Abstract: Giant Axonal Neuropathy (GAN) is a pediatric neurodegenerative disease caused by KLHL16 mutations. KLHL16 encodes gigaxonin, a regulator of intermediate filament (IF) protein turnover. Previous neuropathological studies and our own examination of postmortem GAN brain tissue in the current study revealed significant involvement of astrocytes in GAN. To study the underlying cellular mechanisms, we generated human models of GAN using induced pluripotent stem cells (iPSCs). Skin fibroblasts from seven GAN patients carrying different KLHL16 mutations were reprogrammed to iPSCs, and isogenic controls were derived via CRISPR/Cas9 editing. Neural progenitor cells (NPCs), astrocytes, and brain organoids were generated through directed differentiation. All GAN iPSC lines were deficient for gigaxonin, which was restored in the isogenic clones. While GAN iPSCs displayed normal organization of lamin B1 and keratin IFs, they exhibited patient-specific increased expression and perinuclear bundling of vimentin. Nestin IF morphology was unaffected, but fewer nestin-positive cells were present in GAN NPCs compared to controls. The most dramatic phenotypes were observed in GAN iPSC-astrocytes and brain organoids, which displayed dense perinuclear IF accumulations and abnormal nuclear morphology. GFAP oligomerization and perinuclear aggregation were strongly potentiated in the presence of vimentin, and GAN cells with large perinuclear vimentin aggregates accumulated nuclear KLHL16 mRNA. Doug Bouldin, Family Nurse Practitioner and partner at The Ozzie Smith IMAC Regeneration Center in St. Louis, has been practicing medicine for more than 25 years across the state of Missouri. Since graduating from the University of Missouri-St. Louis, Bouldin has served patients across a broad range of settings, and the experience he has to draw from is derived from several locations across Missouri including St. Luke's Hospital, St. John's Mercy, Lincoln County Medical Center and Barnes Jewish Medical Group. His experience includes clinical and managerial roles that have equipped him to be among the leaders of this groundbreaking effort at the Ozzie Smith IMAC Regeneration Center. Bouldin understands that medicine alone does not always fix the problem. “For years I have known that referring to chiropractic and physical therapy completed the care that I could give to a patient,” Bouldin says. “But now, being able to actually practice it in a medical model where we are accessible to one another under one roof is truly ideal for me.” Bouldin has been a leader amongst his peers, presenting at several conferences and symposiums over the course of his career. He carries a passion for education and is excited about the opportunity to pass it on to patients who have never heard of regenerative medicine techniques. Growing up on his father’s plantation near Clarksdale, Marshall Bouldin, III, dreamed of being a commercial illustrator like his hero, Norman Rockwell. Encouraged by his mother to pursue his love of art, he left Clarksdale in 1939 to attend the School of the Art Institute of Chicago and there began a career that would gain him notoriety around the nation, even as it brought him home again. In this episode, taken from our 1974 oral history interview, Bouldin details his evolution as an artist. During the year and a half spent at the School of the Art Institute of Chicago, felt he learned from more by studying the Institute’s collection of paintings than he did attending class. When WWII broke out, he was forced to leave school. Deemed unfit for military duty due to a birth defect that left him with a limp, he worked as an illustrative draftsman for the Vultee Aircraft Company in Nashville, Tennessee. After the war, Bouldin became the apprentice of a commercial illustrator in Connecticut where he honed his skills as he learned from the best in the business. He soon had his own studio and a New York agent who secured magazine work for him with publications like Colliers and Outdoor Life. It was after attending an exhibition of works by Vincent van Gogh, Bouldin realized that he envied the freedom of expression that differentiated artists from illustrators. He explains why he decided to come home to Clarksdale and become a portrait painter. Throughout his career, Bouldin rejected the stereotypes associated with professional artists. He discusses why it’s important to stay connected to the rest of society. As a portrait painter, he was required to sell his services like any other professional. However, he maintained it was always about making new friends, not money. Of the hundreds of portraits he was commissioned to paint, many of the subjects were famous, including, President Nixon’s daughters, William Faulkner, William Winter and Mike Espy.

Kwasi Boyd-Bouldin (b. 1977) is a Los Angeles based photographer whose work focuses on the urban environment and how a neighborhoods physical composition reflects the lives of it’s inhabitants. He is best known for The Los Angeles Recordings, an ongoing documentary project comprised of photo essays about L.A.’s rapidly changing urban landscape. He has also recently collaborated with KCET in the creation of In Plain Sight, a series photographing locations of police violence and was one of Time Magazine’s 12 African American Photographers to Follow in 2017. Feel free to read the NH 1 article in the show notes, as well as their conversations with the AG. These reps include Pamela Tucker, Amanda Bolden, Joseph Lachance, and Ted Wright. Broadly, I'm wondering why any of them would say anything. Probably in exchange for amnesty, but what did the AG really have one them? Some online communication perhaps. But unless they could track the money, or witnessed the sale, I'm wondering why they said anything at all. I'm disappointed any liberty minded reps would participate so fully. Perhaps there are circumstances beyond my knowledge, and if so, I'm free to change my mind later. At first read, dissapointed. Bouldin and Lachance were pretty detailed in discussing their interactions with Tasker, Bouldin naming Pamela Tucker as someone who she knew purchased drugs from Tasker. Lachance quite forthright about details of his purchases, as well as Tasker's operation. Pamela Tucker is a complete hypocrite. Her official position is against marijuana use. So it's ok to lock peaceful people up for using a plant, but she gets away with it because of her privileged position. On June 9, 2016, the Supreme Court decided Dietz v. Bouldin. Petitioner Rocky Dietz sued respondent Hillary Bouldin for negligence for injuries suffered in an automobile accident. Bouldin removed the case to Federal District Court. At trial, Bouldin admitted liability and stipulated to damages of $10,136 for Dietz’ medical expenses. The only disputed issue remaining was whether Dietz was entitled to more. During deliberations, the jury sent the judge a note asking whether Dietz’s medical expenses had been paid and, if so, by whom. Although the judge was concerned that the jury may not have understood that a verdict of less than the stipulated amount would require a mistrial, the judge, with the parties’ consent, responded only that the information being sought was not relevant to the verdict. The jury returned a verdict in Dietz’ favor but awarded him $0 in damages. After the verdict, the judge discharged the jury, and the jurors left the courtroom. Moments later, the judge realized the error in the $0 verdict and ordered the clerk to bring back the jurors, who were all in the building—including one who may have left for a short time and returned. Over the objection of Dietz’s counsel and in the interest of judicial economy and efficiency, the judge decided to recall the jury. After questioning the jurors as a group, the judge was satisfied that none had spoken about the case to anyone and ordered them to return the next morning. After receiving clarifying instructions, the reassembled jury returned a verdict awarding Dietz $15,000 in damages. On appeal, the Ninth Circuit affirmed. -- The question before the Supreme Court was whether a federal district court can recall a jury it has discharged, or whether the court can remedy the error only by ordering a new trial. By a vote of 6-2, the Supreme Court affirmed the judgment of the Ninth Circuit. Justice Sotomayor delivered the opinion of the Court, which held that a federal district court has a limited inherent power to rescind a jury discharge order and recall a jury in a civil case for further deliberations after identifying an error in the jury's verdict. The district court did not abuse that power here. Justice Sotomayor’s majority opinion was joined by the Chief Justice and Justices Ginsburg, Breyer, Alito, and Kagan. Justice Thomas filed a dissenting opinion, in which Justice Kennedy joined. -- To discuss the case, we have Brad Shannon, who is Professor of Law at Florida Coastal School of Law.
