Inside Out 2 is a 2024 American animated coming-of-age film produced by Pixar Animation Studios for Walt Disney Pictures. The sequel to Inside Out (2015), it was directed by Kelsey Mann (in his feature directorial debut) and produced by Mark Nielsen, from a screenplay written by Meg LeFauve and Dave Holstein, and a story conceived by Mann and LeFauve.[5][6] The film stars Amy Poehler, Phyllis Smith, Lewis Black, Diane Lane, and Kyle MacLachlan reprising their roles from the first film with Tony Hale (replacing Bill Hader as Fear), Liza Lapira (replacing Mindy Kaling as Disgust), Maya Hawke, Ayo Edebiri, Adèle Exarchopoulos, Paul Walter Hauser, and Kensington Tallman (replacing Kaitlyn Dias as Riley) joining the cast. It tells the story of Riley's emotions as they find themselves joined by new emotions that want to take over Riley's head. Inside Out 2 was first announced in September 2022 during the D23 Expo announcement, with Mann, Nielsen, and LeFauve attached as director, producer, and writer, respectively, while Poehler was revealed to reprise her role in the film, along with Smith, Black, Lane, and MacLachlan. Hale, Lapira, and Hawke joined the cast in November 2023, while Edebiri, Exarchopoulos, Hauser, and Tallman's roles were confirmed in March 2024. That same month, Holstein was confirmed to have co-written the screenplay with LeFauve. The film features Pixar chief creative officer Pete Docter's "five to 27 emotions" idea from the first film that Mann pitched during its production to utilize "truthful" worldbuilding. Inside Out 2 premiered at the El Capitan Theatre in Los Angeles on June 10, 2024, and was released in theaters in the United States on June 14, 2024. “Twin Peaks” actor Kyle MacLachlan had a crazy story for his friend. In the 1980s, kingpin Pablo Escobar bribed an entire North Carolina town to let him land airplanes and smuggle cocaine into the US. The little-known story happened in Varnamtown, population 300. Researchers fix the chirality of helical proteinsHello and welcome to the NanoLSI podcast. Thank you for joining us today. In this episode we feature the latest research by Naoki Ousaka, Mark J. MacLachlan and Shigehisa Akine at the Kanazawa University NanoLSI.The research described in this podcast was published in Nature Communications in October 2023 Kanazawa University NanoLSI websitehttps://nanolsi.kanazawa-u.ac.jp/en/Researchers fix the chirality of helical proteinsResearchers at Kanazawa University report in Nature Communications how they can control chirality inversion in α helical peptides.The function of a protein is determined by its structure – prompting great interest in how to manipulate these structures. The structure is defined not just by the sequence of amino acids that make it, but the shape these acids make – the secondary structure – as well as how that shape is then folded. The most common secondary protein structure is the α-helix, which can coil to the right or left. This coiling direction in turn determines how it engages with other chiral structures, which may be the form of a light beam or another molecule. Although molecular components and environmental factors can favor a particular coiling direction over the other, helical molecules tend to flip between the two coil directions. Now Naoki Ousaka, Mark J. MacLachlan and Shigehisa Akine at Kanazawa University in Japan have shown how they can control and fix the coil direction.Helical proteins are chiral molecules, which means that the molecule's shape cannot be fitted into its mirror image. In nature helical proteins often have other chiral components, such as sugars or amino acids, and these will determine which way the protein coils. However, there is a lot of interest in synthesizing artificial helical proteins that have different chemical components and hence functions not found in nature, and these may not have other chiral components. Nonetheless having both types or “enantiomers” of the chiral molecule can be hazardous because of the significant differences in behavior between the two chiral forms, one of which may be benign or even therapeutic while the other is toxic. Hence, there is demand for other ways of selecting and fixing the chirality.So how did they go about this?Ousaka, MacLachlan and Akine synthesized α helical molecules solely from achiral components. They included bulky segments so that the molecule tended towards the larger rings of the α helical structure, as well as side chains of piperidine – molecular components that are common in pharmaceuticals. These side chains can be cross linked to “staple” the molecule into either the righthanded or lefthanded coil, inhibiting flipping between the two – chiral inversion. Finally they added another molecular component, known as an ester – the L-Val-OH residue. This would switch the direction of the coil in response to acidic or basic environments due to preferences in the interaction between oxygen atoms in the ester and the amino acid backbone.The researchers used a range of chiral characterization methods including circular dichroism, nuclear magnetic resonance and liquid chromatography. They found that with the molecule stapled just once, it would slow down the flipping between enantiomers by a factor of 106, although this still occurred over minutes. Changing the solution to acid or alkali also successfully determined which enantiomer was favoured. However, stapling the molecule twice slowed down the chirality inversion by a factor of 1012, so that the molecular chirality was stable for years. This increased energy barrier to chirality inversion could then be overcome by heating the sample to very high temperatures to switch betNanoLSI Podcast website

It gave us a more straightforward Lynch tale, and it recalibrated the Shat The Movies definition of surprise boobs. Janine MacLachlan is on a mission to get ideas heard. She's a communications strategist with deep experience in brand positioning and business storytelling. Janine works with executives and aspiring leaders in the areas of executive visibility, thought leadership and essentially helps them get out of their own way. How many of us would like a little help with that? Growing up in Saginaw, Michigan with three siblings, she admits that they have different childhood memories than she does because while they were outside in the yard playing ball, she was in her room absorbed in her reading. Janine loved books. She also wrote lots of stories and sketched pictures to go along with them. In fact, when her parents downsized their home a few years ago, they gave her a box full of the stories she authored as a kid. In college, she majored in journalism, working as an editor for the university newspaper. Upon graduation, Janine went to work for a public relations agency where she was able to get a wide range of experience. After several job and geographic moves, Janine became the Director of Marketing and Communications for the American Dental Association in Chicago. She remained there for nine years when she decided to start her own communications firm. Today, Janine is a regular contributor to Forbes in the area of leadership strategy, where she has interviewed executives from AbbVie, Microsoft, and 3M, as well as authors, and academics. I have a conversation with Colin Maclachlan who I originally met whilst on a Close Protection course in Scotland. Colin joined the military as a 15 year old originally with the Royal Scots. He then applied for Special Forces selection, passed and successfully became a 22SAS operator. Whilst in the SAS Colin took part in several high profile operations. After military service, Colin pursued various interests as a motivational speaker, author, historian, security consultant and even a video game character!
