Abstract: We developed a platform that utilizes a calcium-dependent luciferase to convert neuronal activity into activation of light sensing domains within the same cell. The platform is based on a Gaussia luciferase variant with high light emission split by calmodulin-M13 sequences that depends on influx of calcium ions (Ca2+) for functional reconstitution. In the presence of its luciferin, coelenterazine (CTZ), Ca2+ influx results in light emission that drives activation of photoreceptors, including optogenetic channels and LOV domains. Critical features of the converter luciferase are light emission low enough to not activate photoreceptors under baseline condition and high enough to activate photosensing elements in the presence of Ca2+ and luciferin. We demonstrate performance of this activity-dependent sensor and integrator for changing membrane potential and driving transcription in individual and populations of neurons in vitro and in vivo. Abstract: Ca2+ plays many critical roles in cell physiology and biochemistry, leading researchers to develop a number of fluorescent small molecule dyes and genetically encodable probes that optically report changes in Ca2+ concentrations in living cells. Though such fluorescence-based genetically encoded Ca2+ indicators (GECIs) have become a mainstay of modern Ca2+ sensing and imaging, bioluminescence-based GECIs--probes that generate light through oxidation of a small-molecule by a luciferase or photoprotein--have several distinct advantages over their fluorescent counterparts. Bioluminescent tags do not photobleach, do not suffer from nonspecific autofluorescent background, and do not lead to phototoxicity since they do not require the extremely bright extrinsic excitation light typically required for fluorescence imaging, especially with 2-photon microscopy. Current BL GECIs perform poorly relative to fluorescent GECIs, producing small changes in bioluminescence intensity due to high baseline signal at resting Ca2+ concentrations and suboptimal Ca2+ affinities. Here, we describe the development of a new bioluminescent GECI, "CaBLAM," which displays a much higher contrast (dynamic range) than previously described bioluminescent GECIs coupled with a Ca2+ affinity suitable for capturing physiological changes in cytosolic Ca2+ concentration. Derived from a new variant of Oplophorus gracilirostris luciferase with superior in vitro properties and a highly favorable scaffold for insertion of sensor domains, CaBLAM allows for single-cell and subcellular resolution imaging of Ca2+ dynamics at high frame rates in cultured neurons. CaBLAM marks a significant milestone in the GECI timeline, enabling Ca2+ recordings with high spatial and temporal resolution without perturbing cells with intense excitation light. Abstract: The ability to manipulate neuronal activity both opto- and chemogenetically with a single actuator molecule presents unique and flexible means to study neural circuit function. We previously developed methodology to enable such bimodal control using fusion molecules called luminopsins (LMOs), where a channelrhodopsin actuator can be activated using either physical (LED driven) or biological (bioluminescent) light. While activation of LMOs using bioluminescence has previously allowed manipulation of circuits and behavior in mice, further improvement would advance the utility of this technique. Thus, we here aimed to increase the efficiency of bioluminescent activation of channelrhodopsins by development of novel FRET-probes with bright and spectrally matched emission tailored to Volvox channelrhodopsin 1 (VChR1). We find that pairing of a molecularly evolved Oplophorus luciferase variant with mNeonGreen significantly improves the efficacy of bioluminescent activation when tethered to VChR1 (construct named LMO7) as compared to previous and other newly generated LMO variants. We proceed to extensively benchmark LMO7 against previous LMO standard (LMO3) and find that LMO7 outperforms LMO3 in the ability to drive bioluminescent activation of VChR1 both in vitro and in vivo, and efficiently modulates animal behavior following intraperitonial injection of fluorofurimazine. In conclusion, we demonstrate a rationale for improving bioluminescent activation of optogenetic actuators using a tailored molecular engineering approach and provide a new tool to bimodally manipulate neuronal activity with increased bioluminescence-driven efficacy. Abstract: Amyotrophic lateral sclerosis (ALS) is a severe neurodegenerative disorder affecting brain and spinal cord motor neurons. Mutations in the copper/zinc superoxide dismutase gene (SOD1) are associated with ~20% of inherited and 1-2% of sporadic ALS cases. Much has been learned from mice expressing transgenic copies of mutant SOD1, which typically involve high-level transgene expression, thereby differing from ALS patients expressing one mutant gene copy. To generate a model that more closely represents patient gene expression, we created a knock-in point mutation (G85R, a human ALS-causing mutation) in the endogenous mouse Sod1 gene, leading to mutant SOD1G85R protein expression. Heterozygous Sod1G85R mutant mice resemble wild type, whereas homozygous mutants have reduced body weight and lifespan, a mild neurodegenerative phenotype, and express very low mutant SOD1 protein levels with no detectable SOD1 activity. Homozygous mutants exhibit partial neuromuscular junction denervation at 3-4 months of age. Spinal cord motor neuron transcriptome analyses of homozygous Sod1G85R mice revealed up-regulation of cholesterol synthesis pathway genes compared to wild type. Transcriptome and phenotypic features of these mice are similar to Sod1 knock-out mice, suggesting the Sod1G85R phenotype is largely driven by loss of SOD1 function. By contrast, cholesterol synthesis genes are down-regulated in severely affected human TgSOD1G93A transgenic mice at 4 months. Our analyses implicate dysregulation of cholesterol or related lipid pathway genes in ALS pathogenesis. The Sod1G85R knock-in mouse is a useful ALS model to examine the importance of SOD1 activity in control of cholesterol homeostasis and motor neuron survival. Copy rights belong to original authors. When city planner and architect Adrian Lipscombe visited a cafe in La Crosse, Wisconsin for a business meeting, little did she know she'd end up moving her family up all the way from Austin, Texas, running the cafe herself, and revitalizing a whole section of the city. In this episode. Nicola takes us through HOW mindfulness supports us “gives us an edge” in EQ, in managing thoughts, and with self-compassion. She also talks about becoming significantly self-aware that in turn helps managing our emotions. Abstract: Voltage-gated CaV2.2 calcium channels are expressed in nociceptors, at pre-synaptic terminals, soma, and axons. CaV2.2 channel inhibitors applied to the spinal cord relieve pain in humans and rodents, especially during pathological pain, but a biological function of nociceptor CaV2.2 channels in processing of nociception, outside pre-synaptic terminals, is not explored. Here, we demonstrate that functional CaV2.2 channels in skin are required for thermal hyperalgesia following intraplantar capsaicin exposure. We provide evidence that CaV2.2 channels at nociceptor free endings release inflammatory signals, ATP and IL-1{beta}. We assess the role of CaV2.2 splice isoforms to capsaicin-induced hyperalgesia measured by thermal and mechanical stimuli. Our data reveal a critical role for peripheral CaV2.2 channels in skin in neurogenic thermal hyperalgesia but not in mechanical hypersensitivity.
