"We tried so many shelters, and we were like rejected. It's like, it's not for migrants, it's not for migrants!” Lebanon is caught in a devastating conflict between the Israeli army and Hezbollah, a powerful Shia Muslim political party and armed group. Critical infrastructure, including hospitals, residential buildings, and shelters, has been destroyed, displacing over a million people across the country. Among those most affected are Lebanon's roughly 250,000 migrant domestic workers, primarily from African and Asian countries including Ethiopia, Bangladesh, and Sierra Leone. Many of these workers, who were already living in precarious conditions under the Kafala system which leaves them heavily reliant on their employers, have now been made homeless and are desperately seeking shelter, food, and safety. For Africa Daily, Muthoni Muchiri speaks with Mariam Bai Sesay, a former domestic worker and Enu, the deputy director of Egna Legna Besidet, an organisation that provides legal aid, housing assistance, and food relief to the workers.

L'actualité est de plus en plus intense en matière de drone. A l'heure où je vous parle, nous sommes dans la phase 2 de l'exercice interarmées ORION (« O2 ») qui mobilise jusqu'à 7 000 militaires de l'ensemble des armées, sur 14 départements du sud de la France. Ceci est consécutif au déclenchement de l'Échelon national d'urgence (ENU), qui a débuté le 9 février 2023. Mais l'ORION, c'est aussi un drone ... un drone un peu particulier, puisqu'il Tethered, autrement dit « attaché »  ou « Filaire » en français. Il est destiné à la sécurité, qu'elle soit civile ou militaire ... Quoi de plus naturel que de se rapprocher de l'équipe de la société ELISTAIR, concepteur du drone ORION. Nitekim Nuh, Hûd, Salih ve Şuayb (a.s.e.) de, Mekke'de vefât etmişlerdir. Bunların, kabirleri, zemzem kuyusu ile Hacerülesved rüknü arasındadır. Zemzem ile rükün arasında yetmiş peygamber diğer bir rivâyete göre de hacca gelip vefât eden peygamberlerden orada 99 peygamber gömülüdür.” Âyet-i kerîmede “O halde (Resûlüm), peygamberlerden azim sahibi olanların sabrettiği gibi sen de sabret!” (Ahkaf s. 35) buyrulur. Burada zikredilen Ulû'l-azm sahibi Peygamberler rivâyete göre: 1. Nûh (a.s.) 2. İbrahim (a.s.) 3. Mûsâ (a.s.) 4. İsâ (a.s.) 5. Peygamber (s.a.v.)'dir. Bir hadîs-i şerîfe göre de peygamberlerin seyyidleri de, Nuh, İbrahim, Mûsâ, İsâ (a.s.e.) ve Hz. Muhammed (s.a.v.) olmak üzere beştir. Hz. Peygamber (s.a.v.) ise, bu beşin seyyididir. Kıyâmet gününde de, âdemoğullarının seyyidi O (s.a.v.)'dir. Öncekilerin ve sonrakilerin Kıyâmet gününde Hâmd sancağı, O (s.a.v.)'e verilecektir. He was fortunate to grow up in the beach community of La Jolla, California and spent his childhood exploring both the land and the sea. As a young boy, he lost his father to suicide, which would deeply inspire his path in life. As a young adult, he had the opportunity to travel extensively and experience many of the world's great surf spots as a professional bodyboarder. He holds a BA in Environmental Policy from San Diego State University and an MA in Leadership Studies from the University of San Diego. He is also the author of The Adventures of Enu, Ashes in the Ocean, and Experience Nature Unplugged: A Guide to Wellness in the Digital Age. In 2012, Sebastian and his wife, Sonya, founded Nature Unplugged with the mission of inspiring wellness in the digital age. There is a growing scientific field called “ecotherapy” that has demonstrated a strong connection between time spent outside in nature and improved well-being. Shifting our attention to the sounds, smells, and beauty is calming for many. On brain scans we can actually see reduced activity in. the parts of the brain that are linked to rumination — or repetitive negative thoughts. In this episode, I speak with Sebastian Slovin, author of Experience Nature Unplugged: A Guide to Wellness in the Digital Age, a new book on how connecting with nature helps reset our brains. We discuss the ways our digital lives are negatively impacting our mental health and how nature is the perfect antidote.About Sebastian SlovinSince Sebastian can remember, nature has been a central part of his life. Since he can remember, nature has been a central part of Sebastian Slovin's life. A professor stood before his philosophy class and had some items in front of him. When the class began, he wordlessly picked up a very large and empty mayonnaise jar and proceeded to fill it with golf balls. He then asked the students if the jar was full. They agreed that it was. The professor then picked up a box of pebbles and poured them into the jar. He shook the jar lightly. The pebbles rolled into the open areas between the golf balls. He then asked the students again if the jar was full. They agreed it was. The professor next picked up a box of sand and poured it into the jar. Of course, the sand filled up everything else. He asked once more if the jar was full. The students responded with an unanimous “yes.” The professor then produced two cups of coffee from under the table and poured the entire contents into the jar effectively filling the empty space between the sand. The students laughed. “Now,” said the professor as the laughter subsided, “I want you to recognize that this jar represents your life. The golf balls are the important things–your family, your children, your health, your friends and your favorite passions–and if everything else was lost and only they remained, your life would still be full. The pebbles are the other things that matter like your job, your house and your car. The sand is everything else–the small stuff. “If you put the sand into the jar first,” he continued, “there is no room for the pebbles or the golf balls. The same goes for life. If you spend all your time and energy on the small stuff you will never have room for the things that are important to you. “Pay attention to the things that are critical to your happiness. Play with your children. Take time to get medical checkups. Take your spouse out to dinner. Play another 18. There will always be time to clean the house and fix the disposal. Take care of the golf balls first–the things that really matter. Set your priorities. The rest is just sand.” One of the students raised her hand and inquired what the coffee represented. The professor smiled. “I’m glad you asked. It just goes to show you that no matter how full your life may seem, there’s always room for a couple of cups of coffee with a friend.”

Katherine Bull completed preclinical studies at Cambridge in 1999 and graduated in Clinical Medicine in 2002 at Oxford. She works on Whole Genome Sequencing in ENU mutagenesis and Autoimmunity under the supervision of Richard Cornall. Every year, about 65 DPhil students start a life changing experience in the Nuffield Department of Medicine in Oxford. The Department offers a highly competitive studentship for our DPhil programme to outstanding candidates of any nationality. The candidates are judged on the basis of their academic and research potential. The NDM Prize Studentship is a four year course which includes full payment of all fees and an annual stipend.

According to recent epidemiological studies, primary aldosteronism is considered to be responsible for almost 10-15% of all cases of arterial hypertension. The genetic background of this common disease, however, has been elucidated only for the rare familial types whereas in the large majority of sporadic cases genetic causes or modifiers still remain unclear. In an attempt to define novel genetic mechanisms of hyperaldosteronism we utilized a random mutagenesis screen after treatment with the alkylating agent N-ethylnitrosourea (ENU) and phenotypically characterized affected mice for their blood aldosterone levels. As the detection method we used a time resolved fluorescent immunoassay which allows the measurement of aldosterone in very small murine plasma volumes. Using this assay we determined the normal aldosterone values for C3HeB/FeJ wild type mice under baseline conditions and following specific stimulation and suppression tests. We could demonstrate the expected increase in aldosterone response upon ACTH stimulation, a high potassium diet and an angiotensin II injection, as well as the decrease after a dexamethasone suppression test, a normal saline load test and a fludrocortisone suppression test. On the molecular level, the expression of aldosterone synthase showed a similar pattern with a fast response to the investigated stimuli. These tests should later be applied to the mouse lines derived from the ENU screen, in order to investigate potential abnormal response to these stimuli in comparison to wild type animals. Furthermore, aldosterone measurement was carried out in more than 2000 F1 offspring (of both genders) of chemically mutated inbred C3HeB/FeJ mice. From these tested F1 offspring, aldosterone levels were consistently elevated (defined as levels above 3 SD over the mean of untreated animals)upon repeated measurement in 9 animals (8 females and 1 male). Further breeding of affected female animals gave rise to F2 pedigrees from which four established lines displayed high aldosterone values. These animals served for a detailed phenotypic characterization and showed an increased aldosterone to renin ratio, low potassium values and normal renal function in line with the presence of primary aldosteronism. In addition, the investigation of their cardiac phenotype showed increased collagen deposits and subsequently cardiac fibrosis, as also observed in patients suffering from primary aldosteronism. In the future, genetic SNP analysis can be performed to identify underlying genetic loci, responsible for this trait. Taken together, these data demonstrate the feasibility of a phenotype-driven mutagenesis screen to detect and establish mutant mouse lines with a high aldosterone phenotype.

Background: Clinical chemical blood analysis including plasma electrolytes is routinely carried out for the diagnosis of various organ diseases. Phenotype-driven N-ethyl-N-nitrosourea (ENU) mouse mutagenesis projects used plasma electrolytes as parameters for the generation of novel animal models for human diseases. Methods: Here, we retrospectively evaluated the use of the plasma electrolytes calcium, chloride, inorganic phosphorus, potassium and sodium in the Munich ENU mouse mutagenesis project where clinical chemical blood analysis was carried out on more than 20,000 G1 and G3 offspring of chemically mutagenized inbred C3H mice to detect dominant and recessive mutations leading to deviations in various plasma parameter levels. Results: We identified a small number of animals consistently exhibiting altered plasma electrolyte values. Transmission of the phenotypic deviations to the subsequent generations led to the successful establishment of mutant lines for the parameters calcium and potassium. Published data from other phenotype-driven ENU projects also included only a small number of mutant lines which were generated according to altered plasma electrolyte levels. Conclusion: Thus, use of plasma electrolytes detected few mouse mutants in ENU projects compared to other clinical chemical blood parameters.

Background: Large-scale mutagenesis screens in the zebrafish employing the mutagen ENU have isolated several hundred mutant loci that represent putative developmental control genes. In order to realize the potential of such screens, systematic genetic mapping of the mutations is necessary. Here we report on a large-scale effort to map the mutations generated in mutagenesis screening at the Max Planck Institute for Developmental Biology by genome scanning with microsatellite markers. Results: We have selected a set of microsatellite markers and developed methods and scoring criteria suitable for efficient, high-throughput genome scanning. We have used these methods to successfully obtain a rough map position for 319 mutant loci from the Tubingen I mutagenesis screen and subsequent screening of the mutant collection. For 277 of these the corresponding gene is not yet identified. Mapping was successful for 80 % of the tested loci. By comparing 21 mutation and gene positions of cloned mutations we have validated the correctness of our linkage group assignments and estimated the standard error of our map positions to be approximately 6 cM. Conclusion: By obtaining rough map positions for over 300 zebrafish loci with developmental phenotypes, we have generated a dataset that will be useful not only for cloning of the affected genes, but also to suggest allelism of mutations with similar phenotypes that will be identified in future screens. Furthermore this work validates the usefulness of our methodology for rapid, systematic and inexpensive microsatellite mapping of zebrafish mutations.

Background/Aim: Phenotype-driven screening of a great pool of randomly mutant mice and subsequent selection of animals showing symptoms equivalent to human kidney diseases may result in the generation of novel suitable models for the study of the pathomechanisms and the identification of genes involved in kidney dysfunction. Methods: We carried out a large-scale analysis of ethylnitrosourea (ENU)-induced mouse mutants for albuminuria by using qualitative SDS-polyacrylamide gel electrophoresis. Results: The primary albuminuria screen preceded the comprehensive phenotypic mutation analysis in a part of the mice of the Munich ENU project to avoid loss of mutant animals as a consequence of prolonged suffering from severe nephropathy. The primary screen detected six confirmed phenotypic variants in 2,011 G1 animals screened for dominant mutations and no variant in 48 G3 pedigrees screened for recessive mutations. Further breeding experiments resulted in two lines showing a low phenotypic penetrance of albuminuria. The secondary albuminuria screen was carried out in mutant lines which were established in the Munich ENU project without preceding primary albuminuria analysis. Two lines showing increased plasma urea levels were chosen to clarify if severe kidney lesions are involved in the abnormal phenotype. This analysis revealed severe albuminuria in mice which are affected by a recessive mutation leading to increased plasma urea and cholesterol levels. Conclusion: Thus, the phenotypic selection of ENU-induced mutants according to the parameter proteinuria in principle demonstrates the feasibility to identify nephropathy phenotypes in ENU-mutagenized mice. Phenotypic and genotypic characterization of the ENU-induced mutant mouse line HST001 for use in nephrological research A dominant mutation was established in the line HST001 within the Munich ENU mouse mutagenesis project. On the genetic background of the inbred strain C3H heterozygous mutant mice showed a pathological increase of the plasma urea concentration. The phenotypic characterization of the mutant line included the measurement of body and organ weights, food intake and a broad spectrum of clinical chemical and hematological parameters as well as histological investigations of inner organs. For those examinations heterozygous mutant mice were mated to C3H mice and the offspring were divided in the two groups of mutant and wildtype littermates according to the concentration of plasma urea. The mutation showed complete phenotypic penetrance in all generations examined. In addition the genomic localization of the causative mutation was determined by linkage analysis. At the age of 4 weeks the body weight of the mutant mice and the wildtype mice showed no difference. After 6 weeks the mutant mice grew more slowly leading to a significantly lower body weight compared to the wildtype littermates. At the age of 26 weeks the body weight of mutant mice was about 20% reduced as compared to controls. Mutants also showed a reduction of the nose-rump-length and the body fat content. Regarding the relative organ weights the mutant mice showed significantly higher values for the spleen and the brain. The clinical-chemical and hematological parameters were determined in the animals at the age of 12, 18 and 24 weeks. The mutant mice had significantly higher plasma concentrations of urea and total protein as well as a higher activity of the alkaline phosphatase. Beside that they showed lower plasma concentrations of triglycerides and glucose and a lower activity of amylase than the wildtype mice. The mutants showed increasing plasma urea levels whereas the levels decreased in the controls. At the age of 24 weeks the mutants showed levels, which were two-fold higher than those of the controls. The histopathological investigation of the kidneys did not reveal any alterations, which could to explain the pathological increase of the plasma urea concentration in the mutant mice. However urea secretion via the kidneys was impaired, since the mutants showed significantly lower urine urea levels compared to the wildtype mice. The investigation of the hematological parameters resulted in the occurrence of altered values in the 18 und 24 weeks old mice. The mutant mice showed reduced numbers 91 of erythrocytes as well as decreased values for hemoglobin concentration, mean corpuscular volume and hematocrit. The hitherto existing results of the linkage analysis of the mutation in the genome using polymorphic microsatellite markers showed that the mutation maps to chromosome 7. This result has to be confirmed and specified by using additional markers and a larger number of animals.

This thesis describes the phenotypic and genotypic analysis of the dominant mutation in the ENU-induced mutant mouse line SMA002. The results were derived from the analysis of heterozygous mutant animals on the C3H genetic background. As the main phenotype of SMA002, an abnormal behavior combined with reduced body weight was revealed. The mutant phenotype showed complete penetrance in all generations analyzed. Abnormal behavior, as the most prominent phenotype was characterized by the occurrence of restlessness and an increased grooming behavior. The grooming behavior was increased fivefold but did not always contain all aspects of the standard repertoire. Pathological examinations of the skin and the analysis of parameters of the immune system indicated that this behavior was not caused by an allergic reaction. Clinical examination of the animals revealed a significant difference in body weight between mutant and control littermates. The body weight of the mutant animals was about 30% lower than control. In addition mutant animals had a significant lower nose-rump length and a significant lower carcass weight. Analysis of the peripheral blood for hematological and clinical chemical parameters as well as subsequent pathological examinations excluded a kidney, liver or pancreas disease as the potential cause of the abnormal phenotype. Decreased values for cholesterol, triglyceride, total protein and potassium in the mutant animals referred to an increased metabolism and a hormonal deviation in these mice. Furthermore, deviations of the IGF-system were found in the mutants. Linkage analysis of the causative mutation was carried out by genome wide polymorphic microsatellite marker analysis. The highest correlation for the mutation was found with the markers D13Mit20 (35cM) and D13Mit253 (37cM) of chromosome 13. This aberrant phenotype of the line SMA002 has not yet been reported. Thus, SMA002 represents a novel mouse model for the analysis of abnormal behavior and growth defects. Subsequent analysis will identify the exact mutation in the line. In addition, further analysis of the phenotype will result in detailed information on the metabolism and hormonal regulation of SMA002.

Background: The ENU Mouse Mutagenesis Project aims at a large-scale, systematic production of mouse mutants using the alkylating agent ethyl-nitrosourea (ENU). Offspring of mutagenized mice are subjected to a multiparameter screen to detect alterations in various phenotypes with the ultimate goal of identifying novel genes relevant for the expression of the phenotype. Using this approach, we have analyzed plasma IgE concentrations to identify mouse mutants with aberrant plasma IgE levels. Methods and Results: ENU-mutagenized male C3HeB/FeJ were mated to wild-type females to produce F1 offspring. F1 animals were analyzed for alterations in their plasma IgE concentrations that showed a dominant mode of inheritance, or bred further to screen for recessive phenotypes. Plasma IgE concentrations were deter mined by ELISA and a normal range for plasma IgE was established using C3HeB/FeJ wild-type animals. So far we have tested 6568 F1 animals. Repeated testing confirmed a stable aberrant IgE phenotype in 124 animals. To confirm the genetic basis of the observed phenotype, these mice were subjected to confirmation crossing. Currently we have established 9 independent m uta nt mouse lines (3 with high plasma IgE and 6 with plasma IgE below detection limit) that have been genetically confirmed and additional 24 variant mouse lines are currently undergoing confirmation testing. Conclusion: ENU mouse mutagenesis allowed us to generate and identify mouse mutants with aberrant plasma IgE levels, which may be used to characterize novel genes involved in IgE regulation and may serve as animal models for IgE-mediated diseases.
