However, Pliant has experienced a stock decline following the halt of its Phase IIb/III study for idiopathic pulmonary fibrosis. Papas, Ph.D., University of Arizona, Tucson, AZ Introduction: Cell therapy has become increasingly studied for therapeutic use in drug delivery and regenerative medicine. Devices intended for implantation of cells require construction of membranes that can safely contain transplanted cells, provide a barrier to the host immune system and an outer vascularizing membrane to induce close vessel formation. Sheep make an excellent model because of their nearly comparable size, physiology, and disease profile with humans, as well as their docility, capacity for repeated blood draws, and ability to tolerate numerous implants during a single operation. In this study, sheep were implanted with devices containing allogeneic primary fetal sheep or xenogeneic Rat-2 fibroblasts to determine cell survival and immune response. Methods: Primary fetal sheep fibroblasts were isolated and cultured in RPMI. Rat-2 fibroblasts (ATCC) were cultured in DMEM. Cells were loaded into devices at different densities and transplanted subcutaneously. Sheep underwent allogeneic or xenogeneic transplantation with empty control and perforated devices transplanted as well. Devices were explanted at days 3, 7, 21 and 38 to evaluate host responses, vascularization, and cell survival through histological evaluation. Results: Sheep fibroblasts survived at different densities, confirming alloprotection with good vascularization and absence of a major foreign body response or extensive inflammation around the implants. Devices overloaded at the highest densities exhibited some host immune responses. Perforated devices exhibited a greater immune response that infiltrated the device. A robust immune response was seen around xenogeneic transplants with no encapsulated cells surviving. Conclusion: Sheep make a valuable translational animal model in cell therapy research. The results of this study concluded alloprotection in immunoisolating devices, allorejection in purposely perforated devices, and host immune response to xenogeneic transplantation, representing a feasible model for translational development of cell therapies. Future work will include continued allogeneic studies while avoiding xenogeneic rejection by incorporating immunomodulatory technologies.

Researchers are discovering how fasting can undo some of the metabolic damage imposed by the overindulgences of the typical Western diet. The human body may be wired to function optimally when exposed to periods of fasting, which reawakens stem cell activity and the body's intrinsic capacity to regenerate. The next frontier in this research field is the brain and the ways in which fasting may help improve age-related cognitive deficits. Abstract: Mycobacterium abscessus (MAB) is a widely disseminated pathogenic non-tuberculous mycobacterium (NTM). Like with M. tuberculosis complex (MTBC), excreted / secreted (ES) proteins play an essential role for its virulence and survival inside the host. ES proteins contain highly immunogenic proteins, which are of interest for novel diagnostic assays and vaccines. Here, we used a robust bioinformatics pipeline to predict the secretome of the M. abscessus ATCC 19977 reference strain and fifteen clinical isolates belonging to all three MAB subspecies, M. abscessus subsp. abscessus, M. abscessus subsp. bolletii, and M. abscessus subsp. massiliense. We found that ~18% of the proteins encoded in the MAB genomes were predicted as secreted and that the three MAB subspecies shared > 85 % of the predicted secretomes. MAB isolates with a rough (R) colony morphotype showed larger predicted secretomes than isolates with a smooth (S) morphotype. Additionally, proteins exclusive to the secretomes of MAB R variants had higher antigenic densities than those exclusive to S variants, independently of the subspecies. For all investigated isolates, ES proteins had a significantly higher antigenic density than non-ES proteins. We identified 337 MAB ES proteins with homologues in previously investigated M. tuberculosis secretomes. Among these, 222 have previous experimental support of secretion, and some proteins showed homology with protein drug targets reported in the DrugBank database. The predicted MAB secretomes showed a higher abundance of proteins related to quorum-sensing and Mce domains as compared to MTBC indicating the importance of these pathways for MAB pathogenicity and virulence. Comparison of the predicted secretome of M. abscessus ATCC 19977 with the list of essential genes revealed that 99 secreted proteins corresponded to essential proteins required for in vitro growth. Abstract: We have revised a previously reported multi-scale model for (ultra) small angle X-ray (USAXS/SAXS) and (very) small angle neutron scattering (VSANS/SANS) of live Escherichia coli based on compositional/metabolomic and ultrastructural constraints. The cellular body is modelled, as previously described, by an ellipsoid with multiple shells. However, scattering originating from flagella was substituted by a term accounting for the oligosaccharide cores of the lipopolysaccharide leaflet of the outer membrane including its cross-term with the cellular body. This was mainly motivated by (U)SAXS experiments showing indistinguishable scattering for bacteria in the presence and absence of flagella or fimbrae. The revised model succeeded in fitting USAXS/SAXS and differently contrasted VSANS/SANS data of E. coli ATCC 25922 over four orders of magnitude in length scale, providing specifically detailed insight into structural features of the cellular envelope, including the distance of the inner and outer membranes, as well as the scattering length densities of all bacterial compartments. Consecutively, the model was also successfully applied to E. coli K12, used for our original modelling, as well as for two other E. coli strains, detecting significant differences between the different strains in terms of bacterial size, intermembrane distance and its positional fluctuations.These findings corroborate the general applicability of our approach to quantitatively study the effect of bactericidal compounds on ultrastructural features of Gram-negative bacteria without the need to resort to any invasive staining or labelling agents. D., Beckham, S., Peleg, A. Y., Greening, C., Li, J., Heinz, E., Lithgow, T. Abstract: Acinetobacter baumannii is a high-risk pathogen due to the rapid global spread of multi-drug resistant lineages. Its phylogenetic divergence from other ESKAPE pathogens means that determinants of its antimicrobial resistance can be difficult to extrapolate from other widely studied bacteria. A recent study showed that A. baumannii upregulates production of an outer-membrane lipoprotein, which we designate BonA, in response to challenge with polymyxins. Here we show that BonA has limited sequence similarity and distinct structural features compared to lipoproteins from other bacterial species. Analyses through X-ray crystallography, small-angle X-ray scattering, electron microscopy, and multiangle light scattering demonstrate that BonA has a dual BON-domain architecture and forms a decamer via an unusual oligomerization mechanism. This analysis also indicates this decamer is transient, suggesting dynamic oligomerization plays a role in BonA function. Antisera recognizing BonA shows it is an outer membrane protein localized to the divisome. Loss of BonA modulates the density of the outer membrane, consistent with a change in its structure or link to the peptidoglycan, and prevents motility in a clinical strain (ATCC 17978). Consistent with these findings, the dimensions of the BonA decamer are sufficient to permeate the peptidoglycan layer, with the potential to form a membrane-spanning complex during cell division.
