On Sunday, Athleague will be the venue for the Intermediate Hurling Club Final between Ballindereen and Tooreen. The sides have met in the final previously in 2017 in a game that was won by the Mayo Champions on a final score of 1-15 to 1-11 and are the defending Connacht Champions after beating Moycullen last year by 1-15 to 0-14.  Ballinderreen became County Intermediate Hurling Champions after a 3-18 to 2-18 win over Tynagh-Abbey/Duniry in the Final played in Duggan Park.  Their joint captains Roy Lane and Kieran Meehan and Manager Keith Connolly spoke to Sean Walsh.

The CDMO Sessions are a series of interviews with leaders at Contract Development and Manufacturing Organisations about what makes their business unique and to share advice for how to create better strategic partnerships. We spoke to the two Vice Presidents from Olon Group responsible for their Biotech and Small Molecules businesses respectively, Andrea Conforto VP for Biotech and Roberta Pachera, VP Small Molecules. Olon Biotech Andrea Conforo explains how Olon use ,icrobial fermentation technology, with two anufacturing sites located in Italy.  “We are very flexible and can offer fermentation capabilities that support the product from early clinical stages up to large commercial production with 100,000 litres of capability” “When partnering we typically create a ‘mirror team' to communicate directly with all the project subject matter experts a partner has. Speed is a key milestone for us, and we try to set reasonable project stages to match the customer's timeline." Olon Small Molecules Roberta Pachera leads the team focusing primarily on production of Intermediates and  final APIs on behalf of innovators or generic companies, from large, mid-sized companies or startups. Roberta highlights how Olon is able to reduce customer time to market while maintaining product quality because ” everything is under one roof”. “We suppor customers through the entire product lifecycle, which means we don't need to go through multiple technology transfers from one company to another. In addition we have  a technology platform which offers a toolbox of small molecules which we can offer to customers.” Roberta gave the example of how they worked during COVID to support a small startup to go from from initial preclinical research, to producing clinical batches to face-to-face clinical studies, all within less than 18 months. Olon have over 300 people in Research and Development, and have forged strong links with academia in Italy, Netherlands and United States which helps them to stay on the cutting-edge.  “We have an innovative R&D team who have, amongst other things, been working on our sustainability impact with the water we use. We now measure the water quality at the start and end of the manufacturing process.” The first step is to make sure the CDMO project team fully understand the goals and drivers of a project in detail so that they can add real value. “We always aim to have an open environment with partners, so that we can better understand why a customer is asking for something. We are the operational experts so with the right information will often make recommendations and propose a better  solution.” says Andrea. A two-way relationship will generally help to reduce costs.  “Our aim is always to reduce the customer's investment as much as possible.  For example, our Downsteam Processing (DSP) purification will work with customers to reuse as much technology as possible from what we have already in-house.  This can dramatically reduce the cost of goods. All this implies a long collaboration, moving from transactional customer relationship to partner.” Roberta adds that there are additional savings that comes from process optimisation when manufacturing large quantities.   “When you are manufacturing large quantities of Intermediates or Active Pharmaceutical Ingredients, it is recommended to use modern tools like Process Modelling to help you reduce production costs.”  To find out more, check out the Olon website

Abstract: The nucleolus is the most prominent membraneless organelle within the nucleus and plays essential roles in rRNA transcription and processing and ribosome assembly. How the structure of the nucleolus is maintained and regulated is poorly understood. Here, we identified two types of nucleoli in C. elegans. Type I nucleoli are spherical, and rRNA transcription and processing factors are evenly distributed throughout the nucleolus. In type II nucleoli, rRNA transcription and processing factors exclusively accumulate in the periphery rim, which is named the nucleolar ring. The hollow vacuole inside the nucleolar ring contains proteins that usually localize in the nucleoplasm but are capable of exchanging contents across the ring. The high-order structure of the nucleolus is dynamically regulated in C. elegans. Faithful rRNA processing is important to maintain the spherical structure of the nucleoli. The depletion of a class of rRNA processing factors, for example, class I ribosomal proteins of the large subunit (RPL), which are involved in 27SA2 rRNA processing, reshaped spherical nucleoli to a ring-shaped nucleolar structure. The inhibition of RNAP I transcription and depletion of two conserved nucleolar factors, nucleolin and fibrillarin, prohibits the formation of the nucleolar ring. We concluded that the integrity of nucleoli is highly dependent on rRNA processing and maturation, which may provide a mechanism to coordinate structure maintenance and gene expression. Abstract: To promote fidelity in nuclear pre-mRNA splicing, the spliceosome rejects and discards suboptimal splicing substrates after they have engaged the spliceosome. Although nuclear quality control mechanisms have been proposed to retain immature mRNPs, evidence indicates that discarded splicing substrates, including lariat intermediates, do export to the cytoplasm, as indicated by their translation and degradation by cytoplasmic nucleases. However, the mechanism for exporting these species has remained unknown. By single molecule (sm) RNA FISH in budding yeast, we have directly observed the nuclear export of lariat intermediates. Further, by crosslinking, export reporter assays, and smRNA FISH, we have demonstrated that the export of lariat intermediates requires the general mRNA export receptor Mex67p and three of its mRNA export adapter proteins, Nab2p, Yra1p, and Nlp3, establishing that both mRNAs and lariat intermediates share the same export machinery. Unexpectedly, the export of lariat intermediates, but not mRNA, requires an interaction between Nab2p and Mlp1p, a nuclear basket component implicated in retaining immature mRNPs, including unspliced pre-mRNA, in the nucleus of budding yeast. Finally, the export of lariat intermediates, like mRNA, relies on the E3 ubiquitin ligase Tom1p and its target sites in Yra1p. Overall, our data indicate that the nuclear basket can promote, rather than antagonize, the export of an immature mRNP. Further, our data imply that the export of discarded lariat intermediates requires both Mlp1p-dependent docking onto the nuclear basket and subsequent Tom1p-mediated undocking, a mechanism our data suggests generalizes to the export of mRNA but in a manner obscured by redundant pathways. Monica Is a group leader at ICIQ (Institut Català d'Investigació Química / Institute of Chemical Research of Catalonia), Tarragona, Spain. Her group focuses on the ‘Development of new chemical transformations: From organometallic mechanistic studies to catalysis'. Prior to her current position, Monica conducted postdoctoral research at the University of Michigan, Ann Arbor, USA, which followed awarding of her Ph.D. from the University of Valladolid, Spain – where she also earnt her B.Sc. and M.Sc.In 2018, Monica was selected as one of the ‘Talented 12' by Chemical & Engineering News (C&EN), the weekly newsmagazine of the American Chemical Society (ACS). She is also involved in outreach activities to increase the visibility of women in Science. You will hear the following terms used during the interview. I've included some descriptions here. NMR – Nuclear Magnetic Resonance subjects samples to a strong magnetic fields and measures the resonance pattern of the nuclei. It is widely used to study the structure and dynamics of organic molecules.Palladium – A chemical element with atomic number 46. It is a rare metal and over 50% of palladium supply is used in catalytic processes, most commonly catalytic converters attached to automobile exhausts.Cobalt A chemical element with atomic number 27. Most commonly produced as a by-product of Copper and Nickel production, the main use for colbalt is in Li-ion batteries, and the production of certain high-strength metal alloys.Transition metal – in chemistry, the term transition metal is defined in one of three ways:Many scientists describe a "transition metal" as any element in the d-block of the periodic table, which includes groups 3 to 12 on the periodic table. In practice, the f-block lanthanide and actinide series are also considered transition metals and are called "inner transition metals".The IUPAC definition defines a transition metal as "an element whose atom has a partially filled d sub-shell, or which can give rise to cations with an incomplete d sub-shell".The expanded IUPAC definition specifies which elements are included. As well as the elements of groups 4 to 11, scandium and yttrium are included Lanthanum and actinium, which are considered group 3 elements, are however classified as lanthanides and actinides respectivelyNucleophile – A chemical species that forms bonds with electrophiles by donating an electron pair.Oxidant – A substance that has the ability to oxidize another substance. Der Große Preis von Russland hatte etwas, was die vielen Jahre zuvor oft vermisst wurde: Action, Spannung und echte Highlights. Lando Norris war der tragische Held des Wochenendes, wurde ihm doch erst kurz vor Schluss durch den einsetzenden Regen der Sieg geklaut. Diesen konnte sich Lewis Hamilton sichern, der damit seinen 100. F1-Sieg feiern durfte. Eine irre Aufholjagd hat Max Verstappen hingelegt, der von 20 auf 2 vorfuhr. Viele Themen, viele spannende Diskussionen erwarten euch in unserer neuen Ausgabe. Kevin Scheuren und Sophie Affeldt haben zu der Nachbetrachtung des Formel-1-Wochenendes in Russland Stefan Ehlen vom Motorsport Network Germany und unsere Hörerin Kim eingeladen. Hamilton: Rekorde für die Ewigkeit? Wer hätte gedacht, dass Lewis Hamilton so schnell nahezu sämtliche F1-Rekorde für sich verbuchen würde? Am Sonntag wurde dann eine echte Schallmauer durchbrochen mit seinem 100. GP-Sieg. Er profitierte von einer richtigen Strategieentscheidung, hatte ein bisschen Glück, aber war dann zur Stelle. Es gibt viele Fans, die ihm seine Rekorde nicht gönnen. "Er fährt ja nur mit dem besten Auto" ist ein gern genommener Spruch, aber ist das wirklich so einfach? Ist es überhaupt fair, dass Hamilton so sehr für seine Klasse kritisiert wird? Norris: Mann für die Zukunft! Das Quartett kürt den Fahrer des Wochenendes. Es gab viele Nominierte: Fernando Alonso, Max Verstappen, Carlos Sainz. Aber am Ende setzte sich klar der Fahrer durch, dem wahrscheinlich alle F1-Fans dieser Welt den Sieg von Herzen gegönnt hätten: Lando Norris! Was der junge Brite in dieser Saison zeigt ist fabelhaft. Er misst sich nicht nur mit dem erfahrenen Teamkollegen Daniel Ricciardo, bis auf das Rennen in Italien hatte er ihn grundsätzlich im Griff. Er fährt routiniert vorne mit, macht natürlich Fehler, aber ist völlig verdient unser Fahrer des Wochenendes. Ist Lando auch eure Wahl? Sotschi: das beste Rennen der bisherigen Saison? Wer hätte gedacht, dass man mal diesen Satz hören würde: der Große Preis von Russland ist das F1-Rennen der Saison. Bisher kam das den meisten Fans nie in den Sinn, diesmal ist alles anders. Es war alles dabei für Actionhungrige und Stragiefüchse. Es wurde gefighted, überholt, es gab Berührungen und auch ein großes Finale. Die Saison 2021 ist vermutlich die beste Saison aller Zeiten, zumindest schickt sie sich an, diese zu sein. Warum der Kurs in Sotschi plötzlich diesmal für eine solche Achterbahn der Gefühle sorgte versuchen wir zu erörtern. Steward-Entscheidungen: Es fehlt die Transparenz! Kim hat ein tolles Thema mitgebracht: die fehlende Einsichtigkeit in die Entscheidungen der Rennkommissare. Warum bekommt welcher Fahrer Strafe XY? Zu häufig müssten Fans in den Unweiten des Reglements blättern, um das halbwegs zu verstehen, was da entschieden wurde. Wir versuchen Möglichkeiten zu besprechen, wie man das Prozedere der Strafenvergabe für alle transparenter und einfacher gestalten könnte. Es ist ein guter Schritt, dass man den Funkverkehr zwischen Teams und Rennleiter Michael Masi geöffnet hat, aber geht da nicht noch mehr? Eure Ideen würden wir auch gerne hören! F1-Fahrer: wie viel eigenen Willen können Teams akzeptieren? Nochmal zurück zu Lando Norris. Es gab nicht wenige Beobachter, die die Schuld für den nicht geholten Sieg eher bei ihm sehen. Er hätte ja auf Intermediates wechseln können, aber tat das nicht. Auch wenn sein Team keine klare Ansage gemacht hat, so muss er sicher aus diesem Fehler lernen und an anderer Stelle klüger handeln. Trotzdem stellt sich die Anschlussfrage, wie viel eigenen Willen die Fahrer überhaupt noch ausgeprägt zeigen dürfen. Häufig übernehmen Teams das Denken, Fahrer führen nur aus. Wird das zuviel? Ist es eigentlich gut so, wie es gelaufen ist, dass ein Fahrer - auch ein junger wie Norris - sich da so entschieden hat? Der Große Preis von Russland hatte etwas, was die vielen Jahre zuvor oft vermisst wurde: Action, Spannung und echte Highlights. Lando Norris war der tragische Held des Wochenendes, wurde ihm doch erst kurz vor Schluss durch den einsetzenden Regen der Sieg geklaut. Diesen konnte sich Lewis Hamilton sichern, der damit seinen 100. F1-Sieg feiern durfte. Eine irre Aufholjagd hat Max Verstappen hingelegt, der von 20 auf 2 vorfuhr. Viele Themen, viele spannende Diskussionen erwarten euch in unserer neuen Ausgabe. Kevin Scheuren und Sophie Affeldt haben zu der Nachbetrachtung des Formel-1-Wochenendes in Russland Stefan Ehlen vom Motorsport Network Germany und unsere Hörerin Kim eingeladen. Hamilton: Rekorde für die Ewigkeit? Wer hätte gedacht, dass Lewis Hamilton so schnell nahezu sämtliche F1-Rekorde für sich verbuchen würde? Am Sonntag wurde dann eine echte Schallmauer durchbrochen mit seinem 100. GP-Sieg. Er profitierte von einer richtigen Strategieentscheidung, hatte ein bisschen Glück, aber war dann zur Stelle. Es gibt viele Fans, die ihm seine Rekorde nicht gönnen. "Er fährt ja nur mit dem besten Auto" ist ein gern genommener Spruch, aber ist das wirklich so einfach? Ist es überhaupt fair, dass Hamilton so sehr für seine Klasse kritisiert wird? Norris: Mann für die Zukunft! Das Quartett kürt den Fahrer des Wochenendes. Es gab viele Nominierte: Fernando Alonso, Max Verstappen, Carlos Sainz. Aber am Ende setzte sich klar der Fahrer durch, dem wahrscheinlich alle F1-Fans dieser Welt den Sieg von Herzen gegönnt hätten: Lando Norris! Was der junge Brite in dieser Saison zeigt ist fabelhaft. Er misst sich nicht nur mit dem erfahrenen Teamkollegen Daniel Ricciardo, bis auf das Rennen in Italien hatte er ihn grundsätzlich im Griff. Er fährt routiniert vorne mit, macht natürlich Fehler, aber ist völlig verdient unser Fahrer des Wochenendes. Ist Lando auch eure Wahl? Sotschi: das beste Rennen der bisherigen Saison? Wer hätte gedacht, dass man mal diesen Satz hören würde: der Große Preis von Russland ist das F1-Rennen der Saison. Bisher kam das den meisten Fans nie in den Sinn, diesmal ist alles anders. Es war alles dabei für Actionhungrige und Stragiefüchse. Es wurde gefighted, überholt, es gab Berührungen und auch ein großes Finale. Die Saison 2021 ist vermutlich die beste Saison aller Zeiten, zumindest schickt sie sich an, diese zu sein. Warum der Kurs in Sotschi plötzlich diesmal für eine solche Achterbahn der Gefühle sorgte versuchen wir zu erörtern. Steward-Entscheidungen: Es fehlt die Transparenz! Kim hat ein tolles Thema mitgebracht: die fehlende Einsichtigkeit in die Entscheidungen der Rennkommissare. Warum bekommt welcher Fahrer Strafe XY? Zu häufig müssten Fans in den Unweiten des Reglements blättern, um das halbwegs zu verstehen, was da entschieden wurde. Wir versuchen Möglichkeiten zu besprechen, wie man das Prozedere der Strafenvergabe für alle transparenter und einfacher gestalten könnte. Es ist ein guter Schritt, dass man den Funkverkehr zwischen Teams und Rennleiter Michael Masi geöffnet hat, aber geht da nicht noch mehr? Eure Ideen würden wir auch gerne hören! F1-Fahrer: wie viel eigenen Willen können Teams akzeptieren? Nochmal zurück zu Lando Norris. Es gab nicht wenige Beobachter, die die Schuld für den nicht geholten Sieg eher bei ihm sehen. Er hätte ja auf Intermediates wechseln können, aber tat das nicht. Auch wenn sein Team keine klare Ansage gemacht hat, so muss er sicher aus diesem Fehler lernen und an anderer Stelle klüger handeln. Trotzdem stellt sich die Anschlussfrage, wie viel eigenen Willen die Fahrer überhaupt noch ausgeprägt zeigen dürfen. Häufig übernehmen Teams das Denken, Fahrer führen nur aus. Wird das zuviel? Ist es eigentlich gut so, wie es gelaufen ist, dass ein Fahrer - auch ein junger wie Norris - sich da so entschieden hat? Het was een enerverende race in Sotsji, waar Max Verstappen vanaf de laatste plek op de grid moest starten. De Nederlander werkte zich knap naar voren en finishte na een regenbui zelfs op de tweede plaats. Lewis Hamilton won de race, maar won daardoor slechts een minimale hoeveelheid punten op de Nederlander. Mercedes probeerde Valtteri Bottas nog in te zetten, maar deze was niet in staat om de Red Bull-coureur achter zich te houden. Uiteindelijk is het Red Bull dat de schade beperkt heeft gehouden en is niet Hamilton, maar Verstappen de grote winnaar na het Grand Prix-weekend in Rusland. In de RN365-podcast wordt de gekke race uitvoerig besproken. In this episode we go over 7 simple, optional steps you can take to learn an instrument quickly and easily.  If you want to skip straight to step 1, then skip to 3 minutes that's when I get into it! :-) Option 1: Learn some songs that you enjoy, this will give you an understanding of chords and song structure Option 2: Start learning scales. (I'll do a full episode on scales). Scales are essentially just a set of musical notes and they give you a lot to work with when songwriting and jamming. Most modern western music is written in the "Major Scale". Look into the Major scale if you wish to understand the fundamentals of music. Option 3: Learn and understand how chords work. Chords are harmonies, stacks of notes that create a sound. Definitely a key concept in music, one of the most important one, along with understanding melodies, I'll do an entire episode on melody creation, and what a melody is. Option 4: Find a group of musicians to play with or form one yourself. It's good to play with other musicians as soon as you can, it's one of the most effective ways to improve, I cannot stress that enough. In the podcast I go over many of the ways you can find / create a music group in the podcast. Ask around, look online, you'll find people in no time. Option 5: Search up free pdf's and informational videos related to your instrument / music theory /songwriting. A simple google search will give you hundreds of thousands of results! Option 6: Find an instructor that can give you real constructive feedback and show you proper technique, help you make adjustments. There are many online and probably some in your area.  Option 7: Purchase books on the topic. People with lots of experience and wisdom have written books on music that are extremely helpful. De GP van Hongarije bracht niet een zege voor Max Verstappen. Ook Lewis Hamilton stond niet op de hoogste trede van het podium, maar Esteban Ocon werd winnaar in Boedapest. Dat had alles te maken met een schuiver van Bottas in de eerste bocht. Daarbij was het einde race voor Norris, Perez, Leclerc en er was schade voor Verstappen. De Nederlander kon wel verder. 'Ik ben geen complotdenker, maar het komt Mercedes allemaal wel heel goed uit hoor', vertelt Tom Coronel in de nieuwste Pitstop tegen presentator Etienne Verhoeff. En Coronel is ook kritisch op Max Verstappen, die als tiende eindigde in Hongarije. 'Bij de herstart had Max met slicks moeten starten. Ik riep het gelijk toen ik zat te kijken, maar wat deed iedereen? Intermediates! Verstappen staat 13e. Hij had niets te verliezen. Als hij bij de herstart al slicks had gehad, stond hij op de grid naast Hamilton. Dat was een fout.' Toch pakte Verstappen dus een punt. 'Je moet ieder punt pakken dat je kunt. Je zult dat ene punt maar te kort komen aan het einde van het jaar.' Daarnaast bespreken Coronel en Verhoeff de zomerstop en het boe-geroep vanaf de tribunes richting Lewis Hamilton. Coronel: 'Ga je schamen als Nederlander als je boe roept naar Hamilton. Abstract: DNA ligase I (LIG1) completes base excision repair (BER) pathway at the last nick sealing step following DNA polymerase (pol) {beta} gap filling DNA synthesis. We previously reported that pol {beta} 8-oxo-2'-deoxyribonucleoside 5'-triphosphate (8-oxodGTP) insertion confounds LIG1 leading to the formation of ligation failure products with 5'-adenylate (AMP) block. Here, we report the mutagenic ligation of pol {beta} 8-oxodGTP insertion products and an inefficient substrate-product channeling from pol {beta} Watson-Crick like dG:T mismatch insertion to DNA ligation by LIG1 mutant with perturbed fidelity (E346A/E592A) in vitro. Moreover, our results revealed that the substrate discrimination of LIG1 for the nicked repair intermediates with preinserted 3'-8-oxodG or mismatches is governed by the mutations at both E346 and E592 residues. Finally, we found that Aprataxin (APTX) and Flap Endonuclease 1 (FEN1), as compensatory DNA-end processing enzymes, can remove 5'-AMP block from the abortive ligation products with 3'-8-oxodG or all possible 12 non-canonical base pairs. These findings contribute to understand the role of LIG1 as an important determinant of faithful BER, and how a multi-protein complex (LIG1, pol {beta}, APTX and FEN1) can coordinate to hinder the formation of mutagenic repair intermediates with damaged or mismatched ends at the downstream steps of the BER pathway. Abstract: TRAP1 is a mitochondrion specific Hsp90, a ubiquitous chaperone family that mediates the folding and maturation of hundreds of 'client' proteins. Through the interaction with client proteins, TRAP1 regulates mitochondrial protein homeostasis, oxidative phosphorylation/glycolysis balance, and plays a critical role in mitochondrial dynamics and disease. However, the molecular mechanism of client protein recognition and remodeling by TRAP1 remains elusive. Here we established the succinate dehydrogenase B subunit (SdhB) from mitochondrial complex II as a client protein for TRAP1 amenable to detailed biochemical and structural investigation. SdhB accelerates the rate of TRAP1 dimer closure and ATP hydrolysis by 5-fold. Cryo-EM structures of the TRAP1:SdhB complex show TRAP1 stabilizes SdhB folding intermediates by trapping an SdhB segment in the TRAP1 lumen. Unexpectedly, client protein binding induces an asymmetric to symmetric transition in the TRAP1 closed state. Our results highlight a client binding mechanism conserved throughout Hsp90s that transcends the need for cochaperones and provide molecular insights into how TRAP1 modulates protein folding within mitochondria. Our structures also suggest a potential role for TRAP1 in Fe-S cluster biogenesis and mitochondrial protein import and will guide small molecule development for therapeutic intervention in specific TRAP1 client interactions. Abstract: Phytochromes are red/far-red light photoreceptors in bacteria to plants, which elicit a variety of important physiological responses. They display a reversible photocycle between the resting (dark) Pr state and the light activated Pfr state, in which light signals are received and transduced as structural change through the entire protein to modulate the activity of the protein. It is unknown how the Pr-to-Pfr interconversion occurs as the structure of intermediates remain notoriously elusive. Here, we present short-lived crystal structures of the classical phytochrome from myxobacterium Stigmatella aurantiaca captured by an X-ray Free Electron Laser 5 ns and 33ms after light illumination of the Pr state. We observe large structural displacements of the covalently bound bilin chromophore, which trigger a bifurcated signaling pathway. The snapshots show with atomic precision how the signal progresses from the chromophore towards the output domains, explaining how plants, bacteria and fungi sense red light. Abstract: -Synuclein (-Syn) amyloid fibrils in synucleinopathies (such as Parkinson's disease (PD), multiple system atrophy (MSA)) are structurally and functionally different, reminiscent of prion-like strains. However, how a single protein can form different fibril polymorphs in various synucleinopathies is not known. Here, we demonstrate the structure-function relationship of two distinct -Syn fibril polymorphs, the pre-matured fibrils (PMF) and helix-matured fibrils (HMF) based on -Syn aggregation intermediates. These polymorphs not only display the structural differences, including their fibril core structure as demonstrated by solid-state nuclear magnetic resonance (NMR) spectroscopy and H/D-exchange coupled with mass spectrometry but also possess different cellular activities such as seeding, cellular internalization, and cell-to-cell transmission. The HMF with a compact core structure exhibits low seeding potency in cells but readily internalizes and transmits from one cell to another. Whereas the less structured PMF lacks the cell-to-cell transmission ability but induces abundant -Syn pathology and triggers the formation of aggresomes in cells. Overall, the study highlights how the conformational heterogeneity in the aggregation pathway may lead to fibril polymorphs with distinct prion-like behavior in PD. You joined a gym. The equipment looks amazing. The energy is off the chart. You're excited about the possibilities. You show up for your first workout, and realize something important: You have no idea what to do. Or maybe that's not your story. Perhaps, you've been showing up at your fitness center for months, but have finally admitted you're not getting any results. You gravitate to the same cardio or strength equipment each workout, not because you think they're working for you, but because they're the only things you know how to do. You've started to wonder what a typical gym workout should be like. Is there a “best” workout routine? With all of the equipment available, how should you plan your workout, anyway? This might come as a surprise, but you're not alone if you feel lost. Most people don’t know what to do at the gym, even if they look like they do in their Lululemon or Under Armour apparel. In this article, I've: shared the secrets to designing an effective exercise program.explained why strength training, and not cardio should be your main focus.provided a beginner and intermediate gym-based workout routine, with sets, reps, and exercise videos. Why Gym Workouts Are Better Than Home-Based Exercise Programs For almost everyone, working out in a gym, or "fitness center," is almost always better than working out at home. First, when you go to a gym, you get out of your home environment. You'll be out of the reach of excuses, interruptions, and temptations that keep you from getting your training sessions done. Second, the energy of being around other people focused on their fitness is infectious. A motivational quote poster and loud music are no match for the inspiration of being around other people doing the same thing you are. Third, most people don't have enough equipment to follow a complete strength training program. A solid home gym would have, at a minimum: Adjustable squat rack with pull-up attachment and safety barsBarbell and weight platesDumbbell setAdjustable benchCable attachment or tubing Buying fancy gym equipment won't motivate you to workout. You need the motivation before you buy the equipment, or the fact that you don't use it will become an irritation, not motivation. Some will read this, and think, "If you need a gym, then why is Beach Body so popular?" "Home entertrainment systems" are easy to sell. They're just what the average person wants to hear. Move from the couch to the floor in front of your TV, and follow this easy 30 minute routine. We've even included sexy models to keep your heart racing. People buy the programs, but that doesn't mean they work long-term. If you want to get and stay fit, be around fit people. They're at the fitness center. The First Rule of VIGOR Training Before delving into the gym workout programs, I need to address one rule and five program design guidelines. I use this rule to determine whether or not to use or recommend an exercise, a nutrition choice, or a lifestyle habit. Like a smart investor who analyzes deals with the lens of "don't lose money," I consider my recommendations with a similar rule: The First Rule of VIGOR Training:Don’t Lose Muscle Muscle loss wreaks havoc on your metabolism, and contributes to the myriad of health problems the average person faces today. Bond cleavage and formation are key steps in chemistry and biochemistry. The present work investigates the generation of diphenylmethyl cations (Ph2CH+) via photoinduced bond cleavage of diphenylmethyl derivatives with a cationic or neutral leaving group. The resulting Ph2CH+ cations and its numerous derivatives serve as reference electrophiles for one of the most extensive reactivity scales covering 40 orders of magnitude. In chapter 1, the focus is on the initial bond cleavage of diphenylmethyltriphenylphosphonium ions (Ph2CH−PPh3+) exhibiting a cationic leaving group. With the help of state-of-the-art quantum chemical and quantum dynamical methods, the reaction mechanism of the bond cleavage is revealed. Using a reduced model system, the potential energy surfaces can be calculated at the ONIOM level of theory along specially designed reactive coordinates. Two competing reaction channels emerge: a homolytic one in the S1 state and a heterolytic one in the ground state. They are connected via an energetically accessible conical intersection which makes an efficient generation of the observed Ph2CH+ cations feasible. In contradiction with the experiment in polar or moderately polar solvents, quantum dynamical calculations for the isolated molecule reveal the formation of Ph2CH• radicals. While electrostatic solvent effects are negligible in this system, dynamic solvent effects emerge as being essential to explain the molecular mechanism. Two methods with increasing complexity to describe the dynamic impact of the solvent environment are developed. The first approach, the dynamic continuum ansatz, treats the environment implicitly. It uses Stokes’ law and the dynamic viscosity of the solvent in combination with quantum chemically and dynamically evaluated quantities to obtain the decelerating force exerted on the dissociating fragments. The ansatz does not require any fitting of parameters. The second method, the QD/MD approach, is based on an explicit treatment of the solvent surrounding. It combines molecular dynamics (MD) simulations of the reactant in a box of solvent molecules with quantum dynamics (QD) calculations of the reactant’s dynamics. In this way, a more detailed microscopic picture of the molecular process can be derived taking into account individual arrangements of the solvent. Both methods unveil the crucial impact of the solvent cage on the bond cleavage mechanism. It hinders the free dissociation in the S1 state and guides the molecular system to the conical intersection. QD simulations including the non-adiabatic coupling around the conical intersection show the formation of Ph2CH+ within ∼400 fs which compares well with the initial rise of the cation absorption in the experiment. Chapter 2 deals with the position of the counterion X– in the ion pairs Ph2CH−PPh3+ X–, PhCH2−PPh3+ X–, and (p-CF3-C6H4)CH2−PPh3+ X– in solution with X– being Cl–, Br–, BF4–, and SbF6–. These structures are essential to clarify the role of oxidizable counterions like e.g. Cl– during the initial bond cleavage in dichloromethane. The structures determined quantum chemically in dichloromethane show a similar counterion position than in the crystal. They are confirmed by the good accordance of the calculated and measured 1H NMR shifts. The C(α)–H···X– hydrogen bonds account for the pronounced counterion-dependent 1H NMR shifts of the C(α)–H in CD2Cl2. The strong downfield shift of the signals increases according to SbF6– < BF4–

Bridged pentavalent carbon structures can be intermediates or transition states of cation rearrangement during SN1 reactions, and short-lived ion pairs explain net stereochemical inversion. The different perspectives of preparative organic chemists and mechanistic organic chemists on reaction yields are illustrated by a study designed to demonstrate that molecular rotation can be rate-limiting in viscous solvents. “Electrophilic” addition to alkenes is the reverse of E2 or E1 reaction, and its mechanisms can be studied by analogous techniques. Because of their unusual acidity very strong base makes it possible to isomerize an internal acetylene to the less stable terminal isomer. Many chemical reactions may be understood in terms of localized bonds, but the special stability of conjugated systems requires considering delocalized orbitals or “resonance.” Equilibrium constants, rates, and regiochemistry in systems involving allylic cations, anions, transition states, and free radicals demonstrate that allylic conjugation is worth about 13 kcal/mole. Regioselection in addition of DCl to 1,3-pentadiene reveals rapid collapse of an allylic ion pair. Allylic substitution of bromine can be favored over Br2 addition by using NBS to control Br2 concentration. Diene conjugation is worth much less than allylic conjugation. This dissertation consists of four chapters, which are self-contained and can be read independently of each other. In Chapter 1, we analyse services trade liberalisation, which is a prerequisite for international production networks to arise. In particular, we study the determinants of services liberalisation undertaken by countries under the General Agreement on Trade in Services (GATS) of 1995. In Chapters 2-4, we analyse different aspects of the patterns of trade in intermediates. In Chapter 2, we assess the determinants of bilateral trade in intermediate goods and services. In Chapter 3, we investigate the role of institutions and transport infrastructure as sources of comparative advantage for intermediate goods. In Chapter 4, we study trade in intermediates in the context of the activities of multinational enterprises by assessing the determinants of the share of intra-firm trade in total trade of intermediates.

Mitochondria contain a complex machinery for the import of nuclear-encoded proteins. Receptor proteins exposed on the outer membrane surface are required for the specific binding of precursor proteins to mitochondria, either by binding of cytosolic signal recognition factors or by direct recognition of the precursor polypeptides. Subsequently, the precursors are inserted into the outer membrane at the general insertion site GIP (general insertion protein. Here we report the analysis of receptors and GIP by crosslinking of translocation intermediates and by coimmunoprecipitation. Surface-accumulated precursors were cross-linked to the receptors MOM19 and MOM72, suggesting a direct interaction of preproteins with surface receptors. We identified three novel mitochondrial outer membrane proteins, MOM7, MOMS, and MOM30 that, together with the previously identified MOM38, seem to form the GIP site and are present in the mitochondrial receptor complex.

We have investigated the properties of nerve cell precursors in hydra by analyzing the differentiation and proliferation capacity of interstitial cells in the peduncle of Hydra oligactis, which is a region of active nerve cell differentiation. Our results indicate that about 50% of the interstitial cells in the peduncle can grow rapidly and also give rise to nematocyte precursors when transplanted into a gastric environment. If these cells were committed nerve cell precursors, one would not expect them to differentiate into nematocytes nor to proliferate apparently without limit. Therefore we conclude that cycling interstitial cells in peduncles are not intermediates in the nerve cell differentiation pathway but are stem cells. The remaining interstitial cells in the peduncle are in G1 and have the properties of committed nerve cell precursors (Holstein and David, 1986). Thus, the interstitial cell population in the peduncle contains both stem cells and noncycling nerve precursors. The presence of stem cells in this region makes it likely that these cells are the immediate targets of signals which give rise to nerve cells.
