C., Dorfman, K. A., Bahiru, M. S., Stephens, A. D. Abstract: Mitosis is an essential process in which the duplicate genome is segregated equally into two daughter cells. CTCF has been reported to be present in mitosis but its importance for mitotic fidelity remains to be determined. To evaluate the importance of CTCF in mitosis, we tracked mitotic behaviors in wild type and two different CTCF CRISPR-based genetic knockdowns. We find that knockdown of CTCF results in prolonged mitoses and failed anaphase segregation via time lapse imaging of SiR-DNA. CTCF knockdown did not alter cell cycling or the mitotic checkpoint, which was activated upon nocodazole treatment. Immunofluorescence imaging of the mitotic spindle in CTCF knockdowns revealed disorganization via tri/tetrapolar spindles and chromosomes behind the spindle pole. Imaging of interphase nuclei showed that nuclear size increased drastically, consistent with failure to divide the duplicated genome in anaphase. Population measurements of nuclear shape in CTCF knockdowns do not display decreased circularity or increased nuclear blebbing relative to wild type. However, failed mitoses do display abnormal nuclear morphologies relative to successful mitoses, suggesting population images do not capture individual behaviors. Thus, CTCF is important for both proper metaphase organization and anaphase segregation which impacts the size and shape of the interphase nucleus. Copy rights belong to original authors. She was always a wonderful teacher in town, very focused on play and learning through doing” -Lowey Bundy Sichol [11:03]“It's pretty cool that schools now are embracing teaching kids about entrepreneurship, and I think it goes back to ‘learn through doing, learn through failure, experimenting and inventing'” -Lowey Bundy Sichol [11:36]“A lot of my books are about how influenced entrepreneurs and business leaders are by their childhood, and the parents who raised them, and I am such a good example of that” -Lowey Bundy Sichol [12:00]“This is what I have learned [about kids in 3rd, 4th, 5th grade]: They are incredibly brave, they are incredibly creative, they are incredibly excited to share their ideas, they are not too cool yet, but they are incredibly bright. It's almost like they are mini entrepreneurs” -Lowey Bundy Sichol [13:56]“It's interesting the philosophy of education for anyone, but for kids, the idea of having applied projects that are practical is very powerful” -Syd Finkelstein [20:23]“I think education, in general, has not done as good a job in entire industries in making it clear why you need to learn” -Syd Finkelstein [20:43]“My purpose is to tell a great story and to teach kids about business and entrepreneurship” -Lowey Bundy Sichol [29:43]“Being able to adapt for the different audience, being able to just persevere throughout the hard times and being brave enough to go against what other people didn't support are some of the common themes” -Lowey Bundy Sichol [38:51]Stay connected:Sydney FinkelsteinWebsite: http://thesydcast.comLinkedIn: Sydney FinkelsteinTwitter: @sydfinkelsteinFacebook: The SydcastInstagram: The SydcastLowey Bundy SicholWebsite: Lowey Bundy Sichol LinkedIn: Lowey Bundy Sichol Twitter: Lowey Bundy Sichol Instagram: Lowey Bundy Sichol Amazon: Lowey Bundy SicholSubscribe to our podcast + download each episode on Stitcher, iTunes, and Spotify. This episode was produced and managed by Podcast Laundry.

Nita Lowey joined Clare Sheridan to discuss her distinguished career in the U.S. House of Representatives. Lowey shared her reflections on the importance of women’s voices in the political process and memories of her historic career. Congresswoman Lowey has served as a U.S. Representative from New York since 1989. In 2018, she became the first woman to chair the House Appropriations Committee. She will retire at the end of this term. Then we turned our attention to the preeminent historian Isabelle Savell, whose comprehensive research documented the history of the women from Rockland County who spent seven decades fighting for the vote. Savell wrote Ladies Lib: How Women Got the Vote, the go-to text for those interested in suffrage history in Rockland County. Savell’s granddaughter, Becky Savell, phoned in to discuss all of the important research that her grandmother did as Senior Historian at the Historical Society of Rockland County; she’ll share fond memories of her grandmother, as well."Isabelle Savell was a lovely, brilliant, beautiful lady, “said Gene Setzer of South Nyack. "She was one of the stalwarts, and one of the finest history researchers we’ve ever had around here—a beautiful writer so useful to the community.”_____Crossroads of Rockland History, a program of the Historical Society of Rockland County, airs on the third Monday of each month at 9:30 am, right after the Steve and Jeff morning show, on WRCR radio 1700 AM and www.WRCR.com. Hey Awesome Person Today's question is the same as the last episode.  Have you ever thought about running for Congress?  My guest is Evelyn Farkas and she is running. The daughter of refugees who fled communist Hungary, Evelyn Farkas grew up in Chappaqua. She has dedicated her life to public service — strengthening America’s national security and defending democracy. For nearly three decades, Evelyn has represented our country defending democracy, advancing human rights, fighting terrorism, and working towards nuclear non-proliferation. She has been a foreign policy leader in the Defense Department, Senate Armed Services Committee, the House Foreign Affairs Committee, and U.S. Marine Corps University’s Command and Staff College. She ran a congressional commission focused on keeping weapons of mass destruction out of the hands of terrorists. She worked in Bosnia just after their bloody war and has advised former Secretary of State Hillary Clinton during her time as a U.S. Senator. As Deputy Assistant Secretary of Defense under President Obama, Evelyn was responsible for U.S. policy toward Russia, Ukraine, and Eurasia. After the 2016 election, she stood up to conservative media to publicly denounce President Trump’s corrupt ties to Russia. I really liked talking with her and I think you will like listening to this conversation. https://evelynforny.com/ Patreon.com/petedominick
