level dependent (BOLD) functional MRI (fMRI) signals throughout the entire brain in response to a task, challenging the idea of sparse and localized brain functions, and highlighting the pervasiveness of potential false negative fMRI findings. In these studies, "whole-brain" refers to gray matter regions only, which is the only tissue traditionally studied with fMRI. However, recent reports have also demonstrated reliable detection and analyses of BOLD signals in white matter which have been largely ignored in previous reports. Here, using model-free analysis and simple tasks, we investigate BOLD signal changes in both white and gray matters. We aimed to evaluate whether white matter also displays time-locked BOLD signals across all structural pathways in response to a stimulus. We find that both white and gray matter show time-locked activations across the whole-brain, with a majority of both tissue types showing statistically significant signal changes for all task stimuli investigated. We observed a wide range of signal responses to tasks, with different regions showing very different BOLD signal changes to the same task. Moreover, we find that each region may display different BOLD responses to different stimuli. Overall, we present compelling evidence that the whole brain, including both white and gray matter, show time-locked activation to multiple stimuli, not only challenging the idea of sparse functional localization, but also the prevailing wisdom of treating white matter BOLD signals as artefacts to be removed. Makoto Rheault-Kihara leads user acquisition at Hopper, one of the fastest-growing travel apps with over 60 million in sales to date. Makoto was previously the Director of Marketing at Cruisehub, an OTA aiming to modernize the cruise industry and lead search engine marketing at Busbud, a bus travel booking platform in 80 countries. Hopper differentiates its paid social strategy by focusing on user acquisition AND long-term trust through investing in paid social and other mobile ad channels. They use Facebook as the primary focus of their paid social ads with TikTok as a growing secondary channel. Paid social is becoming a lot more abstract and therefore more creative. Makoto believes paid TikTok ads must be kept fresh, platform-focused, and generated at a higher frequency. The pandemic did affect the travel industry but Hopper still achieved a 110% YoY growth last year and is on track to do over $1B of business this year. They did this by following a diversification strategy based on customer needs and expanded into hotels and car rentals. Hopper's customer-obsessed culture and self-contained, autonomous teams have helped them grow multiple product arms simultaneously. A weekly catch-up call and an executive team focused on cross-team communication help these siloed teams at Hopper learn from each other's challenges and successes. Sometimes ads that marketers like don't appeal to actual customers and may end up performing worse than some simple customer-oriented ads. Over time, Hopper moved to these customer-centric ad creatives which helped them reduce expensive failures and triple their RPM (revenue per 1000 impressions). At Hopper, Makoto focuses on maximizing the overall contribution margin of a cohort and on-target rate to check their ability to drive revenue from specific target purchases or user actions. Because Hopper diversified across three generic categories (hotels, flights, car rentals), their ad targeting still focuses on great creatives and has not changed much since IDFA deprecation in iOS 14.5. Interested in joining the Hopper team? Click here to check out their open positions! https://www.hopper.com/careers/
Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2022.12.01.518514v1?rss=1 Authors: Edde, M., Theuad, G., Dumont, M., Theberge, A., Valcourt Caron, A., Gilbert, G., Houde, J.-C., Maltais, L., Rheault, F., Spagnolo, F., Barakovic, M., Magon, S., Descoteaux, M. Abstract: Assessing the consistency of quantitative MRI measurements is critical for inclusion in longitudinal studies and clinical trials. Intraclass coefficient correlation and coefficient of variation were used to evaluate the different consistency aspects of diffusion- and myelin-based MRI measures. Multi-shell diffusion and inhomogeneous magnetization transfer datasets were collected from twenty healthy adults at a high-frequency of five MRI sessions. The consistency was evaluated across whole bundles and the track-profile along the bundles. The impact of the fiber populations on the consistency was also evaluated using the number of fiber orientations map. For whole and profile bundles, moderate to high reliability of diffusion and myelin measures were observed. We report higher reliability of measures for multiple fiber populations than single. The overall portrait of the most consistent measurements and bundles drawn from a wide range of MRI techniques presented here will be particularly useful for identifying reliable biomarkers capable of detecting, monitoring and predicting white matter changes in clinical applications and has the potential to inform patient-specific treatment strategies. This week's guest is No Trigger vocalist Tom Rheault. Though the long-running Massachusetts melodic punk project doesn't put out albums very often, they really make it count when they do. The 20-year vets released Dr. Album — only the band's third album ever, and its first since early 2012 — last month, and now they're playing some shows in support of the exceptional Red Scare Industries record. No Trigger performed at Riot Fest last weekend and they're gearing up to play a Punk In Drublic tour stop in their hometown of Worcester this weekend. Before both of those shows (as well as a show with The Lawrence Arms on Sept. 30 and an upcoming appearance at The Fest in October), Rheault spoke to My First Band host Tyler Maas about the making of Dr. Album, getting back on the road and plans for 2023. Along the way, the singer talked about the early impact bands like NOFX and Rancid had on his musical trajectory, playing shows at gymnasiums and all-ages venues (where he regrettably threw pastries everywhere), starting the band that would eventually turn into No Trigger, unforgettable experiences he's had on stage and on tour through the years, and why people should consider micro-dosing. My First Band is sponsored by Mystery Room Mastering and Lakefront Brewery. The show is edited by Jared Blohm. You can listen to My First Band on iTunes, Stitcher, Spotify and wherever else you get podcasts. You can also listen to rebroadcasts of previous My First Band episodes on WMSE every Wednesday from noon to 12:30 p.m. CST. Music used in this episode comes courtesy of Devils Teeth ("The Junction Street Eight Tigers") and No Trigger ("Too High To Die").

Head of User Acquisition @ Hopper: Makoto Rheault-Kihara- Strategic Paid Search Marketing, Massive Growth & The Culture That Empowers It EPISODE SUMMARY: Makoto Rheault-Kihara leads user acquisition at Hopper, one of the fastest-growing travel apps with over 60 million in sales to date. Makoto was previously the Director of Marketing at Cruisehub, an OTA aiming to modernize the cruise industry and lead search engine marketing at Busbud, a bus travel booking platform in 80 countries. Hopper differentiates its paid marketing strategy by focusing on user acquisition AND long-term trust through investing in paid social and other mobile ad channels. They use Facebook as the primary focus of their paid social ads with TikTok as a growing secondary channel. Paid search marketing is becoming a lot more abstract and therefore more creative.
Just go for it. Don't overanalyze everything too much. Put your feelers out and see what your audience is. See what kind of options you have. It's very easy to get caught up in the planning and, by the time you know it, it has been five years and, you still have not launched a company. That's my big advice is to just take it by the reins and see what you can do with it. --- Send in a voice message: https://anchor.fm/the-inventive-journey/message

W., Ambili Vijayakumari, A., Parker, D., Verma, R., Radwan, A., Sun Abstract: White matter bundle segmentation using diffusion MRI fiber tractography has become the method of choice to identify white matter fiber pathways in vivo in human brains. However, like other analyses of complex data, there is considerable variability in segmentation protocols and techniques. This can result in different reconstructions of the same intended white matter pathways, which directly affects tractography results, quantification, and interpretation. In this study, we aim to evaluate and quantify the variability that arises from different protocols for bundle segmentation. Through an open call to users of fiber tractography, including anatomists, clinicians, and algorithm developers, 42 independent teams were given processed sets of human whole-brain streamlines and asked to segment 14 white matter fascicles on six subjects. In total, we received 57 different bundle segmentation protocols, which enabled detailed volume-based and streamline-based analyses of agreement and disagreement among protocols for each fiber pathway. Results show that even when given the exact same sets of underlying streamlines, the variability in the protocols for bundle segmentation is greater than all other sources of variability in the virtual dissection process, including variability within protocols and variability across subjects. In order to foster the use of tractography bundle dissection in routine clinical settings, and as a fundamental analytical tool, future endeavors must aim to resolve and reduce this heterogeneity. Although external validation is needed to verify the anatomical accuracy of bundle dissections, reducing heterogeneity is a step twowards reproducible research and may be achieved through the use of standard nomenclature and definitions of white matter bundles and well-chosen constraints and decisions in the dissection process. S., Poirier, J., Descoteaux, M., Villeneuve, S., PREVENT-AD Research Group Abstract: Beta-amyloid (A{beta}) and tau proteins, the pathological hallmarks of Alzheimer's disease (AD), are believed to spread through connected regions. Combining diffusion imaging and positron emission tomography, we investigated associations between A{beta}, tau and white matter microstructure specifically in bundles connecting brain regions in which AD pathology accumulates. In 126 cognitively normal elderly at risk of AD, we focussed on free-water corrected diffusion measures in the cingulum, posterior cingulum, fornix and uncinate fasciculus. We found higher tissue fractional anisotropy and lower mean and radial diffusivity related to increased A{beta} at the cortical endpoints of the cingulum and fornix. We observed similar but stronger associations in the uncinate fasciculus, but with increased A{beta} and tau at the endpoints of this bundle. This consistent pattern of associations, with opposite directionality to the usual degeneration pattern in symptomatic individuals, suggests more restricted diffusion in bundles vulnerable to preclinical AD pathology. Abstract: Brain atlases have proven to be valuable neuroscience tools for localizing regions of interest and performing statistical inferences on populations. Although many human brain atlases exist, most do not contain information about white matter structures, often neglecting them completely or labelling all white matter as a single homogenous substrate. While few white matter atlases do exist based on diffusion MRI fiber tractography, they are often limited to descriptions of white matter as spatially separate 'regions' rather than as white matter 'bundles' or fascicles, which are well-known to overlap throughout the brain. Additional limitations include small sample sizes, few white matter pathways, and the use of outdated diffusion models and techniques. Here, we present a new population-based collection of white matter atlases represented in both volumetric and surface coordinates in a standard space. These atlases are based on 2443 subjects, and include 216 white matter bundles derived from 6 different state-of-the-art tractography techniques. This atlas is freely available and will be a useful resource for parcellation and segmentation. Copy rights belong to original authors. Visit the link for more info

In her memoir, "My Wild and Precious Life," Susanne Rheault shares her inspiring story of working in rural Africa running AIDS prevention programs and helping to transform a two room orphanage in Tanzania into a home, school and organic farm for 350 children called Precious Project. After experiencing side effects from cancer treatment that affected her skin and brows, chiropractic medicine and wellness specialist, Dr. Doris Dalton, developed a natural skin care and makeup line called DOLL 10.This show is broadcast live on Wednesday's at 12PM ET on W4WN Radio – The Women 4 Women Network (www.w4wn.com) part of Talk 4 Radio (http://www.talk4radio.com/) on the Talk 4 Media Network (http://www.talk4media.com/).
Susie Rheault brings her new book "My Wild and Precious Life: A Memoir of Africa" to us to share a compelling story of finding our life purpose. And sometimes when we have that idea in mind, we might happen to stumble on it, or it simply comes to us. Susie "stumbled" on an orphanage in Tanzania in 2011. She and her husband were seeking for a meaning work, and in 2 small concrete buildings housing 9 AIDS/HIV positive children, they found what they were looking for. While we search for our purpose, or perhaps in addition to it, we may find ourselves drawn to supporting the work of the Precious Project. While it continues as an orphanage, a key part of the work is education, and currently the elementary school has 350 students. And the focus is to build a secondary school. This is highly important work in the world, and is making a huge difference in these children's lives, the leaders of tomorrow. www.preciousproject.org
