Here is Esther following on from sharing her testimony from last week with a message of hope and resurrection.

Out of all of Coltrane's outpourings, this is one of the closest to the Birtwistle, I think. It's not difficult to imagine Trane as the Great God Pan…Or maybe this. This is certainly the acid test for me to see if a piece of Contemporary Classical music is any good.Then again, it's not just Coltrane. One of the most shamefully overlooked players in this field was Sun Ra's tenor player John Gilmore. Abstract: Fluorescent antibodies are a workhorse of biomedical science, but fluorescence multiplexing has been notoriously difficult due to spectral overlap between fluorophores. We recently established proof-of-principal for fluorescence Multiplexing using Spectral Imaging and Combinatorics (MuSIC), which uses combinations of existing fluorophores to create unique spectral signatures for increased multiplexing. However, a method for labeling antibodies with MuSIC probes has not yet been developed. Here, we present a method for labeling antibodies with MuSIC probes. We conjugate a DBCO-Peg5-NHS ester linker to antibodies, a single stranded DNA docking strand to the linker, and finally, hybridize two MuSIC-compatible, fluorescently-labeled oligos to the docking strand. We validate the labeling protocol with spin-column purification and absorbance measurements, which show a degree of labeling of ~9.66 linker molecules / antibody. We demonstrate the approach using (i) Cy3, (ii) Tex615, and (iii) a Cy3-Tex615 combination as three different MuSIC probes attached to three separate batches of antibodies. We incubated MuSIC probe-labeled antibodies with protein A beads to create single and double positive beads that are analogous to single cells. Spectral flow cytometry experiments demonstrate that each MuSIC probe can be uniquely distinguished, and the fraction of beads in a mixture with different staining patterns is accurately measured. Donald Macleod talks to Sir Harrison Birtwistle about his life, inspiration and music. This week Donald Macleod meets Sir Harrison Birtwistle, described as “the most forceful and uncompromisingly original composer of his generation.” We hear his major compositions, broadly in chronological order, and reveal the preoccupations and processes behind a singular music imagination. To begin, we’ll hear about, Birtwistle’s daily working life, and about his early years among what became known as the Manchester school of composers. The premiere of his first opera Punch and Judy at Aldeburgh was infamous - much of the audience – including its commissioner Benjamin Britten – walked out at the interval. Next, we’ll hear about Birtwistle’s time in America and his friendship with Morton Feldman. Modern Masculinity, what is it? It kind of feels like it's a bit of a tricky subject these days and that even the term "masculinity", seems to mean very different things to different people. We have definitely seen some great strides forward in the equality debate in many forms of late, though I am very aware that there is still a long way to go and this is for sure not us wading in on that topic! Instead.... in this week's podcast, @davidbirtwistle and I wanted to talk about what being a modern man is about and what the term means to us. Why is this important? Well, Dave recently mentioned the following to me: • Suicide is still the single biggest killer of men under the age of 45. • Men make up 3/4 of the suicide rate in the UK. Which is crazy but in many ways unsurprising, as a guy it has taken me a long time to feel comfortable about being vulnerable, talking about how I feel, what scares me and even asking for help. So in this podcast, we do just that! By no means are we claiming to be able to help solve the problem or be experts in male counseling, we are just 2 dudes, that have both struggled at times with being a bloke in the modern world. So rather than shy away from the topic, we wanted to have a chat and see if others felt the same, start the conversation amongst our peers and try and share honestly about how we're trying to be modern men. The artist Phyllida Barlow shares her passion for music that reflects her sculpture, in its defiance of convention and delight in surprise. For years Phyllida Barlow was so desperate for people to see her sculptures that she would leave them on the street or in disused factories; or she would install them in friends' houses, using pianos and ironing boards as plinths. Initially overlooked by museums and galleries, she was in her sixties when she found widespread recognition - in the last decade she's been invited to exhibit all over the world, and has became a Royal Academician, a CBE, and the recipient of numerous awards. Her 2014 exhibition at Tate Britain was unforgettable - she filled the cavernous Duveen Galleries with huge, gravity-defying pieces made out of timber and scrap materials which appeared to be about to topple over or to be on the point of collapse. And in 2017 she received the ultimate accolade of representing Great Britain at the Venice Biennale. She talks to Michael Berkeley about finding success in later life, how she juggled life as a teacher, artist and mother of five, and the challenges of constructing monumental installations. She chooses music by Birtwistle, Wagner, Janacek, Webern, and Messiaen, pieces which reflect her fascination with size, scale, texture and unexpected beauty. She is regularly engaged as a concerto soloist nation-wide; other concerti in the recent seasons include Rachmaninoff 2nd concerto with Dmitry Sitkovetsky and Greensboro Symphony, Rachmaninoff 2nd with Vallejo Symphony, Gershwin Project with Daniel Meyer and the Erie Symphony, and Clara Schumann at Wintergreen Festival, Beethoven 3rd with Evanston Symphony. She has been featured on WQXR, WNYC, WFMT and many international television broadcasts, and has performed in major venues such as Carnegie Hall’s Weill Concert Hall, Metropolitan Museum of Art, Paris’ Salle Cortot, Chicago’s Orchestra Hall, Moscow’s Tchaikovsky Hall and in many important festivals such as Verbier, Portland International, Music in the Mountains, Brevard, Taos, International Keyboard Festival at Mannes, Bargemusic, and Chautauqua.

Fellow Lancastrians Harrison Birtwistle and bass John Tomlinson talk about three-decades of creative collaboration, including the operatic roles Birtwistle, alongside librettist David Harsent, created especially for Tomlinson. The first was in 1991: the Green Knight and Bertilak in 'Gawain' and the second, the title role in 'The Minotaur' from 2007. But their collaboration goes back further still – to the recording of Birtwistle’s first opera in 1979.
Sir Alan Moses is a distinguished lawyer who sat as a judge for almost 20 years, latterly in the Court of Appeal. He resigned last autumn to become the first Chairman of the new Press Standards Organisation, IPSO, the successor to the Press Complaints Commission. It's a challenging, and indeed highly controversial role. Alongside this he has spent 6 years as Chairman of Spitalfields Music, and is a dedicated concert goer, and a member of the Parliament Choir. In Private Passions, Sir Alan curates a playlist of great choral works: Bach, Monteverdi, Schubert, Donizetti, and a Handel oratorio, Saul. He introduces a little-known work by Birtwistle which was written for his wife, Dinah, and he chooses a French chanson by Brassens in tribute to his mother, a French teacher. Pianist Joanna MacGregor celebrates Harrison Birtwistle’s The Triumph of Time, an orchestral work she describes as “sculpted, dream-like and mesmeric”; Paul Griffiths remembers the London premiere and how it marked a new departure for the composer, and Birtwistle himself reflects on the subject of time in music.
Richard Rodney Bennett is a contemporary of Birtwistle and Maxwell Davies but his musical life has pursued a very different path. From his childhood onwards music was there for him. His mother was a pupil of Holst while his father wrote children's books and ballad lyrics. But his frailty meant that the young Richard was sent to boarding school, so he hardly knew him. Bennett's musical mind was inquisitive from the start and after reading about her he approached the composer Elisabeth Lutyens for lessons. She invigorated him further. Soon after he went to the Royal Academy of Music but this didn't give him the stimulus he needed although it was there that he met one of his best friends Cornelius Cardew. Together they wanted to find out about the new music which was being written in the 1940s and 50s. For a while he was the only pupil of Pierre Boulez, and with Cardew he visited Darmstadt in Germany where the new music supremos of the era met and had their works performed. His prowess as a pianist meant he was called upon to play some of the more challenging music by Boulez, Stockhausen and others. But in parallel with this he was writing film scores and continuing to play jazz with friends. So already at the age of twenty his musical life was eclectic to say the least. In the late 50s and 60s his compositional career burgeoned with commissions and performances all over the world. His film scores included Far from the Madding Crowd, Nicholas and Alexandra and Murder on the Orient Express all of which earned him Oscar nominations. In 1979 after the breakdown of a love affair and with the pressure of responsibilities in the music world proving too much, Bennett moved to New York where he has lived ever since.Now 75 Bennett enjoys his life spent between New York and London, singing with his regular collaborator Clare Martin. In conversation with Norman Lebrecht Richard Rodney Bennett talks frankly about his life, his reasons for the different musical directions he has taken and why he no longer composes nor is interested in new music.
