In this episode, we will learn from Jill Maglio, PhD, OTR/L. Dr. Maglio is an occupational therapist with over 15 years of experience using circus as an educational, therapeutic, and community-building tool. She has collaborated with service recipients and governmental and non-governmental organizations to improve occupational participation for individuals and communities worldwide. Jill has trained professionals in Australia, Europe, Asia, Central America, and the USA in the methodology she has developed. Jill has collected evidence, tested methods, and created programs for political and environmental refugees underpinned by occupational science and social circus research. R., Loiselle, A. E. Abstract: Tendon injuries are a major clinical problem, with poor patient outcomes caused by abundant scar tissue deposition during healing. Myofibroblasts play a critical role in the initial restoration of structural integrity after injury. However, persistent myofibroblast activity drives the transition to fibrotic scar tissue formation. As such, disrupting myofibroblast persistence is a key therapeutic target. While myofibroblasts are typically defined by the presence of SMA+ stress fibers, SMA is expressed in other cell types including the vasculature. As such, modulation of myofibroblast dynamics via disruption of SMA expression is not a translationally tenable approach. Recent work has demonstrated that Periostin-lineage (PostnLin) cells are a precursor for cardiac fibrosis-associated myofibroblasts. In contrast to this, here we show that PostnLin cells contribute to a transient SMA+ myofibroblast population that is required for functional tendon healing, and that Periostin forms a supportive matrix niche that facilitates myofibroblast differentiation and persistence. Collectively, these data identify the Periostin matrix niche as a critical regulator of myofibroblast fate and persistence that could be targeted for therapeutic manipulation to facilitate regenerative tendon healing. Dans ce 9ème épisode, Marion Leménager, étudiante au doctorat en biologie végétale, nous transporte dans le temps ainsi que dans les Antilles, vaste archipel tropical qu'on qualifie de véritable laboratoire d'évolution naturelle. Marion y étudie un groupe de plantes de la famille des Gesnériacées dans lequel plusieurs espèces ont évolué indépendamment mais présentent une stratégie de pollinisation commune de type généraliste, soit un ensemble d'adaptations pour la pollinisation par plusieurs types de pollinisateurs. Marion tente de comprendre pourquoi est-il avantageux pour ces espèces d'opter pour une stratégie de pollinisation généraliste plutôt que spécialiste, notamment en étudiant la forme des fleurs à l'aide de modèles 2D et 3D. Qu'est-ce qui caractérise les stratégies de pollinisation des plantes? Quels en sont les exemples? Qu'est-ce que la convergence évolutive? Abstract: Flexor tendon injuries are common and heal poorly owing to both the deposition of function- limiting peritendinous scar tissue and insufficient healing of the tendon itself. Therapeutic options are limited due to a lack of understanding of the cell populations that contribute to these processes. Here, we identified a bi-fated progenitor cell population that originates from the epitenon and goes on to contribute to both peritendinous fibrosis and regenerative tendon healing following acute tendon injury. Using a combination of genetic lineage tracing and single cell RNA-sequencing (scRNA-seq), we profiled the behavior and contributions of each cell fate to the healing process in a spatio-temporal manner. Branched pseudotime trajectory analysis identified distinct transcription factors responsible for regulation of each fate. Finally, integrated scRNA-seq analysis of mouse healing with human peritendinous scar tissue revealed remarkable transcriptional similarity between mouse epitenon- derived cells and fibroblasts present in human peritendinous scar tissue, which was further validated by immunofluorescent staining for conserved markers. Combined, these results clearly identify the epitenon as the cellular origin of an important progenitor cell population that could be leveraged to improve tendon healing. One of her favourite bakeries, Genny shared a few of her fav pastries such as his Almond Croissant (Alix mentioned it's his #1 seller at the beginning!) and his seasonal croissants such as pumpkin spice, blueberry, and cherry. She also shared that his “croque monsieur”, which is a savoury item, was also one of her favs!Alix shared that he was inspired by his grandmother's baking but initially, his first passion was music! A classically trained pianist, Alix also played the trombone and wrote his own music. He then pursued his passion for baking by enrolling in Culinary School. Ryan Loiselle is a non-binary queer person (using he/him or they/them pronouns) and has been exploring radical identity and spirituality since their early 20s. While that journey has involved enculturation as a gay male and initiation into some more organized systems their experience has always been foregrounded by deep interest in expressing and celebrating their own queerness and discovering the inherent magic that can come out of liminal being. Under the name Sister Sharin' Dipity, they became fully-professed member of the Sisters of Perpetual Indulgence in 2003 and continues that work today as the current Prioress of the Portland Sisters. In 2018, he was initiated as a Brother of the Unnamed, an ecstatic spiritual path for men who love men that focuses on service and personal work with ancestors and the deities connected to that path. In his secular life, Ryan is a licensed professional counselor and gender therapist in private practice in Portland Oregon where they also serve as the Clinical Supervisor for Brave Space, a counseling center devoted to supporting a transgender, non-binary and other gender expansive individuals and their families. Episode Highlights Ryan tells us who the Sisters of Perpetual Indulgence (SPI) are, how they began and what they do today. He also tells us how he first discovered them and his path to joining them. They share their ideas about the connection of the Sisters to the Radical Faeries. Ryan talks about how being a Sister shaped his spiritual life and his own queer identity. BetterU on the move in India with partnerships, products and personnel. BetterU (BTRU:TSXV) has been taking some heat in 2019 for delays of a very significant financing, which quite frankly has been out of their hands. However, while funders get themselves sorted out, BTRU has been on the move in a big way with new content partners, updating their product offering with some head snapping features and the addition of some great personnel.
