Jsem čtyři sta kilometrů před Ósakou, což je teoreticky tak půlden, pět až deset aut. Teoreticky. V Japonsku od včerejšího, tedy pondělního večera lije, stopovat se nedá. Sedím v nějaké autodílně, kde jsem dostal už dvě kávy - sice plechovkové, ale teplé - a podle všeho mám dost času na článek.Nevím přesně, kde se nacházím, musím mrknout do mapy, takže moment. Jakási vesnice Kushirocho kousek nad městem Masuda, mezi rýžovými poli na břehu Japonského moře: jihozápad ostrova Honšú.Do Masudy mě přivezl mládenec jménem Sakamoto Takumi, který jel přesně opačným směrem, ale otočil to a dal si se mnou čtyřicet kilometrů jenom tak - z radosti, že má v autě cizice. A za společnou fotku.V té Masudě jsem si konečně koupil boty, protože celé poslední dva měsíce chodím v jedněch a těch samých, ostudně jetých sandálech. S novými křuskami na nohou jsem potom kráčel tmou a ptal se sebe sama, co je vlastně cílem. “Netuším,” odpovídala stopařská část mého já té příčetnější. Ve skutečnosti jsem šel vstříc nějakému poslednímu zážitku dne, který se ovšem nedostavil - tato část Japonska je klidná, oproti celé mé předchozí Asii úplně mrtvá, je tady božské ticho. Lidé najednou nežijí na ulici, po setmění jsou všichni zavření doma.Můj nevyřčený plán, že se vnutím k někomu aspoň pod pergolu, kde dostanu drink a nabiju veškerou elektroniku, neměl šanci vyjít; během své hodinové túry jsem vůbec nikoho nepotkal.Naštěstí je Japonsko cestovatelsky luxusní, nádherně jednoduchou zemí. Takové autobusové zastávky, jaké stojí tady na zapadlé vsi, nemají ve světě obdoby, jsou to vlastně čisťounké chatky. Takže tam. Nemaje žádnou funkční elektroniku, poslepu jsem vytáhl spacák a dlouho nemohl usnout, protože to takhle bylo naposledy. Už za pár hodin začne, vypadá to, obyčejný život.Po svítání jsem netušil, jakým směrem se vydat, ale nové boty mě loužemi dovedly až ke garáží lomeno autodílně, kde už dvě hodiny sedím, pozoruju nekončící déšť a vzpomínám na předchozí dny.Zajímavé je, že co bylo včera, připadá mi jako před týdnem, a co předevčírem, to jako před rokem.

An underrated rock album coming from a seiyuu artist that deserves more attention. In 2021, Toshiki Masuda impressed me with his most consistent album to date, "Origin". This rock album with indie influences goes back into Masuda's origins as a rock fan and showers listeners with quite the nostalgic and motivational set of tunes. While this album sold a lot at the time of release, it is barely mentioned as one of the best rock albums by a male seiyuu so, I come here to talk to you about it. Did you enjoy this episode? We are always thrilled to be joined by special guests here at Sake On Air, but it is rather rare that we have a chance to interview a 15th-generation kuramoto. This week we're thrilled to share our recent interview with Junichi Masuda, newly appointed CEO of Tokubee Masuda Shoten, makers of Tsukinokatsura, a brand of which we are all collectively huge fans.This week our regular hosts Rebekah Wilson-Lye and Sebastien Lemoine speak with Masuda-san about his sake brewery: Tokubee Masuda Shoten - a historical innovator in the industry - their brand “Tsukinokatsura”, and what changes we can expect to see with his new vision for the company. On a more personal note, we get to hear about what it's been like to have grown up as part of such an iconic brewing family, the unique pressures, and exciting opportunities.Of course, we also dive into Kyoto, or Fushimi to be precise, where the brewery is located, and the brewery's connection to the sake community there. Whether it is involvement in local events, being part of the Fushimi Sake Association, or organizing rice planting and harvesting activites for the local community, the Masuda family's dedication to the region has us excited for where Masuda-san plans to take the brewery in the years ahead.With big events including the 60th anniversary of nigori sake (which the brewery is responsible for creating!) in 2024, as well as the brewery's 350th anniversary in 2025, there are plenty of good reasons for Tokubee Masuda Shoten to celebrate. From Kamakura, Japan, Takuji Masuda has been a progenitor of surf culture since the 1980s. After traveling the surfing world and competing professionally with the Oxbow Longboard team, whose members included Joel Tudor, Duane Desoto, and others, Masuda pursued an interest in storytelling and cultural production. In the 1990s, through his reverence of surf and skate magazines and inspiration from the booming technological innovations of the era, he established his own magazine, Super X Media, published in English, Japanese, and French, influential as a cross-cultural hub for the surf-skate industry. Masuda more recently pursued filmmaking, including his documentary 2016 film Bunker77. In this episode, he sits down with show host Jamie Brisick to talk about how surfing allows you to design a lifestyle that reflects who you are, the fashion industry, art and culture in Los Angeles, Pipeline, what drew him to the Bunker Spreckels story, and the joy of making friends through surfing.

I am so excited to introduce my guest, Masuda Mohamadi. She is a yoga teacher and an author. Abstract: A lack of the mitochondrial tRNA taurine modifications mediated by mitochondrial tRNA translation optimization 1 (Mto1) was recently shown to induce proteostress in embryonic stem cells. Since erythroid precursors actively synthesize the hemoglobin protein, we hypothesized that Mto1 dysfunctions may result in defective erythropoiesis. Hematopoietic-specific Mto1 conditional knockout (cKO) mice were embryonic lethal due to niche-independent defective terminal erythroid differentiation. Mechanistically, mitochondrial oxidative phosphorylation complex-I was severely defective in the Mto1 cKO fetal liver and this was followed by cytoplasmic iron accumulation. Overloaded cytoplasmic iron promoted heme biosynthesis and enhanced the expression of embryonic hemoglobin proteins, which induced an unfolded protein response via the IRE1 -Xbp1 signaling pathway in Mto1 cKO erythroblasts. An iron chelator rescued erythroid terminal differentiation in the Mto1 cKO fetal liver in vitro. The new point of view provided by this novel non-energy-related molecular mechanism may lead to a breakthrough in mitochondrial research. Copy rights belong to original authors. Abstract: Exon-skipping therapy mediated by antisense oligonucleotides (ASOs) is expected to provide a therapeutic option for Duchenne muscular dystrophy (DMD). ASOs for exon skipping reported so far target a single continuous sequence in or around the target exon. In the present study, we investigated ASOs for exon 44 skipping (applicable to approximately 6% of all DMD patients) to improve activity by using a novel ASO design incorporating two connected sequences. Phosphorodiamidate morpholino oligomers targeting two separate sequences in exon 44 were created to simultaneously target two splicing regulators in exon 44, and their exon 44 skipping was measured. NS-089/NCNP-02 showed the highest skipping activity among the oligomers. NS-089/NCNP-02 also induced exon 44 skipping and dystrophin protein expression in cells from a DMD patient to whom exon 44 skipping is applicable. We also assessed the in vivo activity of NS-089/NCNP-02 by intravenous administration to cynomolgus monkeys. NS-089/NCNP-02 induced exon 44 skipping in skeletal and cardiac muscle of cynomolgus monkeys. In conclusion, NS-089/NCNP-02, an ASO with a novel connected-sequence design, showed both in vitro and in vivo exon-skipping activity. Copy rights belong to original authors. Abstract: Cre/LoxP technology has revolutionized genetic studies and allowed for spatial and temporal control of gene expression in specific cell types. The field of microglial biology has particularly benefited from this technology as microglia have historically been difficult to transduce with virus or electroporation methods for gene delivery. Here, we interrogate four of the most widely available microglial inducible Cre lines. We demonstrate varying degrees of recombination efficiency and spontaneous recombination, depending on the Cre line and loxP distance. We also establish best practice guidelines and protocols to measure recombination efficiency in microglia, which could be extended to other cell types. There is increasing evidence that microglia are key regulators of neural circuit structure and function. Microglia are also major drivers of a broad range of neurological diseases. Thus, reliable manipulation of their function in vivo is of utmost importance. Identifying caveats and benefits of all tools and implementing the most rigorous protocols are crucial to the growth of the field of microglial biology and the development of microglia-based therapeutics. Layer a recession on top of this and new investments must be made under increased scrutiny to match the future uncertainty. “What we are thinking about now is making sure we are hiring carefully. We are more cognizant of the spending and hiring now that we're in the mid to late 2022”Keith shares that as a software company, typically you will experience less margin pressure, and therefore can discount. Look for the best possible people for your stage of company: A players are different for different types and stages of a businesses. “I try to look for the best possible people for the company. Everybody is always trying to find the best people or the A players. No one wants to hire a C player. And there are many A players who are perfect for an early stage and some who are perfect for a large public company. The United Nations estimates around half of Afghanistan's population – nearly 20 million people – faces acute hunger. The ongoing humanitarian crisis in Afghanistan follows the end of America's twenty-year war and the withdrawal of all US troops in August 2021. In February, the Biden administration decided to freeze nearly $10 billion in assets belonging to the Afghan central bank in order to prevent money going into the hands of the Taliban. Are US policies exacerbating the humanitarian crisis in Afghanistan, and what does freezing of assets mean to accomplish? This week, the Eurasia Group Foundation's Mark Hannah is joined by freelance journalist Matthieu Aikins and the co-founder of Unfreeze Afghanistan Masuda Sultan to better understand the current state of Afghanistan, President Biden's decision to freeze Afghanistan assets in response to the Taliban's takeover, and the ongoing plight of Afghans. Matthieu Aikins is a contributing writer for the New York Times Magazine, and the author of The Naked Don't Fear the Water: An Underground Journey with Afghan Refugees.
