GUEST: John Ronald, London (ON, Canada)Welcome back!  It has been a while but are back with a bang, having our good friend John Ronald from The Robarts Research Institute in London, Ontario as our guest. John's group combines advances in molecular and synthetic biology with a multimodal imaging perspective to build new tools for early detection and treatment of cancer, as well as non-invasive monitoring of cell, gene and genome therapies. One of John's passions is gene reporter imaging, which is also the main focus of this edition. The paper up for discussion comes from Cynthia Dunbar's lab at NIH, Bethesda which was recently published in Cell Stem Cell. We discover their approach to non-invasively follow engraftment and maturation of pluripotent stem cells in rhesus macaques with myocardial infarctions, discuss some of the technicalities of engineering autologous cells, and fall in love with the beautiful RNAScope images that are presented. We discuss what makes a good imaging reporter, why one size doesn't always fit all, and a future where not just cells, but activation states can be imaged. Along the way, John also shares advices received from the wonderful Sanjiv Sam Gambhir on how to improve the chances of getting your grants funded and that supposedly crazy ideas should not be discarded... they might even lead to high impact publications. And of course, true to our tradition, we enjoy a special cocktail. For this edition the “Angry Canadian”, obviously. For the curious listeners: a good measure of Whisky, 2 tablespoons of fine Canadian Maple syrup, a few drops of bitters and 1 teaspoon lemon juice, then top up with soda water.Selected Publication: Lin Y, Sato N, Hong S, Nakamura K, Ferrante EA, Yu ZX, Chen MY, Nakamura DS, Yang X, Clevenger RR, Hunt TJ, Taylor JL, Jeffries KR, Keeran KJ, Neidig LE, Mehta A, Schwartzbeck R, Yu SJ, Kelly C, Navarengom K, Takeda K, Adler SS, Choyke PL, Zou J, Murry CE, Boehm M, Dunbar CE. Leo and his wife Kim are heading up an Acts 29 initiative in London (ON) called, Sojourn Church, that we at Harvest have the privilege of supporting. For more information, please go to sojournchurch.ca On Sunday, Leo will bring us the third sermon in our “Invisible God, Visible Faith” series, preaching a message entitled, “The Proclamation of Faith” based on Colossians 1:24-2:5.  In this passage, Paul highlights his commitment to the full Gospel and his desire to see the Colossian church grow together as mature disciples.  Of course, there are many ways that we might apply this in our own context as we consider our own commission to serve God by revealing his truth despite the personal cost, and as we consider, the importance of developing confident and mature believers who are united in the truth of God's Word. We look forward to hearing what God has laid on Leo's heart this week. In episode 22, we continued digging deeper into some topics from season 1 and 2.  There's something decidedly different about next week's federal election: it's the first time we're going in with our eyes wide open. To the existence of racism and discriminatory systems, to the ugly history of this country's residential schools and to the need for leadership that is going to bring real change. A throwback to episode 14 touches on the anti-Islamic hate that resulted in a heinous act of terror in London ON and still persists. And in episode 15, Ben Borne really opened our eyes to the realities and barriers that Indigenous Canadians face to this day – and the systems that need to be rebuilt. This episode will give you chills. Tune in and take notes.

She survived, but was only the first of more than fifty women to suffer the same in London. This wasn't Jack The Ripper – he wouldn't come along for another 100 years. This was the Monster of London. (The Monster of London) *** On his first day walking to the bus stop alone before school, Etan Patz was abducted – and became one of the first children whose face ended up on a milk carton. (The Boy On The Milk Carton) *** If you are left-handed, you might be offended by one of the stories I have in this episode. But then, you lefties are all sinister anyway, right? (The Sinister Left-Handers) *** In 1827, young Maria Marten slipped into a red barn to meet her secret lover. It was the last time she would be seen alive. (The Red Barn Murder) *** It sounds like the makings of a paranormal slasher film – a woman is followed home one day by a paranormal entity – an entity that is angry because it used to be a human that was murdered. (The Haunting of Leah Lewis) *** Imagine your business is failing and you need to drum up customers, so you hire a consultant. “No problem,” he says. “All I need is a few things to get started… do you happen to have have any problems chopping off your closest living relative's hand?” Welcome to Muti magic that some say still takes place even today. (Murderous Muti Magic) *** Edward Howard Bell was a murderer on the run and evaded capture for close to twenty years. When finally captured, authorities found out they didn't just have a murderer… but a serial killer, who he claims had sent eleven young girls to heaven. The Pairs are a fantastic female fronted folk/pop group from London ON featuring classically trained vocalists Hillary Watson, Renée Coughlin and Noelle Coughlin with a wonderful sound driven by sweet harmonies, and joyful rhythms. With support from their other band member, former punk rock drummer, Steve Plimmer, they really put on a great show – they will have you up on your feet dancing before you realize what’s happening! The Pairs have just released their second album “Noise” which features twelve self-written songs that tell stories about this beautiful, and, at times, crazy and chaotic journey we call life, and all its conflicting emotions… the joy, the sadness – the comedy and the tragedy. The Pairs sat down with Folk Roots Radio at Folk Music Ontario for a fun conversation about their music and three live songs from the new album. For more information about The Pairs, visit https://thepairsmusic.com. Music: The Pairs “Wrong Side Of Life” (Live), “Everything I Call Home” (Live) and “Put The Gun Down” (Live).
