D., Sanjay, A. Abstract: Despite the remarkable regenerative capacity of skeletal tissues, nonunion of bone and failure of fractures to heal properly presents a significant clinical concern. Stem and progenitor cells are present in bone and become activated following injury; thus, elucidating mechanisms that promote adult stem cell-mediated healing is important. Wnt-associated adult stem marker Lgr6 is implicated in the regeneration of tissues with well-defined stem cell niches in stem cell-reliant organs. Here, we demonstrate that Lgr6 is dynamically expressed in osteoprogenitors in response to fracture injury. Using an Lgr6-null mouse model, we find that Lgr6 expression is necessary for maintaining bone volume and efficient postnatal bone regeneration in adult mice. Skeletal progenitors isolated from Lgr6-null mice have reduced colony-forming potential and reduced osteogenic differentiation capacity due to attenuated cWnt signaling. Lgr6-null mice consist of a lower proportion of self-renewing stem cells. In response to fracture injury, Lgr6-null mice have deficient proliferation of periosteal progenitors and reduced ALP activity. Further, analysis of bone regeneration phase and remodeling phase of fracture healing in Lgr6-null mice showed impaired endochondral ossification and reduced mineralization. We propose that in contrast to not being required for successful skeletal development Lgr6-positive cells have a direct role in endochondral bone repair. Copy rights belong to original authors. Abstract: In previous research, miR-148a-3p deficiency was observed in bone malformation in hemifacial microsomia. Herein, in this article, we probed into the role of miR-148a-3p in bone physiology by utilizing miR-148a knock-out (KO) mice. Compared with wild-type (WT) or heterozygotic (HE) littermates, miR-148a knock-out mice manifested lower body weight, bone dysplasia with increased bone mass. Through in-vitro experiments, in terms of miR-148a-3p overexpression (miRNA mimic transfection) and knockout (primary cells from WT and KO littermates), we found that miR-148a-3p can suppress osteogenesis, either in the ALP activity or bone nodules formation. Afterward, by means of proteomics, combined with RNA-sequencing and prediction databases of microRNA targets (miRDB and TargetScan), nine candidate genes targeted by miR-148a-3p were identified. Among them, only Itga11 was regulated by mRNA degradation, while the others were modulated via post-transcriptional inhibition. Based on several online databases (GenePaint, BioGPS, STRING), Integrin Subunit Alpha 11 (Itga11) was suggested to play an essential role in osteogenesis and it was confirmed as one direct target of miR-148a-3p by dual-luciferase reporter assay. Meanwhile, gene set enrichment analysis (GSEA) indicated activation of PI3K-Akt signaling pathway and WNT signaling pathway in miR-148a KO mice. The thereafter western blot confirmed that PI3K/Akt/GSK3/{beta}-catenin signaling pathway was involved. Taken together, we demonstrated that miR-148a-3p can inhibit osteogenesis by targeting Itga11 via PI3K/Akt/GSK3/{beta}-catenin pathway. Copy rights belong to original authors. In this episode of the Epigenetics Podcast, we caught up with Susanne Mandrup from the University of Southern Denmark to talk about her work on the role of enhancer communities in adipocyte differentiation. The Laboratory of Susanne Mandrup focuses on the effect of enhancers and enhancer communities on the differentiation of mesenchymal stem cell into adipocytes and osteoblasts. The team has shown that there is significant cross-talk between enhancers and that these form communities of highly interconnected enhancers. Inactive enhancers are then activated by association with these pre-existing enhancer networks to facilitate gene expression in adipocyte differentiation. Kip Mangira was a Cutco Sales Professional in Vector Marketing's Midwest Region. He was born on September 2nd, 1995, in Akron, Ohio, with Osteogenesis imperfecta (brittle-bone disease), which confined him to a wheelchair for his entire life. Still, he had an indomitable spirit, and was not afraid to take on the challenge of selling Cutco Cutlery, starting in 2013. He won numerous awards for his exemplary work ethic and sales success, including the Silver Cup for finishing #1 in the Nation in sales in his competitive category in 2019. Known for his generosity and compassion, Kip became of the most-loved figures in the Cutco/Vector sales organization. He passed away on November 9th, 2021 in a road accident in Cozumel, Mexico. A study by Stanley Yung-Chuan Liu, Christian Guilleminault, Leh-Kiong Huon, Audrey Yoon found that Objective Distraction osteogenesis maxillary expansion (DOME) is a reliable method to expand the nasal floor and hard palatal vault in adults with obstructive sleep apnea (OSA). DOME results in a reduction in the apnea-hypopnea index (AHI) and subjective report of improved nasal breathing.A narrow maxilla with high arched palate characterizes a phenotype of obstructive sleep apnea (OSA) patients that is associated with increased nasal resistance and posterior tongue displacement. Current maxillary expansion techniques for adults are designed to correct dentofacial deformity. We describe distraction osteogenesis maxillary expansion (DOME) tailored to adult patients with OSA with narrow nasal floor and high arched palate without soft tissue redundancy. DOME is performed with placement of maxillary expanders secured by mini-implants along the midpalatal suture. This minimizes the maxillary osteotomies necessary to re-create sutural separation for reliable expansion at the nasal floor and palatal vault. We report the safety and efficacy profile of the first 20 patients at Stanford who underwent DOME.Airway and Sleep group employs the DOME technique for improving airway and combatting the effects of obstructive sleep apnea. Bladen Pietro, otherwise known as Bastrin, from North Carolina, was born with Osteogenesis imperfecta (OI), a genetic disorder that prevents the body from building strong bones, also called brittle bone disease.
THE ORIGINS OF THE MAJOR ORGANS-There are two major modes of bone formation or osteogenesis, and both involve the transformation of a preexisting mesenchymal tissue into bone tissue. The direct conversion of mesenchymal tissue into bone is called intramembranous ossification. This process occurs primarily in the bones of the skull. In other cases, the mesenchymal cells differentiate into cartilage, and this cartilage is later replaced by bone. The process by which a cartilage intermediate is formed and replaced by bone cells is called endochondral ossification.

Welcome back to our weekend Cabral HouseCall shows! This is where we answer our community's wellness, weight loss, and anti-aging questions to help people get back on track! Check out today's questions: Bria: I am desperate for help for my sister. Please!! For two years, she’s been unable to eat almost anything except a few foods. One mistaken ingredient, and she’s vomiting for 72 hours with a raging migraine. Even a sip of water causes her to vomit so she can’t keep anything down during this period of time, not medicine, not electrolytes, nothing. She knows she has Cibo, and bought the CBO protocol from your site. One pill sent her over the edge and into severe vomiting for three days. Please help! Western medicine doctors call it acid reflux but we know it’s not tha. How can she get the Cibo out of her system if she can’t even take one pill? The number of days of missed work, the amount of money spent, and the helplessness of her husband and her family is very difficult to watch. She recently saw a functional medicine practitioner in Anaheim California, but hasn’t been able to even start that lingual medicine yet. We need guidance. Thank you. Bria Jeanette: (I felt I couldn’t ask this question on the Facebook page due to privacy for my daughter) I have searched the list of podcast topics and have not located any information on osteogenesis imperfector. My daughter was clinically diagnosed with OI type 1 twenty years ago and has had ten fractures predominantly involving leg bones. Prior to 2011 she had not endure pain other than when she broke. In March 2011 she had to move 4 hours away to attend university and that very weekend she had to be hospitalised for an absus removal. Soon after she started having back pain and has endured 8 years of constant pain while studying a pharmacy degree. Over this time Ok she has taken anti inflammatories of varying strengths since it started in 2011. Plus a paracetamol every day at a full dose. She was on tramadol from May 2011 - October 2018 and now Is taking palexia. She took omeprazole along side the anti inflammatory as well. It seems as a result of the medication she has an ulcer and has regular bouts of diarrhoea. She lost 10kg in 2018 and was hoping with this she would lessen the pain which has only happened a little. When she tries to exercise the pain gets worse. As a mum I hate to see her in pain. In 2012 we took her to an alternative practitioner who had her on water and fruit for six months, but it made no difference and consequently she is very sceptical of all things alternative. As an wellness client I am starting to experience success Please help me with some ideas for my daughter. I decided to take 80,000IU Serra for a cyst located on the inframammary fold of the breast as it appears Serra may be extremely helpful for eliminating cysts. Unfortunately, upon using Serra on two separate occasions, it caused vomiting (the second time I took only one tablet). I contacted the company to ask if this could possibly be a detox/herxheimer reaction or if generally it does not agree with the body, unfortunately they were unhelpful. Having done some further research, Serra can result in nausea and may be best to start off with a lower potency, even though the majority of Serra start at 40,000iu. I did a podcast search and you briefly mention Serrapeptase on podcast 617, the notes link is missing so not sure of the brand suggested but wondered if you had any thoughts? Perhaps it can conflict with some people and I should just discontinue? (May also have underlining candida/thyroid issues). Thank you always for all you do. I tend to not eat/drink anything during a flight (unless I'm flying business it may be a light meal and bottled mineral water). I tend to have peppermint tea (at least two cups) before the flight. Even on a sleeper service flat bed and not eating or drinking anything may cause some bloating. I never touch the water/tea/coffee on a flight due to the hygiene. Even after a flight I can occasionally be left bloated for a couple of days. Does an aggravated/inflamed stomach during a flight specifically indicate something such as candida or is it simply the air pressure can be hard on a sensitive stomach? Is there anything you would recommend that would prevent an uncomfortable stomach during a flight? Thank you very much. Osteogenesis imperfecta (OI) is a rare disorder that causes bones to break easily. Children with OI may sustain broken bones from seemingly minor injuries where children without the disorder may not have a fracture. The motion analysis center located inside Shriners Hospitals for Children, provides motion analysis to children and uses the data collected to help doctors provide the most effective care for each patient, including those with osteogenesis imperfecta (OI).In this inspiring segment, learn about gait analysis for children, OI and the exciting new research being done at Shriners Hospital for Children with the guest, Peter Smith, MD. He is also a is a board-certified psychotherapist, and his a great book called, "Get off Your 'BUT': How to end self-sabotage and stand up for yourself." Expected to die at birth, Sean Stephenson has faced an army of reasons to give up and countless opportunities to embrace pity. He suffered more than 200 bone fractures by age eighteen, reached a height of only three feet, and is permanently confined to a wheelchair. Despite his challenges, he took a stand for a quality of life that has inspired millions of people around the world, including Tony Robbins, Richard Branson, and President Clinton. Sean has had more life adventures than most people who are twice his age. He worked on Capitol Hill for a U.S. Congressman and in the White House alongside President Clinton. This episode is better than beans on toast! Sam Renke, an up and coming British Actress with a disability and Director Max Barber were so much fun to chat with that I ended up breaking down the interview into two episodes! I just couldn't bare to cut any of our silly yet informative dialogue.
Background: Mutations in the FKBP10 gene were first described in patients with Osteogenesis imperfecta type III. Two follow up reports found FKBP10 mutations to be associated with Bruck syndrome type 1, a rare disorder characterized by congenital contractures and bone fragility. This raised the question if the patients in the first report indeed had isolated Osteogenesis imperfecta or if Bruck syndrome would have been the better diagnosis. Methods: The patients described here are affected by severe autosomal recessive Osteogenesis imperfecta without contractures. Results: Homozygosity mapping identified FKBP10 as a candidate gene, and sequencing revealed a base pair exchange that causes a C-terminal premature stop codon in this gene. Conclusions: Our study demonstrates that FKBP10 mutations not only cause Bruck syndrome or Osteogenesis imperfecta type III but can result in a severe type of isolated Osteogenesis imperfecta type IV with prenatal onset. Furthermore, it adds dentinogenesis imperfecta to the spectrum of clinical symptoms associated with FKBP10 mutations.
