BUFFALO, NY — October 23, 2025 — A new #research paper was #published in Volume 17, Issue 9 of Aging-US on September 11, 2025, titled “Roles of plasminogen activator inhibitor-1 in aging-related muscle and bone loss in mice.” In this study led by first author Takashi Ohira and corresponding author Hiroshi Kaji from Kindai University Faculty of Medicine, researchers found that female mice lacking the gene for plasminogen activator inhibitor-1 (PAI-1) were protected from age-related muscle weakness and bone thinning. This suggests that PAI-1 could be a potential target for future treatments to reduce frailty in aging populations. As the global population continues to age, more people are affected by conditions such as sarcopenia and osteoporosis. These disorders involve the progressive loss of skeletal muscle mass and bone density, leading to reduced mobility, a greater risk of falls, and a lower quality of life. To investigate the role of PAI-1 in aging, researchers compared young (6-month-old) and aged (24-month-old) male and female mice, with and without the PAI-1 gene. They found that PAI-1 levels increased with age in both sexes. However, only female mice lacking the PAI-1 gene experienced a significant reduction in age-related muscle and bone loss. Female mice without PAI-1 maintained stronger grip strength and greater muscle mass in their lower limbs. They also showed less cortical bone loss in their femurs and tibias. In contrast, male mice did not experience the same benefits, despite also showing increased levels of PAI-1 with age. These results suggest that PAI-1 contributes to aging-related decline in a sex-specific manner. “The present study found that lower limb muscle mass, gastrocnemius and soleus muscle tissue weights, and grip strength were significantly lower in 24-month-old male and female wild-type mice than in their 6-month-old counterparts.” PAI-1 plays key roles in blood clotting, inflammation, and cellular senescence—a process in which aging cells release harmful molecules that affect nearby tissues. One of these molecules, interleukin-6 (IL-6), is a major driver of inflammation. The researchers found that aged female mice lacking PAI-1 had lower IL-6 levels in both muscle and blood, suggesting that PAI-1 may contribute to muscle and bone loss by promoting inflammation. These protective effects were also not associated with changes in muscle protein turnover or reductions in fibrous tissue, reinforcing the idea that PAI-1's impact is likely driven by inflammatory signaling. This study highlights PAI-1 as a promising therapeutic target for slowing or preventing age-related declines in muscle and bone health, particularly in women. Since postmenopausal women are especially vulnerable to osteoporosis and frailty, a better understanding of how PAI-1 contributes to aging could lead to new strategies for maintaining strength and mobility in later life. Further research is needed to explore how PAI-1 interacts with other age-related biological changes and why its effects differ between sexes. Story at-a-glance Bone issues were once seen only among the elderly, but now young adults are also developing osteopenia, stress fractures, arthritis, and other signs of skeletal decline Poor sleep, chronic stress, sedentary habits, ultraprocessed diets, and harmful habits like smoking and alcohol use are key drivers behind the rising rates of bone loss in younger populations Calcium, vitamin D, magnesium, and vitamin K2 are all essential for bone health. Without proper balance, calcium can't be absorbed, used effectively, or kept out of arteries Diets high in ultraprocessed foods strip the body of key nutrients and promote inflammation. Excess iron also damages bone cells and increases fracture risk Weight-bearing exercises like resistance training, walking, and jumping stimulate bone growth and strengthen the skeletal structure at the molecular level

STRONGER BONES LIFESTYLE: REVERSING THE COURSE OF OSTEOPOROSIS NATURALLY
Dr. Evan Hirsch is a world-renowned fatigue expert and the creator of the EnergyMD Method, a clinically proven 4-step process to naturally resolve chronic fatigue and long COVID. In this deeply important conversation, we uncover how chronic fatigue, long COVID, and yes—bone loss—are all connected. If you've been diagnosed with osteoporosis or osteopenia and you're exhausted, overwhelmed, or scared of breaking a bone, this one's for you. These aren't isolated issues. They're symptoms of deeper dysfunction in the body. Evan and I walk through what I always talk about—the root causes—and why the traditional approach to bone health is flat-out missing the mark.We talk about toxins. We talk about trauma. We talk about the nervous system. And we talk about how your bones are not weak—they're depleted. From heavy metals to hormone disruptors to emotional patterns you inherited, this episode is going to open your eyes. If you're trying to take back control of your body and your bones, you'll want to hear how Evan's method has helped thousands of people get their energy back and rebuild from the inside out. Because bone health isn't just about density—it's about strength, quality, and resilience. And it starts here.Key TakeawaysThe Toxic 5 Framework – Dr. Hirsch breaks down how hidden toxins—like mold, metals, and chronic infections—are silently wrecking your bones and your energy.Bone Loss and Toxic Burden – Your bones are doing their best to protect you. When they break down, they're releasing toxins stored over decades. It's not a defect—it's survival. Why Lab Tests Often Miss the Mark – Just because your labs say "normal" doesn't mean your body is. If you're tired, inflamed, and losing bone, something deeper is going on.The 4-Step EnergyMD Process – We walk through Evan's practical approach: identify your causes, restore what's missing, prep your detox channels, and remove the junk. The Nervous System-Bone Health Connection – You can't rebuild a body that feels unsafe. Nervous system dysregulation blocks healing—even when you're doing everything else "right."You're Not Broken—You're Depleted – This is not your fault. And it's not yours forever. You can rebuild stronger bones and reclaim your energy—naturally, and on your terms.Where to Find Our GuestDr. Evan Hirsch's Website: http://www.energymdmethod.comFREE Facebook Group: http://energymdmethod.com/fbgroupDr. Hirsch's Facebook: https://www.facebook.com/DrEvanHirsch/Dr. Hirsch's Instagram: https://www.instagram.com/evanhirschmd/Links & ResourcesStronger Bones Lifestyle CommunityJoin a supportive space to strengthen your bones naturally with live sessions and masterclasses. In this episode of the Optimal Body Podcast, Doc Jen and Doctor Dom, both doctors of physical therapy, explore the benefits and drawbacks of whole body vibration plates. They discuss claims about these devices enhancing muscle gain, weight loss, and bone health, while emphasizing the need for critical evaluation of research. The conversation highlights the importance of balance and proprioception, especially as we age, and the sensory benefits of being barefoot. When we face bone loss, whether that be osteopenia or osteoporosis, we're rarely told about lifestyle factors that could make a difference. Today we'll delve into why we might suffer bone loss, and what we can do about it. Taking our very foundation – the strength of our skeleton – into our own hands is a step worth taking, and our guide in doing so is Debi Robinson. Her expertise in yoga instruction and functional health coaching -- plus her own experience breaking a hip -- make her the ultimate expert. When you hear what's connected to our bone health, I guarantee you'll be floored (safely!) Even if your bones are in good shape, learning how to keep them that way is definitely worth knowing.
