A new technology that blends bioluminescence with cellular machinery may shine some light on the details of their comings and goings and interactions that can be hazy.Scientists had the bright idea to take that same enzyme that makes fireflies glow and tie it to RNA, the molecule that reads the genetic information in DNA. The intent is to reduce the number of animals used, refine methods to be efficient and humane and replace animals with other models where possible.This cc: Life Science episode is sponsored content, courtesy of MediLumine.I talked to Stephen Marchant, Founder and CEO of MediLumine about the importance of animal research, how imaging in small animals is different from humans and innovations that support the 3Rs of laboratory animal sciences.    While the FDA no longer requires the testing of all new drugs in animal models prior to clinical trials, for some drugs, the requirement may stand. Animal research has been essential for many advances in human health and will surely remain relevant in the quest to discover new treatments in the future.As compassionate beings, we want to help our fellow humans whenever possible. When a child has cancer or a mother with a diagnosis of early onset Alzheimer's tells her children that at some point she may not recognize them (or they her), no options will be left unexplored to look for and find solutions and those options likely include some research using animal models to better understand disease. The animal technicians and scientists working in laboratory animal sciences have a passion for developing therapies and treatments for human disease but also for taking care of animals. For those interested to know more, Stephen recommends the GetReal Podcast by Dr Cindy Buckmaster to truly appreciate the reality of this and what it is like to work with laboratory animals in the life sciences.In vivo imaging is used to better understand disease progression or response to treatment, and it is one aspect of research that isn't likely to be replaced soon. The development of novel contrast agents allows scientists to visualize structures such as tumors or vasculature much deeper in the organism thus precluding the use of visual inspections and use of a caliper to measure tumor size. This breakthrough is built in to MediLumine's tag line ‘Vision without Sacrifice'.      Imaging structures in a mouse, whether by MRI or PET scan is very different from imaging in humans. The structures are (obviously) much smaller. What I hadn't realized previously is how the small size requires longer measurements to get the desired resolution. Typical contrast agents like CT contrast agents used in the clinic are rapidly cleared making it more difficult for small animal micro-CT systems to generate high resolution images with these contrast agents. Stephen explained how the development of contrast agents like Fenestra HDVC, allows improved imaging of mouse organs with in vivo micro-computed tomography (CT).Depending on the goal of an experiment, modalities like MRI or CT can provide good resolution which allows researchers to calculate, for example, the volume of an object such as a tumor. Other modalities, such as optical imaging, are preferred when sensitivity is important. Bioluminescent reporters can sensitively be detected in vivo with the trade-off being resolution.…with optical imaging, it's a very sensitive modality. So you might be able to see something at lower concentrations, for example, with bioluminescent imaging. If you look at some of the tumor studies, in some of the publications, we see even a few days after the injection of tumor cells, we're able to see signals, but we're not necessarily able to localize them very well. So, for example, if you have, let's say, a signal in the right lobe of the liver, you would see something coming with optical imaging, but you wouldn't necessarily be able to localize it and say precisely exactly where it is.Without these in vivo imaging methods, understanding the biology of tumor progression would require a larger cohort of animals, with a requirement to euthanize some fraction at various time points to locate tumors and measure their size. That's the reduction element of the 3 Rs.Refinement is also possible. Stephen shared an example of how new contrast agents in microCT allow the study of fatty liver disease that couldn't be done previously with terminal studies and histology.As for replacement, the development of organoids, small collections of differentiated cells, offer an alternative to some animal assays while more closely reflecting an in vivo condition than monolayer cell cultures. Abstract: We developed a platform that utilizes a calcium-dependent luciferase to convert neuronal activity into activation of light sensing domains within the same cell. The platform is based on a Gaussia luciferase variant with high light emission split by calmodulin-M13 sequences that depends on influx of calcium ions (Ca2+) for functional reconstitution. In the presence of its luciferin, coelenterazine (CTZ), Ca2+ influx results in light emission that drives activation of photoreceptors, including optogenetic channels and LOV domains. Critical features of the converter luciferase are light emission low enough to not activate photoreceptors under baseline condition and high enough to activate photosensing elements in the presence of Ca2+ and luciferin. We demonstrate performance of this activity-dependent sensor and integrator for changing membrane potential and driving transcription in individual and populations of neurons in vitro and in vivo. Abstract: Ca2+ plays many critical roles in cell physiology and biochemistry, leading researchers to develop a number of fluorescent small molecule dyes and genetically encodable probes that optically report changes in Ca2+ concentrations in living cells. Though such fluorescence-based genetically encoded Ca2+ indicators (GECIs) have become a mainstay of modern Ca2+ sensing and imaging, bioluminescence-based GECIs--probes that generate light through oxidation of a small-molecule by a luciferase or photoprotein--have several distinct advantages over their fluorescent counterparts. Bioluminescent tags do not photobleach, do not suffer from nonspecific autofluorescent background, and do not lead to phototoxicity since they do not require the extremely bright extrinsic excitation light typically required for fluorescence imaging, especially with 2-photon microscopy. Current BL GECIs perform poorly relative to fluorescent GECIs, producing small changes in bioluminescence intensity due to high baseline signal at resting Ca2+ concentrations and suboptimal Ca2+ affinities. Here, we describe the development of a new bioluminescent GECI, "CaBLAM," which displays a much higher contrast (dynamic range) than previously described bioluminescent GECIs coupled with a Ca2+ affinity suitable for capturing physiological changes in cytosolic Ca2+ concentration. Derived from a new variant of Oplophorus gracilirostris luciferase with superior in vitro properties and a highly favorable scaffold for insertion of sensor domains, CaBLAM allows for single-cell and subcellular resolution imaging of Ca2+ dynamics at high frame rates in cultured neurons. CaBLAM marks a significant milestone in the GECI timeline, enabling Ca2+ recordings with high spatial and temporal resolution without perturbing cells with intense excitation light. My fingers shook. The rough rock walls around me didn't afford many hand holds. My feet pressed against the sides to keep me from falling down this cold, dark shaft. My knees were scraped, my hands bloody. I lifted my head again to look at the scrap of light far above me. No one was coming. My arms started shaking, my feet were slipping. All I could think was, "Help." "Let go." Did I hear the words bounce off the rock shaft, or did I just think them? An empty place moaned in my chest where I wished hope was. The light above me grew dim as my vision blurred. I could only hold myself up for so long. I'd tried climbing up every way I could think, but every time I tried, I just seemed to fall farther and farther...down this dark hole. "Let go." There it was again. I couldn't think straight. How long had it been since I'd had a drink of water? My breath rattled. I strained to push myself up, but I had no strength left. Was that crackling I heard? My right foot shifted—and I plummeted a few feet deeper. Pain seared my left arm where the rock wall tore at my skin. “Why?” I moaned. "Let yourself fall." Okay, three times. Maybe that was me, maybe it wasn't. But what other options did I have? Either wait for my muscles to give out completely or pull my limbs close to my body and hope I didn't lose any. I shuddered. “Are you sure about this?” I asked the voice. "Trust me," it replied. I let out a sigh...then let myself fall. Fall. Fall. SPLASH! I bobbed up to the surface with a gasp—I was floating. I didn't even need to kick to keep my head above the...was this water? It glowed a soft blue-green. Wait. I leaned my head back and let my feet float up to a laying-down position. The cave ceiling was covered in blue-green stars. “Bioluminescent,” I breathed. Beautiful. And the ceiling was moving—or I was. That was more likely. Downstream I floated, watching the beauty above me. My muscles weren't shaking anymore. I even felt relaxed. My arm didn't hurt—what? I lifted my arm from the water and saw new skin where it had been bloody and raw. A laugh bubbled out of me and echoed off the cavern walls. "Let me carry you." The voice was comforting, and I felt myself drifting to sleep. When I woke up, I heard the trickling of the current, drips from stalactites, and the voice whispered, "It's time." Then I felt myself being moved to the rocky bank of the underground river. I pushed myself up out of the water with surprising ease. "This way." I turned and saw a glowing path that shimmered along the cave floor. One step at a time, I followed the path carefully. The path started to grow dimmer. "No!" I dropped to my knees in dismay. "Look up." The voice was gentle. Then I noticed the path wasn't darker, the cave was getting lighter. I rushed forward as the light grew brighter, until I could see an opening in the rock. I ran into the warm light of day. • Hannah Howe • Have you ever felt like you were stuck in that dark hole? • We all have times when we get in trouble, either because of our sin or simply because we live in a broken world. Throughout the Bible, God makes it clear that we cannot save ourselves, but He wants to save us. There is nothing we could do to earn God's kindness, yet, through Jesus, He generously gives us mercy and makes us new. When do you find it difficult to accept God's kindness? • Oftentimes, the way out is not taking matters into our own hands or trying harder but humbly acknowledging that we can't do it...and leaning into God's help. And He is so good—along the way He brings healing to our weary souls. How might God be inviting you to rely on Him today? When I said, “My foot is slipping!” Your loving kindness, Yahweh, held me up. Shadows are climbing the faces of the majestic floating mountains, while a symphony of crickets and other, less identifiable nocturnal creatures call out into the darkness. The trill of a banshee, the coo of some celestial dove…they are both familiar and unfamiliar, endearing and intriguing. As darkness settles onto every surface, the entire planet seems to take its cue to come alive, radiance emanating from its very core. Bioluminescent flora and fauna emerge from shadowy hiding places, and threads of blue and green light seep through the surface of the hard earth, like veins connecting every moment to the next, every space of time to something greater, something whole and beautiful. (7:00) The heartbeat of Pandora pulses in sovereign rhythm into the night, from the base of the floating mountains in this Swotu Wayä, this “sacred place of song.” A connection between the physical and metaphysical, music cannot be bound by tangible restraints. Here, nature's song is a tranquil ebb and flow that is harmonious and complete, boundless and free. Beneath the stars and the luminous glow of this spiritual space, we are wrapped in an enclave of Magic and light. Welcome to wander as we please through this otherworldly Eden, we amble through trails of the Mo'ara Valley, where wildlife zigzags and climbs over every rock and bridge. The echo of creatures settles into a steady cadence Here, pulling us into a trance that calls us deeper and deeper into the realm of their enigma. (12:45) Then, out of the depths of our lullaby emerges a celestial chorus of voices, a cinematic orchestra that seems to shower from the stars. We have reached the heart of civilization, the gathering place known as Satu'li Canteen. Once a mess hall to feed the expats from Earth who came to live on Pandora, it represents a gift of friendship and a symbol of camaraderie and respect. Both the indigenous Na'vi and the humans can find their cultures reflected Here, in everything from the art and artifacts displayed throughout to the delicious cuisine. (15:14) Still, the night beckons, and our appetites are allayed only by our intensifying curiosity. The rush of a waterfall can only mean that we have found ourselves in the wake of the extraordinary floating mountains. Powerful and somehow reassuring, the majesty of these mountains calls to mind the vastness of the world and all of the natural wonders it holds. Mountains, rivers, lush valleys and endless oceans…our connection to them is as limitless as we desire, their gift to us their presence. In return, we can offer the promise that we will honor and cherish them as the Na'vi people so reverently care for this place they call home. (16:29) As we pass the Alpha Centauri Expeditions' Flight of Passage, we hear the almost robotic inner workings of the operation that brings civilians like ourselves to this planet, to be immersed in its story from the back of a banshee. And while an aerial view of Pandora by daylight is enticing for the bravest and boldest, to be engaged in the crux and soul of this space at nighttime is serenity unlike anything we can experience elsewhere. The delicate hush endures as pathways and bridges reach further into the jungles of Pandora. Our wonder is met by a wave of calm, a sense that the sounds that we hear are friendly, rather than fierce. Somehow, we feel seen by the night, personally covered and protected by its cloak of inky stillness. And as the ethereal bioluminescence is reflected on the faces of those around us, permitting us to see the curious wanderers not so unlike ourselves, we understand. (18:04) The Na'vi have a phrase that serves as a greeting, but that means so much more: “Oel ngati kameie,” “I see you.” Like those we meet under the floating mountain or beside the giant Flasca Reclonada, it speaks not only to someone's physical presence, but their spiritual presence as well. When we see those who come to this place, those who seek the renewal of quiet moments of joy with the zeal and excitement that we do, we recognize something in them that does not need to be spoken in order to be true. (22:45) We see their soul, their insatiable wanderlust, their desire to find Magic in the everyday. Whether Na'vi or human, tangible being or intangible darkness, Magic has the power to connect us in ways that cannot be held or measured. Like the suspension of twilight just before nightfall, like the pendulum caught mid-swing, we are met with a moment of stillness. Here, we can truly see and delight in the spaces surrounding us, the people who share in them With us, and the Magic that binds us together.

Officially, this episode is on the amazing glowing algae living in the waters of three of Puerto Rico's bays, most notably Puerto Mosquito on Vieques, one of Puerto Rico's smaller islands. Listener and boriqueño native Roberto Cancel describes swimming in the bay on a dark night, surrounded by glowing blue waters. But most of the episode is devoted to perhaps the most important event in world history: 1493. Not 1492, but 1493. That's the year when Christopher Columbus returned to the Americas, not as an explorer, but as a conqueror. We discuss (and really only scratch the surface of) the impact of this second voyage. Today on the Mushroom Hour Podcast we have the privilege of speaking with Brian Perry PhD, Associate Professor of Biology at California State University, East Bay. He received his Master's Degree from San Francisco State University under the guidance of Dr. Dennis E. Desjardin, and his Ph.D. from Harvard University where he studied with Dr. Donald H. Pfister. Brian has been studying fungi since 1995, and has published over 45 papers in scientific journals. In addition to studying the evolution of fungal bioluminescence, he also conducts research on mushrooms and other fungi of Vanuatu, the assembly and biogeography of island fungal communities, endophytic fungi of Hawaiian plants, and the systematics of Mycena and allied genera. Alan is a mycologist who has undertaken extensive field and lab research on the mushrooms of California and Mexico. He has been collecting mushrooms for 17+ years and has focused on the taxonomy and photography of Mexican mushrooms for the past 11+ years. Our epic journey brings us back to the dawn of the new millennium when a young Alan Rockefeller first became spellbound by fungi. A few years into his mycology explorations, he had a chance encounter on shroomery.org with a biologist in Mexico who was posting amazing fungal finds. Alan made a connection and began traveling to Mexico each year to seek out rare fungi. Since he began these adventures, he has played a central role in mapping fungal diversity throughout different climate ranges in Mexico. This includes some of the most in-depth field observations in the world when it comes to those incredible bioluminescent and infamous psilocybin containing fungi. We'll cover it all - how bioluminescence occurs in biology, the evolutionary significance of bioluminescent enzymes and psilocybin compounds in fungi and how Alan goes about finding the rarest varieties of these fascinating organisms. The epitome of the self-taught naturalist and citizen scientist we'll learn the secrets of how Alan gained his vast taxonomic knowledge. Just "showing up" and pursuing his interests he has been able to collaborate with leading academics in the field of mycology. Alan then reveals how we can all contribute to the science of mycology by making good observations in the field and cataloging them effectively for posterity. We get a masterclass on cataloging field finds including how to take good photos in the field, record relevant observations, preserve dried specimens and even how we can sequence the genome of our fungal finds. This is an incredible opportunity to soak up wisdom and experience from a legend in the field of citizen mycology. TIPS FOR VISITING PUERTO RICO As you know Puerto Rico was hit with a category 5 hurricane in late 2017. Don’t let that knowledge stop you from visiting though, the island has made great progress in rebuilding. Plus your tourism dollars will add to the money it takes for the island to continue to rebuild. There are many areas of the island to stay, though if this is something you are worried about, the southwest coast has the least amount of damage from the hurricane. The beaches there are very calm, and it is highly recommended that if you are traveling with younger kids and you want to hit some beaches you should head over to the southwest side of the island. The Combate Beach Resortis a great locally owned hotel to stay in if you are thinking of staying on the southwest side of the island. Close to the beach with easy access. There are a few other hotels you can stay but most of this area of the island is Airbnb or rentals. The outer islands are very beautiful and since you have to take a ferry or a small plane to get to them, they are far less crowded. The islands are inhabited, but not with many people, making visiting the pristine beaches an experience you won’t get many other places. If you are looking to experience bioluminescent bays, Puerto Rico has three of them. There is Vieques (or Mosquito Bay as called by tourists), which is the brightest, then Laguna Grande in Fajardo and also La Parguera. San Juan is a great place to stay as your vacation base. You can get to a lot of different places in about an hour. The El Conquistador Resortis a great resort to stay for families. They have a large water park as well as a spa and other activities. La Concha Resortis also a great resort for families to stay along with the Ritz-Carlton Reservewhich is a more luxury hotel that has just reopened and houses an amazing spa. About an hour drive from San Juan is an area called Guavate which is well known for their restaurants serving pork, playing live music and dancing. If you want to experience some true local authentic food and entertainment you should definitely check it out. The food is also not expensive, a family of four could eat to a meal for less than $40. Visiting El Yunque Rainforestis a fun place for both tourists and local to visit. It is only about 45 minutes from San Juan. They still have some trails closed but if you have a family that loves outdoors and likes hiking, this is a must see adventure. Old San Juan is a beautiful place to visit and if you do some research there may be some gallery shows or events going on that you can explore. There are some small islands off of Fajardo that you may want to look into visiting. They have catamaran tours and snorkeling available, and you may see some sealife like sea turtles. Ponce is a beautiful town to visit with its great boardwalk and art museums. There is a great mix of history and culture in the town. If you are comfortable driving you should definitely think about renting a car. It will definitely give you more freedom to go places when you want to or even just leaving to go to your next destination that you want to explore instead of waiting for a ride. A lot of the beaches have shacks where you can get great food. Make sure you try the empanadas, which can be stuffed with octopus, meat, sea food, or even just cheese and sauce making a kind of fried pizza. Alcapurrias is another must try dish, which everyone makes a little differently, but the dough is typically made out of green plantains which is stuffed with meat or crab and then fried. Roasted pork is also a must try food! Kasaltais a great bakery in Ocean Park that has some great sandwiches! Lot 23is an area that has been turned into a food truck park and has a variety of food and picnic benches to eat. Everyone can get something different and enjoy as a family. If your family is adventurous you should definitely look into rental properties or even Airbnbs. As well as renting a car to be able to get around as you please. BEST PLACE TO TAKE FAMILY PHOTO Jessica recommends a sunset shot with your family if you make it to the southwest side of the island. Though if you don’t make it to the southwest you can get some great shots at El Morroin Old San Juan. Although fireflies and other glowing critters have been well researched, fungi that glow are not nearly as well understood. Often referred to as "foxfire" or "fairy fire", their glow was first documented way back in ancient times, but researchers are still figuring out what it's all about. Dr Siouxsie Wiles is a passionate science communicator, microbiologist and bioluminescence enthusiast; or as her TedX profile describes her: “that pink-haired science lady”. Siouxsie leads the Bioluminescent Superbugs Group at the University of Auckland, where she studies infectious diseases by making bacteria glow in the dark. In addition to this incredibly cool research, Siouxsie communicates science as a blogger on Sciblogs, in her own podcast and produces explainer animations for YouTube.   For her science communication efforts, Siouxsie has been recognised with tonnes of very important accolades – the New Zealand Association of Scientists Science Communication Award, the Prime Minister's Science Media Communication Prize and the Royal Society of New Zealand Callahan medal. As proof of her rising celebrity status in the field, this weekend she’s delivering a TedX talk in Christchurch on her research.
